This paper reports the results of a study carried out to evaluate the potentiality of combining selective solvent extraction and linear gradient elution reversed-phase high performance liquid chromatography (RP-HPLC) for characterizing soil organic matter on the basis of polarity of its constituting organic compounds. Such approach comprises the sequential extraction of soil organic matter with organic solvents of increasing polarity in a Soxhlet extractor and the subsequent separation of each extract by RP-HPLC. Accordingly, each soil sample has been subjected to cycles of sequential extraction with n-hexane, dichloromethane, ethyl acetate, and methanol. Each sample extracted by one of the four solvents have been dried in a rotary evaporator device at 40°C and then dissolved again in the proper volume of extraction solvent to obtaining sample solutions of concentration ranging from 5.0 to 15 mg ml−1. These sample solutions have been subjected to RP-HPLC separation using a Supelcosil LC-ABZ column that has been eluted by a linear acetonitrile gradient in water, having the same profile for all samples. The study has evidenced the possibility of producing highly repeatable chromatographic profiles, which are correlated to the polarity of organic solvents employed for extracting the organic matter in the Soxhlet extractor.

The effectiveness of extracting p,p′-DDT and p,p′-DDE from aged contaminated soil samples by means of pressurized liquid extraction (PLE) was evaluated. Two soil samples, which were contaminated more than 10 years ago, were used in the investigation. The static extraction time was optimised and then validated against the total sum of target analytes obtained from multiple sequential extractions. The PLE results were also compared with Soxhlet extraction (SOX). PLE for 3×10 min at 100 °C was proven to be more exhaustive than SOX in the determination of p,p′-DDE from both soil samples. In the case of p,p′-DDT, PLE was found to be equally as exhaustive as SOX. Additionally, most of the previous PLE investigations used hazardous organic solvents such as n-hexane, toluene and dichloromethane mixed with acetone, whereas in this investigation the less toxic solvent combination n-heptane/acetone has been employed.  相似文献

The objective of this work was to investigate the extraction of Ilex paraguariensis leaves by means of three extraction techniques: pressurized liquid extraction (PLE, also called accelerated solvent extraction – ASE), maceration, and sonication. Samples of mate tea leaves were collected from an experiment conducted under agronomic control at Indústria e Comércio de Erva‐Mate Bar?o LTDA, Brazil. Six solvents with increasing polarities (n‐hexane, toluene, dichloromethane, ethyl acetate, acetone, and methanol) were used in this investigation. Chemical analysis of the extracts was performed by GC coupled with a mass spectrometer detector. The identification and quantification were accomplished by coinjections of certified standards. The results showed that no significant differences in the qualities of the extracts were noticed regarding the extraction methods. On the other hand, the PLE technique was found to be more effective for the extractions of caffeine, phytol, palmitic, and stearic acid. The use of PLE led to a significant decrease in the total extraction time, amount of solvent consumption, and manipulation of samples compared to maceration and ultrasound‐assisted extraction methods.  相似文献

Capillary supercritical fluid chromatography has been directly coupled with supercritical fluid extraction using modified carbon dioxide. The mixed fluids were prepared with a single pump on-line mixing system. The most important step in the SFE-SFC interface was the elimination of the modifier solvent. This was achieved by use of a coupled trap, 0.1 mm i.d. and 0.53 mm i.d. capillary tubing connected in series, with the collected solutes refocused on the second (0.53 mm i.d.) trap before transfer into the separation column. This enabled complete elimination of various modifier solvents and high efficiency collection of the solutes. The effect of the modifier on trapping efficiency was investigated using methanol, ethanol, dichloromethane, hexane, and toluene at a variety of concentrations. n-Eicosane was, for example, trapped quantitatively by modified carbon dioxide containing up to 13 % (w/w) methanol. The use of the technique has been demonstrated by selective extraction of n-paraffins, fatty acid methyl esters, and alcohols from a silica matrix; the effect of different modifiers on the extraction of a mixture of pesticides from soil has also been investigated.  相似文献

The binding properties of three p-tert-butyldihomooxacalix[4]arene tetraketone derivatives (tert-butyl 2b, adamantyl 2c and phenyl 2d) in the cone conformation and one derivative (methyl 2a) in a partial cone conformation, towards alkali and alkaline earth metal cations have been established by extraction studies of metal picrates from water into dichloromethane, stability constant measurements in methanol and acetonitrile, and by 1H NMR spectrometry. Transport experiments of metal picrates through a dichloromethane membrane were also performed. The results are compared to those obtained with closely-related calix[n]arene derivatives (n = 4 and 5) and discussed in terms of the substituents, size and conformational effects. Methylketone 2a is a poor binder for all the cations studied, due to its partial cone conformation. Ketones 2b, 2c and 2d show high extraction and complexation levels for the alkali cations, with similar profiles and preference for K+ and Na+ (plateau selectivity). Towards alkaline earth cations, these ketones show a strong peak selectivity for Ba2+ in extraction, but a plateau selectivity for Ca2+, Sr2+ and Ba2+ in complexation. The nature of the substituent attached to the ketone function has some influence on their binding properties, with phenylketone 2d being a slightly weaker binder than ketones 2b and 2c. 1H NMR titrations confirm the formation of 1:1 complexes between the ketones and the cations studied, also indicating that they should be located inside the cavity defined by the phenoxy and carbonyl oxygen atoms. Ketones 2b, 2c and 2d show transport rates that do not follow, in general, the same trends observed in extraction and complexation.  相似文献

A nontarget screening method was developed based on D-optimal designs for ultra-high performance supercritical fluid chromatography with positive and negative electrospray ionization mode mass spectrometry. A mixture of organic contaminants such as pesticides, steroids, surfactants, phenolic and fatty acids, and polycyclic aromatic hydrocarbon derivatives, was used for the optimization. An aprotic mixture of dichloromethane and acetone [3:1] performed overall best as the injection solvent. The highest peak capacities (n) were accomplished at the shallowest gradient (1%B/min), ammonium formate (n = 378 in negative ionization mode), or ammonium acetate (n = 327 in positive ionization mode) in methanol as the modifier. Capillary voltage, make-up solvent flow rate, water, and additive concentration were the most significant factors for improving peak intensity: higher peak intensities were obtained at lower additive concentrations (5mM ammonium formate), and with 5% water in positive ionization mode. Conversely, water had detrimental effects in negative ionization mode. The optimized method was used to quantify organic contaminants in 17 freshwater sediment samples from Copenhagen, Denmark. Out of 50 monitored contaminants, 35 were detected in at least one sample. Further, the method has a potential for target and nontarget screening analysis of organic contaminants in solid matrices.  相似文献

Free fatty acids were derivatized as amides (DFFA) by reaction with (R)‐(+)‐1‐phenylethylamine, using a simple, fast and robust reaction scheme. A HPLC method with diode array and ESI MS detection was developed for the analysis of the derivatized substances. Six fatty acids were used in the method development: myristic, linoleic, palmitic, oleic, margaric and stearic acids. Under these conditions the elution of the DFFA are well resolved with retention times raging from 6.9 to 16.0 min. Fatty acids were extracted from cemetery soil and from adipocere formation experimental soils using a Soxhlet extraction, using as solvent ether/dichloromethane (1:1). Each DFFA is characterized by three m/z peaks: molecular weight of the substance; molecular weight of a dimer of the substance; the molecular weight of the dimer plus the atomic mass of sodium. The analysis of soil samples detected the six fatty acids used in the method developed plus palmitoleic and pentadecanoic. Beside this set of eight fatty acids other 13 fatty acids were detected in trace quantities or only in some soils and some were tentatively assigned as: 10‐hydroxystearic, myristoleic, heptadecenoic and arachidic acids.  相似文献

In this paper, we present an optimized procedure for metabolomic analysis of endogenous metabolites in mouse fibroblast (L929) cell line using gas chromatography/time-of-flight mass spectrometry with multivariate statistics. The optimization of metabolite extraction was performed using three solvents: methanol, water, and chloroform, and then followed by methoxymation and silylation. This method was subsequently validated using 29 reference standards and cell line samples. The intra- and inter-day relative standard deviations (RSDs) of the standard compounds were lower than 15.0 and 25.0 %, respectively. As for most of the tested metabolites in cell line samples, RSDs were below 20.0 % for reproducibility and stability, respectively. We applied this approach in metabolomic study of L929 cells obtained from TiO2 nanoparticle-induced cytotoxicity model samples (n = 5) and control samples (n = 5). Metabolite markers associated with TiO2 nanoparticle-induced cytotoxicity were identified and validated by statistical methods and reference standards. Our work highlights the potential of this method for cell metabolomic study.

Soxhlet (methanol) and SFE extraction with carbon dioxide in the presence of modifiers at different temperatures (100–200°C) for the extraction of atrazine and its main metabolites from a soil sample were compared. The most effective extraction conditions for both atrazine and its metabolites (i.e. deethylatrazine and deisopropylatrazine) were Co2 modified with 20% molar methanol-trifluoroacetic acid (MeOH-TFA) (TFA 0.65M in MeOH) at 100°C, leading to an extraction efficiency comparable with that of Soxhlet extraction with MeOH for atrazine and ca. 20% higher for its main metabolites. The relative standard deviation (RSD) of SFE was lower than that obtained by Soxhlet extraction, probably because of less interference in the cGC-NPD determination. All the other modifiers evaluated (acetone, triethylamine, and methanol) were less effective than MeOH-TFA for the extraction of atrazine and its metabolites from a soil sample, even at high molar concentrations (20%) and use of higher extraction temperatures (200°C). These results indicate the importance of matrix effects and the need of the selection of an appropriate modifier in order to obtain quantitative extractions by SFE.  相似文献

High resolution capillary gas chromatography–mass spectrometry has been used in combination with various sample preparation methods to analyze a wide range of low molecular weight organic contaminants in municipal wastewaters. Volatile organic compounds were extracted using a purge and trap concentrator connected directly to a wide bore DB-624 capillary column. Because of the high organic load in wastewater samples, this method was preferred to the more sensitive closed loop stripping technique which suffered from competition effects and insufficient adsorbent capacity. Volatile and semi-volatile base/neutral compounds and acids were extracted with dichloromethane at pH 11 and pH 2, respectively, or were recovered using a simultaneous distillation-extraction procedure. Acidic compounds were subsequently derivatized using diazomethane methylation. Both fractions were separated with a 30m DB-5ms capillary column. Specific ionic substances (LAS, NTA, EDTA) were isolated by solid-phase extraction then derivatized with diazomethane (LAS) or acetyl chloride in n-butanol (NTA, EDTA) before GC–MS analysis. This array of techniques enabled the identification of more than 150 organic contaminants in the influent of a municipal wastewater treatment plant in northern suburban Paris. The individual concentrations of these contaminants ranged from < 0.1 μg/l to 5.7 mg/l.  相似文献

A new rapid, sensitive and validated HPLC method has been developed for the determination of methylxanthines and their metabolites in asthmatic patients. The method was initiated by using spiked urine samples on a silica monolithic column as a novel packing material. The mobile phase consisted of 10 mM potassium dihydrogen phosphate buffer/methanol (87.5:12.5 v/v), at a flow rate 1 mL/min. Detection was set at 274 nm. The LOQ for all the compounds ranged from 14 to 41 ng/mL. Excellent linearity was achieved over the studied range of concentration with correlation coefficients 0.9991–0.9998 (n = 6). The developed method was validated by precision and accuracy with RSD <2.55%. On extraction of the drugs and metabolites from the urine samples high recoveries were achieved ranging from 82.06 to 98.34% w/w on RP18 cartridges and methanol/chloroform (20:80 v/v) as the extraction solvent. This method has advantages over other methods using conventional C18 packings.  相似文献

This paper describes the use of graphene oxide/silica modified with nitro‐substituted tris(indolyl)methane as a solid‐phase extraction sorbent for the determination of organic acids. The resultant graphene oxide/silica modified with nitro‐substituted tris(indolyl)methane was characterized by FTIR spectroscopy and adsorption experiments. Solid‐phase extraction parameters such as sorbent type, sample solution pH, sample loading rate, eluent salt concentration, eluent methanol concentration, elution rate, sample loading, and elution volume were optimized. The method showed good precision, accuracy, sensitivity, and linear response for organic acids analysis over a concentration range of 1–100 μg/L for benzoic acid, p‐methoxybenzoic acid, and salicylic acid and 5–100 μg/L for the remaining organic acids (cinnamic acid, p‐chlorobenzoic acid, and p‐bromobenzoic acid) with coefficients of determination (r2) of higher than 0.9957. Limits of detection from 0.50 to 1.0 μg/L for six organic acids were achieved. The developed method was successfully applied to determine organic acids in real samples.  相似文献

Liquid – liquid extraction with N-methyl-2-pyrrolidone, combined with solid-phase extraction on silica gel using n-pentane and dichloromethane as solvents, has been used for the isolation and concentration of the aromatic fraction from lubricating oil. The identification of individual compounds in this aromatic fraction was performed by capillary gas chromatography–mass spectrometry (CGC-MS) and by determination of retention indices. Quantitative results were obtained by flame ionization detection (CGC-FID). Eighty four compounds, predominantly naphthalenes, phenanthrenes, biphenyls, fluorenes, dibenzo-thiophenes, and their alkylated derivatives, have been identified in a commercial lubricating oil. The total amount of the aromatic fraction did not exceed 0.04 % of the oil.  相似文献

The ergosterol content of soil can be used as an indicator of fungal activity. A method has been developed for the extraction and determination of ergosterol in organic soils, as part of a study to assess the correlation between fungal activity and the sequestration of metal pollutants. The moisture content of the soil affected the extraction process. Four consecutive extractions with methanol removed >95% of the ergosterol that can be obtained from the fresh sample (63% moisture) by exhaustive extraction. By freeze drying the soils prior to extraction (a) up to 35% more ergosterol was extracted after a single extraction, (b) >90% of the recoverable ergosterol was collected in two extractions and (c) the repeatability of the extraction was improved. Storage of soil extracts in the absence of light prevents degradation of ergosterol. A previously reported method for determination of ergosterol by HPLC has been improved by modification of the eluant composition. With 46% methanol/46% acetonitrile/8% dichloromethane, ergosterol was eluted with good resolution approximately 8 min after injection of 20 mul of the extract. The detection limit of the HPLC method was 0.5 mug/ml ergosterol, equivalent to 0.06 mug/g in 25 g fresh soil. Changes in ergosterol contents of peaty soil treated with fungicide, and in samples of the peaty podzol and a humus iron podzol in the vicinity of fungal fruiting bodies, have been determined.  相似文献

A novel and sensitive dispersive liquid–liquid microextraction method based on the solidification of the floating organic drop combined with high‐performance liquid chromatography and ultraviolet detection was used for the determination of atorvastatine in blood serum samples. The chromatographic separation of atorvastatin was carried out using methanol as the mobile phase organic modifier. Various parameters affecting the extraction efficiency were optimized, such as the kind and volume of extraction solvent (1‐undecanol) and disperser solvent (acetonitrile), pH, and the extraction time. The calibration curve was linear in the range of 0.2–6000 μg/L of atorvastatin (r2 = 0.995) with a limit of detection of 0.07 μg/L. The relative standard deviation for 100 μg/L of atorvastatin in human plasma was 8.4% (n = 4). The recoveries of plasma samples spiked with atorvastatin were in the range of 98.8–113.8%. The obtained results showed that the proposed method is fast, simple, and reliable for the determination of very low concentrations of atorvastatin in human plasma samples.  相似文献

An ultrasound-assisted solvent extraction procedure has been optimised to speed up total polycyclic aromatic hydrocarbons (T-PAHs) extraction from mussel soft tissue. The T-PAHs releases have been evaluated by spectrofluorimetry (excitation and fluorescence emission wavelengths of 300 and 382 nm, respectively, and using chrysene as calibrant). Variables such as sonication time, ultrasound frequency, n-hexane volume, dichloromethane volume, number of repeated extractions with n-hexane and number of repeated extraction with dichloromethane were simultaneously studied by applying a Plackett-Burman design (PBD) approach. Results showed that ultrasound frequency and n-hexane and dichloromethane volumes were statistically significant variables (confidence interval of 95%). These last two variables were finally optimised by using central composite designs (CCD), yielding optimum n-hexane and dichloromethane volumes of 2.5 and 6.5 ml, respectively. The lowest T-PAHs releasing at high ultrasound frequency (35 kHz) led to choice the lowest ultrasound frequency (17 kHz) to perform the extraction. Variables such as sonication time and number of repeated extraction with n-hexane or dichloromethane were statistically non-significant and they were fixed at 10 min and the extraction with n-hexane and dichloromethane were performed once. The limit of detection was 0.021 μg g−1 (referred to dried mass), the repeatability of the overall method was 4.7% (n = 9) and the analytical recoveries were between 98 and 105%. The proposed method was finally applied to 16 mussel samples (Mytilus galloprovincialis) from Ría de Arousa estuary (Galicia, northwest Spain).  相似文献

Ondansetron, a 5‐hydroxytryptamine type 3 (5‐HT3) receptor antagonist, is regarded as an excellent candidate to treat chemotherapy‐ and radiotherapy‐induced nausea and vomiting. To better understand the metabolic profiles of ondansetron in human urine, the metabolites were analyzed using liquid chromatography/mass spectrometry (LC/MSn). Urine samples were collected after oral administration of 8 mg ondansetron to healthy volunteers. Then samples were treated by solid‐phase extraction and detected with LC/MSn. Besides ondansetron, in human urine, a total of 19 metabolites including 13 new metabolites were detected and identified via comparing the retention time and product ion spectra with those of reference standards isolated and characterized. The results showed that ondansetron was metabolized via hydroxylation, glucuronidation, sulfation and minor N‐demethylation in human. LC/MSn was demonstrated to be useful and sensitive in the metabolic study of ondansetron.  相似文献

A quick, easy, cheap, effective, rugged, and safe procedure was designed to extract pesticide residues from fruits and vegetables with a high percentage of water. It has not been used extensively for the extraction of phthalate esters from sediments, soils, and sludges. In this work, this procedure was combined with gas chromatography with mass spectrometry to determine 16 selected phthalate esters in soil. The extraction efficiency of the samples was improved by ultrasonic extraction and dissolution of the soil samples in ultra‐pure water, which promoted the dispersion of the samples. Furthermore, we have simplified the extraction step and reduced the risk of organic solvent contamination by minimizing the use of organic solvents. Different extraction solvents and clean‐up adsorbents were compared to optimize the procedure. Dichloromethane/n‐hexane (1:1, v/v) and n‐hexane/acetone (1:1, v/v) were selected as the extractants from the six extraction solvents tested. C18/primary secondary amine (1:1, m/m) was selected as the sorbent from the five clean‐up adsorbents tested. The recoveries from the spiked soils ranged from 70.00 to 117.90% with relative standard deviation values of 0.67–4.62%. The proposed approach was satisfactorily applied for the determination of phthalate esters in 12 contaminated soil samples.  相似文献

A method using derivatization and supercritical fluid extraction coupled with gas chromatography was developed for the analysis of dimethylarsinate, monomethylarsonate and inorganic arsenic simultaneously in solid matrices. Thioglycolic acid n‐butyl ester was used as a novel derivatizing reagent. A systematic discussion was made to investigate the effects of pressure, temperature, flow rate of the supercritical CO2, extraction time, concentration of the modifier, and microemulsion on extraction efficiency. The application for real environmental samples was also studied. Results showed that thioglycolic acid n‐butyl ester was an effective derivatizing reagent that could be applied for arsenic speciation. Using methanol as modifier of the supercritical CO2 can raise the extraction efficiency, which can be further enhanced by adding a microemulsion that contains Triton X‐405. The optimum extraction conditions were: 25 MPa, 90°C, static extraction for 10 min, dynamic extraction for 25 min with a flow rate of 2.0 mL/min of supercritical CO2 modified by 5% v/v methanol and microemulsion. The detection limits of dimethylarsinate, monomethylarsonate, and inorganic arsenic in solid matrices were 0.12, 0.26, and 1.1 mg/kg, respectively. The optimized method was sensitive, convenient, and reliable for the extraction and analysis of different arsenic species in solid samples.  相似文献

Abstract

Meclofenamic acid is extracted to dichloromethane together with the internal standard diclofenac sodium. After evaporation of the organic solvent the residue is dissolved in the chromatographic eluent and analyzed by liquid chromatography. The acids are separated on a column packed with Spherisorb ODS with methanol - phosphate buffer as the eluent and detected at 280 nm. A possible metabolite of meclofenamic acid was also detected in the chromatograms. The detection limit for meclofenamic acid in plasma was 0.361 μmol/L (0.107 μg/ml) for 1.5 ml sample size. The conditions of quantitative extraction of meclofenamic acid and diclofenac to dichloromethane are given.  相似文献
