A new pyrrolidinone, nigrospine (1), a new indole alkaloid, nigrospin A (2) and two new citrinins, nigrospins B and C (3-4) were isolated from a culture broth of the marine-derived fungal strain Nigrospora oryzae SCSGAF 0111. Their structures were determined by spectroscopic analysis, and the absolute configurations of 1 and 2 were determined by the Mosher ester technique. Compound 1 contains a rare 2,3-dihydro-benzofuran[2,3-c]2-pyrrolidone skeleton.  相似文献

Four new C22N2Lycopodium alkaloids, senepodines B-E (2-5), consisting of an octahydroquinoline ring and a quinolizidine ring have been isolated together with senepodine A (1) from the club moss Lycopodium chinense. The relative and absolute stereochemistry of 2-5 were determined by combination of NOESY correlations and chemical transformation, while the absolute configuration of 1 was assigned by exciton chirality method.  相似文献

Paecilomycine A (1), B (2), and C (3) were isolated from cultivated fruiting bodies of Paecilomyces tenuipes (Isaria japonica), which is a popular entomopathogenic fungus used in folk medicine and health foods in China, Korea, and Japan. The structures of 1-3 were deduced from their spectroscopic data and their absolute configurations were elucidated by preparing their MPA esters. Compound 1 showed activity in the neurotrophic factor biosynthesis in glial cells.  相似文献

Five new oxindole alkaloids, gelegamines A-E (1-5), were isolated from the roots of Gelsemium elegans. Their structures were extensively elucidated on the basis of spectroscopic analysis. Among them, the epoxy ring (C-19/C-20) of gelegamine A (1) was assigned as α-orientation by ROESY experiment and DFT method at B3LYP/6-31g(d) level, and gelegamine B (2) is the first humantenine-type alkaloid with 19-(E) ethylidene configuration. The absolute configurations of gelegamines A-E (1-5) were established on biosynthetic consideration coupled with CD experiments.  相似文献

Six new naturally occurring diacetylenic spiroacetal enol ethers, Lactiflodiynes A-F (1-6), together with five known congeners (7-11), were isolated from the whole plant of Artemisia lactiflora (Compositae). The structures were elucidated by extensive spectroscopic methods, X-ray crystallography, chemical transformations, and CD. The absolute configuration of Lactiflodiyne A (1) was determined to be 2R, 5S, 6S, and 7R by an X-ray crystallographic diffraction experiment using Mo Kα radiation with the absolute parameter of 0.01(8). In combination with CD, the absolute configurations of compounds 2-11 were confirmed by chemical transformations using 1 as the starting material.  相似文献

Nine new steroids, sclerosteroids A-I (1, 5, 6, 8-13), along with 18 known metabolites (2-4, 7, 14-27), were isolated from the soft coral Scleronephthya gracillimum. These structures were elucidated on the basis of detailed spectroscopic analysis. The absolute configurations of sugar moieties in steroidal glycosides 10-13 were determined by HPLC analysis of the o-tolylthiocarbamate derivatives of the liberated sugars from hydrolysis of these steroidal giycosides. Cytotoxic and anti-inflammatory activities of these compounds were measured in vitro.  相似文献

Three new norcembrane-based diterpenoids, leptocladolides A (1), B (4) and C (5), along with five known metabolites 6-10, have been isolated from the dichloromethane extract of a Taiwanese soft coral Sinularia leptoclados. Furthermore, a chemical investigation on the dichloromethane extract of S. parva has resulted in the isolation of two new related isomers, 1-epi-leptocladolide A (2) and 7E-leptocladolide A (3), in addition to 1 and 7. The structures of new metabolites 1-5 were elucidated on the basis of extensive spectroscopic analyses and their relative stereochemistries were determined by NOESY experiments. The new metabolites 1 and 3 have been shown to exhibit significant cytotoxic activity against KB and Hepa59T/VGH cancer cell lines.  相似文献

Two novel dimeric pyrrole-aminoimidazole (PAI) alkaloids (−) donnazoles A-B (1-2) were isolated from the marine sponge Axinella donnani collected off the Mauritius Island. Their structures were elucidated mainly by 2D NMR. Both compounds bear the crucial substituted cyclopentane of ‘pre-axinellamine’, the hypothetical common intermediate of all dimeric PAIs. Additionally, the essential trans-6,7 ring junction of palau′amine’s congeners is already established in 1 and 2. The absolute configurations of 1 and 2 were deduced via NOE correlations in combination with the comparison of their circular dichroism data with the one of (−) sceptrin. The absolute configuration of (−) donnazoles A (1) and B (2) is coherent with the absolute configuration of the known dimeric members of PAI alkaloids.  相似文献

Three new furanocoumarins, named feroniellins A (1), B (2), and C (3), were isolated from the roots of Feroniella lucida. Compounds 1-3 are novel structures having an oxolane, oxane, and oxepane moiety. Their overall structures and configurations were determined by spectral and chemical methods. A new cembradiene diterpenoid 1, together with the known antiplasmodial cembradiene 2, was isolated from the sea whip Eunicea sp. collected from Santa Marta Bay on the Colombian Caribbean sea. The structures and absolute stereochemistry of 1-2 were determined by extensive spectroscopic analysis, chemical transformation of 2 to the new diterpenoid 3, and by using modified Mosher’s method. Tabularisins A-D (1-4), a new class of phragmalins incorporating a cyclopropanyl ring, were isolated from Chukrasia tabularis. Compounds 1 and 2 are also the first examples of phragmalins with an 8,9,11-ortho ester. The structures of 1 and 3 were confirmed by single crystal X-ray studies. Two novel Daphniphyllum alkaloids with unprecedented skeletons, namely paxdaphnines A (1) and B (2), have been isolated from the seeds of Daphniphyllum paxianum. Paxdaphnine A (1) is a 19-nor-Daphniphyllum alkaloid with highly caged skeleton, and paxdaphnine B (2) is the first 1,19-bisnor-Daphniphyllum alkaloid. The relative structures of 1 and 2 were elucidated by spectral methods, and their unique biosynthetic pathway postulated. The absolute structure of 1 was determined by X-ray diffraction of the iodide derivative (1a) of 1, and the absolute stereochemistry of 2 was proposed by correlation with the biosynthetic pathway for 1.  相似文献

Chemical investigation of the dichloromethane extract of the Red Sea marine sponge Lamellodysidea herbacea led to the isolation of seven new polychlorinated derivatives 3-6 and 8-10 in addition to the known dysidamide 1, dysidamide B 2 and dysidamide C 7. Their structures were established by extensive NMR spectroscopic data. The absolute configuration of compound 9 was determined by X-ray crystallographic diffraction analysis. Dysidamide 1 exhibited neurotoxic effects towards both mesencephalic and cortical murine neurones at 0.8 μg/ml.  相似文献

Four new Stemona alkaloids, sessilifoliamides A-D (1-4), were isolated from the roots of Stemona sessilifolia, along with five known alkaloids, stenine (5), 2-oxostenine (6), stemoninoamide (7), tuberostemonone (8), and neotuberostemonol (9). The structures and absolute configurations of the new alkaloids were determined by the spectral studies (HRMS, IR, 1H, 13C, and 2D NMR), single-crystal X-ray analyses, and chemical correlations. Three new microviridins namely, SD1684 (1), SD1634 (2), and SD1652 (3), were isolated from the hydrophilic extract of Microcystis aeruginosa. The planar structures of compounds 1-3 were determined by homonuclear and inverse-heteronuclear 2D-NMR techniques as well as by high-resolution mass spectrometry. The absolute configuration of the asymmetric centers was studied using Marfey's method for HPLC. Compounds 1-3 contain l-threo-β-hydroxy aspartic acid as a building block of the peptide chain. This is the first example where microviridins contain non-proteinogenic amino acid in their structure. Compound 2 is a mild serine protease inhibitor.  相似文献

Three new unstable metabolites, (6E,10Z)-2′-O-methylmyxalamide D (1), 2′-O-methylmyxalamide D (2) and (6E)-2′-O-methylmyxalamide D (3) were isolated from the myxobacterium Cystobacter fuscus. The planar structures were elucidated by spectroscopic analyses to be geometrical isomers of a polyene amide related to a myxobacterial metabolite, myxalamide D (4). Their absolute stereochemistry was determined by synthesis of degradation products. Five new unsaturated 2-amino-3-alcohols, crucigasterins A-E (2-6), were isolated together with known related compound 7 from the Mediterranean ascidian Pseudodistoma crucigaster and characterised as diacetyl derivatives (2a-6a) by spectroscopic methods. The threo-relative configuration of the amino alcohol portion was inferred by NOE analysis of the oxazolidinone derivative of crucigasterin A (2) as well as by 13C NMR comparison with synthetic threo and erythro model compounds. The co-occurring metabolites were assumed to have the same relative configuration as 2 by comparison of the diagnostic carbon value of C-1. The absolute stereochemistry of compound 7 that had not been previously reported was determined by applying the modified Mosher’s method on the corresponding N-acetyl derivative. The same absolute configuration was suggested for the other co-occurring crucigasterins by biogenetic considerations. Antibacterial and antifungal activities of selected crucigasterins were also evaluated.  相似文献
