Precise measurement of low enrichment of stable isotope labeled amino‐acid tracers in tissue samples is a prerequisite in measuring tissue protein synthesis rates. The challenge of this analysis is augmented when small sample size is a critical factor. Muscle samples from human participants following an 8 h intravenous infusion of L‐[ring‐13C6]phenylalanine and a bolus dose of L‐[ring‐13C6]phenylalanine in a mouse were utilized. Liquid chromatography tandem mass spectrometry (LC/MS/MS), gas chromatography (GC) MS/MS and GC/MS were compared to the GC‐combustion‐isotope ratio MS (GC/C/IRMS), to measure mixed muscle protein enrichment of [ring‐13C6]phenylalanine enrichment. The sample isotope enrichment ranged from 0.0091 to 0.1312 molar percent excess. As compared with GC/C/IRMS, LC/MS/MS, GC/MS/MS and GC/MS showed coefficients of determination of R2 = 0.9962 and R2 = 0.9942, and 0.9217 respectively. However, the precision of measurements (coefficients of variation) for intra‐assay are 13.0%, 1.7%, 6.3% and 13.5% and for inter‐assay are 9.2%, 3.2%, 10.2% and 25% for GC/C/IRMS, LC/MS/MS, GC/MS/MS and GC/MS, respectively. The muscle sample sizes required to obtain these results were 8 µg, 0.8 µg, 3 µg and 3 µg for GC/C/IRMS, LC/MS/MS, GC/MS/MS and GC/MS, respectively. We conclude that LC/MS/MS is optimally suited for precise measurements of L‐[ring‐13C6]phenylalanine tracer enrichment in low abundance and in small quantity samples. This paper reports on the instrumental and experimental prerequisites of the combination of high temperature gas chromatography (HTGC) with electron impact- (El) and chemical ionization (Cl) mass spectrometry (HTGC/EI/CIMS). It describes a newly developed high temperature GC/MS interface in detail and discusses the particular problems arising in HTGC/MS. The capabilities of the system are demonstrated by direct analyses of nickel- and vanadyl porphyrins, extracted from Julia Creek oil shale. The use of CI/NH3 in the GC/MS analysis of metalloporphyrins is demonstrated for the first time.  相似文献

Ion mobility spectrometry (IMS) is a well-known analytical method for the detection of CWAs and explosives since many years. Coupling IMS to GC pre-separation, new application fields in medicine and biology could be opened, dealing with complex and humid mixtures. However, identification of unknowns in such a complex sample is challenging and can only be achieved by parallel GC/MS analysis, thus obtaining a proposal for the responsible compound for validation via reference substances by GC/IMS again. The available adsorption tools for such accompanying GC/MS analysis have their particular drawbacks (e.g. problematic quantification for SPME, high sample volumes for adsorption tubes). Therefore miniaturised adsorption needles (NeedleTrap) were applied to both GC/IMS and GC/MS for validation of their reproducibility. It could be demonstrated that the needles can even be used for appropriate quantification when the adsorbent and the sample volume are adapted properly to the concentration range, the compounds of interest and humidity of the sample. The method is very flexible with regard to the concentration range by variation of the sample volume (e.g. 20 mL for pptV, 10 mL for lower ppbV or 1 mL for ppmV) and with regard to the compounds of interest by application of common adsorption materials optimised for the relevant substance group. Such materials are available commercially in a broad variability. Therefore, the miniaturised adsorption needles are a helpful complementary sampling method for any GC/MS or GC/IMS investigations.  相似文献

The aim of the present study was to set‐up a Fast gas chromatography/mass spectrometry method for the analysis of cholesterol oxidation products (COPs). A silylated mixture of seven oxysterol standards was injected into a Fast GC/MS system. A capillary GC column (10 m×0.1 mm internal diameter×0.1 μm film thickness) coated with 95% dimethyl‐ and 5% diphenyl‐polysiloxane, was used. The method gave a fast (total analysis time=3.5 min) and satisfactory resolution (R>1.2) of the COPs standards, with a good repeatability and sensitivity, similar to those of conventional GC/MS; recoveries were tested on mice liver. Fast GC/MS method suitability for COPs analysis in food was also tested on an oxidized sardine fillet, which had been previously saponified and purified by NH2 solid‐phase extraction (SPE); a good repeatability and sensitivity was also obtained. The analytical performance of the Fast GC/MS method for the determination of COPs, together with the consequent significant reduction of the analysis time and consumables, demonstrates that Fast GC/MS represents a valid alternative to conventional GC/MS and evinces the great potential of such an analytical technique, which could be applied for both food and biological samples.  相似文献

In the present study, different MS methods for the determination of human muscle protein fractional synthesis rate (FSR) using [ring‐13C6]phenylalanine as a tracer were evaluated. Because the turnover rate of human skeletal muscle is slow, only minute quantities of the stable isotopically labeled amino acid will be incorporated within the few hours of a typical laboratory experiment. GC combustion isotope ratio MS (GC‐C‐IRMS) has thus far been considered the ‘gold’ standard for the precise measurements of these low enrichment levels. However, advances in liquid chromatography‐tandem MS (LC‐MS/MS) and GC‐tandem MS (GC‐MS/MS) have made these techniques an option for human muscle FSR measurements. Human muscle biopsies were freeze dried, cleaned, and hydrolyzed, and the amino acids derivatized using either N‐acetyl‐n‐propyl, phenylisothiocyanate, or N‐methyl‐N‐(tert‐butyldimethylsilyl)trifluoroacetamide (MTBSTFA) for GC‐C‐IRMS, LC‐MS/MS, and GC‐MS/MS analysis, respectively. A second derivative, heptafluorobutyric acid (HFBA), was also used for GC‐MS/MS analysis as an alternative for MTBSTFA. The machine reproducibility or the coefficients of variation for delta tracer‐tracee‐ratio measurements (delta tracer‐tracee‐ratio values around 0.0002) were 2.6%, 4.1%, and 10.9% for GC‐C‐IRMS, LC‐MS/MS, and GC‐MS/MS (MTBSTFA), respectively. FSR determined with LC‐MS/MS compared well with GC‐C‐IRMS and so did the GC‐MS/MS when using the HFBA derivative (linear fit Y = 1.08 ± 0.10, X + 0.0049 ± 0.0061, r = 0.89 ± 0.01, P < 0.0001). In conclusion, (1) IRMS still offers the most precise measurement of human muscle FSR, (2) LC‐MS/MS comes quite close and is a good alternative when tissue quantities are too small for GC‐C‐IRMS, and (3) If GC‐MS/MS is to be used, then the HFBA derivative should be used instead of MTBSTFA, which gave unacceptably high variability. Fast atom bombardment combined with mass spectrometry (FAB/MS), high resolution FAB/MS, FAB tandem mass spectrometry (MS/MS), and gas chromatography/mass spectrometry (GC/MS) were used to determine the composition of the resinous material recovered from the wrappings of an Egyptian mummy believed to be from the Roman period (100–350 A.D.). FAB/MS and MS/MS studies identified several oxidation products of abietic acid as the principal resin components, indicating that one or more species of coniferous trees were used by the Egyptians as a source of the resin. GC/MS studies also identified several n-alkanes with carbon numbers from C19 to C33 in the sample. The relative amounts of these n-alkanes, along with characteristic trace metals, indicate that bitumen, an asphalt native to the region, was added to the resin. The presence of this ancient source of carbon in this sample explains the inconsistent date assigned to the mummy by carbon-14 analysis.  相似文献

Anionic living polymerization methods, using organometallic initiators such as butyllithium reagents, have proven useful for, inter alia, styrene polymerization and are amenable to subsequent functionalization of poly(styryl)lithium in the termination step. In this study, general methods for the selective preparation of small styrene oligomers and termination of the intermediate oligo(styryl)lithium anions were investigated. The crude reaction mixtures were analyzed directly by tandem gas chromatography/mass spectrometry (GC/MS). Of the carbon- and silicon-based electrophiles tested, chloro(chloroalkyl)silanes, Cl-SiR2(CH2)nCl in particular, were shown by GC/MS to be regioselective end-capping reagents, thus allowing subsequent transformation to the primary amine. The combined GC/MS data allow not only an estimate of the degree of functionalization, but also the identification of by-products, thus providing insight into the end-capping process that otherwise might be difficult to access. © 1995 John Wiley & Sons, Inc.  相似文献

Studies have shown that the administration of androstenedione (ADIONE) significantly increases the urinary ratio of testosterone glucuronide to epitestosterone glucuronide (T/E) – measured by gas chromatography/mass spectrometry (GC/MS) – in subjects with a normal (≈1) or naturally high (>1) initial values. However, the urinary T/E ratio has been shown not to increase in subjects with naturally low (<1) initial values. Such cases then rely on the detection of C6‐hydroxylated metabolites shown to be indicative of ADIONE administration. While these markers may be measured in the routine GC/MS steroid profile, their relatively low urinary excretion limits the use of gas chromatography/combustion/isotope ratio mass spectrometry (GC/C/IRMS) to specifically confirm ADIONE administration based on depleted 13C content. A mass spectrometry strategy was used in this study to identify metabolites of ADIONE with the potential to provide compound‐specific detection. C4‐hydroxylation was subsequently shown to be a major metabolic pathway following ADIONE administration, thereby resulting in urinary excretion of 4‐hydroxyandrostenedione (4OH‐ADIONE). The Japanese pepper (Zanthoxylum piperitum DC.) is an attractive plant that is highly palatable and benefits human health. There are several lineages of pepper plants in Japan. However, the classification of each lineage by analyzing its volatile compounds and studies on the effects of differences in volatile compounds on human flavor perception have not been performed in detail. Herein, we conducted gas chromatography (GC) and GC/mass spectrometry (GC/MS) analysis of volatile compounds and sensory evaluation of flavor by an analytical panel using 10 commercially available dry powdered Japanese pepper products from different regions. GC and GC/MS analysis detected limonene, β-phellandrene, citronellal, and geranyl acetate as the major volatile compounds of Japanese peppers. The composition of volatile compounds showed different characteristics depending on the growing regions, and cluster analysis of composition classified the products into five groups. The sensory evaluation classified the products into four groups, and the results of both classifications were in good agreement. Our results provide an important basis for proposing cooking and utilization methods that take advantage of the unique characteristics of each lineage based on scientific evidence.  相似文献

Gas chromatography (GC) is an analytical tool very useful to investigate the composition of gaseous mixtures. The different gases are separated by specific columns but, if hydrogen (H2) is present in the sample, its detection can be performed by a thermal conductivity detector or a helium ionization detector. Indeed, coupled to GC, no other detector can perform this detection except the expensive atomic emission detector. Based on the detection and analysis of H2 isotopes by low‐pressure chemical ionization mass spectrometry (MS), a new method for H2 detection by GC coupled to MS with an electron ionization ion source and a quadrupole analyser is presented. The presence of H2 in a gaseous mixture could easily be put in evidence by the monitoring of the molecular ion of the protonated carrier gas. Copyright © 2013 John Wiley & Sons, Ltd.  相似文献

Using silicon tetrachloride (SiCl4), tert-butanol (t-BuOH), ethanol (EtOH) and NH3, the hybridized silicon ethoxide 3-tert-butoxide (Si(OEt)x(OBut)4-x) was synthesized and the configuration of the material was investigated by FT-IR,1 H and 13C NMR and gas chromatogram/mass spectrum (GC/MS) techniques. The results confirm that both the ethoxy and the tert-butoxy groups have been attached to silicon atoms. Furthermore, the alkoxy group types and their relative amounts in the alkoxide were also determined by 1H and 13C NMR and GC/MS.  相似文献

NN-Bis(dimethylsilyl)tetramethylcyclodisilazane (NSCDSN) was selected for plasma polymerization as a model monomer representing N-silyl-substituted cyclodisilazanes. Owing to the aromatic character resulting from the strong (d-p) coupling between silicon and nitrogen atoms, these compounds manifest the highest thermal stability among the organosilicones. GC/MS examination of a low-molecular-weight fraction evaporated from the plasma polymer revealed the presence of various monomer derivatives having mostly the general structure of N-silyl-substituted cyclodisilazane (Si4N2) units. GC/MS and ATR/IR studies have shown that the Si4N2 skeleton displays a high resistance to plasma fragmentation and that it was incorporated as such into the polymer film. A radical mechanism for plasma polymerization was postulated assuming the formation of and propagation precursors. The low value found for the polar component of the surface free energy confirmed the contribution of Si4N2 units to the polymer film. TGA investigation showed the rate and degree of thermal decomposition of plasma-polymerized (PP) NSCDSN to be lower than those of plasma polymers from N-hydrogen-substituted silazanes. Vacuum pyrolysis, at 800°C, converted the polymer film into a glassy, dense, and almost inorganic material with a strong adhesion to the metal substrates.  相似文献

An analytical method for simultaneous in situ ethylation, of organolead, organotin and organomercury compounds in aqueous samples was developed using a new derivatisation agent, bromomagnesium tetraethylborate (BrMgEt4B). The determination of lead, tin and mercury compounds was done by species‐specific isotope dilution, derivatisation and GC–inductively coupled plasma MS (GC‐ICP‐MS) or by GC‐MS. The recovery and accuracy of the derivatisation were evaluated. The effect of pH and the relative quantity of derivatisation agent were studied.  相似文献

An analytical method for differentiation of primary, secondary, and tertiary amines using exhaustive trifluoroacetylation prior to GC/MS has been developed. Using the conditions described in the report, most primary amines add two and secondary amines add one trifluoroacetyl group. In general, tertiary amines do not react. GC retention indices and relative GC/MS total ion current response factors for 102 trifluoroacetyl derivatives are reported. Examples of the application of the method to petroleum and coal liquid products are provided. Because of the limited thermal stability of the derivatives of primary amines, the method is applicable only to distillates boiling below 370 °C (700 °F).  相似文献

A quality control procedure for Fused Silica Capillary Column Gas Chromatography/Mass Spectroscopy (FSCC GC/MS) was tested and shown to produce similar response factors at five Spectroscopy (FSCC GC/MS) was tested and shown to produce similar response factors at five laboratories. The average relative standard deviation (RSD) of interlaboratory response factors for fifty three compounds of environmental interest determined at each laboratory at nominal injected weights of 20, 100, and 200 ng was found to be 18.9% for four of the five laboratories which participated in this work. These data demonstrate that similar response factors can be attained in routine GC/MS measurements when adequate quality control is maintained.  相似文献

Summary A sensitive GC method was developed to analyse O2 (10–400ppm) in argon, by using Ar as carrier gas. A sampling method was developed for lamp analysis, and the O2 impurity content in the gas filling of incandescent lamps was measured. The analysis of low volatile compounds by (GC)2/MS using high-temperature stable glass-capillary columns and a commercially available GC/quadrupole MS instrument is described. It will be shown that no special modifications or procedures are required in analyzing organic compounds exceeding carbon numbers of 50. Operational characteristics have been determined for fused silica capillary column (FSCC) GC/MS as applied to “extractable” priority polutants. Chromatographic data show excellent relative retention time (RRT) intralaboratory precision and interlaboratory accuracy when multiple internal standards are empolyed. Potential chromatographic problems, such as column overload and “double peaking”, are addressed. Response factor relative standard deviations (RSD) at 50 ng for most of the extractable priority pollutants over the long term indicated precise determination (i.e. RSD generally ≤ 10%). Linearity was demonstrated over two orders of magnitude for FSCC GC/MS analysis of compounds with relatively low and high RF (response factor) values. Potential quantitative problems, such as saturation, are discussed. For certain aromatic priority pollutants interlaboratory RF agreement was observed. This was noted as perhaps the most important property of FSCC GC/MS analysis when the multiple internal standard approach is utilized. Determinations of extractable priority pollutants are directly compared for paced column GC/MS and FSCC GC/MS analysis of separate and composited extracts. For six extracts analyzed in triplicate, the latter configuration was shown to produce more consistent results. In view of the superior analysis logistics of composite extract FSCC GC/MS analysis, this approach was established as the preferred method for the analysis of priority pollutants classified as extractable.  相似文献
