In this study, we developed a fluorescence assay for the highly sensitive and selective detection of Hg2+ and Pb2+ ions using a gold nanoparticle (Au NP)-based probe. The Hg–Au and Pb–Au alloys that formed on the Au NP surfaces allowed the Au NPs to exhibit peroxidase-mimicking catalytic activity in the H2O2-mediated oxidation of Amplex UltraRed (AUR). The fluorescence of the AUR oxidation product increased upon increasing the concentration of either Hg2+ or Pb2+ ions. By controlling the pH values of 5 mM tris–acetate buffers at 7.0 and 9.0, this H2O2–AUR–Au NP probe detected Hg2+ and Pb2+ ions, respectively, both with limits of detection (signal-to-noise ratio: 3) of 4.0 nM. The fluorescence intensity of the AUR oxidation product was proportional to the concentrations of Hg2+ and Pb2+ ions over ranges 0.05–1 μM (R2 = 0.993) and 0.05–5 μM (R2 = 0.996), respectively. The H2O2–AUR–Au NP probe was highly selective for Hg2+ (>100-fold) and Pb2+ (>300-fold) ions in the presence of other tested metal ions. We validated the practicality of this simple, selective, and sensitive H2O2–AUR–Au NP probe through determination of the concentrations of Hg2+ and Pb2+ ions in a lake water sample and of Pb2+ ions in a blood sample. To the best of our knowledge, this system is the first example of Au NPs being used as enzyme-mimics for the fluorescence detection of Hg2+ and Pb2+ ions.  相似文献

A rapid,sensitive,selective and reliable strip assay based on DNA-functionalized gold nanoparticles for Hg2+ detection has been developed,with a detection limit 5 nmol/L.The measurement principle was based on thymine-Hg2+-thymine(T-Hg2+-T) coordination chemistry and streptavidin-biotin interaction.The major advantages of this assay are that results can be read visually without any instrument in less than 10 min and that it does not require any sample pretreatment.  相似文献

Equilibrium dialysis and atomic absorption analysis were used to obtain adsorption isotherms and determine the stoichiometric binding constants of Cu(II) and Cd(II) ions to DNA from Spirulina platensis in solutions. The stoichiometric constants of Cu(II) and Cd(II) ions with DNA from S. platensis in 3 mM NaCI are 15.56⋅104 and 14.40⋅104, respectively. Effect of ionic strength and DNA GC content on binding constants of Cu(II)- and Cd(II)-DNA complexes were studied out. It was showed that the binding constants of Cu(II)- and Cd(II)-DNA complexes decrease with increase of ionic strength. The empirical dependences of logK on the GC content has been derived for Cd(II)- and Cu(II)-DNA complexes.  相似文献

A colorimetric nanoprobe-mercury-specific DNA-functionalized gold nanoparticles (Au-MSD) was developed for sensing Hg(2+). The new mercury-sensing concept relies on measuring changes in the inhibition of "non-crosslinking" aggregation of Au-MSD-induced by the folding of mercury-specific DNA strand through the thymine-Hg(2+)-thymine (T-Hg(2+)-T) coordination. In the absence of Hg(2+), a high concentration of MgCl(2) (50 mM) results in a rapid aggregation of Au-MSD because of the removal of charge repulsion. When Hg(2+) is present, the particles remain stable due to the folding of MSD functionalized on the particle surface. The assay enables the colorimetric detection of Hg(2+) in the concentration range of 0.1-10 μM Hg(2+) ions with a detection limit of 60 nM, and allows for the selective discrimination of Hg(2+) ions from the other competitive metal ions. Toward the goal for practical applications, the sensor was further evaluated by monitoring Hg(2+) in fish tissue samples.  相似文献

We have investigated the effect of the folding of DNA aptamers on the colloidal stability of gold nanoparticles (AuNPs) to which an aptamer is tethered. On the basis of the studies of two different aptamers (adenosine aptamer and K+ aptamer), we discovered a unique colloidal stabilization effect associated with aptamer folding: AuNPs to which folded aptamer structures are attached are more stable toward salt-induced aggregation than those tethered to unfolded aptamers. This colloidal stabilization effect is more significant when a DNA spacer was incorporated between AuNP and the aptamer or when lower aptamer surface graft densities were used. The conformation that aptamers adopt on the surface appears to be a key factor that determines the relative stability of different AuNPs. Dynamic light scattering experiments revealed that the sizes of AuNPs modified with folded aptamers were larger than those of AuNPs modified with unfolded (but largely collapsed) aptamers in salt solution. From both the electrostatic and steric stabilization points of view, the folded aptamers that are more extended from the surface have a higher stabilization effect on AuNP than the unfolded aptamers. On the basis of this unique phenomenon, colorimetric biosensors have been developed for the detection of adenosine, K+, adenosine deaminase, and its inhibitors. Moreover, distinct AuNP aggregation and redispersion stages can be readily operated by controlling aptamer folding and unfolding states with the addition of adenosine and adenosine deaminase.  相似文献

A novel and sensitive electrochemical DNA biosensor has been developed for the detection of DNA hybridization. The biosensor was proposed by using copper(II) complex of Luteolin C30H18CuO12 (CuL2) as an electroactive indicator based on silver nanoparticles and multi-walled carbon nanotubes (Ag/MWCNTs) modified glassy carbon electrode (GCE). In this method, the 4-aminobenzoic acid (4-ABA) and Ag nanoparticles were covalently grafted on MWCNTs to form Ag/4-ABA/MWCNTs. The proposed method dramatically increased DNA attachment quantity and complementary ssDNA detection sensitivity for its large surface area and good charge-transport characteristics. DNA hybridization detection was performed using CuL2 as an electroactive indicator. The CuL2 was synthesized and characterized using elemental analysis (EA) and IR spectroscopy. Cyclic voltammetry (CV) and fluorescence spectroscopy were used to investigate the interaction between CuL2 and ds-oligonucleotides (dsDNA). It was revealed that CuL2 presented high electrochemical activity on GCE, and it could be intercalated into the double helices of dsDNA. The target ssDNA of the human hepatitis B virus (HBV) was quantified in a linear range from 3.23 × 10−12 to 5.31 × 10−9 M (r = 0.9983). A detection limit of 6.46 × 10−13 M (3σ, n = 11) was achieved.  相似文献

Platinum nanoparticles were used in combination with multi-walled carbon nanotubes (MWCNTs) for fabricating sensitivity-enhanced electrochemical DNA biosensor. Multi-walled carbon nanotubes and platinum nanoparticles were dispersed in Nafion, which were used to fabricate the modification of the glassy carbon electrode (GCE) surface. Oligonucleotides with amino groups at the 5′ end were covalently linked onto carboxylic groups of MWCNTs on the electrode. The hybridization events were monitored by differential pulse voltammetry (DPV) measurement of the intercalated daunomycin. Due to the ability of carbon nanotubes to promote electron-transfer reactions, the high catalytic activities of platinum nanoparticles for chemical reactions, the sensitivity of presented electrochemical DNA biosensors was remarkably improved. This article report microwave assisted green method for the synthesis of silver nanoparticle from Brassica oleracea var. Italica (BI) extract. The synthesized silver nanoparticle (AgNP-BI) was characterized by various analytical techniques. Here we are reporting three methods for the sensing of Ni (II) from the synthesized AgNP-BI. First one is the detection of Ni (II) ion based on changes in the absorbance resulting from the complex formation of the Ni (II) ion with AgNP-BI. The second one is the fluorescent sensing of Ni (II) ion using AgNP-BI by the changes in the fluorescence intensity. The third one is the electrochemical sensing of Ni (II) ion in which silver nanoparticle attached to the platinum electrode surface. The above-mentioned methods exhibit outstanding selectivity towards Ni (II) ion. The practical application of the AgNP-BI was also carried out for the trace determination of Ni (II) ions. The limit of detection was found to be 0.932 µM using differential pulse voltammetry (DPV).  相似文献

The negatively charged ruthenate(II) complex [Ru(bpy)(PPh3)(CN)3]? and gold nanoparticles (AuNPs) were used for detecting lysozyme (LYS). The luminescence of the ruthenate(II) complex is quenched by AuNPs, and this induces the aggregation of AuNPs and a color change from red to blue. After addition of lysozyme, the positively charged lysozyme and the negatively charged ruthenate(II) complex bind each other by electrostatic interaction firstly. This prevents AuNPs from aggregation and quenches the emission of the ruthenate(II) complex. Its luminescence and the degree of aggregation of the AuNPs can be used to quantify LYS. The fluorometric calibration plot is linear in the 0.01 to 0.20 μM LYS concentration range, and the calibration plot is linear between 0.02 and 0.20 μM of LYS. The color of the solution can be easily distinguished by bare eyes at 0.08 μM or higher concentration of LYS. The applicability of the method was verified by the correct analysis of LYS in chicken egg white.

Trimercaptotriazine-modified gold nanoparticles exhibit strong SERS effects, yielding vibrational profiles very sensitive to the presence of heavy metal ions. Because of the contrasting response observed for selected vibrational bands in the SERS profiles, they provide useful nanoprobes for Hg2+ and Cd2+ ions, allowing direct quantitative assays by employing relative peak intensity ratios instead of using internal standards.  相似文献

We have developed a simple, low-cost, paper-based probe for the selective colorimetric detection of copper ions (Cu(2+)) in aqueous solutions. The bovine serum albumin (BSA)-modified 13.3-nm Au nanoparticle (BSA-Au NP) probe was designed to detect Cu(2+) ions using lead ions (Pb(2+)) and 2-mercaptoethanol (2-ME) as leaching agents in a glycine-NaOH (pH 12.0) solution. In addition, a nitrocellulose membrane (NCM) was used to trap the BSA-Au NPs, leading to the preparation of a nanocomposite film consisting of a BSA-Au NP-decorated membrane (BSA-Au NPs/NCM). The BSA-Au NPs probe operates on the principle that Cu deposition on the surface of the BSA-Au NPs inhibits their leaching ability, which is accelerated by Pb(2+) ions in the presence of 2-ME. Under optimal solution conditions (5 mM glycine-NaOH (pH 12.0), Pb(2+) (50 μM), and 2-ME (1.0 M)), the Pb(2+)/2-ME-BSA-Au NPs/NCM enabled the detection of Cu(2+) at nanomolar concentrations in aqueous solutions by the naked eye with high selectivity (at least 100-fold over other metal ions). In addition, this cost-effective probe allowed for the rapid and simple determination of Cu(2+) ions in not only natural water samples but also in a complex biological sample (in this case, blood sample).  相似文献

A test strip for detection of Hg(2+) in aqueous solution based on the DNA-functionalized gold nanoparticles (DNA-AuNPs) was developed and evaluated. When Hg(2+) ions were introduced, the biotinylated DNA(2) hybridized with thiolated DNA(1) functionalized on the AuNPs (DNA(1)-AuNPs) to form mismatch complexes through thymine-Hg(2+)-thymine (T-Hg(2+)-T) coordination. The formed mismatch complexes and excess DNA(1)-AuNPs could be captured on the test line formed by streptavidin and the control line formed by DNA(3)-BSA, respectively. Two red lines appeared due to the accumulation of AuNPs, enabling visual detection of Hg(2+) with a detection limit of about 6 nM. The assay results can be obtained within 5 min. The results show that the test strip has excellent sensitivity and selectivity for detection of Hg(2+); thus it holds a great potential for rapid, on-site and real time detection of Hg(2+).  相似文献

DNA modified nanoparticles (AuNPs) are an established and widely used type of nucleotide sensor. We sought to improve the design by applying short rigid DNA duplexes near the surface of the AuNPs forming a so called double-anchored AuNP sensor, and compared it with other conventional DNA modified AuNPs. We developed a simple, non-enzymatic approach for the colorimetric detection of glucose based on a gold nanoparticles (Au NPs) assisted silver mirror reaction (AuSMR). The linear range of the concentration of glucose is from 0.04 mM to 1 mM, and the lowest concentration that can be distinguished by the naked eye is 10 nM. We have developed a colorimetric method for the determination of Pb(II) ions. It is based on the use of gold nanoparticles and a guanine-rich synthetic oligonucleotide. On addition of Pb(II), the color of the solution turns from red to blue. The ratio of the UV-vis absorption at 630?nm and 525?nm is proportional to the concentration of Pb(II) ions in the range from 10 to 100?nM, and the detection limit is 20?nM. Other metal ions do not interfere if present in up to a 10-fold molar excess. The method was successfully applied to the detection of Pb(II) in lake water and urine. The recovery in case of spiked samples is 92%. The results show that this method is sensitive, simple and fast.

In this study, new xanthine biosensors, XO/Au/PVF/Pt and XO/Pt/PVF/Pt, based on electroless deposition of gold(Au) and platinum(Pt) nanoparticles on polyvinylferrocene(PVF) coated Pt electrode for detection of xanthine were presented. The amperometric responses of the enzyme electrodes were measured at the constant potential, which was due to the electrooxidation of enzymatically produced H2O2. Compared with XO/PVF/Pt electrode, XO/Au/PVF/Pt and XO/Pt/PVF/Pt exhibited excellent electrocatalytic activity towards the oxidation of the analyte. Effect of Au and Pt nanoparticles was investigated by monitoring the response currents at the different deposition times and the different concentrations of KAuCl4 and PtBr2. Under the optimal conditions, the calibration curves of XO/Au/PVF/Pt and XO/Pt/PVF/Pt were obtained over the range of 2.5 × 10?3 to 0.56 mM and 2.0 × 10?3 to 0.66 mM, respectively. The detection limits were 7.5 × 10?4 mM for XO/Au/PVF/Pt and 6.0 × 10?4 mM for XO/Pt/PVF/Pt. The effects of interferents, the operational and the storage stabilities of the biosensors and the applicabilities of the proposed biosensors to the drug samples analysis were also evaluated.  相似文献

The present work proposes the use of Agave sisalana (sisal fiber) as an natural adsorbent for ions Pb(II) and Cd(II) biosorption from natural waters. The flame atomic absorption spectrometry was used for quantitative determination and study of the ions Pb(II) and Cd(II) adsorption on the solid phase. The Fourier transform infrared spectroscopy (FT IR) was used to investigate the sisal structure and the specific BET surface area was analyzed. The biosorption potential of sisal as biosorbent for the removal of the ions Pb(II) and Cd(II) from aqueous solution was investigate considering the followings parameters: pH, biomass amount and contact time. Langmuir and Freundlich isotherms were used to evaluate adsorption behavior of the ions on this solid phase. The results showed that sisal has a surface area to adsorption of 0.0233 m2 g− 1, and the OH and CO functional groups are the main involved in the biosorption. The best interpretation for the experimental data was given by Freundlich isotherm that proposes a monolayer sorption with a heterogeneous energetic distribution of active sites, accompanied by interactions between sorbed molecules. The maximum monolayer biosorption capacity was found to be 1.85 mg g− 1 for Cd (II) and 1.34 mg g− 1 for Pb (II) at pH 7 and 296 K. This phase solid can be used for biosorption of cadmium and lead in polluted natural waters.  相似文献

We report on a novel method for visual detection silver(I) ion. It is based on the finding that Ag(I) ions are rapidly reduced by hydroquinone to form a shell of silver on the surface of gold nanoparticles (AuNPs) which act as catalysts for this reaction. This leads to a color change from red to yellow which can be seen with bare eyes. This scheme is sensitive and highly specific for Ag(I) ions. The detection limits are 5 μM for visual inspection and 1 μM for photometric readout, respectively. The method was successfully applied to the determination of Ag(I) ions in spiked lake water and soil.
