重离子诱导的质粒DNA双链断裂分布研究   总被引:1,自引:0,他引:1  
利用能量为7.2MeV/u氖离子束辐照体外质粒DNA:pUC18,采用恒场凝胶电泳结合多功能荧光成像系统研究了pUC18双链断裂片段的分布。证实了双链断裂片段分布的非随机性,结果还发现DNA断裂后片段的交联现象,而且交联片段的分布也是非随机的。DNA is considered to be the most important and sensitive target in biological systems. In addition to the base damage, DNA strand breaks are the major lesion in the genome due to exposure to ionizing radiation. Mutation can be introduced to DNA as a result of enzymatic processing of DNA lesions or post irradiation replication. However, the mechanisms of radiation induced mutations are not well clarified at the molecular level. To study the effect on the simple plasmid DNA of heavy ion is even predominant or more feasible. Plasmid pUC18 DNA was prepared and irradiated by neon beam (7.199 MeV/u). The fragment distributions were determined by quantifying the ethidium bromide fluorescence. It can be seen that the shape of the intensity distributions is vastly different for the used radiation Dose. The distribution produced shows an excess of fragments particularly in 3 000 and 10 000 Gy the size range between 20—40 kbp and 20—50 bp. This clustering of double stranded fragments might be influenced by the higher order chromatin structure of genomic DNA. If so, DNA loop structures could correspond to the size range for which we observed DSB clustering. Further studies aim at elucidating the heterogeneity of DSB induction within the genome and investigate the influence of chromatin structure on the non random fragment distribution.  相似文献

Choosing ^7 Li and ^12C heavy ions respectively with different linear energy transfer (LET) values, purified plasmid DNA samples in aqueous solution are irradiated with various doses. The atomic force microscopy (AFM) is used for analysis of DNA fragments induced by both the kinds of heavy ions. There is a change of three forms of DNA, i.e. supercoiled, open circular and linear form, as the dose is observed. The distribution function of DNA fragment length is obtained for the first time and fitted with the Tsallis entropy statistical theory. The result indicates that AFM is a useful tool for analysis of the short fragment of DNA, high-LET heavy ion radiation induces DNA double strand breaks (DSBs) more effectively, and the distributions of the DSBs are more local and dense in comparison with low-LET radiation.  相似文献

Irradiation caused DNA single-strand breaks in S180 cells in the presence oflaser-hematoporphyrin derivative(HPD).The number of single strand breaks was 3.69 ×10~(10)break dolton~(-1).The analysis of base composition of DNA showed that the effect of laser-HPDirradiation on guanine was the highest,being 5-12 times as high as those of the three others(adenine,cytosine and thymine).It was different from the nature of DNA damage caused by γ-ray irradiation.  相似文献

本工作是基于蒙特卡罗模拟软件FLUKA对高能强流重离子加速器(HIAF)高能辐照终端感生放射性进行初步研究。该终端可运行质子最高能量为9.3 GeV,最大流强是1.45×1012 pps(particle per second)。研究内容包括:(1)预测高能辐照终端内活化物质的放射性活度特性;(2)预测不同冷却时间高能辐照终端内残余剂量率分布。研究结果表明,HIAF正常运行时高能辐照终端内的感生放射性主要受束流垃圾桶活化产生的放射性核素影响。当加速器连续运行100天冷却4小时,垃圾桶表面残余剂量率为2.375 mSv·h-1。终端内空气中13N和15O动态饱和比浓度大于其对应的导出空气浓度。冷却水中13N和15O的活度大于对应的ALImin。该研究是HIAF辐射防护基础研究以及加速器环境影响评价的一项重要内容。The Monte Carlo code FLUKA was used to predict the induced radioactivity of high-energy irradiation terminal of HIAF. The maximum energy of proton is 9.3 GeV, and the maximum current is 1.45×1012 pps (particle per second). In this study we were to predict:(1) the activity properties of activated substances in the experimental terminal; (2) the residual dose rate distribution in the experimental terminal at different cooling time. The results indicate that the induced radioactivity in the high energy irradiation terminal of the HIAF is mainly affected by the radionuclide induced in the beam dump. The residual dose rate on the surface of the beam dump is 2.375 mSv·h-1, after 100 d irradiation and 4 h cooling. The dynamic saturation ratio of 13N and 15O induced in the air inside the terminal is higher than its corresponding derived air concentration. The activity of 13 N and15O induced in cooling water is higher than its ALImin. This study is a part of radiation protection basic research and environmental impact assessment for HIAF.  相似文献

采用重离子辐照模拟方法和正电子湮没寿命测量技术研究了钨辐射损伤随辐照剂量的变化。20,60和90dpa(每个原子的位移次数)辐照损伤水平的实验结果表明,辐照在钨中产生单空位、双空位、位错和空位团等缺陷;随辐照剂量的增大,单空位、双空位和位错浓度增加,空位团的尺度和浓度都随之增大。Radiation damage in W has been studied as a function of irradiation dose by heavy ion simulation and positron annihilation lifetime measurement. The experimental results of 20, 60 and 90 dpa irradiations illustrate that the mono-and di-vacancies, dislocations and vacancy clusters are produced by the irradiation. The concentrations of the mono-and di-vacancies and dislocations and both the concentration and size of the vacancy clusters or voids all increase with the increasing of the irradiation dose.  相似文献

A pinned photodiode complementary metal–oxide–semiconductor transistor(CMOS) active pixel sensor is exposed to ~(60)Co to evaluate the performance for space applications. The sample is irradiated with a dose rate of 50 rad(SiO_2)/s and a total dose of 100 krad(SiO_2), and the photodiode is kept unbiased. The degradation of dark current, full well capacity,and quantum efficiency induced by the total ionizing dose damage effect are investigated. It is found that the dark current increases mainly from the shallow trench isolation(STI) surrounding the pinned photodiode. Further results suggests that the decreasing of full well capacity due to the increase in the density, is induced by the total ionizing dose(TID) effect, of the trap interface, which also leads to the degradation of quantum efficiency at shorter wavelengths.  相似文献

The Raman spectra of DNA in different levels of vitamin C with 10- and 30-min ultraviolet (UV) radiations were reported. The intensity of UV radiation was 18.68 W/m2. The experimental results proved that vitamin C could alone prevent UV radiation from damaging DNA, but the effects depended on the concentration of vitamin C. When the concentration of vitamin C was about 0.08-0.4 mmol/L, vitamin C decreased UV radiation-induced DNA's damage. When the concentration of vitamin C exceeded 0.4 mmol/L, vitamin C accelerated DNA's damage instead. Maybe the reason is that when DNA in aqueous solution is radiated by UV, free radicals come into being, and vitamin C can scavenge free radicals, so vitamin C in lower concentration can protect DNA. The quantity of free radicals is finite, when vitamin C is superfluous, free radicals have been scavenged absolutely and vitamin C is residual. Vitamin C is a strong reductant. Ta2O5 films axe deposited on fused silica substrates by conventional electron beam evaporation method. By annealing at different temperatures, Ta2 O5 films of amorphous, hexagonal and orthorhombic phases are obtained and confirmed by x-ray diffractometer (XRD) results. X-ray photoelectron spectroscopy (XPS) analysis shows that chemical composition of all the films is stoichiometry. It is found that the amorphous Ta2 O5 film achieves the highest laser induced damage threshold (LIDT) either at 355 or 1064nm, followed by hexagonal phase and finally orthorhombic phase. The damage morphologies at 355 and 1064nm are different as the former shows a uniform fused area while the latter is centred on one or more defect points, which is induced by different damage mechanisms. The decrease of the LIDT at 1064nm is attributed to the increasing structural defect, while at 355nm is due to the combination effect of the increasing structural defect and decreasing band gap energy.  相似文献

The testing techniques and experimental methods of the 60Co gamma irradiation effect on AlGaN/AlN/ GaN high electron mobility transistors (HEMTs) are established. The degradation of the electrical properties of the device under the actual radiation environment are analyzed theoretically, and studies of the total dose effects of gamma radiation on AlGaN/AlN/GaN HEMTs at three different radiation bias conditions are carried out. The degradation patterns of the main parameters of the AlGaN/AlN/GaN HEMTs at different doses are then investigated, and the device parameters that were sensitive to the gamma radiation induced damage and the total dose level induced device damage are obtained.  相似文献

<正>The DNA conformational transition depends on both the DNA sequences and environment such as solvent as well as electrolyte in the solution.This paper uses the AMBER8 package to investigate the electrolyte concentration influence on the dynamics of the A→B conformational transition of DNA duplex d(CGCGAATTCGCG)_2.The results from the restrained molecular dynamics(MD) simulations indicate that the total energies of the systems for A-DNA are always higher than those for B-DNA,and that the A→B conformational transition in aqueous NaCl solution is a downhill process.The results from the unrestrained MD simulations,as judged by the average distance between the C5' atoms(average helical rise per ten base pair),show that the concentrated NaCl solution slows down the A→B conformational transition.This observation can be well understood by analyses of the difference between the counterion distributions around A-DNA and B-DNA.  相似文献

Radiation induced offstate leakage in the shallow trench isolation regions of SIMC 0.18 μm nMOSFETs is studied as a function of dose rate. A "true" dose rate effect (TDRE) is observed. Increased damage is observed at low dose rate (LDR) than at high dose rate (HDR) when annealing is taken into account. A new method of simulating radiation induced degradation in shallow trench isolation (STI) is presented. A comparison of radiation induced offstate leakage current in test nMOSFETs between total dose irradiation experiments and simulation results exhibits excellent agreement. The investigation results imply that the enhancement of the leakage current may be worse for the dose rate encountered in the environment of space.  相似文献

DNA–RNA hybrid(DRH) plays important roles in many biological processes. Here, we use a thermodynamic theory to analyze the free energy and unpeeling properties of the overstretching transition for the DRH molecule and compare the results with double-helix DNA. We report that the RNA strand of DRH is easier to get unpeeled than the DNA strand while the difficulty in unpeeling the double helix DNA lies in between. We also investigate the sequence effect, such as GC content and purine content, on the properties of unpeeling the DRH. Further, to study the temperature effect, the forcetemperature phase diagram of DRH and DNA are calculated and compared. Finally, using a kinetic model, we calculate the force–extension curves in the DRH stretching and relaxation process under different pulling rates and temperatures. Our results show that both pulling rate and temperature have important influences on the stretching and relaxation kinetics of unpeeling the DRH. Putting all these results together, our work provides a comprehensive view of both the thermodynamics and kinetics in DRH overstretching.  相似文献

利用激光扫描共聚焦显微技术和碱性单细胞凝胶电泳技术对经过离子注入后的黑松花粉粒内细胞核的直接损伤效应进行了观察鉴定。 研究结果表明, 离子注入后可以直接损伤细胞核结构, 导致细胞核裂解。 细胞核的损伤程度与注入离子的剂量密切相关, 即细胞核DNA分子的损伤程度随着注入离子剂量的增加而提高。 The damage of pollen nuclei of Pinus thunbergii induced directly by ion implantation were measured by the laser confocal microscopy technique and the single cell gel electrophoresis test. The results showed that ion implantation caused the nuclei structure to be damaged, leading to the nuclei degradation. The results of statistical analysis showed that the damage grade of nuclei was very correlative with the ion dosage. The damage degree of DNA in the nuclei at the level of single cells was increased with the increase of the ion implantation dose.  相似文献

The irradiation effects of 0.28-2.80 MeV protons on GalnP/GaAs/Ge solar cells have been analysed, and then correlated with the displacement damage dose. The results of I-V and spectral response measurements, combined with the SRIM-derived vacancies produced rates, show that the degradation of the solar cells is largely determined by the displacement damage of the GaAs sub-cell. Thus the SRIM-derived NIEL values for protons in the GaAs sub-cell are used to calculate the displacement damage dose. It is shown that the irradiation effects of the solar cells caused by protons at different energies are correlated well with the aid of displacement damage dose.  相似文献

Taking the interaction between a DNA damage repair module, an ATM module, and a P53--MDM2 oscillation module into account, this paper presents a mathematical model of a P53 oscillation network triggered by a DNA damage signal in individual cells. The effects of the DNA damage signal and the delay time of P53-induced MDM2 expression on the behaviours of the P53 oscillation network are studied. In the oscillatory state of the P53--MDM2 oscillator, it is found that the pulse number of P53--P oscillation increases with the increase of the initial DNA damage signal, whereas the amplitude and the period of P53--P oscillation are fixed for different initial DNA damage signals, and the period numbers of P53--P oscillations decrease with the increase of time delay of MDM2 expression induced by P53. These theoretical predictions are consistent with previous experimental results. The combined negative feedback of P53--MDM2 with the time delay of P53-induced MDM2 expression causes oscillation behaviour in the P53 network.  相似文献

探讨了正常皮肤对重离子辐照急性损伤反应的耐受性, 为重离子治癌临床应用提供安全性检测的实验依据。实验前10 min, 实验猪肌肉注射复方氯胺酮1.2 mg/kg进行麻醉, 然后在兰州重离子研究装置(HIRFL)辐照终端利用12C6+束照射, 辐照剂量分别为0, 12, 21和27 Gy, 辐照分3次完成, 剂量率约为1.2 Gy/min, Bragg峰区照射, 辐照后每隔7 d对照射野拍照并活检取样, 做HE组织病理学观察。不同剂量12C6+离子束辐照实验猪皮肤后, 皮肤外观反应随辐照剂量增大而加快, 表现为肿胀和色素沉积等; 皮肤组织结构的变化明显, 上皮细胞排列紊乱、 萎缩、 空泡变性; 基本恢复正常所需时间也越长, 且都存在明显的剂量效应关系。结果表明, 辐照剂量范围为0—27 Gy时, 重离子对正常皮肤的辐照是安全的。The tolerance of the normal skin to the acute radiation injury reaction induced by heavy ion beams has been studied experimentally. The experimental pigs were injected with 1.2 mg/kg ketamine in 10 min before irradiation and were irradiated with 0, 12 , 21 and 27 Gy 12C6+ ion at a dose rate of 1.2 Gy/min at the Heavy Ion Research Facility in Lanzhou(HIRFL). The total radiation dose was finished by 3 times at Bragg Peak Region of Heavy Ion Beams.The radiation fields of skin were taken photo and performed biopsy. The contaneous tissues of radiation fields were stained by HE and examined histopatholo gical changes every seven days after irradiation. The results indicated that the cutaneous appearance reaction became more faster with radiation dosage rising and presented with swollen, melanin forming and so on after irradiated by the carbon ions at different dosage. The Pathological examination showed noticeable changes in histological and structural of experimental pigs skin, such as atrophy, vacuolation, denaturation and arranged irregularly in epithelial cells. Furthermore, the time for return to normality became longer with the increasing of radiation dosage. All indexes demonstrated correlation between the does and effects. It is concluded that the irradiation of heavy ion beams to normal skin is security when the radiation dose range is about 0—27 Gy.  相似文献

国产ZIRLO合金是CAP1400重要的燃料包壳管候选材料之一,因此需要在使用前对该材料进行辐照性能考验。研究了国产ZIRLO合金的辐照性能随辐照温度和损伤剂量的变化。辐照实验在中国原子能科学研究院HI-13串列加速器上,采用80 MeV的Ni离子进行,辐照产生的损伤采用透射电子显微镜、能量色散谱和纳米压痕法等方法表征。国产ZIRLO合金在300℃经10 dpa损伤剂量辐照后观察到均匀分布的析出颗粒,同样剂量在700℃辐照析出颗粒数目减少,析出相的主要成分为Nb。辐照后出现硬化现象,辐照损伤剂量越大,硬化程度越高;随着辐照温度升高,辐照硬化呈减小趋势。The domestic ZIRLO is a promising candidate of cladding materials for CAP1400. It is necessary to test its radiation properties before its use. In this paper, the radiation properties of the domestic ZIRLO have been studied as functions of irradiation temperature and dose respectively. The experiment was performed at the HI-13 tandem accelerator of China Institute of Atomic Energy by using 80 MeV Ni ions at different temperature and different dose. The transmission electron microscopy, electron dispersive spectroscopy and nano-indentation were used to characterize the radiation damage. The experimental results show that the precipitates are uniformly distributed at 10 dpa and 300℃, while at 700℃ the number of precipitates is reduced. Among all the precipitates, Nbisthe dominant element. Hardening phenomenon was also observed after irradiation, the higher the radiation dose, the higher the degree of hardening, and it illustrates a decreasing tendency with the increasing of the irradiation temperature.  相似文献

The effects of 400 keV helium ion irradiation dose and temperature on the microstructure of the Ti_3SiC_2 ceramic were systematically investigated by grazing incidence x-ray diffraction, scanning electron microscopy, and transmission electron microscopy.The helium irradiation experiments were performed at both room temperature(RT) and 500℃ with a fluence up to 2.0 × 10~(17) He~+/cm~2 that resulted in a maximum damage of 9.6 displacements per atom.Our results demonstrate that He irradiations produce a large number of nanometer defects in Ti_3SiC_2 lattice and then cause the dissociation of Ti_3SiC_2 to TiC nano-grains with the increasing He fluence.Irradiation induced cell volume swelling of Ti_3SiC_2 at RT is slightly higher than that at 500℃, suggesting that Ti_3SiC_2 is more suitable for use in a high temperature environment.The temperature dependence of cell parameter evolution and the aggregation of He bubbles in Ti_3SiC_2 are different from those in Ti_3AlC_2.The formation of defects and He bubbles at the projected depth would induce the degradation of mechanical performance.  相似文献

The nickel-base alloy is one of the leading candidate materials for generation IV nuclear reactor pressure vessel.To evaluate its stability of helium damage and retention,helium ions with different energy of 80 keV and 180 keV were introduced by ion implantation to a certain dose(peak displacement damage 1-10 dpa).Then thermal desorption spectroscopy(TDS)of helium atoms was performed to discuss the helium desorption characteristic and trapping sites.The desorption peaks shift to a lower temperature with increasing dpa for both 80 keV and 180 keV irradiation,reflecting the reduced diffusion activation energy and faster diffusion within the alloy.The main release peak temperature of 180 keV helium injection is relatively higher than that of 80 keV at the same influence,which is because the irradiation damage of 180 keV,helium formation and entrapment occur deeper.The broadening of the spectra corresponds to different helium trapping sites(He-vacancies,grain boundary)and desorption mechanisms(different Hen Vm size).The helium retention amount of 80 keV is lower than that of 180 keV,and a saturation limit associated with the irradiation of 80 keV has been reached.The relatively low helium retention proves the better resistance to helium bubbles formation and helium brittleness.  相似文献
