An approach to determine the photomorphogenic effect of light (white or continuous far-red) on the development of rhythmic enzyme activity in Chenopodium rubrum L. is described. Previous results, obtained from mature seedlings grown in white light, demonstrated stable oscillations with periods ranging between 12 and 15 h for all of the enzymes tested. The present results, obtained during deetiolation, were complicated by the presence of a higher frequency component with a period of about 6 h. When the various oscillating components were defined, the analysis showed: (1) the enzymes of the Krebs cycle (malate and isocitrate dehydrogenase), the closely associated glutamate dehydrogen-ase, and the glycolytic pathway ((NAD) glyceraldehyde-3-phosphate dehydrogenase) had a dominant period in the range of 12–15 h, (2) those of the oxidative pentose phosphate pathway were either weakly circadian (glucose-6-phosphate dehydrogenase) or apparently arhythmic (6-phosphogluconate dehydrogenase), (3) the (NADP) glyceraldehyde-3-phosphate dehydrogenase from the Calvin cycle was circadian when kept in continuous darkness but becomes 15 h when placed in light, and (4) only the Calvin cycle enzyme is affected by light in the level of its activity and in its oscillatory behavior.  相似文献

Abstract— The biological effects of single and 4-time irradiation of primary human embryo fibroblasts with 4 J/cm2 polarized light emitted by a halogen light source were investigated. The functional state of the plasma membrane was examined by means of lectin-binding and polycationized ferritin-binding techniques. It was established that the Con A binding of the cells did not change, whereas the number of negatively charged binding sites increased to a significant degree in relation to the untreated (control) samples and cell cultures exposed to diffuse (non-polarized) light. The micromorphological examinations showed no ultrastructural deviations. The quantitative increase of negative surface charges may be regarded as an indication of the biological effect of polarized light exerted on the cell membrane. The modifying effect of polarized light on the survival of E. coli exposed to the ionizing radiation was manifested in decreased anoxic radiation response.  相似文献

Abstract— The photobinding of phenothiazine derivatives (chlorpromazine, fluphenazine, promazine and promethazine) was studied on four different types of biological membranes (microsomes, myelin and synaptosomes from rat brain as well as human erythrocytes). The photoreaction was performed by ultraviolet irradiation of the tritiated compounds in their long wavelength absorption band (313 nm) and bound photoproducts were analysed by autoradiography of the proteins separated by polyacrylamide gel electrophoresis. The specificity of binding is low, however, a 34000 dalton band is intensely labeled on synaptic membranes with chlorpromazine and fluphenazine. All the phenothiazines bind on erythrocyte membrane proteins and specially on band 4.2 and on a peptide located before actin on the electrophoresis gel. These results show the generality of the phenothiazine photobinding on membrane proteins. These photobinding properties can be used for the identification and localization of some of these proteins.  相似文献

Abstract— An aqueous effluent produced during the retorting of oil shale has been shown to induce a significant genotoxic response in cultured Chinese hamster (CHO) cells following activation by near ultraviolet light (UVA). In this report the light-activated responses induced by this complex mixture were compared between two DNA excision repair deficient mutants and their parental strain, CHO-AA8-4. The mutants, UV-5 and UV-135, were hypersensitive to both the cytotoxic and mutagenic effects of concurrent exposures to the retort process water and UVA. Repair proficiency appeared to render AA8-4 relatively insensitive to low doses of UVA treatment, whereas in the two mutants a linear dose-response in the induction of 6-thioguanine resistant (6TG®) mutations was observed even at the lower UVA doses examined. Filter DNA alkaline elution methods were utilized to demonstrate that both single-strand breaks and DNA-DNA interstrand crosslinks were induced in CHO DNA following process water and UVA treatment. Results were also obtained which indicated that the inability to repair DNA-DNA crosslinks contributed significantly to the hypersensitive response seen in the excision repair mutants following the photoactivation of this complex mixture.  相似文献

水溶性煤基酸分为从含有水溶性煤基酸的低价煤中抽提得到的水溶性煤基酸和从不含有水溶性煤基酸的低阶煤中氧化得到的水溶性煤基酸。考察了从武川风化煤和霍林河风化褐煤通过两步催化氧化法得到的水溶性煤基酸和从晋城风化煤中得到的水溶性煤基酸对高效、高毒性的杀虫剂--久效磷生物活性的影响。结果表明:(1)三种煤基酸都能增加久效磷的生物活性;(2)所用水溶性煤基酸的理想浓度为0.0025%;(3)根据毒力曲线,久效磷、武川煤基酸-久效磷和霍林河-久效磷的致死中浓度为9.94g/ha,5.99g/ha,6.62g/ha,相对毒力系数分别为:100,165.9,150.2;(4)大田实验结果证实了水溶性煤基酸对久效磷的增效作用。  相似文献

–The adverse effects of ultraviolet-B (UV-B) radiation on the eye have been suspected but only recently have they been quantified by epidemiologic studies in humans. These studies became possible as methods to qualify individual ocular UV-B exposure were developed. A consistent dose-dependent association between ocular UV-B exposure and two common types of cataract (cortical and posterior subcapsular) has been shown. Although suggested by experimental studies, at present, evidence does not exist to link UV-B exposure to senile macular degeneration. Ocular exposure to UV-B is also associated with several corneal changes, pterygium, climatic droplet keratopathy, and acute photokeratitis (snow blindness). Significant reduction in ocular UV-B exposure results from the use of eyeglasses and hats.  相似文献

Abstract— Near-ultraviolet light (365.5 nm) reduces the ability of Escherichia coli B/r and B8-1, to form colonies on nutrient agar after irradiation. This lethal effect is distinct from that obtained after far-u.v. irradiation (253.7 nm) because the far-u.v. sensitive and resistant strains are equally susceptible to near-u.v. Variation in susceptibility to ultraviolet light during growth is more marked for near-u.v. than for far-u.v. The number of survivors after near-u.v. irradiation of log phase cells is affected by several post-irradiation treatments; more cells survive if growth immediately after irradiation occurs at higher temperatures (unlike far-u.v.). Also, the presence of acriflavine and caffeine in the nutrient agar decreases the number of survivors (in common with far-u.v.).  相似文献

Abstract— Lysozyme undergoes conformational changes when exposed to visible light in the presence of methylene blue. Ultracentrifugation, gel filtration and end group analysis showed that no peptide bond was cleaved. About 30 per cent of the tryptophan residues were, however, modified. The enzymic activity decreased by 50 per cent. The sedimentation coefficient and helical content decreased; the extinction in the u.v. region between 185 to 210 nm increased. The photooxidized enzyme was more susceptible to trypsinolysis than the native enzyme.  相似文献

Abstract Merocyanine derivatives were prepared by structural alterations at the barbituric acid or chalcogenazole moieties. The photophysical properties of the dyes were markedly influenced by the presence of selenium rather than sulfur as a substituent at position 2 of the barbiturate. In methanol, quantum yields of both triplet state (φτ) and singlet oxygen sensitization (φΔ) were increased by over an order of magnitude, with a concomitant decrease in fluorescence, when selenium was present in the molecule. Photoisomerization, one of the dominant deactivation pathways in the sulfur- or oxygen-containing analogues, was completely absent in the selenium-containing derivatives. Efficient triplet state formation was observed for selenium-containing derivatives incorporated into L1210 cells by diffuse reflectance laser flash photolysis. Cytotoxicity studies, camed out using clonogenic assays on L1210 leukemia cells, showed a good correlation with φτ and φΔ, measured in solution. Experimental evidence provided by this paper supports a triplet state-, and probably singlet oxygen-, mediated phototoxic mechanism. Photoisomerization or singlet state mechanisms can be discounted.  相似文献

Abstract— Effects of 8-methoxypsoralen plus near-UV light on the nematode Caenorhabditis elegans were studied as a novel example of photosensitized actions at the individual level. Either the eggs or the worms were illuminated with near-UV light in the presence of 8-methoxypsoralen. The treatment decreased hatchability of the eggs depending on light fluence and concentration of the sensitizer. Inhibition of growth and premature death were observed when the larvae in the second stage were illuminated in the presence of 8-methoxypsoralen. When young adults were treated before the beginning of egg-laying, they grew to lay eggs, but the total number of eggs deposited per hermaphrodite was decreased and the life span was shortened.  相似文献

Abstract-The acute effects of low-fluence ultraviolet light, primarily between wavelengths 280 and 320 nm (ultraviolet B) on T-lymphocyte subsets were assessed in virro and in vivo by quantitative cytofluorometric analysis. In virro, 90J/m2 of ultraviolet irradiation of human mononuclear cells produced a significant increase in the OKT4 (helper-inducer)/OKT8 (suppressor-cytotoxic) ratio (P>0.008), due primarily to a decrease in the OKT8 subset. In vivo, 24 h after whole-body UV irradiation of human subjects with one half of a minimal erythemal dose of ultraviolet B the OKT4/OKT8 ratios again increased (P > 0.002), again with somewhat greater effects on the OKT8 subset. These results indicate that suberythemal exposure to UV light increases OKT4/OKT8 ratios in normal human subjects because of differential effects on lymphocyte subsets. They suggest that casual exposure to sunlight mediates immunologic changes in normal human subjects.  相似文献

Abstract— The effect of human serum components on the photodynamic activity of zinc phthalocyanine (ZnPc) toward Chinese hamster fibroblasts (lineV–79) was studied. Photodynamic activities were correlated with cellular uptake of radiolabeled [65Zn]ZnPc, which allowed corrections to be made for the amount of sensitizer present in the cells at the time of irradiation and to express photodynamic efficiences on a cellular dye concentration basis. All serum components, with the exception of high-density lipoproteins, inhibit uptake of ZnPc byV–79 cells, when compared to incubation of ZnPc with the same cells in serum-free medium. High-density lipoproteins increased ZnPc uptake by 23%, but the photodynamic efficiency corrected for the cellular ZnPc concentration was unaffected. Very low-density lipoprotein and globulins decreased ZnPc cell uptake but likewise did not affect the cellular photodynamic efficiency of the dye. In contrast low-density lipoprotein and albumin, while inhibiting ZnPc cell uptake, increased the cellular photodynamic efficiency of ZnPc, suggesting that these proteins facilitate localization of the dye at cellular targers sensitive to photodynamic damage and vital to cell survival. We conclude from these results that association of ZnPc with serum components can have important, and widely differing, effects on both degree of uptake and cellular distribution of the photosensitizer.  相似文献

Abstract The hydrophobic photosensitizer acridine plus near-ultraviolet light damages both plasma membranes and outer membranes in Escherichia coli. Two lines of evidence are presented that outer membrane proteins are affected by acridine plus near-ultraviolet light treatment and that the effect is selective for certain proteins. First, analysis of outer membrane proteins on sodium dodecylsulfate polyacrylamide gels revealed that some protein bands are diminished upon treatment while others remain unaltered. New bands also appear, suggesting degradation or crosslinking reactions. Second, bacteriophage adsorption studies showed that treatment of E. coli F cells with acridine plus near-ultraviolet light causes a loss in functionality of the receptor for phage T5. Treatment of E. coli ABU57 cells under comparable conditions has no discernable effect on the functionality of the receptor for phage BF23.  相似文献
