Summary We carried out a comprehensive theoretical study on the alkaline hydrolysis of the bicyclic system of penicillins (a four-member ring fused to a thiazolidine ring) on the basis of aB AC2 mechanism. We assayed the MINDO/3, MNDO, and AM1 semi-empirical calculation methods in order to determine their suitability for studying -lactam rings.Both the geometric and the energetic results obtained for the different intermediate states were compared with literature values — chiefly those determined byab initio methods — with which they proved to be very consistent.The conformation of the carboxyl group at position 3 was found to be rather significant to the determination of the energy of the different reaction maxima and minima.  相似文献

The pharmacological properties of penam, 3-cepheme, and semisynthetic penicillins were predicted by means of the ORAKUL automated system. A comparative evaluation of the similarity between the structures of these compounds and the structures of 8800 biologically active substances in the data base of the system made it possible to uncover the high probability of the manifestation of anti-inflammatory, analgesic, antitumorigenic, antiallergic, and anticoagulant activity by structural analogs of -lactam antibiotics.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 4, pp. 555–564, April, 1992.  相似文献

Kinetic study of the reactions of amoxicillin (I), ampicillin (II) and cephlaxin (III) with p-dimethylaminobenzaldehyde (DAB) in weakly acidic EtOH/H2O solution has been investigated using spectrophotometric method. Relatively slow reversible reactions of first order with respect to the antibiotic have been found. A derived equation for detecting the existence of reversibility from the linearity has been introduced. The effect of anionic surfactants (sodium dodecyl sulfate, SDS) on the kinetic of these reactions in aqueous solution has been studied. The presence of 0.005 M of SDS increases the rate constants by 4.3, 2 and 3.3 times for I, II and III, respectively. The consequence of the rate constants have a similar order in absence and presence of SDS; III > II > I. The rate constants pass through maxima with increasing SDS concentration followed by a gradual but steady decrease in the rate as the surfactant concentration increases further. Multiple linear regression method has been performed to evaluate the binding constants of each drug and DAB with SDS from the resulted kinetic data. The results suggest using multiple linear correlation method for such calculations, which is more accurate, reliable and less time consuming. The calculated binding constants between these drugs with SDS are following the consequence I > II > III which is related to the differences in their substitutions. The kinetic results were employed for spectrophotometric microdetermination of these drugs (I–III) in aqueous solution. The method was based on the reaction of β-lactam with an excess of DAB in presence of SDS and HCl (pH 2) at a wavelength 410 nm. The results indicate that the presented method is simple, precise and accurate. This method is applied to bulk antibiotics and some of their pharmaceutical preparations.  相似文献

Unique block co-polymer P(MAn-St-NIPAm) has been successfully synthesized by reversible addition fragmentation chain transfer radical polymerization protocol. Based on the tunable hydrophobic/hydrophilic properties of the block co-polymer, a new open tubular capillary electrochromatography (OT-CEC) system has been constructed with the prepared block co-polymer as the coating and applied in analysis of β-lactam antibiotics in serum samples.  相似文献

In this article, we analyze the results of a molecular dynamics simulation in aqueous solution of the N-methylazetidinone molecule, often used to model β-lactam antibiotics. The radial distribution functions (RDFs) corresponding to the most interesting atoms, in terms of reactivity, are presented. We focus our study on the effect of a polar environment on the molecule. The solvent structure around the system is compared to the structure of β-lactam-water complexes, as obtained in a previous study of reaction mechanisms for the neutral and alkaline hydrolyses of N-methylazetidinone. Two types of complexes have been considered which are related to different hydrolysis mechanisms having similar energy barriers at the rate-limiting step of the reaction path. In the first type, the β-lactam-water interaction takes place through the oxygen carbonyl atom and there is agreement between the maxima of the RDFs obtained here and the ab initio structure of the complexes previously reported. In the second type, the interaction takes place through the nitrogen atom and we do not predict a coordination layer around the β-lactam nitrogen atom. The results suggest that in aqueous solution hydrolysis of the carbonyl group is the most probable starting point for the overall hydrolysis reaction. β-Lactam antibiotics, including penicillins and cephalosporins, are commonly used in veterinary medicine. Illegal use and abuse of β-lactams could cause allergy and selected bacterial resistance. BlaR-CTD, the carboxy-terminal of penicillin-recognizing protein BlaR from Bacillus licheniformis ATCC 14580, was utilized in this study to develop a receptor-based ELISA for detection and determination of β-lactam antibiotics in milk, beef, and chicken. This assay was based on directly competitive inhibition of binding of horseradish peroxidase-labeled ampicillin to the immobilized BlaR-CTD by β-lactams. The assay was developed as screening test with the option as semiquantitative assay, when the identity of a single type of residual β-lactam was known. The IC50 values of 15 β-lactam antibiotics, including benzylpenicillin, ampicillin, amoxicillin, dicloxacillin, oxacillin, nafcillin, cefapirin, cefoperazone, cefalotin, cefazolin, cefquinome, ceftriaxone, cefotaxime, cefalexin, ceftiofur and its metabolite desfuroylceftiofur were evaluated and ranged from 0.18 to 170.81 μg L?1. Simple sample extraction method was carried out with only phosphate-buffered saline, and the recoveries of selected β-lactam antibiotics in milk, beef, and chicken were in the range of 53.27 to 128.29 %, most ranging from 60 to 120 %. The inter-assay variability was below 30 %. Limits of detection in milk, beef, and chicken muscles with cefquinome matrix calibration were 2.10, 30.68, and 31.13 μg kg?1, respectively. This study firstly established a rapid, simple, and accurate method for simultaneous detection of 15 β-lactams in edible tissues, among which 11 β-lactams controlled by European Union could be detected below maximum residue limits.

Multi-drug-resistant forms of the Gram-negative pathogen Acinetobacter baumannii are an emerging threat to human health and further complicate the general problem of treating serious bacterial infections. Meeting this challenge requires an improved understanding of the relationships between the structures of major therapeutic targets in this organism and the activity levels exhibited against it by different antibiotics. Here we report the first crystal structures of A. baumannii penicillin-binding proteins (PBPs) covalently inactivated by four β-lactam antibiotics. We also relate the results to kinetic, biophysical, and computational data. The structure of the class A protein PBP1a was solved in apo form and for its covalent conjugates with benzyl penicillin, imipenem, aztreonam, and the siderophore-conjugated monocarbam MC-1. It included a novel domain genetically spliced into a surface loop of the transpeptidase domain that contains three conserved loops. Also reported here is the first high-resolution structure of the A. baumannii class B enzyme PBP3 in apo form. Comparison of this structure with that of MC-1-derivatized PBP3 of Pseudomonas aeruginosa identified differences between these orthologous proteins in A. baumannii and P. aeruginosa. Thermodynamic analyses indicated that desolvation effects in the PBP3 ligand-binding sites contributed significantly to the thermal stability of the enzyme-antibiotic covalent complexes. Across a significant range of values, they correlated well with results from studies of inactivation kinetics and the protein structures. The structural, biophysical, and computational data help rationalize differences in the functional performance of antibiotics against different protein targets and can be used to guide the design of future agents.  相似文献

β-Lactam peptides were envisioned as conformational constraints in antigenic peptides (APs). Three different β-lactam tripeptides of varying flexibility were prepared in solution and incorporated in place of the central part of the altered melanoma associated antigenic peptide Leu(27)-Melan-A(26-35) using solid phase synthesis techniques. Upon TFA cleavage from the solid support, an unexpected opening of the β-lactam ring occurred with conservation of the amide bond. After adaptation of the solid phase synthesis strategy, β-lactam peptides were successfully obtained and both opened and closed forms were evaluated for their capacity to bind to the antigen-presenting class-I MHC HLA-A2 protein system. None of the closed β-lactam peptides bound to HLA-A2, but their opened variants were shown to be moderate to good HLA-A2 ligands, one of them being even capable of stimulating a Melan-A-specific T cell line.  相似文献

The high-yielding synthesis of enantiomerically pure epicatechin gallate analogues where the A and/or B-ring hydroxylation is reduced or altered has been achieved by optimising routes to the catechin stereochemistry. The B-ring analogues were synthesised by using an electrophilic ring closure onto an enantiomerically enriched epoxide as a key step. The A and B-ring hydroxyl-deleted analogues were synthesised through a Mitsunobu cyclisation. For the B-ring analogues, the anti- (catechin) stereochemistry was converted to the syn- (epicatechin) stereochemistry by a known oxidation/reduction protocol. Absolute stereochemistry was derived from either a Sharpless epoxidation or asymmetric dihydroxylation.  相似文献

A simple and very effective approach to versatile carbohydrate β-lactam synthons of predictable absolute configuration has been developed. The procedure, which is based on the indium-mediated reaction of imines and bromoesters, was applied to the enantioselective synthesis of 3-monosubstituted and 3-disubstituted β-lactams from readily available carbohydrates. The opening of the β-lactamic ring gave rise to the corresponding sugar-derived β-amino acids. Transformation of the β-lactams into the corresponding azetidines was also achieved.  相似文献

Antibiotic drug resistance among hospital and community acquired methicillin resistant Staphylococcus aureus (MRSA) has dramatically eroded the efficacy of current therapeutics. We describe a chemical genetic strategy using antisense interference to broadly identify new drug targets that potentiate the effects of existing antibiotics against both etiological classes of MRSA infection. Further, we describe the resulting chemical genetic interaction networks and highlight the prominent and overlapping target sets that restore MRSA susceptibility to penicillin, cephalosporins, and carbapenems. Pharmacological validation of this approach is the potent synergy between a known inhibitor to a member of this genetic potentiation network (GlmS) and a broad set of β-lactam antibiotics against methicillin resistant Staphylococci. Developing drug-like leads to these targets may serve as rational and effective combination agents when paired with existing β-lactam antibiotics to restore their efficacy against MRSA.  相似文献

A sensitive and reliable method using capillary HPLC with UV-diode array detection (DAD) has been developed and validated for the trace determination of residues of 10 β-lactam antibiotics of human and veterinary use, in milk, chicken meat and environmental water samples. The analytes included ampicillin, amoxicillin, penicillin V, penicillin G, cloxacillin, oxacillin, dicloxacillin, nafcillin, piperacillin and clavulanic acid. Legal levels are regulated by the EU Council regulation 2377/90 in animal edible tissues for these compounds. For food analysis, a solid-phase extraction (SPE) procedure consisting in a tandem of Oasis HLB and Alumina N cartridges was applied for off-line preconcentration and cleanup. For water analysis, the first step was only necessary. The limits of detection for the studied compounds were between 0.04–0.06 μg l−1 for water samples and 0.80–1.40 μg l−1 (or μg kg−1) in the case of foods derived from animals. Average recoveries for fortified samples at different concentration levels ranged between 82.9% and 98.2%, with relative standard deviations (RSDs) lower than 9%. The method showed the advantages of capillary HPLC for the detection of these widely applied antibiotics in different samples at very low concentration levels.  相似文献

Herein, we report the development of mild, organocatalyzed routes to novel β-lactam carbapenem derivatives through Michael type CC bond forming reactions.
