建立固相萃取-离子交换色谱法测定浓缩苹果汁中苯菌灵、多菌灵和噻菌灵的残留量。样品直接用水稀释后,于80 ℃下将苯菌灵完全转化为多菌灵,再经SCX固相萃取柱富集,采用LC-SCX离子交换色谱柱(25 cm×4.6 mm,5 μm)分离,二极管阵列检测器检测,以0.1 mol/L KH2PO4溶液(pH 2.5)-乙腈(体积比为70 ∶30)为流动相,在1.0 mL/min下等度洗脱,于282 nm波长下检测。在0.02~2.0 mg/L范围内,多菌灵和噻菌灵的峰面积与其浓度呈良好的线性关系,最低检出限均可达到0.004 mg/kg,回收率为94.2%~100.4%,相对标准偏差低于4.2%。该方法简便、快速、灵敏、准确,可用于浓缩苹果汁中苯菌灵、多菌灵和噻菌灵残留量的检测。  相似文献

The presented paper deals with a new methodology for direct determination of propranolol in human plasma. The methodology described is based on sequential injection analysis technique (SIA) coupled with solid phase extraction (SPE) column based on restricted access materials (RAM). Special RAM column containing 30 μm polymeric material—N-vinylacetamide copolymer was integrated into the sequential injection manifold. SIA–RAM system was used for selective retention of propranolol, while the plasma matrix components were eluted with two weak organic solutions to waste.

Due to the acid–basic and polarity properties of propranolol molecule and principles of reversed-phase chromatography, it was possible to retain propranolol on the N-vinylacetamide copolymer sorbent (Shodex MSpak PK-2A 30 μm (2 mm × 10 mm)). Centrifuged plasma samples were aspirated into the system and loaded onto the column using acetonitrile–water (5:95, v/v), pH 11.00, adjusted by triethylamine. The analyte was retained on the column while proteins contained in the sample were removed to waste. Interfering endogenous substances complicating detection were washed out by acetonitrile–water (15:85), pH 11.00 in the next step. The extracted analyte was eluted by means of tetrahydrofuran–water (25:75), pH 11.00 to the fluorescence detector (emission filter 385 nm). The whole procedure comprising sample pre-treatment, analyte detection and column reconditioning took about 15 min. The recoveries of propranolol from undiluted plasma were in the range 96.2–97.8% for three concentration levels of analyte. The proposed SIA–RAM method has been applied for direct determination of propranolol in human plasma.  相似文献

A simple high-performance liquid chromatographic (HPLC) method was developed and validated for the quantification of mizoribine in human serum. After the addition of 70% perchloric acid and 3-methylxanthine (50 microg/mL, internal standard) to human serum, the samples were mixed and centrifuged at 12,000 rpm (1432 g) for 10 min. The supernatant was injected onto a C(18) column eluted with a mobile phase of 20 mm Na2HPO4 and methanol (93:7, v/v, pH 3) containing 0.04% octanesulfonic acid and detected utilizing an ultraviolet detector at 275 nm. The linear calibration curve was obtained in the concentration range of 0.1-4.0 microg/mL and the lower limit of quantification was 0.1 microg/mL. This method was validated with selectivity, linearity, precision and accuracy. In addition, the method was successfully applied to estimate the pharmacokinetic parameters of mizoribine in Korean subjects following an oral administration of 100 mg mizoribine (two Bredinine 50 mg tablets). The maximum serum concentration (C(max)) of 2.30 +/- 0.83 microg/mL was reached 2.27 +/- 0.66 h after an oral dose. The mean AUC(0-12 h) and the elimination half-life (t(1/2)) were 13.2 +/- 4.79 microg h/mL and 3.10 +/- 0.74 h, respectively.  相似文献

The potential of online trace enrichment on a highly apolar short column in LC was evaluated for the determination of pyrethroids in river water. Twelve millilitres of water samples, modified with 8 mL ACN (ACN/water 40:60, v/v), were passed through 50 x 4.6 mm ID first separation column packed with 5 microm Hypersil Elite C18. Pesticides were preconcentrated in this column while the matrix background was eluted to waste. Separation of pesticides was performed on a 3.5 microm symmetric C18 column (250 x 4.6 mm ID) with an ACN step gradient as mobile phase and fluorescence detection was used after postcolumn derivatization by using UV light. The use of photochemically induced fluorescence for detection improved sensitivity and selectivity. Quantification limits ranged from 0.05 to 0.1 microg/L and pesticide recoveries at two concentration levels (0.1 and 0.5 microg/L) were between 93.1 and 118.6%, with RSD between 2.5 and 7.5% (n = 3) in river water samples. No matrix effect was detected.  相似文献

This article explores the use of capillary electrochromatography for the analysis of chamomile (Chamomilla recutita L.) extracts. After a thorough study of analytical parameters such as mobile and stationary phase composition, applied voltage, and temperature, a methodology to determine 11 bioactive phenolic compounds (coumarins: herniarin, umbelliferone; phenylpropanoids: chlorogenic acid, caffeic acid; flavones: apigenin, apigenin-7-O-glucoside, luteolin, luteolin-7-O-glucoside; flavonols: quercetin, rutin and flavanone: naringenin) in chamomile extracts was proposed. The method was performed in a Hypersil SCX/C18 column with pH 2.8 phosphate buffer at 50 mmol L(-1) containing 50% acetonitrile (pH adjusted before the addition of the organic solvent). All compounds were separated in less than 7.5 min under isocratic conditions. Figures of merit include linearity (peak area versus apigenin concentration) from 50.0-1000 microg/mL (r2=0.995), and intra-day precision of retention time and peak area better than 1.3% CV and 15%, respectively. The limits of detection and quantification for apigenin were 35.0 microg/mL and 150.0 microg/mL, respectively. This article also describes an NMR 1H study, carried out to monitor a new clean-up procedure for extracts containing propyleneglycol, whose components are poorly retained in conventional octadecyl silica cartridges.  相似文献

建立了气相色谱-脉冲火焰光度法(GC-PFPD)测定水产品中二硫氰基甲烷(MBT)残留的分析方法。用二氯甲烷-正己烷溶液(1∶1,v/v)在超声条件下提取水产品中的MBT,然后用中性氧化铝固相萃取小柱对提取物进行净化和富集,通过HP-5MS石英毛细管柱(30 m×0.32 mm×0.25μm)分离,最后采用配有脉冲火焰光度检测器的气相色谱仪进行测定,外标法定量。MBT在1.0~20.0 mg/L范围内线性关系良好,相关系数为0.997 1;检出限为0.1 mg/kg;加标回收率为65.6%~97.6%,精密度为6.32%~12.8%(n=7)。该法能很好地满足水产品中药物残留检测的需求。  相似文献

建立了测定土壤中杀鼠灵、杀鼠醚、溴敌隆、氟鼠灵、溴鼠隆5种香豆素类灭鼠药残留量的柱后衍生荧光检测高效液相色谱方法。样品在加入内标物氯杀鼠灵后用丙酮-氨水-甲醇(体积比为100:3:100)混合液提取,浓缩的提取液用5 mL正己烷-氯仿(体积比为3:1)混合液溶解,NH2固相萃取小柱净化,用15 mL 50 mmol/L四丁基磷酸二氢铵甲醇溶液洗脱分析物,移除溶剂,用甲醇-0.25%(体积分数)乙酸水溶液(体积比为3:2)混合液溶解,过滤后,经高效液相色谱分离,以甲醇-氨水-水(体积比为1:1:8)混合液为衍生试剂进行柱后衍生,采用荧光检测器检测。杀鼠灵、杀鼠醚、溴敌隆、氟鼠灵、溴鼠隆5种鼠药在0.02~10.00 mg/L范围内线性关系良好,相关系数均大于0.999,定量限(LOQ,S/N=10)为2.2~18.5 μg/L。在0.1~0.3 mg/kg添加水平内,5种灭鼠药的回收率为94.6%~118.0%,相对标准偏差(RSD)为0.8%~10.2% (n=3)。实验结果表明该方法灵敏、准确,重复性好。  相似文献

A precise method is presented for determination of residual spiramycin (SP) in chicken eggs and tissues by high-performance liquid chromatography (HPLC). The sample preparation was performed by homogenizing with a mixture of acetonitrile and n-hexane (5:4, v/v) to minimize the fat amount followed by ultra-filtration using a MolCutII(R). The extracts containing SP were free from interfering compounds when examined by the normal-phase HPLC using a LiChrosorb(R) NH(2) column and a mobile phase of acetonitrile-water (85:15, v/v) with a photo-diode array detector. The average recoveries from spiked SP (0.1, 0.5 and 1.0 ppm) were in excess of 89.0% with coefficients of variation between 1.4 and 2.4%. The limit of detection was 0.1 ppm.  相似文献

建立了同时测定茶叶中噻嗪酮、甲胺磷、乙酰甲胺磷和三唑磷4种农药残留量的测定方法。在45 ℃加温条件下,用乙酸乙酯-正己烷混合溶剂提取及活性炭色谱柱净化,用不同配比的乙酸乙酯-正己烷混合液梯度洗脱待测组分,以DB-210毛细管色谱柱分离、氮磷检测器测定。结果表明,上述4种农药在10 min内能很好地分离;样品加标回收率(n=3)为73.4%~96.9%。方法的变异系数为2.49%~3.35%,茶叶(干重)中4种农药的定量检测下限为7.0~12.0 μg/kg。  相似文献

A column high-performance liquid chromatography (HPLC) method was developed for the determination of glucosamine in dosage forms. Glucosamine was derivatized by addition of a solution containing orthophthaldialdehyde. The HPLC separation was achieved on a Spherimage 80 ODS2 column (250 x 4 mm id, 5 microm particle size) using an isocratic mobile phase containing phosphate buffer-methanol (90 + 10, v/v, pH 6.50) and methanol-tetrahydrofuran (97 + 3, v/v) in proportions of 85 + 15 at a flow rate of 1 mL/min, followed by fluorescence detection. The method was validated for specificity, linearity, accuracy, precision, limit of detection (LOD), and limit of quantitation (LOQ). The detector response for glucosamine HCI was linear over the concentration range of 0.1-20 microg/mL with a correlation coefficient of 0.9980. The accuracy was between 99.4 and 100.8%. The LOD and the LOQ were 0.009 and 0.027 microg/mL, respectively. The method was applied to determination of glucosamine in solid dosage forms.  相似文献

建立了固相萃取-超高效液相色谱-电喷雾串联三重四极杆质谱(SPE-UPLC-ESI-MS/MS)联用技术分析水中9种微囊藻毒素的方法。样品经SPE提取和净化后,以Waters ACQUITY UPLCTM BEH C18色谱柱为分离柱,以含0.1%甲酸乙腈和含0.1%甲酸水作为流动相进行梯度洗脱,电喷雾离子源电离、正离子多反应监测模式质谱进行定性和定量分析。9种微囊藻毒素在0.1~50 μg/L或0.5~100 μg/L质量浓度范围内线性良好,相关系数为0.9990~0.9998,方法的检出限(以3倍信噪比计)为0.1~0.5 ng/L;高、中、低3个添加水平的回收率为75.8%~109%,相对标准偏差为0.49%~10.0%。结果表明,该方法灵敏、准确,检测范围广,分析速度快。应用该方法检测了杭州市两处水库水样中的微囊藻毒素,分别检出了3种和8种微囊藻毒素。  相似文献

A liquid chromatography electrospray mass spectrometry (LC/ESI/MS) method has been developed to identify and quantify microcystins in fish liver and intestine. Microcystins (MCs) were extracted from 500 mg sample with methanol-water (85:25, v/v) and the extracts concentrated to 250 microl. The parameters were optimized by a full factorial 2(3) design. Neither laborious pre-treatment nor clean up were necessary. MCs were separated using conventional C18 column and an acetonitrile-acidified water (pH 3) gradient. Negative samples (without MCs) were discriminated by liquid chromatography diode array detection (LC/DAD). The limits of detection (LOD) and the limits of quantification (LOQ) resulted equal for MC-RR, MC-YR, and MC-LR and were 0.1 and 0.5 microg g(-1), respectively. MCs recoveries at three levels in spiked samples (0.5-3.0 microg g(-1)) were > 92%, with relative standards deviations (RSDs) < 16% for liver samples and > 68% with RSDs < 18% for intestine samples. The proposed method was applied to determine MC-LR in exposed fish to evaluate the bioaccumulation risk. The results showed the transference of MC-LR from cyanobacterial cells to fish tissues.  相似文献

Salvianolic acid B is an herbal ingredient isolated from Salvia miltiorrhiza. An in vivo microdialysis sampling method coupled to high-performance liquid chromatography has been developed for continuous monitoring of protein-unbound salvianolic acid B in rat blood and bile. Microdialysis probes were inserted into the jugular vein/right atrium and bile duct of Sprague-Dawley rats, and a dose of 100 mg/kg salvianolic acid B was then administered via the femoral vein. Dialysates were collected and directly injected into a liquid chromatographic system. Salvianolic acid B was eluted using a microbore reversed-phase ODS 5 microm (150 mm x 1 mm I.D.) column. Isocratic elution of salvianolic acid B was achieved within 10 min using the liquid chromatographic system. The chromatographic mobile phase consisted of acetonitrile-methanol-20 mM monosodium phosphoric acid (pH 3.5) (10:30:60, v/v/v) containing 0.1 mM 1-octanesulfonic acid with 0.05 ml/min. The wavelength of the UV detector was set at 290 nm. Salvianolic acid B in both blood and bile dialysates was adequately determined using the liquid chromatographic conditions described, although the blank bile pattern was more complex. The retention times of salvianolic acid B in rat blood and bile dialysates were found to be 7.2 min. Peak-areas of salvianolic acid B were linear (r2 > 0.995) over a concentration range of 0.1-50 microg/ml. In vivo recoveries of microdialysis probes of salvianolic acid B in rat blood and bile averaged 22 +/- 2% and 41 +/- 1%, respectively. This study indicates that salvianolic acid B undergoes hepatobiliary excretion.  相似文献

High-performance thin-layer chromatography with diode array scanning (TLC-DAD) and high-performance column liquid chromatography with a diode array detector (HPLC-DAD) were used to screen water samples for pesticides. Atrazine, clofentezine, chlorfenvinphos, hexaflumuron, terbuthylazine, lenacyl, neburon, bitertanol, and metamitron were enriched from canal water samples by solid-phase extraction (SPE) on octadecyl silane (C18)/styrene-divinylbenzene-1, C18, C18 Polar Plus, and cyanopropyl (CN) cartridges. Recovery rates were high for all extraction materials except CN, for which values for all pesticides were lower. SPE was used for both preconcentration and fractionation of the analytes. Analytes were eluted by means of methanol and dichloromethane. Methanol eluates were analyzed by HPLC-DAD and dichloromethane eluates by TLC-DAD. The method was validated for precision, repeatability, and accuracy. Calibration graphs were linear between 0.1 and 50.0 microg/mL for all pesticides, and correlation coefficient (r) values were between 0.9994 and 1.000 as determined by HPLC-DAD. Calibration graphs were linear between 0.1 and 1.5 microg/spot for all pesticides, and r values were between 0.9899 and 0.9987 as determined by TLC-DAD. The limit of detection was between 0.04 and 0.23 microg/spot for TLC-DAD and 0.02 and 0.45 microg/mL for HPLC-DAD.  相似文献

A simple and sensitive high-performance liquid chromatographic method for the determination of Therminol 66 thermal heating fluid in glycerin and fatty acids is developed. Sample solutions dissolved in methanol-tetrahydrofuran (50:50, v/v) are injected directly into a reversed-phase C18 column and eluted with a methanol and water mixture (88:12, v/v). The concentration of the thermal heating fluid is monitored by fluorescence detection at 257 nm (excitation) and 320 nm (emission). The calibration graph obtained from various concentrations of the thermal heating fluid in the methanol and tetrahydrofuran mixture is linear (correlation coefficient = 0.999), and the limit of detection is 0.01 microg/mL. Spiked glycerin containing 0.1 to 1.0 microg/g of the thermal heating fluid also gives good linearity with a mean recovery of 95.3%. The mean intra- and interassay precision are 1.80-6.51% and 5.71-9.03%, respectively, at the 0.1-microg/g level. The method is simple and does not require any pretreatment step, thus it is ideal for quality assurance purposes.  相似文献

On-line coupling continuous-flow liquid membrane extraction (CFLME) with HPLC, a novel automatic system was developed for the determination of sulfonylurea herbicides in water. After an automatic trace-enrichment process by CFLME, which is the combination of continuous flow liquid-liquid extraction and support liquid membrane (SLM) extraction, the target analytes were concentrated in 50 microl of 0.2 M Na2CO3-NaHCO3 (pH 10.0) buffer. The concentrated sample solutions were injected directly onto a C18 analytical column with a valve, and detected at 240 nm with a diode array detector. Metsulfuron methyl (MSM), and DPX-A 7881 were baseline separated with a mobile phase consisting of methanol and 67 mM KH2PO4-Na2HPO4 (pH 5.91) buffer (45+55, v+v) at a flow-rate of 1.0 ml min(-1). With an enrichment time of 10 min and enrichment sample volume of 20 ml, the enrichment factors and detection limits are 100 and 0.05 microg l(-1) for MSM, and 96 and 0.1 microg l(-1) for DPX-A 7881, respectively. The linear range and precision (RSD) are 0.1-50 microg l(-1) and 7.0% for MSM, and 0.2-50 microg l(-1) and 9.2% for DPX-A 7881, respectively. This proposed method was applied to determine MSM and DPX-A 7881 in seawater, tap water, and bottled mineral water with spiked recoveries in the range of 83-95% for MSM and 88-100% for DPX-A 7881, respectively.  相似文献

A comprehensive two-dimensional liquid chromatographic system (2D SCX/RP) is con- structed with a 10-port-2-way valve using strong cation exchange chromatography (Hypersil SCX, 100 mm×4.6 mm I.D.) followed by reversed phase chromatography (Hypersil BDS C18, 15 mm×4.6 mm I.D.) to separate the complex peptides from globin peptic hydrolysate. After the sample was loaded on the SCX column, the phosphate buffer (pH 4.0) was used to elute the peptides. Then, elutes flowed through the interface and the peptides focused on the head of the trapping columns (Hypersil BDS C18, 15 mm×4.6 mm I.D.) but salt passed into the waste. After the valve was switched, the samples were flushed with a backward flow into the RP analytical column. The peptides on the SCX were eluted with 12 discontinuous steps linearly increasing salt concentrations. The peptides enriched on the trapping column were desalted and separated by the RP columns. The resolution and the resolved peaks of the 2D SCX/RP system were greatly increased and the total peak capacity reached as high as 2280.  相似文献

A solid-phase extraction (SPE) method for sample clean-up followed by a reversed-phase high-performance liquid chromatography (HPLC) procedure for the assay of five antidepressant drugs (trazodone, doxepin, desipramine, maprotiline and imipramine) is reported. The drugs were recovered from plasma buffered at a suitable pH using C18 Bond-Elut cartridges and mixtures of methanol-aqueous buffer as washing and elution solvents. The recoveries of the drugs using other sorbent materials (C8, C2, cyclohexyl, cyanopropyl and phenyl Bond Elut and copolymer HLB waters cartridges) were also examined. The selectivity of SPE was examined by using spiked plasma samples and the CH cartridge gave rise to the cleanest extracts. Cyclohexyl cartridges were conditioned successively with 2 ml of methanol and 1 ml of acetic acid-sodium acetate buffer (0.1 M, pH 4.0). Plasma sample was buffered at pH 4.0 and then applied to the sorbent. The washing step was performed subsequently with 1.5 ml of acetate buffer (0.1 M, pH 4.0), 100 microl of acetonitrile and 1 ml of methanol-acetate buffer (30:70, v/v). Finally, the analytes were eluted with 0.5 ml of methanol-acetate buffer (70:30, v/v). The extract was evaporated to dryness, reconstituted in mobile phase, and chromatographed on a reversed-phase C18 column with ultraviolet detection at 215 nm. The recoveries of trazodone, doxepin, desipramine, maprotiline and imipramine from spiked plasma samples using the CH cartridge were 58 2, 84 3, 83 3, 83 3 and 82 2%, respectively. The within-day and between-day repeatabilities were lower than 6% and 9%, respectively. The linearity of calibrations for the five antidepressants was between 0.005 and 2 microg/ml. The limits of detection were 1 ng/ml for trazodone, doxepin and desipramine and 2 ng/ml for maprotiline and imipramine.  相似文献

The accuracy, repeatability, and reproducibility characteristics of a method for measuring levels of zearalenone (ZON) in botanical root products, soybeans, grains, and grain products were determined by an AOAC single-laboratory validation procedure. Replicates of 10 test portions of each powdered root product (black cohosh, ginger, ginseng), brown rice flour, brown rice grain, oat flour, rice bran, soybeans, and wheat flour at each spiking level (ZON at 0, 50, 100, and 200 microg/kg) were analyzed on 3 separate days. Test samples were extracted with methanol-water (75 + 25, v/v). The extracts were centrifuged or filtered, diluted with phosphate-buffered saline (PBS) containing 0.5% Tween 20, and filtered; the filtrates were applied to an immunoaffinity column containing antibodies specific for ZON. After the column was washed with methanol-PBS (15 + 85, v/v) containing 0.5% Tween 20 and then with water, the toxin was eluted from the column with methanol, and the eluate was diluted with water. The eluate containing the toxin was then subjected to RPLC with fluorescence detection. All commodities that were found to contain ZON at < 10 microg/kg were used for the recovery study. The average within-day and between-days recoveries of ZON added at levels of 50-200 microg/kg ranged from 82 to 88% and from 81 to 84%, respectively, for all test commodities. The total average of within- and between-day SD and RSDr values for all test commodities ranged from 2.5 to 7.3 microg/kg and from 4.6 to 6.2%, respectively. HorRat values were <1.3 for all matrixes examined. The tested method was found to be acceptable for the matrixes examined.  相似文献

建立了化妆品中7种磺胺(磺胺醋酰、磺胺吡啶、磺胺甲基嘧啶、磺胺二甲嘧啶、磺胺甲氧嘧啶、磺胺间甲氧嘧啶、磺胺甲基异唑)和甲硝唑及氯霉素的高效液相色谱测定方法。样品经0.1%甲酸水溶液-乙腈(体积比为8∶2)混合液超声提取后进行液相色谱分析。方法的定量检测限为3~80 μg/g,7种磺胺在20~200 μg/mL时,甲硝唑及氯霉素在40~400 μg/mL时方法的线性关系良好(r≥0.9993)。加标回收率为83.8%~105.3%(7种磺胺的添加水平为50 μg/mL和150 μg/mL,甲硝唑及氯霉素的添加水平为100 μg/mL和300 μg/mL),其相对标准偏差均小于5%。  相似文献
