A novel poly(methacrylic acid-ethylene glycol dimethacrylate) (MAA-EGDMA) monolith microextraction method coupled with CZE was proposed for rapidly determining a mixture of opiates comprising heroin, 6-monoacetylmorphine, morphine, codeine, papaverine, and narcotine in human urine. The extraction device contained a regular plastic syringe, the poly(MAA-EGDMA) monolithic capillary tube (530 microm id x 3 cm) and a plastic pinhead, which connected the monolithic capillary tube and the syringe without leakage. In the polymer monolith microextraction, the sample solution was ejected via the monolithic capillary tube by a programmable syringe pump, followed by desorption with an aliquot of appropriate solution, which was collected into a vial for the subsequent analysis by CZE. The best separation was achieved using a buffer composed of 0.1 M disodium hydrogen phosphate (adjusted to pH 4.5 with 1 M hydrochloric acid) and 20% methanol v/v with temperature and voltage of 25 degrees C and 25 kV, respectively. By applying electrokinetic injection with field-enhanced sample stacking, detection limits of 6.6-19.5 ng/mL were achieved. Excellent method of reproducibility was found over a linear range of 80-2000 ng/mL. 相似文献

A method based on in-tube solid-phase microextraction and capillary zone electrophoresis (CZE) was proposed for simultaneously determining four amphetamines (amphetamine, methamphetamine, 3,4-methylenedioxyamphetamine, and 3,4-methylenedioxymethamphetamine) in urine. A poly(methacrylic acid-ethylene glycol dimethacrylate) monolithic capillary column, which can provide sufficient extraction efficiency, was introduced for the extraction of amphetamines from urine samples. The hydrophobic main chains and acidic pendant groups of the monolithic column make it a superior material for extraction of basic analytes from aqueous matrix. After extraction, the samples were analyzed by CZE. The best separation was achieved using a buffer composed of 0.1 M disodium hydrogen phosphate (adjusted to pH 4.5 with 1 M hydrochloric acid) and 20% methanol v/v, with a temperature and voltage of 25 degrees C and 20 kV, respectively. By applying electrokinetic injection with field-amplified sample stacking, detection limits of 25-34 microg/L were achieved. Excellent method of reproducibility was found over a linear range of 0.1-5 mg/L. Determination of these analytes from abusers' urine sample was also demonstrated. 相似文献

A poly (methacrylic acid-ethylene glycol dimethacrylate) monolithic capillary column was prepared for in-tube solid-phase microextraction. Comparing with the commonly used open tubular extraction capillary, which cannot provide sufficient extraction efficiency since the ratio of its coating volume to that of the capillary void volume is relatively small, the monolithic column with greater phase ratio combined with convective mass transfer provides the possibility to improve the extraction efficiency with shorter capillary. As to poly (methacrylic acid-ethylene glycol dimethacrylate), its hydrophobic main chains and acidic pendant groups make it a superior material for extraction of basic analytes from aqueous matrix.An on-line monolithic capillary column solid phase microextraction (SPME) method was developed for determination of theobromine, theophylline and caffeine in serum samples. The high extraction efficiency was obtained for all the three analytes, yielding the detection limits of 12, 8 and 6.5 ng/mL by UV detection, respectively. Excellent method reproducibility (R.S.D. < 2.9%) was found over a linear dynamic range of 0.05-2 μg/mL in serum sample. In-tube solid-phase microextraction (SPME) based on a poly(methacrylic acid-ethylene glycol dimethacrylate) monolithic capillary column was investigated for the extraction of amphetamine, methamphetamine and their methylenedioxy derivatives. The monolithic capillary column showed high extraction efficiency towards target analytes, which could be attributed to its larger loading amount of extraction phase than conventional open-tubular extraction capillaries and the convective mass transfer procedure provided by its monolithic structure. The extraction mechanism was studied, and the results indicated that the extraction process of the target analytes was involved with hydrophobic interaction and ion-exchange interaction. The polymer monolith in-tube SPME-HPLC system with UV detection was successfully applied to the determination of amphetamine, methamphetamine and their methylenedioxy derivatives in urine samples, yielding the detection limits of 1.4 - 4.0 ng/mL. Excellent method reproducibility (RSD < 2.9%) was found over a linear range of 0.05-5 microg/mL, and the time for the whole analysis was only approximately 25 min. A hydroxylated poly(glycidyl methacrylate-co-ethylene dimethacrylate) (GMA-co-EDMA) monolithic capillary was prepared and investigated for in-tube solid-phase microextraction (SPME). The polymer monolith was synthesized by in-situ polymerization of GMA and EDMA in the presence of dodecanol and toluene as the mixed porogenic solvents. After polymerization, glycidyl groups were hydrolyzed with sulfuric acid to produce diol groups at the surface of the porous monolith. To investigate the extraction mechanism, several groups of model analytes (including neutral, acidic and basic) were selected to perform extractions. The resulting monolith showed high extraction selectivity towards polar compounds, which resulted from the enhancement of dipole-dipole and hydrogen bonding interactions relative to hydrophobic interactions. The equilibrium extraction time profiles were also monitored for those model compounds to assess the extraction capacity of the monolithic capillary. Moreover, the hydroxylated poly(GMA-co-EDMA) monolithic capillary exhibited satisfactory reproducibility and stability. Finally, the in-tube SPME-HPLC method, based on the developed monolithic capillary as the extraction media, was successfully applied to the determination of five polar organic contaminants in lake water. 相似文献

A restricted access material (RAM), alkyl-diol-silica (ADS), was used to prepare a highly bio-compatible solid-phase microextraction (SPME) capillary for the automated and direct in-tube extraction of several benzodiazepines from human serum. The bifunctionality of the ADS extraction phase prevented fouling of the capillary by protein adsorption while simultaneously trapping the analytes in the hydrophobic porous interior. This the first report of a restricted access material utilized as an extraction phase for in-tube SPME. The approach simplified the required apparatus in comparison to existing RAM column switching procedures, and more importantly eliminated the excessive use of extraction solvents. The biocompatibility of the ADS material also overcame the existing problems with in-tube SPME that requires an ultrafiltration or other deproteinization step prior to handling biological samples, therefore further minimizing the sample preparation requirements. The calculated oxazepam, temazepam, nordazepam and diazepam detection limits were 26, 29, 22 and 24 ng/ml in serum, respectively. The method was linear over the range of 50-50 000 ng/ml with an average linear coefficient (R2) value of 0.9998. The injection repeatability and intra-assay precision of the method were evaluated with five injections of a 10-microg/ml serum sample (spiked with all compounds), resulting in an average RSD<7%. Novel polymer monolith microextraction (PMME) using a poly(methacrylic acid-ethylene glycol dimethacrylate) (poly(MAA-EGDMA)) monolith in conjunction with capillary zone electrophoresis (CZE) was developed for the determination of several angiotensin II receptor antagonists (ARA-IIs) in human urine. The extraction device consisted of a regular plastic syringe (1 mL), a poly(MAA-EGDMA) monolithic capillary (2 cm x 530 microm I.D.) and a plastic pinhead connecting the former two components seamlessly. The extraction was achieved by driving the sample solution through the monolithic capillary tube using a syringe infusion pump, and for the desorption step, an aliquot of organic solvent was injected via the monolithic capillary and collected into a vial for subsequent analysis by CZE. The best separation was realized at 25 kV using a buffer that consisted of 50% acetonitrile and 50% buffer solution (v/v) containing 10 mM disodium hydrogenphosphate (adjusted to pH 2.3 with 1M hydrochloric acid). The method was successfully applied to the determination of telmisartan (T), irbesartan (I) and losartan (L) in urine samples with candesartan (C) as internal standard, yielding the detection limit of 15-20 ng/mL. Close correlation coefficients (R>0.999) and excellent method reproducibility were obtained for all the analytes over a linear range of 0.08-3 microg/mL. 相似文献

A hydroxylated poly(glycidyl methacrylate-co-ethylene dimethacrylate) (GMA-co-EDMA) monolithic capillary was prepared for polymer monolith microextraction (PMME). Coupled to HPLC with UV detection, this extraction medium was successfully applied to establish a simple and fast method for the analysis of clenbuterol (CLB) in urine. To obtain optimum extraction performance, the effects of pH value and ionic strength of the sample matrix on extraction efficiency were investigated. The linearity of the method was evaluated over a concentration range of 10-2000 ng/mL and the correlation coefficient (R2 value) was 0.9985. The detection limit and quantification limit were 2.3 and 7.7 ng/mL, respectively. Good reproducibility of the method was obtained, yielding the intra- and interday RSDs less than 5.1 and 9.1%, respectively. Moreover, the hydroxylated poly(GMA-co-EDMA) monolithic capillary exhibited good preparation reproducibility and long-term extraction life. When applied to the determination of CLB in urine samples, an effective removal of interfering compounds was achieved and recoveries were in the range of 87.6-106%. The determination of CLB from one real sample including pretreatment, extraction, and analysis could be finished within 30 min. 相似文献

An octyl-functionalized hybrid silica monolithic column was developed for in-tube solid-phase microextraction (SPME) to perform on-line preconcentration coupled to capillary high-performance liquid chromatography (microHPLC) analysis. A hybrid silica monolithic column functionalized with octyl groups was conveniently synthesized by a two-step acid/base-catalyzed hydrolysis/co-condensation of tetraethoxysilane (TEOS) and n-octyltriethoxysilane (C8-TEOS). The size of through-pores as well as the carbon content can be adjusted by changing the ratio of TEOS to C8-TEOS in the polymerization mixture. The extraction characteristics of the monolithic column prepared under optimized fabrication conditions were studied by using polycyclic aromatic hydrocarbons (PAHs) as the analytes. The sample volume that could be injected into the system was increased up to 1mL with simultaneous increase of column efficiency, when hybrid silica monolithic column was used as a precolumn. Good linear calibration curves (R>0.999) were obtained, and the limits of detection (signal-to-noise ratio, S/N=3) for the analytes were found to be between 2.4 and 8.1ng/mL with a UV absorbance detector, which are 299-456 times lower than those obtained without preconcentration. A method for the determination of endocrine disruptors, bisphenol A and 17alpha-ethinylestradiol, in environmental water samples was developed using in-tube solid-phase microextraction followed by liquid chromatography and fluorescence detection. A poly(acrylamide-vinylpyridine) monolithic capillary column was applied as the extraction media in view of its greater phase ratio than open-tubular capillaries and thus higher extraction efficiency. After optimizing the extraction conditions, bisphenol A and 17alpha-ethinylestradiol were extracted directly from water samples in a wide dynamic linear range of 0.5-1000 ng mL(-1), with the detection limits obtained as 0.064 and 0.12 ng mL(-1), respectively. The precision of the method was satisfactory with the intraday and interday RSD values smaller than 7.2%. Environmental water samples of different sources were successfully analyzed with the presented method and the monolithic capillary was proved to be robust and reusable in analyzing real water samples. 相似文献

A configuration of in-tube solid-phase microextraction (SPME) coupled to HPLC was constructed by using a pump and a six-port valve combined with a PEEK tube as the pre-extraction segment. The extraction capillary was fixed directly on the HPLC six-port valve to substitute for the sample loop. The whole system could be handled easily to perform accurate on-line extraction, and the possible inaccurate quantification caused by sample/mobile phase mixing when using an autosampler could be eliminated.A β-cyclodextrin coated capillary, prepared by sol-gel method, was used as the extraction capillary for in-tube SPME. Three non-steroidal anti-inflammatory drugs, ketoprofen, fenbufen and ibuprofen, were employed to evaluate the extraction performance of the capillary. After optimizing the extraction conditions, satisfactory extraction efficiency was obtained and detection limits for ketoprofen, fenbufen and ibuprofen in diluted urine samples were 38, 18 and 28 ng/mL, respectively. The extraction reproducibility was evaluated with intra-day and inter-day precision, and the R.S.D.s obtained were lower than 4.9 and 6.9%, respectively. A novel direct high-performance liquid chromatographic (HPLC) assay for the simultaneous determination of three salicylate glucuronide conjugates and other salicylate metabolites in human urine has been developed. Salicylate glucuronide conjugates were purified by HPLC from the urine of a volunteer after oral administration of aspirin and identified by selective hydrolysis with beta-glucuronidase and with sodium hydroxide. This method gave high reproducibility with coefficients of variation less than 10%. The total urinary recovery of salicylic acid after a single 1.2-g dose of soluble aspirin was greater than 90%. This assay has been successfully used to re-evaluate the capacity-limited pharmacokinetics of salicylic acid in humans. 相似文献

A solid-phase microextraction (SPME) method coupled to high-performance liquid chromatography with diode array detection (HPLC-DAD) for the analysis of six organochlorine fungicides (nuarimol, triadimenol, triadimefon, folpet, vinclozolin and penconazole) in wine was developed. For this purpose, polydimethylsiloxane-divinylbenzene-coated fibers were utilized and all factors affecting throughput, precision, and accuracy of the SPME method were investigated and optimized. These factors include: matrix influence, extraction and desorption time, percentage of ethanol, pH, salt effect and desorption mode. The performed analytical procedure showed detectability ranging from 4 to 27 microg l(-1) and precision from 2.4 to 14.2% (as intra-day relative standard deviation, RSD) and 4.7-25.7% (as inter-day RSD) depending on the fungicide. The results demonstrate the suitability of the SPME-HPLC-DAD method to analyze these organochlorine fungicides in red wine. 相似文献

Hyperlink robust biocompatible solid-phase microextraction (SPME) devices were prepared using continuous bed (monolithic) restricted-access media (RAM) as the SPME capillary insert. The RAM-based SPME approach was able to simultaneously separate proteins from a biological sample, while directly extracting the active components of caffeine, paracetamol and acetylsalicylic acid from the drug NeoCitramonum. The devices were interfaced with a CZE system and fully automated analysis for sample preconcentration, desorption, separation and quantification of analytes was evaluated. Comparative study of in-line coupled SPME-CZE using RAM and RP capillary inserts was carried out. 以[2-(丙烯酰氧基)乙基]三甲基氯化铵(DAC)为单体,乙二醇二甲基丙烯酸酯(EDMA)为交联剂在注射器中制备聚合物整体柱,用其固相萃取尿液中溴西泮(BRZ)、劳拉西泮(LRZ)和地西泮(DZP)3种苯二氮(卄卓)类药物(BZDs),并采用高效液相色谱法(HPLC)分析。实验考察了整体柱聚合时间及固相萃取条件(淋洗溶液、洗脱溶剂种类和体积)对BZDs萃取效率的影响。结果表明,仅聚合4 h得到的整体柱对BZDs吸附效率为100%。取尿液样品4 mL上样,用4 mL H2O冲洗,1 mL乙酸乙酯洗脱,采用高效液相色谱分析。在最优条件下,3种BZDs在4.0~1000 ng/mL范围内线性关系良好(r=0.999),检出限(S/N=3)和定量限(S/N=10)分别为1.0~1.2 ng/mL和3.3~4.0 ng/mL;在10、25和50 ng/mL加标水平下回收率为81.4%~102%,日内(n=3)和日间(n=3)相对标准偏差分别为1.2%~4.5%和2.5%~8.3%。该整体柱可对尿液中3种BZDs有效净化,且富集达12~15倍。方法构筑的聚合物整体柱制备简单,萃取高效,可成功用于尿液中3种BZDs的分析。 相似文献

Solid-phase microextraction (SPME) coupled with high-performance liquid chromatography (HPLC) for the determination of triazine is described. Carbowax/templated resin (CW/TPR, 50 μm), polydimethylsiloxane/divinylbenzene (PDMS/DVB, 60 μm), polydimethylsiloxane (PDMS, 100 μm), and polyacrylate (PA, 85 μm) fibers were evaluated for extraction of the triazines. CW/TPR and PDMS/DVB fibers were selected for further study. Several parameters of the extraction and desorption procedure were studied and optimized (such as types of fibers, desorption mode, desorption time, compositions of solvent for desorption, soaking periods and the flow rate during desorption period, extraction time, temperature, pH, and ionic strength of samples). Both CW/TPR and PDMS/DVB fibers are acceptable; a simple calibration-curve method based on simple aqueous standards can be used. The linearity of this method for analyzing standard solution has been investigated over the range 5-1000 ng mL−1 for both PDMS/DVB and CW/TPR fibers. All the correlation coefficients in the range 5-1000 ng mL−1 were better than 0.995 except Simazine and Atratone by CW/TPR fiber. The R.S.D.s range from 4.4% to 8.8 % (PDMS/DVB fiber) and from 2.4% to 7.2% (CW/TPR fiber). Method-detection limits (MDL) are in the range 1.2-2.6 and 2.8-3.4 ng mL−1 for the two fibers. The preparation of monolithic poly(butyl methacrylate-co-ethylene dimethacrylate) capillary columns using photoinitiated in situ polymerization within 200 microm i.d. capillaries and their application for microHPLC separations of proteins have been studied. The low resistance to flow characteristic of monolithic columns, enabled the use of very high flow rates of up to 100 microL/min representing a flow velocity of 87 mm/s. Very good separations of a model protein mixture consisting of ribonuclease A, cytochrome c, myoglobin, and ovalbumin was achieved in less than 40 s using a very simple single step gradient of the mobile phase. Interestingly, no effect of the pore size on the separations of proteins was observed for these monolithic columns within the size range of 0.66-2.2 microm. The monolithic microHPLC columns are found very robust and no changes in the long term separation performance and back pressure were observed. 相似文献

In-tube solid-phase microextraction (SPME) based on a poly(acrylamide-vinylpyridine-N,N'-methylene bisacrylamide) monolithic capillary was investigated and on-line coupled to HPLC for the determination of trace analytes in aqueous samples. The polymer monolith was conveniently synthesized in a fused silica capillary by in situ polymerization method. Several groups of analytes including non-steroidal anti-inflammatory drugs, phenols, non-peptide angiotensin II receptor antagonists and endocrine disrupting chemicals were extracted by the monolithic capillary. High extraction efficiency was achieved for the analytes investigated and great improvement of the limits of detection were obtained in comparison to that of direct chromatographic analysis and strong hydrophobic and ion-exchange interactions between the analytes and the polymer were confirmed. The newly developed monolithic capillary showed excellent reusability and high stability under extreme pH conditions during extraction. The possibility of applying the established method to water sample analysis was also demonstrated. 相似文献

In present study, a convenient, sensitive and environmentally friendly method for the determination of five benzoylurea insecticides (BUs) in water and juice samples was developed. To extract trace benzoylurea insecticides effectively, poly(methacrylic acid-co-ethylene dimethacrylate) monolith was prepared and used as the sorbent of multiple monolithic fiber solid-phase microextraction (MMF-SPME). The influences of preparation conditions of monolith and extraction parameters of MMF-SPME on BUs were studied thoroughly. Under the optimized conditions, the combination of MMF-SPME with high performance liquid chromatography-diode array detection (MMF-SPME-HPLC-DAD) showed expected analytical performance for target analytes. The limits of detection (S/N = 3) of the developed method were 0.026–0.075 μg L−1 in water and 0.053–0.29 μg L−1 in juice samples. Good linearity was obtained for analytes with the correlation coefficients (R2) above 0.99. Satisfactory repeatability and reproducibility was achieved, with relative standard deviations (RSD) of both less than 10%. Finally, the established MMF-SPME-HPLC-DAD method was successfully applied for the determination of BUs residues in juice and environmental water samples. Recoveries obtained for the determination of BUs in spiking samples ranged from 65.1% to 118%, with RSD below 10% in all cases. 相似文献
