贵州兴仁煤砷中毒患者的死因调查与防治   总被引:1,自引:0,他引:1  
运用高分辨率等离子体质谱法(HRICP-MS)测定了贵州兴仁居民燃用煤中砷含量,并调查了煤砷中毒死者详细资料。结果显示,当地居民燃用煤中砷仍然高度富集,从1976年到2003年,贵州兴仁砷中毒患者共死亡265人,其中死于癌症人数最多,死于肝硬化、腹水的病人其次,其中大部分患者死亡时间集中在1992年以后,说明贵州兴仁煤砷中毒的远期危害逐渐暴露,煤砷中毒的预防与防治应从各个方面全面进行。  相似文献

Abstract— Over a period of 5 years, we performed 29 experiments in which survival curves of UV-irradiated adenovirus were determined using fibroblast strains from 10 normal persons and from 7 persons having Cockayne's syndrome. In all of these, the survival of UV-irradiated adenovirus 5 was less when assayed using monolayers of fibroblasts from Cockayne's syndrome patients than from normal persons. Survival curves using normal fibroblasts were, within error, straight lines on a log survival vs. linear fluence plot. Survival curves obtained using Cockayne's syndrome fibroblasts showed 2 components: an initial sensitive component, reflecting the behavior of approx. 75% of the infected cells, followed by a component having normal sensitivity. In the 28 experiments that were considered reliable, 58 curves were done using Cockayne's fibroblasts, 41 using normal human fibroblasts. Although experimental variation was encountered, there was no individual case in which sensitivity as measured using Cockayne's was equal to (or less than) the sensitivity measured using normal fibroblasts.  相似文献

Abstract— The use of hematoporphyrin derivative (HpD) has previously been demonstrated to be beneficial in clinical cancer therapy. This paper describes cell culture studies used to examine HpD phototherapy in Chinese hamster ovary cells (line CHO). Survival curves have been obtained for both direct HpD toxicity and HpD induced photoinactivation. Examination of HpD induced photoinactivation as a function of stage in the cell growth cycle has also been performed, as has the quantitative measurement of HpD uptake in cells (using 3H-HpD) as a function of cellular incubation time, serum concentration in the incubation medium, and cell cycle position. In the absence of light, no toxicity was observed for HpD incubation levels of up to 400 μg/m/ when incubations times were 3 h or less. Exposure of cells to light alone (> 590 nm, 4.0 mW/cm2) for 9 min was also found to be completely nontoxic. Survival curves obtained for exponentially growing cells labeled with various concentrations of HpD and subsequently illuminated with red light exhibited a threshold or shoulder region at short exposure times followed by exponential killing at longer exposure times. The cell cycle response curves for HpD induced photoinactivation of synchronized CHO cells was nearly flat, indicating no variation in sensitivity for cells treated at time periods from 6 to 15 h after mitosis. Additon of serum to the incubation medium resulted in improved plating efficiency and reproducible survival curves but decreased cellular uptake of HpD.  相似文献

The results of historical cohort studies of three arsenic poisoning incidents in Japan are presented. The first episode was in a small mountainous community near an arsenic mine and refinery, at Toroku, where patients with chronic arsenism were certified by application of the Pollution Health Damage Compensation Law. The second area was in a small town, Namiki-cho, near Nakajo-machi, where wells were poisoned by arsenic produced in an arsenic(III) sulfide factory. As to the third cohort, eight residents of Nishikawa-machi who ingested well-water suffered arsenic poisoning about 35 years ago. The standardized mortality ratios were used in analyzing these data. Excesses of cancer mortality, especially lung cancer, were observed among the subjects in these areas. In Japan, there are many arsenic poisoning episodes, involving for example soy-sauce poisoning, powdered-milk poisoning and other incidents associated with arsenic mines. Thus it is necessary to clarify the chronic effects of arsenic in these areas.  相似文献

Arsenic exposure to humans is pervasive, and, increasingly, studies are revealing adverse health effects at ever lower doses. Drinking water is the main route of exposure for many individuals; however, food can be a significant source of arsenic to an individual, especially if their diet is rice-based. Infants are particularly susceptible to dietary exposure, since many first foods contain rice and they have a low body mass. Here we report on arsenic concentration and speciation in infant formulas and first foods. Speciation is essential for food analysis because of the much greater toxicity of inorganic arsenic species and the possibility that arsenic in food (unlike water) may be present in either inorganic or organic forms. Infant milk formulas were low in total arsenic (2.2-12.6 ng g(-1), n=15). Non-dairy formulas were significantly higher in arsenic than dairy-based formulas. Arsenic in formula was almost exclusively inorganic and predominantly arsenic(V). Arsenic concentration in purees (n=41) and stage 3 foods (n=18) ranged from 0.3-22 ng g(-1). Rice-fortified foods had significantly higher total arsenic concentrations than non rice-based foods. Again arsenic speciation was predominantly inorganic; arsenic(III) was the main species with lower concentrations of DMA and arsenic(V) also present. These data confirm that infants are exposed to arsenic via diet, and suggest that careful attention to diet choices may limit this.  相似文献

Sheep on the island of North Ronaldsay (Orkney, UK) feed mostly on seaweed, which contains high concentrations of dimethylated arsenoribosides. Wool of these sheep contains dimethylated, monomethylated and inorganic arsenic, in addition to unidentified arsenic species in unbound and complexed form. Chromatographic techniques using different separation mechanisms and detectors enabled us to identify five arsenic species in water extracts of wool. The wool contained 5.2 ± 2.3 µg arsenic per gram wool. About 80% of the arsenic in wool was extracted by boiling the wool with water. The main species is dimethylarsenic, which accounted for about 75 to 85%, monomethylated arsenic at about 5% and the rest is inorganic arsenic. Depending on the separation method and condition, the chromatographic recovery of arsenic species was between 45% for the anion exchange column, 68% for the size exclusion chromatography (SEC) and 82% for the cation exchange column. The SEC revealed the occurrence of two unknown arsenic compounds, of which one was probably a high molecular mass species. Since chromatographic recovery can be improved by either treating the extract with CuCl/HCl (CAT: 90%) or longer storage of the sample (CAT: 105%), in particular for methylated arsenic species, it can be assumed that labile arsenic–protein‐like coordination species occur in the extract, which cannot be speciated with conventional chromatographic methods. It is clear from our study of sheep wool that there can be different kinds of ‘hidden’ arsenic in biological matrices, depending on the extraction, separation and detection methods used. Hidden species can be defined as species that are not recordable by the detection system, not extractable or do not elute from chromatographic columns. Single noble metal permanent modifiers such as, Rh, Ir, and Ru, as well as mixed tungsten plus noble metal (W-Rh, W-Ru, W-Ir) permanent modifiers thermally deposited on the integrated platform of transversally heated graphite atomizer were employed for the determination of arsenic in sludges, soils, sediments, coals, ashes and waters by electrothermal atomic absorption spectrometry. Microwave digests of solid samples and water samples were employed for obtaining the analytical characteristics of the methods with different permanent modifiers. The performance of the modifiers for arsenic determination in the real samples depended strongly on the type of permanent modifier chosen. The single noble metal (Rh, Ir and Ru) permanent modifiers were suitable for the analyte determinations in simpler matrices such as waters (recoveries of certified values 95-105%), but the analyte recoveries of certified values in sludges, soils, sediments, coals, and ashes were always lower than 90%. On the other hand, for the determination of arsenic, using W-Rh, W-Ru, and W-Ir permanent modifiers presented recoveries of certified values within 95-105% for all the samples. Long-term stability curves obtained for the determination of arsenic in environmental samples with different permanent modifiers (Rh, Ir, Ru, W-Rh, W-Ir, W-Ru) showed that the improvement in the tube lifetime depends on the tungsten deposit onto the platform. The tungsten plus noble metal permanent modifier presents a tube lifetime of at least 35% longer when compared with single permanent modifier. The results for the determination of As employing different permanent modifiers in the samples were in agreement with the certified reference materials, since no statistical differences were found after applying the paired t-test at the 95% confidence level.  相似文献

A direct flow-injection atomic-absorption spectrometric (FIA-AAS) method for the assessment of inorganic arsenic compounds and their metabolites was developed and statistically evaluated by the estimation of the method evaluation function (MEF), which provides detailed information on the analytical performance of the method, i.e., the average combined uncertainty and the magnitude of potential systematic errors. The method evaluation study demonstrated that the use of standard addition was a necessity to obtain an acceptable method performance at low concentrations typical for low dose exposure. In contrast the use of calibration curves resulted in a method with reduced sensitivity and high systematic error. The developed method, using standard addition, had a limit of detection (2.9 microg/l.) sufficiently low for the determination of hydride-generating arsenic species in urine from non-exposed and low exposed persons. Organoarsenicals such as arsenobetaine and arsenocholine are not detected by this method. Hence, the contribution of these compounds derived from a diet containing seafood does not affect the monitoring of inorganic arsenic compounds after occupational or environmental exposure. The high capacity of the FIA-AAS system (three minutes per sample measured by standard addition) together with the low limit of detection makes this method suitable for biological monitoring of inorganic arsenic exposure even though standard addition is required.  相似文献

We investigated the effects of nanosecond pulsed electric fields (nsPEF) on three human cell lines and demonstrated cell shrinkage, breakdown of the cytoskeleton, nuclear membrane and chromosomal telomere damage. There was a differential response between cell types coinciding with cell survival. Jurkat cells showed cytoskeleton, nuclear membrane and telomere damage that severely impacted cell survival compared to two adherent cell lines. Interestingly, disruption of the actin cytoskeleton in adherent cells prior to nsPEF exposure significantly reduced cell survival. We conclude that nsPEF applications are able to induce damage to the cytoskeleton and nuclear membrane. Telomere sequences, regions that tether and stabilize DNA to the nuclear membrane, are severely compromised as measured by a pan-telomere probe. Internal pore formation following nsPEF applications has been described as a factor in induced cell death. Here we suggest that nsPEF induced physical changes to the cell in addition to pore formation need to be considered as an alternative method of cell death. We suggest nsPEF electrochemical induced depolymerization of actin filaments may account for cytoskeleton and nuclear membrane anomalies leading to sensitization.  相似文献

Arsenic in drinking water affects millions of people around the world. While soluble arsenic is commonly measured, the amount of particulate arsenic in drinking water has often been overlooked. We report here determination of the acid-leachable particulate arsenic and soluble arsenicals in well water from an arsenic-poisoning endemic area in Inner Mongolia, China. Water samples (583) were collected from 120 wells in Ba Men, Inner Mongolia, where well water was the primary drinking water source. Two methods were demonstrated for the determination of soluble arsenic species (primarily inorganic arsenate and arsenite) and total particulate arsenic. The first method used solid phase extraction cartridges and membrane filters to separate arsenic species on-site, followed by analysis of the individual arsenic species eluted from the cartridges and filters. The other method uses liquid chromatography separation with hydride generation atomic fluorescence detection to determine soluble arsenic species. Analysis of acidified water samples using inductively coupled plasma mass spectrometry provided the total arsenic concentration. Arsenic concentrations in water samples from the 120 wells ranged from <1 to ∼1000 μg L−1. On average, particulate arsenic accounted for 39 ± 38% (median 36%) of the total arsenic. In some wells, particulate arsenic was six times higher than the soluble arsenic concentration. Particulate arsenic can be effectively removed using membrane filtration. The information on particulate and soluble arsenic in water is useful for optimizing treatment options and for understanding the geochemical behavior of arsenic in groundwater.  相似文献

The CISORP Water Sorption Analyser has been used to characterise a selection of solid samples at relative humidities from 0 to 100% and at ambient pressure. The analysis reveals many interesting features about the samples and shows the scope of the equipment. Hysteresis due to porosity and differences in the physical properties of similar chemical samples show up clearly in isotherm curves. Kinetic curves reveal features such as the level of stability of dehydrated food products, changes in the hydration states of salts, and the effect of adding powdered excipient on the water sorption behaviour of a pharmaceutical compound. Kinetic curves were also used to compare the water sorption behaviour of two types of wood found inside a pine cone, and to determine equilibrium moisture sorption by calculation. It was shown that many samples take up moisture irreversibly under the experimental conditions such as amorphous sucrose and other freeze-dried samples, as well as unstable crystalline forms of compounds. Wet samples such as soaked brick and archaeological wood from a well dry out irreversibly even at 100% RH. Recording isotherms at different temperatures allows the calculation of enthalpies of water sorption. If these are compared with the enthalpy of water condensation the two processes can be compared quantitatively. We examined the effects of daily (chronic) exposure to artificial UVB radiation on the survival and reproduction of Daphnia   magna over two generations. Control and experimental animals in each generation (parental and F1) were exposed to 16 h of UVA radiation and photosynthetically active radiation daily. In addition, experimental animals were exposed to 6 h of UVB during the middle of the light period. Survival and reproduction were followed for 12 days for each individual. Survival and production of F1 were significantly lower in the UVB exposed parental generation Daphnia than in controls. F1 exposure to UVB significantly decreased F1 survival and reproduction. Reproduction was lowest in UVB exposed F1 animals whose parents were also exposed to UVB. Adverse effects of UVB on offspring production may be magnified in successive generations suggesting that short-term experiments could underestimate the impact of increased UVB exposure on populations.  相似文献

Arsenic is toxic to humans with the lethal dose being approximately 1 mg/kg/day. At much lower long-term exposures, arsenic is hypothesized to increase the risk of certain cancers. We have developed an irradiation position for the neutron activation analysis (NAA) of nail specimens for arsenic, in support of a case-control study involving New Hampshire residents consuming well water above the EPA Safe Drinking Water Standard of 0.050 ppm. Arsenic is bound to nail keratin through sulfhydryl groups proportional to intake providing a convenient means of integrating arsenic intake in population-based studies. Our objective was to develop the necessary facilities and procedures by which relatively small samples (i.e. 20 to 100 mg) could be accurately analyzed for arsenic, so that affordable nutritional epidemiology investigations, requiring large numbers of samples (>1000 in this case), could be undertaken. A high-flux reflector position, with minimal axial variation throughout the fuel cycle, suitable for pneumatic-tube irradiations, was characterized by measurement of the neutron flux distribution (thermal and epithermal) within the irradiation capsule over time. Results from application of the method to a case-control study of basal and squamous cell skin cancer will be presented.  相似文献

Abstract— For E. coli B/r u.v.-irradiated while in logarithmic growth, the nature of the dose-response curve was strongly dependent on both pre- and post-irradiation conditions of growth. Survival curves for cells grown in nutrient medium, or minimal medium with glucose, and plated immediately after irradiation, demonstrated an initial insensitive or 'shoulder' region provided the plating medium was such that no derepression was required of operons controlling inducible enzyme systems. If, however, such derepression was called for, survival curves were of exponential form. Delay in plating resulted in the return of the survival curve to the shouldered form even when 'shift-down' media were used.
Of those cells grown before u.v.-irradiation in minimal media and plated thereafter with the same sugar as carbon source, only those grown with glucose (or lactose) demonstrated the shouldered survival curve.  相似文献

A 1 m diameter water lens was used to focus solar radiation, giving an 8-fold concentration of the total spectrum and a cytocidal flux similar to that of laboratory UV sources. Survival curves for human melanoma cells were similar for sunlight and 254 nm UV, in that D q, was usually larger than D o. An xeroderma pigmentosum lymphoblastoid line was equally sensitive to both agents and human cell lines sensitive to ionizing radiation (lymphoblastoid lines), crosslinking agents or monofunctional alkylating agents (melanoma lines) had the same 254 nm UV and solar survival responses as appropriate control lines. Two melanoma sublines derived separately by 16 cycles of treatment with sunlight or 254 nm UV were crossresistant to both agents. In one melanoma cell line used for further studies, DNA strand breaks and DNA-protein crosslinking were induced in melanoma cells by sunlight but pyrimidine dimers (paper chromatography) and DNA interstrand crosslinking (alkaline elution) could not be detected. The solar fiuence response of DNA repair synthesis was much less than that from equitoxic 254 nm UV, reaching a maximum near the D o value and then declining; semiconservative DNA synthesis on the other hand remained high. These effects were not due to changes in thymidine pool sizes. Solar exposure did not have a major effect on 254 nm UV-induced repair synthesis.  相似文献

In the manufacture of integrated circuits involving semiconductors, various hazardous materials including arsenic compounds are used. This paper present an evaluation of the biological monitoring of arsenic in the urine, blood and hair of semiconductor workers. Eighty-two blood samples were obtained from research and development workers in a semiconductor factory. Thirty-one samples of urine were obtained from the same group of worker before and after a work period (one shift). Seventy-three hair samples were similarly obtained from the workers. The concentration of arsenic in each sample was determined by arsine (AsH3) generation flame atomic absorption spectrometry after wet ashing of the sample. The mean concentration of arsenic in hair in these semiconductor workers was significantly higher than that of controls. The mean concentration of arsenic in hair from workers with a longer employment duration was higher than from those with a shorter employment duration. The mean concentration of arsenic in hair from workers engaged in epitaxial growth processes was higher than from workers engaged in other processes. The concentrations of arsenic in hair from semiconductor workers correlated with the duration of their employment. The mean concentration of arsenic in urine before a work a period (shift) was not significantly different from that of controls. The mean concentration of arsenic in urine after a work period was in fact lower than that of controls. The mean concentration of arsenic in blood was not significantly different from that of controls. The determination of arsenic in hair is therefore considered useful for the evaluation of relatively long-term exposure to arsenic in semiconductor workers.  相似文献

Human being have been using inorganic arsenic for a long time. Many reports on arsenic poisoning have been published: eg case reports, examination reports, post mortem reports, and epidemiological studies. Several aspects and features of arsenic poisoning are discussed in this report. Methods of inferring arsenic-related disease, disease classification according to acute or chronic criteria, exposure route, interaction, confounding factors, and the oxidation state of arsenic are all discussed. The effects of arsenic are classified into skin, respiratory system, gastrointestinal tract, liver, cardiovascular system, nervous system, and bone marrow effects. Carcinogenicity is an important chronic effect of arsenic poisoning, so special attention is paid to it in this review. In Japan, there have been many incidents of arsenic poisoning. In this review, we often use data from these cases, such as the Morinaga powdered-milk poisoning case, the Ube soy-sauce poisoning case, the Toroku mine incident, and the Nakajo well-water poisoning case. We emphasize here the necessity of planning follow-up studies and total health care for patients exposed to arsenic.  相似文献

The determination of arsenic(III) and arsenic(V), as inorganic arsenite and arsenate, was investigated by CE with capacitively coupled contactless conductivity detection (CE-C(4)D). It was found necessary to determine the two inorganic arsenic species separately employing two different electrolyte systems. Electrolyte solutions consisting of 50 mM CAPS/2 mM L-arginine (Arg) (pH 9.0) and of 45 mM acetic acid (pH 3.2) were used for arsenic(III) and arsenic(V) determinations, respectively. Detection limits of 0.29 and 0.15 microM were achieved for As(III) and As(V), respectively by using large-volume injection to maximize the sensitivity. The analysis of contaminated well water samples from Vietnam is demonstrated.  相似文献

The size, diffusional properties, and dynamics of reverse water-in-oil nanoemulsions, or reverse micelles (RMs), have been widely investigated because of interest in this system as a model for biological compartmentalization. Here, we have employed fluorescence lifetime correlation spectroscopy (FLCS) to reveal the dynamics and sizes of aerosol-OT (AOT)/isooctane RMs using a fluorescent xanthene derivative called Tokyo Green II (TG-II). The dye undergoes a partition and a shift in its tautomeric equilibrium such that the TG-II anion remains in the inner micellar aqueous core, and the neutral quinoid form lies in the interfacial region. By applying FLCS, we specifically obtained the lifetime filtered autocorrelation curves of the anionic TG-II, which shows a characteristic lifetime of approximately 4 ns. Analysis of the FLCS curves provides the diffusion coefficient and hydrodynamic radius of the RMs as well as micelle dynamics in the same experiment. The FLCS curves show dynamics in the microsecond time range, which represents an interconversion rate that changes the distribution of the TG-II neutral and anionic forms in the hydrophobic interface and the water core.  相似文献

Pico-second time-resolved time-correlated single photon counting (TCSPC) technique under the total internal reflection (TIR) condition has been used to study the photophysical properties of Coumarin 343 (C343) dye molecules adsorbed at the water/1,2-dichloroethane (DCE) interface. The fluorescence decay profile of C343 under TIR condition at the water/DCE interface was non-exponential and fitted to the double exponential decay function with the fluorescence lifetimes 0.3 and 3.6 ns, which proved the existence of two different forms of C343 species having largely different lifetimes at the interface. The longer fluorescence lifetime component of C343 at the interface is attributed to the emission from the monomeric form of the dye molecules and the shorter lifetime component is due to the aggregation of dye molecules. The penetration depth dependence of decay curves indicated no change in the fluorescence lifetime components, however, the amplitude corresponding to the lifetime of aggregate increased and the amplitude corresponding to the lifetime of monomer decreased with the decrease in penetration depth of the aqueous phase from the interface. Aggregation is significant in the interfacial layer. The decrease in monomer lifetime at the interface compared to that in the bulk solution is interpreted in terms of excitation energy migration between the dye molecules.  相似文献
