Synthesis of 1-(2-Fluorophenyl)pyrazoles by 1,3-Dipolar Cycloaddition of the Corresponding Sydnones

3-Arylsydnones bearing fluorine and bromine atoms on the benzene ring were synthesized from N-nitroso-2-fluorophenylglycines and characterized by NMR spectroscopy. These were employed further in synthesis of the corresponding 1-(2-fluorophenyl)pyrazoles by 1,3-dipolar cycloaddition reaction with dimethyl acetylenedicarboxylate (DMAD) as activated dipolarophile. The sydnones as reaction intermediates were characterized by single crystal X-ray diffraction analysis showing interesting features such as halogen bonding as an important interaction in modeling the crystal structure.     

Crystal Structures of 5-Nitro- and 5-Ethoxycarbonyl-Substituted 6-Phenyl-4-(3-fluorophenyl)-3,4-dihydro-(1H)-pyrimidin-2-ones

The crystal structures of 5-nitro- and 5-ethoxycarbonyl-substituted 6-phenyl-4-(3-fluorophenyl)-3,4-dihydro-(1H)-pyrimidin-2-ones were determined by X-ray diffraction analysis. Conformational peculiarities of the structures have been revealed and compared with data for other biologically active dihydropyrimidinones.     

Synthesis,crystal and molecular structure of 2-(3-chloropropyl)-6-methyl-4-phenylquinazoline 3-oxide

The acylation of the syn isomer of the oxime of 2-amino-5-methylbenzophenone with 4-chlorobutyryl chloride gives a mixture of the anti isomer of the 4-chlorobutyryloximine of 2-(4-chlorobutyryl)amino-5-methylbenzophenone and 2-(3-chloropropyl)-6-methyl-4-phenylquinazoline 3-oxide. The crystal and molecular structure of this oxide was established by X-ray diffraction structural analysis. The molecule is planar. Synthesis of 3-alkylamino-3-(2-hydroxyaryl)-1-polyfluoroalkylprop-2-en-1-ones and 2-polyfluoroalkyl-4H-chromen-4-imines

Condensation of Schiff"s bases (prepared from 2-hydroxy- or 2-hydroxy-5-methylacetophenones and primary amines) with ethyl polyfluoroalkanoates in the presence of LiH in THF affords 3-alkylamino-3-(2-hydroxyaryl)-1-polyfluoroalkylprop-2-en-1-ones, which undergo cyclization in acidic media into 2-polyfluoroalkyl-4H-chromen-4-iminium salts. When neutralized with aqueous ammonia, the latter give 2-polyfluoroalkyl-4H-chromen-4-imines in high yields.     

2-Polyfluoroalkylchromones. 9. Synthesis and structures of 5-(2-hydroxyaryl)-7-polyfluoroalkyl-1,4,8-triazabicyclo[5.3.0]dec-4-enes

2-Polyfluoroalkylchromones reacted with diethylenetriamine at 20 °C to form the corresponding 1,4,8-triazabicyclo[5.3.0]dec-4-ene derivatives. The crystal structures of 5-(2-hydroxyphenyl)-7-trifluoromethyl-1,4,8-triazabicyclo[5.3.0]dec-4-ene and 1-(2-aminoethyl)-7-(2-hydroxy-5-methoxyphenyl)-5-(1,1,2,2-tetrafluoroethyl)-2,3-dihydro-1H-1,4-diazepine were established by X-ray diffraction analysis.     

Synthesis of 1-(2-polyfluoroacylaminophenyl)-3-polyfluoroalkylpropane-1,3-diones and 2-polyfluoroalkyl-4-quinolones

The condensation of 2-aminoacetophenone with R^FCO₂Et (R^F = CF₃, CF₂H, CF₂CF₂H) in the presence of LiH in THF or Bu^tOK in Bu^tOH affords either 2-polyfluoroalkyl-4-quinolones or 1-(2-polyfluoroacylaminophenyl)-3-polyfluoroalkylpropane-1,3-diones, depending on the ratio of the initial reactants. The latter are hydrolyzed in an acidic medium to produce 2-polyfluoroalkyl-4-quinolones. N-Methyl-2-trifluoromethyl-4-quinolone was synthesized from 2-aminoacetophenone, CF₃CO₂Et, and MeI in the presence of Bu^tOK.

Substituted 6-aminopyrano[2,3-c]pyrazoles were synthesized by the two-component condensation of arylidenemalononitriles and substituted 5-pyrazolones or three-component condensation of aromatic aldehydes, malononitrile, and substituted 5-pyrazolones. It was established by X-ray crystallographic analysis that pyranopyrazoles exist in the 2H and not the 1H tautomeric form.     

Synthesis of 1-[1-bromo-2-benzoyl(2-thenoyl)vinyl]-1,1-dimethylhydrazinium bromides from 1-bromo-2-benzoyl(2-thenoyl)acetylenes and 1,1-dimethylhydrazine

1-[1-Bromo-2-benzoyl(2-thenoyl)vinyl]-1,1-dimethylhydrazinium bromides were synthesized by the reactions of 1-aroyl-2-bromoacetylenes withN,N-dimethylhydrazine in acetonitrile at 20°C. 4-Aryl(benzyl)sulfonyl-2-chloro-5-polyfluoroalkyl-1,2,3-triazoles. The first monocyclic N-chloro-1,2,3-triazoles

Treatment of 4-aryl(benzyl)sulfonyl-5-polyfluoroalkyl-v-triazoles with NaOCl gave the 4-aryl(benzyl)-sulfonyl-2-chloro-5-polyfluoroalkyl-v-triazole derivatives which contain a chlorine atom only on the N₍₂₎ atom of the heterocycle. The structure of 2-chloro-5-(1,1,2,2,3,3-hexafluoropropyl)-4-(p-tolyl-sulfonyl)-2H-[1,2,3]triazole has been established by X-ray structural investigation. The presence of a highly polarized N—Cl bond with a positive halogen atom causes the N-chlorotriazoles to react with KCN and KF as strong acids to form the potassium salts of the triazoles and to form 4-arylsulfonyl-2-(2-chloro-1-ethoxyethyl)-5-polyfluoroalkyl-2H-[1,2,3]triazoles with vinyl ethyl ether. 1-( p -Fluorophenyl)-2-(2′-pyridyl)ethanol and 1-( p -fluorophenyl)-2-(2′-pyridyl)ethene obtained from the condensation reaction of 2-picoline and p -fluorophenylaldehyde under catalyst-and solvent-free conditions

The novel intermediate 1-(p-fluorophenyl)-2-(2′-pyridyl)ethanol or 2-[2′-(1-hydroxy-1-(p-fluorophenyl)ethyl]pyridine and the corresponding novel dehydration compound 1-(p-fluorophenyl)-2-(2′-pyridyl)ethene or 2-[p-fluorophenylvinyl]pyridine were obtained from the condensation reaction of p-fluorophenylaldehyde and 2-picoline under catalyst-and solvent-free conditions. The intermediate 1-(p-fluorophenyl)-2-(2′-pyridyl)ethanol was obtained at 42 h reaction time and temperature of 120°C, respectively. ¹H-NMR, IR spectroscopic data of the 1-(p-fluorophenyl)-2-(2-pyridyl)ethanol clearly showed the presence of the-CH₂-CHOH-group. The compound was obtained as a white powder with m.p. 121–122°C and a yield of 8%. For 1-(p-fluorophenyl)-2-(2-pyridyl)ethene, the reaction conditions were similar, but the reaction temperature was increased to yield the double bond in the 1-(p-fluorophenyl)-2-(2′-pyridyl)ethene. At the reaction temperature of 140°C, the compound was a slightly brown powder with a m.p. of 78°C and yield of 18%. ¹H-NMR, IR spectroscopic data for the 1-(p-fluorophenyl)-2-(2′-pyridyl)ethene showed the presence of a double bond in trans configuration (-CH=CH-), characteristic of a styrylpyridine.
