New symmetrical 4-[2-chloro-2-(4-chloro-6-methyl-2-oxo-2H-pyran-3-yl)vinyl]-substituted 1,4-di-hydropyridines were synthesized in moderate to good yields via the modified Hantzsch reaction of β-dicarbonyl compounds with (Z)-3-chloro-3-(4-chloro-6-methyl-2-oxo-2H-pyran-3-yl)acrolein in the presence of an excess amount of NH4OAc. Also, the reaction of β-dicarbonyl compounds with (Z)-3-chloro-3-(4-chloro-6-methyl-2-oxo-2H-pyran-3-yl)acrolein in the presence of enamino esters and ketones was performed, and asymmetrical 4-[2-chloro-2-(4-chloro-6-methyl-2-oxo-2H-pyran-3-yl)vinyl]-substituted 1,4-dihydropyridines were obtained in moderate to good yields at room temperature. The calcium channel blocking activity of these compounds was assessed. They demonstrated moderate to weak effects, although one compound had a comparable effect (IC50 =1.40×10−7 M) with respect to the reference drug Nifedipine.  相似文献

6-Chloro-2(2H)-pyranone, which can be prepared in high yield from commercially available trans-glutaconic acid, undergoes facile Pd/Cu-catalyzed reaction with various 1-alkynes to give rise to the corresponding 6-(1-alkynyl)-2(2H)-pyranones in moderate to good yields. These last hitherto unknown compounds have been used as direct precursors to 6-alkyl- and 6-[(Z)-1-alkenyl]-2(2H)-pyranones.  相似文献

Some new azo sulfa drugs 3-cyano-4,6-diphenyl-1-[4-(N-substituted)sulfamyl]phenylazo-2-pyridinone dyes (1-6) and 3-cyano-4,6-diphenyl-1-[4-(N-substituted)sulfamyl]phenylazo-2-pyridinethione dyes (1′-6′) were synthesized by coupling of 4-(N-substituted)sulfamylbenzene diazonium salts with 3-cyano-4,6-diphenyl-2-pyridinone and/or with 3-cyano-4,6-diphenyl-2-pyridin-ethione. The corresponding iron (1a-6a, 1′a-6′a), copper (1b-6b, 1′b-6′b) and mercury (1c-6c, 1′c-6′c) chelates wvere also prepared. All synthesized compounds were screened in vitro for antibacterial and antifungal activities.  相似文献

Condensation of 4-hydroxy-3,3-dimethyl- and 4-hydroxy-3,3-pentamethylenebutan-2-ones with ethyl trifluoroacetate in the presence of LiH in hexane afforded 2-hydroxy-5,5-dimethyl-and 2-hydroxy-5,5-pentamethylene-2-trifluoromethyltetrahydro-4-pyranones, whose behavior in reactions withN-nucleophiles is analogous to that of unsymmetrical polyfluorinated β-diketones. 2,3-Alkadienoates as Dienophiles, Application in the Synthesis of (+)-(R)-Lasiodiplodin Methyl 2, 3-alkadienoates 2 are shown to react at 80° with l, 1-dimethoxy-3trimethylsilyloxy-l, 3-butadiene (1) to give the adducts 3 in good yields. Rearrangement of 3 , catalyzed by p-toluenesulfonic acid or by sodium methoxide, affords the 6-substituted methyl 4-hydroxy-2-methoxybenzoates 4 (R ? H, CH3, C6H5). An analogous reaction sequence starting with (-)-(11 R)-dodeca-2, 3-dien-11-olide ((-) -6 ) and 1 leads, via the adduct (R)-7 , to (+)-( R )-lasiodiplodin ((+) ?8 ) with properties identical to those of the natural product. The allene lactone (-) -6 was prepared by an intramolecular Wittig condensation of (R) ?5 , produced from (–)-(R)-9-hydroxydecanoic acid.  相似文献

The reactions of 1-substituted 2-nitro-3-phenylaminoprop-2-en-1-ones with cyanothioacetamide afforded the corresponding 6-substituted 3-cyano-5-nitropyridine-2(1H)-thiones, which were used for the synthesis of 6-substituted 3-cyano-2-methylthio-5-nitropyridines and 7-substituted 4-hydroxy-8-nitropyrido[2",3":4,5]thieno[2,3-b]pyridin-2(1H)-ones.  相似文献

We report a highly enantioselective Morita-Baylis-Hillman (MBH) reaction of isatins and acrolein to provide enantiomerically enriched 3-substituted 3-hydroxyoxindoles, which could serve as valuable synthetic building blocks. This is also the first time that a ketone has been used as the electrophile and acrolein as the nucleophile in a highly enantioselective catalytic asymmetric MBH reaction. Hatakeyama's catalyst, β-isocupreidine (1), turned out to be a powerful catalyst for this transformation.  相似文献

The lactone of 2, 6-dichloro-5-(hydroxymethyl)pyrimidine-4-carboxylic acid is synthesized, and various 2-chloro-6-amino and 2, 6-diamino derivatives are prepared from it by ammonolysis. It is shown that treatment of these compounds with alkaline reagents leads to lactone ring opening when there is a primary or secondary amino group at position 6, while the lactone ring is unaffected if there is a tertiary amino group at that position.A study is made of the action of dilute solutions of hydrochloric acid on the amino derivatives obtained by opening the lactone ring, and their reverse conversion to the corresponding 2, 6-substituted lactones of 5-(hydroxymethyl)pyrimidine-4-carboxylic acid is demonstrated.For Part III see [1].  相似文献

Lewis acid derived by mixing Tf2CH2 and Me3Al was found to be an effective catalyst system for the catalytic DA reaction of less reactive α,β-unsaturated lactone derivative with cyclopentadiene (CP). In this catalyst system, Tf2CHAlMe2 is an active species and an excess amount of Me3Al plays an important role to lower the catalyst loading. Substituent effect of the lactone framework on π-facial selectivity was also examined. In the reactions of both γ-substituted 5-membered lactone derivatives and γ- or δ-methylated 6-membered lactone derivatives with CP, selective attack on the anti face of γ- or δ-substituent was observed. On the other hand, in the cases of γ- or ?-methylated 7-membered lactone derivatives, CP favorably attacked on the syn face.  相似文献

The Synthesis of Some 2H-Pyran-2-one Derivatives Derivatives of 6-unsubstituted 2H-pyran-2-one have been synthesized by several different methods. The 4-chloro-2H-pyran-2-one ( 9 ) is the most important, since it serves as starting material for the synthesis of different 4-substituted 2H-pyran-2-ones (Scheme 2). Also described are simple transformations of cumalic-acid derivatives producing 5-(2,2-dichlorovinyl)-2H-pyran-2-one ( 23 ), 2-oxo-2H-pyran-2-carbonitrile ( 26 ), and 4,5-bis(trifluoromethyl)-2H-pyran-2-one ( 32 ) (Scheme 3 and 4).  相似文献

The methods of synthesis of 1,4-benzodioxin-2(3H)-one have been reexamined. Frequently quoted in the literature the method of Ghosh has been found to give impure (2-hydroxyphenoxy)acetic acid rather than the lactone. Using various methods some simple derivatives of the title lactone substituted in the benzene ring have been prepared and characterized. The bromination of the lactone with NBS gives predominantly 7-bromo-1,4-benzodioxin-2(3H)-one under electrophilic conditions, while 6-bromo-1,4-benzodioxin-2(3H)-one is obtained as a major isomer under photolytic conditions.  相似文献

Deprotonation then electrophilic quench of the lactone derived from the Diels-Alder addition adduct of anthracene and maleic anhydride gave α-substituted lactones in good yield. Of particular note was the reaction with chlorotrimethylsilane which gave only the C-silylated product. Flash vacuum pyrolysis (FVP) of the alkylated products afforded 3-substituted furan-2(5H)-ones in good overall yield.  相似文献

Reactions of 4-ethoxycarbonylamino-1H-1,5-benzodiazepine-3-carbonitrile (2) with aliphatic primary amines gave 1-substituted 4-(2-aminoanilino)pyrimidin-2(1H)-one-5-carbonitriles 3. Analogous reactions of 2 with aromatic primary amines afforded 2-(2′-anilino-1′-cyanovinyl)benzimidazoles 5 and 6. Upon treatment with triethylamine, 3 underwent intramolecular cyclization to give 3-substituted 5-aminopyrimidino[4,5-b]-[1,5]benzodiazepin-2(3H,11H)-ones 8 . Heating of 3 with p-toluenesulfonic acid in ethanol gave 2-substituted pyrimidino[1,6-a]benzimidazol-1(2H)one-4-carbonitriles 9 . Reactions of 2 with hydrazines were also described. Mechanistic pathways are proposed to account for the products.  相似文献

N-Substituted amides of 2-cyanopenta-2E,4-dienoic acid were synthesized by condensation ofN-substituted cyanoacetamides with acrolein in a dioxane-DMSO solution in the presence of Zn(OAc)2·2H2O as a condensing agent. X-ray diffraction study of 3-methylanilide of 2-cyanopenta-2E,4-dienoic acid (2d) demonstrated that the crystal structure of this compound is similar to that of 2-cyanopenta-2E,4-dienoic acid studied previously. However, the presence of themeta-tolyl substituent in molecule2d apparently results in the fact that the β-structure, which is typical of 2-cyanopenta-2E,4-dienoic acid, does not exist in the crystalline phase of2d. Synthesis of pyrrolidinyl-spirooxindoles fused to sugar lactone has been achieved by a one pot three component 1,3-dipolar cycloaddition (1,3-DC) reaction. A unique dipolarophile (α,β-unsaturated lactone) derived from d-glucose/d-galactose reacted with azomethine ylide generated in situ from isatin/N-substituted isatin and secondary amino acids (sarcosine/proline/piperidine-2-carboxylic acid) to give the corresponding cycloadducts in good yield. The cycloaddition was found to be highly regio- and diastereoselective.  相似文献

Three new eremophilane sesquiterpenes were isolated from the MeOH extract of the roots of Senecio nemorensis. Their structures were identified as 8α‐hydroxy‐6β‐(isobutanoyloxy)‐1‐oxoeremophila‐7(11),9‐dieno‐12,8β‐lactone ( 1 ), 6β,8β‐dimethoxy‐1‐oxoeremophila‐7(11),9‐dieno‐12,8α‐lactone ( 2 ), and 10α‐hydroxy‐6β‐(isobutanoyloxy)‐1‐oxoeremophila‐7(11),8‐dieno‐12,8‐lactone ( 3 ), respectively, based on spectroscopic data, including IR, EI‐MS, HR‐ESI‐MS, and 1D‐ and 2D‐NMR data.  相似文献

DFT calculations (B3LYP/6-31+G**) have been carried out on 106 tautomers and conformers of NH-azoles bearing CO2H and CO2CH3 groups. The following azoles systems have been studied: 2-substituted pyrroles, 2-substituted indoles, 2-substituted imidazoles, 2-substituted benzimidazoles, 4(5)-substituted imidazoles, 3(5)-substituted pyrazoles, 3-substituted indazoles (1H and 2H), 3,4(5)-substituted-1,2,3(5)-triazoles, 2,3(5)-substituted-1,2(3),4-triazoles, 4(5)-1,2,3,4(5)-tetrazoles. The cyclization reaction between N-substituted dithiocarbamates ( 1 ) and N-substituted N-chloromethylcarbamoyl chlorides ( 2 ) gives 3,5-disubstituted tetrahydro-2-thioxo-2H-1,3,5-thiadiazin-4-ones ( 3 ). In order to decide among the theoretically possible structures 3-5 , the compounds 6a,b , containing a thioxo group instead of an oxo group as in 3a,b , as well as the S-oxide derivative of 3a was also established by X-ray structure determination.  相似文献
