In the course of our studies on the development of anti-plaque agents for prevention of dental caries, we investigated effects of some of tea preparations and their individual components on the glucan synthesis catalyzed by glucosyltransferase (GTF) from Streptococcus mutans. Extracts of green tea and black tea, and polyphenol mixtures showed appreciable inhibition in the synthesis of insoluble glucan. Among the components isolated from tea infusions, theaflavin and its mono- and digallates had potent inhibitory activities at concentrations of 1-10 mM against GTF. (+)-Catechin, (-)-epicatechin and their enantiomers had moderate inhibitory activities at these concentrations, while galloyl esters of (-)-epicatechin, (-)-epigallocatechin and (-)-gallocatechin had increased inhibitory activities.  相似文献

To figure out the changed biological characteristics of a fluoride-resistant strain compared with those of the parent sensitive strain of Streptococcus mutans(S,mutans) with the help of differential proteomics analysis,sixtyseven proteins with obvious differential expression were identified in fluoride-resistant strain.Among them are the important virulence factors causing caries,including glucosyltransferase-S,glucosyltransferase-I,glucosyltransferase-Si,major cell-surface adhesin Pac,F-ATPase β-subunit,Fth,molecular chaperone DnaK,chaperonin GroEL,and the lactate dehydrogenase.We obtained the differential proteomics expression profiles of the fluoride-resistant strain(UA159-FR) and its parental strain(UA159) of S.mutans.Several important virulence factors bringing about caries were identified to have differential expressions,among which the expression of lactate dehydrogenase in fluoride-resistant strain was enhanced compared to that in its parent strain.The results could provide the basis for analvsing the functions of proteins encoded by caries-related genes from the fluoride-resistant S.mutans strain.  相似文献

Bacterial adhesion to protein-coated surfaces is mediated by an interplay of specific and nonspecific interactions. Although nonspecific interactions are ubiquitously present, little is known about the physicochemical mechanisms of specific interactions. The aim of this paper is to determine the influence of ionic strength on the adhesion of two streptococcal strains to fibronectin films. Streptococcus mutans LT11 and Streptococcus intermedius NCTC11324 both possess antigen I/II with the ability to bind fibronectin from solution, but S. intermedius binds approximately 20x less fibronectin than does the S. mutans strain under identical conditions. Both strains as well as fibronectin films are negatively charged in low ionic strength phosphate buffered saline (PBS, 10x diluted), but bacteria appear uncharged in high ionic strength PBS. Physicochemical modeling on the basis of overall cell surface properties (cell surface hydrophobicity and zeta potentials) demonstrates that both strains should favor adhesion to fibronectin films in a high ionic strength environment as compared to in a low ionic strength environment, where electrostatic repulsion between equally charged surfaces is dominant. Adhesion of S. intermedius to fibronectin films in a parallel plate flow chamber was completely in line with this modeling, while in addition atomic force microscopy (AFM) indicated stronger adhesion forces upon retraction between fibronectin-coated tips and the cell surfaces in high ionic strength PBS than in low ionic strength PBS. Thus, the dependence of the interaction on ionic strength is dominated by the overall negative charge on the interacting surfaces. Adhesion of S. mutans to fibronectin films, however, was completely at odds with theoretical modeling, and the strain adhered best in low ionic strength PBS. Moreover, AFM indicated weaker repulsive forces upon approach between fibronectin-coated tips and the cell surfaces in low ionic strength PBS than in high ionic strength PBS. This indicated that the dependence of the interaction on ionic strength is dominated by electrostatic attraction between oppositely charged, localized domains on the interacting surfaces, despite their overall negative charge. In summary, this study shows that physicochemical modeling of bacterial adhesion to protein-coated surfaces is only valid provided the number of specific interaction sites on the cell surfaces is low, such as on S. intermedius NCTC11324. Nonspecific interactions are dominated by specific interactions if the number of specific interaction sites is large, such as on S. mutans LT11. Its ionic strength dependence indicates that the specific interaction is electrostatic in nature and operative between oppositely charged domains on the interacting surfaces, despite the generally overall negatively charged character of the surfaces.  相似文献

Biofilm–material interactions are increasingly recognized as critical to success of some materials/devices and failure of others. We use a model system of dental monomers, salivary pellicles, and oral biofilms to demonstrate for the first time that degree of conversion of cross‐linked dimethacrylate polymers alters biofilm metabolic activity. This response is due primarily to leachable release (not surface chemistry) and is complex, with no changes in some biofilm measurements (i.e., biomass), and time‐ and leachable‐dependent responses in others (i.e., metabolic activity). These results highlight the need for considering biofilm‐material interactions when designing/evaluating new materials.

Isolation and characterization of the surface active components from the crude biosurfactant produced by Streptococcus thermophilus A was studied. A fraction rich in glycolipids was obtained by the fractionation of crude biosurfactant using hydrophobic interaction chromatography. Molecular (by Fourier transform infrared spectroscopy) and elemental compositions (by X-ray photoelectron spectroscopy) were determined. Critical micelle concentration achieved was 20g/l, allowing for a surface tension value of 36mJ/m(2). Moreover, this glycolipid rich fraction was found to be an anti-adhesive and antimicrobial agent against several bacterial and yeast strains isolated from explanted voice prostheses. Further purification steps should be carefully analyzed as each purification step will increase the costs and decreases the amounts of biosurfactants recovered.  相似文献

The rheology of cellulose microfibril suspensions from TEMPO-oxidized pulp was investigated. The suspension showed a pseudo-plastic and thixotropic behavior, slowly evolving with time under a given shear rate. The viscosity was proportional to the concentration up to the critical concentration of 0.23%. Above it, the viscosity followed a power law with exponents from 2 to 6 depending on the shear rate, and the system showed shear thinning behavior and behaved gel-like. Below this concentration, the system was more Newtonian. Birefringence measurement of 0.44% and 0.78% suspension showed that microfibrils alignment saturated at a small shear rate with a Herman’s orientation parameter below 0.65 probably due to the interconnection of microfibrils.  相似文献

New 5'-nucleotidase inhibitors named NF-86I, NF-86II were recently isolated from the seeds of Areca catechu L. NF-86I and NF86II showed inhibitory effects on the growth of Streptococcus mutans MT8148(c) and Streptococcus mutans MT6715(g), respectively. In addition, these inhibitors could inhibit insoluble glucan formation from sucrose. NF-86I and NF-86II were found to be polyphenolic substances. Some polyphenols such as tannic acid bind non-specifically to proteins (tannic activity). The 5'-nucleotidase inhibitors that we isolated did not show any such activity. However, the growth inhibitory activity and the inhibitory effect on water-insoluble glucan production were equal to tannic acid. It is therefore considered that these inhibitors bind specifically to the bacterial cell surface. Our findings suggest that the 5'-nucleotidase inhibitors NF-86I and NF-86II may be useful anti-plaque preventing agents.  相似文献

Streptococcus mutans (S. mutans) forms part of the commensal microflora and is deemed to be the major pathogen responsible for the generation of dental caries. The enzyme, sortase A enzyme, modulates the surface properties and cariogenicity of S. mutans. Curcumin has been reported to be an inhibitor of Staphylococcus aureus sortase A. In this study, inhibition of a purified S. mutans UA159 sortase A by curcumin was evaluated. Curcumin exerted strong inhibitory activity with a half maximal inhibitory concentration (IC50) of 10.2?±?0.7 μM which was lower than the minimum inhibitory concentration of 175 μM and the minimum bactericidal concentration of 350 μM. These results indicated that curcumin is a S. mutans UA159 sortase A inhibitor and therefore represents as a promising anticaries agent.  相似文献

Autoionization of Rydberg states of HfF, prepared using the optical-optical double resonance technique, holds promise to create HfF(+) in a particular Zeeman level of a rovibronic state for an electron electric dipole moment search. We characterize a vibronic band of Rydberg HfF at 54 cm(-1) above the lowest ionization threshold and directly probe the state of the ions formed from this vibronic band by performing laser-induced fluorescence on the ions. The Rydberg HfF molecules show a propensity to decay into only a few ion rotational states of a given parity and are found to preserve their orientation qualitatively upon autoionization. We show empirically that we can create 30% of the total ion yield in a particular ∣J(+), M(+) state and present a simplified model describing autoionization from a given Rydberg state that assumes no angular dynamics.  相似文献

Antibiotic resistance has prompted the search for new agents that can inhibit bacterial growth. We recently reported on the antibiofilm activities of nanosized ZnO and CuO nanoparticles (NPs) synthesized by using sonochemical irradiation. In this study, we examined the antibacterial activity of ZnO and CuO NPs in a powder form and also examined the antibiofilm behavior of teeth surfaces that were coated with ZnO and CuO NPs using sonochemistry. Free ZnO and CuO NPs inhibited biofilm formation of Streptococcus mutans . Furthermore, by using the sonochemical procedure, we were able to coat teeth surfaces that inhibited bacterial colonization.  相似文献

Membrane proteins are rarely identified in two-dimensional electrophoretic (2-DE) proteomics maps. This is due to low abundancy, poor solubility, and inherent hydrophobicity leading to self-aggregation during the first dimension. In this study, membrane proteins from the Gram-positive bacterium Streptococcus mutans were solubilized using three different methods and evaluated by 2-DE. In the first method, the extraction was performed using sodium dodecyl sulfate (SDS) followed by solubilization with a chaotropic buffer and precipitation with methanol/chloroform. The second method was based on temperature-dependent phase partitioning using Triton X-114 followed by purification using the ReadyPrep 2-D clean-up kit from Bio-Rad. The third method involved extraction using the organic solvents trifluoroethanol (TFE) and chloroform, which produced three separate phases. The upper aqueous phase, enriched with TFE, gave the highest overall protein yield and best 2-DE resolution. Protein spot identification by nanoelectrospray quadrupole time of flight (QTOF)-tandem mass spectrometry (MS/MS) revealed known membrane and surface-associated proteins. This is the first report describing the successful solubilization and 2-D electrophoresis of membrane proteins from a Gram-positive bacterium.  相似文献

The utilization of carrier-free ionic18F is a convenient method for studying the effects of very low extracellular fluoride concentrations on the metabolism of the cariogenic bacterium Streptococcus mutans. We have developed a method for the preparation of carrier-free aqueous solutions of ionic18F with high specific activity (about 707 Bq/ml). Fluoride distribution and fluoride binding experiments showed that submicromolar fluoride concentrations have already a negative effect on the metabolism of Streptococcus mutans.  相似文献
