This investigation describes the synthesis of a dual stimuli-responsive, amphiphilic ABC tri-block copolymer (BCP) based on the functional monomers via RAFT polymerization. In this case, ABC-type BCP was prepared based on N-isopropylacrylamide, n-butyl acrylate, and 4-vinylpyridine in DMF solvent using cyanomethyl dodecyl trithiocarbonate as the RAFT agent and azobisisobutyronitrile as a thermal initiator in a subsequent macro-RAFT approach, respectively. The BCPs were characterized by SEC, 1H-NMR, FTIR spectroscopy, and DSC analyses. Temperature and pH-dependent properties of the smart BCP micelles in aqueous medium were investigated using dynamic light scattering. Transmission electron microscopic images were taken at cryogenic and dry conditions to study the morphology of molecular assemblies of block copolymers in an aqueous medium. The phase and topographical images were captured by atomic force microscopy to understand the assembly of block copolymers in solvents of different polarities. The morphology of BCP micelles was transformed from flower-like to spherical in the presence of solvents with different polarities (H2O or CHCl3).  相似文献

Polymer-based nanoparticles show substantial promise in the treatment and diagnosis of cancer and other diseases. Herein we report an exploration of the cellular uptake of tailored, low dispersity segmented 1D nanoparticles which were prepared from an amphiphilic block copolymer, poly(dihexylfluorene)-b-poly(ethyleneglycol) (PDHF13-b-PEG227), with a crystallizable PDHF core-forming block and a ‘stealth’ PEG corona-forming block with different end-group functionalities. Segmented C–B–A–B–C pentablock 1D nanofibers with varied spatially-defined coronal chemistries and a selected length (95 nm) were prepared using the living crystallization-driven self-assembly (CDSA) seeded-growth method. As the blue fluorescence of PDHF is often subject to environment-related quenching, a far-red BODIPY (BD) fluorophore was attached to the PEG end-group of the coronal B segments to provide additional tracking capability. Folic acid (FA) was also incorporated as a targeting group in the terminal C segments. These dual-emissive pentablock nanofibers exhibited uptake into >97% of folate receptor positive HeLa cells by flow cytometry. In the absence of FA, no significant uptake was detected and nanofibers with either FA or BD coronal groups showed no significant toxicity. Correlative light and electron microscopy (CLEM) studies revealed receptor-mediated endocytosis as an uptake pathway, with subsequent localization to the perinuclear region. A significant proportion of the nanofibers also appeared to interact with the cell membrane in an end-on fashion, which was coupled with fluorescence quenching of the PDHF core. These results provide new insights into the cellular uptake of polymer-based nanofibers and suggest their potential use in targeted therapies and diagnostics.

The azobenzene‐based amphiphilic copolymers have drawn significant attention as a kind of multi‐responsive smart materials. The demand on deeper investigation of how the azobenzene group influences the micelles formation and light‐responsive behavior on molecular level is very urgent. In this article, multi‐responsive block copolymers, poly (acrylic acid)‐block‐poly[4'‐[[(2‐Methacryloyloxy)ethyl]ethylainino]azobenzene‐co‐poly (ethylene glycol) methyl ether methacrylate] (PAA‐b‐P (AzoMA‐co‐PEGMA)), with pH‐, light‐ and reduction‐responsiveness were synthesized by the monomers of AzoMA, PEGMA and acrylic acid via reversible addition‐fragmentation chain transfer polymerization (RAFT). The amphiphilic block copolymer presented aggregation‐induced emission effect, and it was pH, light, and reduction responsive. The results showed that the micelle size decreased with the decreasing of pH within a certain range. However, the particle size of micelles increased significantly when the pH was 4. Once adding reduction agent, the micelles were disassembly. Fluorescent molecule of Nile red was selected as a hydrophobic guest molecule to study the properties of encapsulating and releasing abilities of block copolymer micelles for guest molecules. The results showed that the loading capacity of three kinds of copolymer micelles was closely related to the aggregates formed by the hydrophobic block, mainly azobenzene block. Besides, the block copolymer micelles could release a certain amount of Nile red under the irradiation of UV light, the reduction with Na2S2O4 as reductant, and the exposure to alkaline environment. The mechanism of how the different status of azobenzene group influenced the self‐assembly and multi‐responsive behavior was explored on molecular level.  相似文献

A series of ABA amphiphilic triblock copolymers possessing polystyrene (PS) central hydrophobic blocks, one group with “short” PS blocks (DP = 54–86) and one with “long” PS blocks (DP = 183–204) were synthesized by atom transfer radical polymerization. The outer hydrophilic blocks were various lengths of poly(oligoethylene glycol methyl ether) methacrylate, a comb‐like polymer. The critical aggregation concentrations were recorded for certain block copolymer samples and were found to be in the range circa 10−9 mol L−1 for short PS blocks and circa 10−12 mol L−1 for long PS blocks. Dilute aqueous solutions were analyzed by transmission electron microscopy (TEM) and demonstrated that the short PS block copolymers formed spherical micelles and the long PS block copolymers formed predominantly spherical micelles with smaller proportions of cylindrical and Y‐branched cylindrical micelles. Dynamic light scattering analysis results agreed with the TEM observations demonstrating variations in micelle size with PS and POEGMA chain length: the hydrodynamic diameters (DH) of the shorter PS block copolymer micelles increased with increasing POEGMA block lengths while maintaining similar PS micellar core diameters (DC); in contrast the values of DH and DC for the longer PS block copolymer micelles decreased. Surface‐pressure isotherms were recorded for two of the samples and these indicated close packing of a short PS block copolymer at the air–water interface. The aggregate solutions were demonstrated to be stable over a 38‐day period with no change in aggregate size or noticeable precipitation. The cloud point temperatures of certain block copolymer aggregate solutions were measured and found to be in the range 76–93 °C; significantly these were ∼11 °C higher in temperature than those of POEGMA homopolymer samples with similar chain lengths. © 2008 Wiley Periodicals, Inc. Precise control over the morphology and dimensions of block copolymer (BCP) micelles has attracted interest due to the potential of this approach to generate functional nanostructures. Incorporation of liquid crystalline (LC) block can provide additional ways to vary micellar morphologies, but the formation of uniform micelles with controllable dimensions from LC BCPs has not yet been realized. Herein, we report the preparation of monodisperse cylindrical micelles with a LC poly(2‐(perfluorooctyl)ethyl methacrylate (PFMA) core via a fragmentation‐thermal annealing (F‐TA) process, resembling the “self‐seeding” process of crystalline BCP micelles. The average length of the cylinders increases with annealing temperature, with a narrow length distribution (Lw/Ln<1.1). We also demonstrate the potential application of the cylinders with LC cores as a cargo‐carrier by the successful incorporation of a hydrophobic fluorescent dye tagged with a fluorooctyl group.  相似文献

Preparing well-defined semiconducting nanostructures from conjugated polymers is of paramount interest for organic optoelectronic devices. Several studies have demonstrated excellent structural and size control from block copolymers (BCPs) containing non-conjugated blocks via crystallization-driven self-assembly (CDSA); however, the precise control of their size and shape remains a challenge due to their poor solubility, causing rapid and uncontrolled aggregation. This study presents a new type of fully conjugated BCP comprising two polyacetylene derivatives termed poly(cyclopentenylene-vinylene) to prepare semiconducting 1D nanofibers. Interestingly, the widths of nanofibers were tuned from 12 to 32 nm based on the contour lengths of their crystalline core blocks. Their lengths could also be controlled from 48 nm to 4.7 μm using the living CDSA. Monitoring of the growth kinetics of the living CDSA revealed the formation of micron-sized 1D nanofibers in less than 20 min. The rapid CDSA enabled us to watch real-time growth using confocal fluorescence microscopy.

New fully conjugated block copolymers formed semiconducting 1D nanofibers with excellent structural and size control. The rapid living CDSA enabled us to watch the real-time video of the whole self-assembly process.  相似文献

Living crystallization-driven self-assembly (CDSA) has emerged as an efficient route to generate π-conjugated-polymer-based nanofibers (CPNFs) with promising applications from photocatalysis to biomedicine. However, the lack of efficient tools to endow CPNFs with morphological stability and surface tailorability becomes a frustrating hindrance for expanding application spectrum of CPNFs. Herein, a facile strategy to fabricate length-controllable OPV-based (OPV = oligo(p-phenylenevinylene)) CPNFs containing a cross-linked shell with high morphological stability and facile surface tailorability through the combination of living CDSA and thiol–ene chemistry by using OPV5-b-PNAAM32 (PNAAM = poly(N-allyl acrylamide)) as a model is reported. Uniform fiber-like micelles with tunable length can be generated by self-seeding of living CDSA. By taking advantage of radical thiol–ene reaction between vinyls of PNAAM corona and four-arm thiols, the shell of micelles can be cross-linked with negligible destruction of structure of vinylene-containing OPV core. The resulting micelles show high morphological stability in NaCl solution and PBS buffer, even upon heating at 80 °C. The introduced extra thiol groups in the cross-linked shell can be further employed to install extra functional moieties via convenient thiol-Michael-type reaction. Given the negligible cytotoxicity of resulting CPNFs, this strategy opens an avenue to fabricate various CPNFs of diverse functionalities for biomedicine.  相似文献

Self-assembly of crystalline-coil block copolymers (BCPs) in selective solvents is often carried out by heating the mixture until the sample appears to dissolve and then allowing the solution to cool back to room temperature. In self-seeding experiments, some crystallites persist during sample annealing and nucleate the growth of core-crystalline micelles upon cooling. There is evidence in the literature that the nature of the self-assembled structures formed is independent of the annealing time at a particular temperature. There are, however, no systematic studies of how the rate of cooling affects self-assembly. We examine three systems based upon poly(ferrocenyldimethylsilane) BCPs that generated uniform micelles under typical conditions where cooling took pace on the 1–2 h time scale. For example, several of the systems generated elongated 1D micelles of uniform length under these slow cooling conditions. When subjected to rapid cooling (on the time scale of a few minutes or faster), branched structures were obtained. Variation of the cooling rate led to a variation in the size and degree of branching of some of the structures examined. These changes can be explained in terms of the high degree of supersaturation that occurs when unimer solutions at high temperature are suddenly cooled. Enhanced nucleation, seed aggregation, and selective growth of the species of lowest solubility contribute to branching. Cooling rate becomes another tool for manipulating crystallization-driven self-assembly and controlling micelle morphologies.

The interaction of amphiphilic block copolymers comprising an anionic block (polyacrylate or polymethacrylate) and a hydrophobic block (polystyrene, poly(butyl acrylate) or polyisobutylene) with lightly crosslinked poly(N,N-diallyl-N,N-dimethylammonium chloride) is studied for the first time. It is shown that the cationic hydrogel can sorb anionic amphiphilic block copolymers via electrostatic interaction with the corona of block copolymer micelles. The rate of sorption of block copolymer polyelectrolytes is significantly lower than the rate of sorption of linear polyions and is controlled by the lengths of the hydrophilic and hydrophobic blocks and the flexibility of the latter blocks. The sorption of amphiphilic block copolymers is accompanied by their self-assembly in the polycomplex gel and formation of a continuous hydrophobic layer impermeable to water and the low-molecular-mass salt dissolved in it.  相似文献

Cylindrical block copolymer micelles with a crystalline poly(ferrocenyldimethylsilane) (PFDMS) core and a long corona-forming block are known to elongate through an epitaxial growth mechanism on addition of further PFDMS block copolymer unimers. We now report that addition of the semicrystalline homopolymer PFDMS(28) to monodisperse short (ca. 200 nm), cylindrical seed micelles of PFDMS block copolymers results in the formation of aggregated structures by end-to-end coupling to form micelle networks. The resulting aggregates were characterized by dynamic light scattering (DLS), transmission electron microscopy (TEM), and atomic force microscopy (AFM). In some cases, a core-thickening effect was also observed where the added homopolymer appeared to deposit and crystallize at the core-corona interface, which resulted in an increase of the width of the micelles within the networks. No evidence for aggregation was detected when the amorphous homopolymer poly(ferrocenylethylmethylsilane) (PFEMS(25)) was added to the cylindrical seed micelles whereas similar behavior to PFDMS(28) was noted for semicrystalline polyferrocenyldimethylgermane (PFDMG(30)). This suggested that the crystallinity of the added homopolymer is critical for subsequent end-to-end coupling and network formation to occur. We also explored the tendency of the cylindrical seed micelles to form aggregates by the addition of PI-b-PFDMS (PI = polyisoprene) block copolymers (block ratios 6:1, 3.8:1, 2:1, or 1:1), and striking differences were noted. The results ranged from typical micelle elongation, as reported in previous work, at high corona to core-forming block ratios (PI-b-PFDMS; 6:1) to predominantly end-to-end coupling at lower ratios (PI-b-PFDMS; 2:1, 1:1) to form long, essentially linear structures. The latter process, especially for the 2:1 block copolymer, led to much more controlled aggregate formation compared with that observed on addition of homopolymers.  相似文献

We report the design and demonstration of a dual-stimuli-responsive block copolymer (BCP) micelle with increased complexity and control. We have synthesized and studied a new amphiphilic ABA-type triblock copolymer whose hydrophobic middle block contains two types of stimuli-sensitive functionalities regularly and repeatedly positioned in the main chain. Using a two-step click chemistry approach, disulfide and o-nitrobenzyle methyl ester groups are inserted into the main chain, which react to reducing agents and light, respectively. With the end blocks being poly(ethylene oxide), micelles formed by this BCP possess a core that can be disintegrated either rapidly via photocleavage of o-nitrobenzyl methyl esters or slowly through cleavage of disulfide groups by a reducing agent in the micellar solution. This feature makes possible either burst release of an encapsulated hydrophobic species from disintegrated micelles by UV light, or slow release by the action of a reducing agent, or release with combined fast-slow rate profiles using the two stimuli.  相似文献

The solubilization sites provided by micelles formed by a diblock copolymer with one neutral hydrophobic block, polystyrene, and one charged hydrophilic block, poly(acrylic acid) or poly(methacrylic acid), have been studied by fluorescence quenching of pyrene by polar and nonpolar quenchers. Pyrene solubilized into these micelles is distributed between the inner corona and the micelle core. The fraction of pyrene residing in the inner corona is almost unity for star micelles, where the corona-forming blocks are larger than the core-forming blocks, and around 0.5 for crew-cut micelles where the opposite situation prevails. The kinetics of the quenching process depends on the pyrene location, i.e. is static in the micelle core, and largely dynamic in the inner corona at low quencher concentration. The rate constants for fluorescence quenching by nitromethane are shown to increase with increasing pH.  相似文献

Well‐defined amphiphilic block copolymers were prepared by ring opening metathesis polymerization and their stimuli responsive behavior of formed micelles in aqueous solution was investigated. The hydrophobic core of the micelles consists of either a poly[5,6‐bis(ethoxymethyl)bicyclo[2.2.1]hept‐2‐ene]‐block with a glass transition Tg at room temperature or a poly[endo,exo[2.2.1]bicyclohept‐5‐ene‐2,3‐diylbis (phenylmethanone)] with a Tg of 143 °C. For the polyelectrolyte shell, the precursor block poly[endo,exo[2.2.1]bicyclohept‐5‐ene‐2,3‐dicarboxyclic tert‐butylester] was transformed into the free acidic block by cleavage of the tert‐butyl groups using trifluoroacetic acid. Micellar solutions were prepared by dialysis, dissolving the copolymers in dimethyl sulfoxide which was subsequently replaced by water. All polymers form micelles with radii between 10 and 20 nm at a pH‐value below 5, where the carboxylic acid groups are in the protonated state. The block copolymer micelles show a strong increase of the hydrodynamic radius with increasing pH‐value, due to the repulsion among the formed carboxylate anions resulting in a stretching of the polymer chains. In this state, the micelles exhibit responsive behavior to ionic strength where a contraction of the micelles is observed as the carboxylate charges are balanced by sodium ions, whereas no changes of the hydrodynamic radius on addition of salt are observed at low pH. © 2009 Wiley Periodicals, Inc. Microphase separated epoxy-based materials modified with an amphiphilic poly(styrene-block-ethylene oxide) diblock copolymer (PS-b-PEO) with low amount of PEO-block as well as ternary systems modified with this block copolymer and containing via sol–gel in situ synthesized TiO2 nanoparticles were prepared and characterized. The obtained results indicate that block copolymer had enough amount of PEO-block in order to achieve microphase separated materials for a high range of PS-b-PEO contents, morphologies changing from spherical micelles to long wormlike micelles passing through vesicles upon increasing copolymer amounts. In the case of 20 wt.% inorganic/organic epoxy-based materials, addition of synthesized TiO2 nanoparticles into PS-b-PEO-(DGEBA/MCDEA) system led to location of the nanoparticles in PEO-block/epoxy-rich confined between two microphase separated PS-block-rich phases. Designed highly transparent multiphase inorganic/organic epoxy-based materials possess interesting specific properties such as high UV shielding efficiency and high water repellence.  相似文献

Amphiphilic block and statistical copolymers of vinyl ethers (VEs) with pendant glucose residues were synthesized by the living cationic polymerization of isobutyl VE (IBVE) and a VE carrying 1,2:5,6‐di‐O‐isopropylidene‐D ‐glucose (IpGlcVE), followed by deprotection. The block copolymer was prepared by a two‐stage sequential block copolymerization, whereas the statistical copolymer was obtained by the copolymerization of a mixture of the two monomers. The monomer reactivity ratios estimated with the statistical copolymerization were r1 (IBVE) = 1.65 and r2 (IpGlcVE) = 1.15. The obtained statistical copolymers were nearly uniform with the comonomer composition along the main chain. Both the block and statistical copolymers had narrow molecular weight distributions (weight‐average molecular weight/number‐average molecular weight ∼ 1.1). Gel permeation chromatography, static light scattering, and spin–lattice relaxation time measurements in a selective solvent revealed that the block copolymer formed multimolecular micelles, possibly with a hydrophobic poly(IBVE) core and a glucose‐carrying poly(VE) shell, whereas the statistical copolymer with nearly the same molecular weight and segment composition was molecularly dispersed in solution. The surface properties of the solvent‐cast films of the block and statistical copolymer were also investigated with the contact‐angle measurement. © 2001 John Wiley & Sons, Inc. The synthesis, characterization and self-assembly of a novel amphiphilic block copolymer containing a poly(N-vinylpyrrolidone) as a segment of hydrophilic and poly(4-vinyl benzene chloride) (PVBC) arms are reported. The copolymer was characterized by FT-IR spectroscopy 1H NMR. The composition and the molecular weights of the block copolymers were established using gel permeation chromatography and 1H NMR. The water-soluble fraction of poly(N-vinyl-2-pyrrolidone) (PVP)/PVBC block copolymers formed micelles which were investigated at 25 °C in water at 5 mg/ml concentration using a tensiometer. The morphology of micelles in aqueous solution was determined by the AFM, SANS, and SAXS methods.  相似文献

The water content-dependent supramolecular structure formation of polystyrene-block-poly(acrylic acid) (PS-b-PAA) copolymer in the presence of a fourth-generation amine-terminated poly(amido amine) dendrimer (PAMAM) is investigated by dynamic light scattering, turbidity measurements, and transmission electron microscopy. The solvent system for this study is a mixture of dioxane/THF and water. A very complex turbidity profile is observed with increasing water content in the system and is explained by the presence of various aggregated structures based on strong interactions between the amine-containing dendrimers and the poly(acrylic acid) blocks of the polymer. The onset of the self-assembly of single chains of PS-b-PAA (primary structure) into single and multiple dendrimer core inverse micelles (secondary structure) is detected as very low water contents of cw < 2% wt (cwc). These micelles consist of dendrimers coated with PAA blocks, which are connected to the corresponding PS chains that form the corona. Further addition of water leads to an association of these micelles into compound multiple dendrimer core inverse micelles (tertiary structure) in the range of cw = approximately 6 to approximately 10% wt. At still higher water content, some of the acrylic acid chains of the block copolymer move from the vicinity of the dendrimer to the outside of the aggregates, resulting in a decrease in the size of the formed structures and the acquisition of progressively increasing hydrophilic character of the aggregates. Multiple dendrimer core inverse onion micelles are formed, which agglomerate into compound multiple dendrimer core inverse onion micelles at cw = approximately 12 to approximately 18% wt. Above this water content, vesicular structures are formed. The complexity is unusual for block copolymer systems and illustrates the importance of strong interactions in structure formation.  相似文献

Ultrasmall gold particles have been prepared inside the micelles of polystyrene-block-poly(ethylene oxide) and polystyrene-block-poly(2-vinylpyridine) in toluene. Starting point was the formation of a thermodynamically stable dispersion of HAuCl4 or LiAuCl4 in the inverse micelles of the block copolymer which were treated with hydrazine or pyrrole. Analysis of the effect of the reduction agent on the stability of the micelles yielded a simple model for the transformation process involving coagulation of the swelling micelles. Kinetic control of the different steps, i.e., reduction, mineralization, coagulation, film formation, allowed to prepare thin films in which highly uniform gold particles were arranged in yet unknown order. When pyrrole was employed for the reduction, the gold monocrystals got embedded in a shell of polypyrrole.  相似文献

We present here the synthesis of two kinds of amphiphilic block copolymers, a diblock copolymer MPEG‐b‐PTAm and a triblock copolymer MPEG‐b‐PLA‐b‐PTAm, which can self‐assemble into micelles with nitroxyl radicals‐containing PTAm segment in the core. The structure of the block copolymers was characterized by 1H NMR and GPC. Dynamic laser light scattering and transmission electron microscopy were used to study the micellar behavior of the two block copolymers in aqueous solution. The micelles carrying nitroxyl radicals in the core can generate electron paramagnetic resonance, which is stable for a period of time up to 8 min even in the presence of reducing reagent such as ascorbic acid. The enhanced stability against the reducing agent was ascribed to the inaccessibility of the nitroxyl radical core placed in the interior of the micelles. Poly(acrylic acid-b-styrene) (PAA-b-PS) amphiphilic block copolymers were synthesized by consecutive telomerization of tert-butyl acrylate, atom transfer radical polymerization (ATRP) of styrene, and hydrolysis. The resulting block copolymers were characterized by 1H NMR and GPC. These amphiphilic block copolymeric micelles were prepared by dialysis against water. Transmission electron micrograph (TEM) and laser particle sizer measurements were used to determine the morphology and size of these micelles. The results showed that these amphiphilic block copolymers formed spherical micelles with average size of 140–190?nm. The critical micelle concentration (CMC) and the kinetic stability of these micelles were investigated by fluorescence technique, using pyrene as a fluorescence probe. The observed CMC value was in the range of 0.075–0.351?mg/L. Kinetic stability studies showed that the stability of micelles increased with the decrease of the pH value of the solution.  相似文献
