Two on-line SPE-LC-ESI-MS/MS methods have been developed for the rapid determination and confirmation of 18 polar pesticides and nine transformation products (TPs) in water samples. Given the very different physico-chemical characteristics of the analytes, it was not feasible the simultaneous determination of all selected compounds in only one method. Thus, it was necessary to use heptafluorobutyric acid and formic acid in order to obtain good retention in the SPE cartridge for basic and acidic analytes, respectively. The developed analytical methodology based on the direct injection of 2 mL of water sample in the system allowed the quantification of all analytes at the 25 ng/L level (LOQ) with limits of detection normally lower than 5 ng/L. Satisfactory recoveries (70-110%) were obtained for most compounds in ground and surface water samples. Some exceptions were found mainly in surface water, due to the ion suppression produced by the higher amount of matrix interferents in these samples. The acquisition of two MS/MS transitions for each compound allowed the reliable confirmation of positive findings even at the LOQ level. The developed methodology was applied to real ground and surface water samples showing the interest of including TPs in monitoring methods, as several of them were found at concentrations higher than that of parent compounds.  相似文献

The potential of three mass spectrometry (MS) analyzers (triple quadrupole, QqQ; time of flight, TOF; and quadrupole time of flight, QTOF) has been investigated and compared for quantification, confirmation and screening purposes in pesticide residue analysis of fruit and vegetable samples. For this purpose, analytical methodology for multiresidue determination of 11 pesticides, taken as a model, has been developed and validated in nine food matrices for the three mass analyzers coupled to ultra high pressure liquid chromatography. In all cases, limits of quantification around 0.01 mg/kg were reached, fulfilling the most restrictive case of baby‐food analysis. Regarding absolute sensitivity, the lower limits of detection were obtained, as expected, for QqQ (100 fg), whereas slightly higher limits (300 fg) were obtained for both TOF and QTOF. Confirmative capacity of each analyzer was studied for each analyte based on the identification points (IPs) criterion, useful for a comprehensive comparison. QTOF mass analyzer showed the highest confirmatory capacity, although QqQ normally led to sufficient number of IPs, even at lower concentration levels. The potential of TOF MS was also investigated for screening purposes. To this aim, around 50 commercial fruits and vegetables samples were analyzed, searching for more than 400 pesticides. TOF MS proved to be an attractive analytical tool for rapid detection and reliable identification of a large number of pesticides thanks to the full spectrum acquisition at accurate mass with satisfactory sensitivity. This process is readily boosted when combined with specialized software packages, together with theoretical exact mass databases. Several pesticides (e.g. carbendazim in citrus and indoxacarb in grape) were detected in the samples. Further unequivocal confirmation of the identity was performed using reference standards and/or QTOF MS/MS experiments. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

Omeprazole is one of the most consumed pharmaceuticals around the world. However, this compound is scarcely detected in urban wastewater and surface water. The absence of this pharmaceutical in the aquatic ecosystem might be due to its degradation in wastewater treatment plants, as well as in receiving water. In this work, different laboratory‐controlled degradation experiments have been carried out on surface water in order to elucidate generated omeprazole transformation products (TPs). Surface water spiked with omeprazole was subjected to hydrolysis, photo‐degradation under both sunlight and ultraviolet radiation and chlorination. Analyses by liquid chromatography coupled to quadrupole time‐of‐flight mass spectrometry (LC–QTOF MS) permitted identification of up to 17 omeprazole TPs. In a subsequent step, the TPs identified were sought in surface water and urban wastewater by LC–QTOF MS and by LC coupled to tandem mass spectrometry with triple quadrupole. The parent omeprazole was not detected in any of the samples, but four TPs were found in several water samples. The most frequently detected compound was OTP 5 (omeprazole sulfide), which might be a reasonable candidate to be included in monitoring programs rather than the parent omeprazole. Liquid chromatography (LC) coupled to hybrid quadrupole time-of-flight (QTOF) mass spectrometry (MS) is a useful analytical tool in the elucidation and confirmation of transformation products (TPs)/metabolites of pesticides with a wide range of polarity, in both environmental and biological samples. Firstly, the versatility of LC allows the determination of very distinct TPs/metabolites as chromatographic conditions can be easily changed and optimized depending on the analytical problem. Secondly, the mass accuracy provided by the TOF analyser allows the assignment of a highly probable empirical formula for each compound and the differentiation between nominal isobaric compounds. Finally, the possibility of performing MS/MS spectra with accurate mass measurements can been used for the final characterization of the TPs/metabolites detected and for the differentiation of isomeric compounds. In this study, the insecticide diazinon was used as model compound, and its photodegradation and metabolism have been investigated by LC-QTOF-MS. On one hand, environmental spiked water was irradiated with a mercury lamp for 9 days, sampling 3-mL aliquots approximately every 12 h. On the other hand, both in vitro and in vivo metabolism experiments were carried out with different substrate concentrations and incubation times. After centrifugation, and protein precipitation in the in vitro and in vivo studies, 50-μL aliquots of both environmental and biological samples were directly injected into the LC electrospray ionization QTOF system. The most important transformation processes were found to be hydrolysis of the ester moiety, hydroxylation in the aromatic ring or in one of the alkylic groups, oxidation of the sulfur atom on the P=S cleavage or a combination of these processes, with the highest number of compounds being found in the photodegradation study. Very polar compounds, such as diethyl phosphate and diethyl thiophosphate, were detected after direct injection of the aqueous sample, which was feasible owing to the characteristics of the LC. In MS mode, mass errors were below 3 mDa, leading to an empirical formula for each compound. MS/MS spectra with accurate mass were used for the final elucidation of the compounds detected.  相似文献

The presence of pharmaceuticals in the environment is a matter of major concern because of their wide consumption and their potential negative effect on the water quality and living organisms. After human and/or veterinary consumption, pharmaceuticals can be excreted in unchanged form as the parent compound and/or as free or conjugated metabolites. These compounds seem not to be completely removed during wastewater treatments and might finally arrive to surface and ground waters. Consequently, both parent pharmaceuticals and metabolites are target analytes to be considered in analytical methodologies. The satisfactory sensitivity in full-acquisition mode, high-resolution, exact mass measurements and MS/MS capabilities of hybrid quadrupole time-of-flight (QTOF) mass spectrometry make of this technique a powerful analytical tool for the identification of organic contaminants. In this study, the use of QTOF-MS with the aid of specialised processing-data application managers has allowed the retrospective analysis of pharmaceuticals metabolites in urban wastewater without the need for additional injection of sample extracts. Around 160 metabolites have been investigated in wastewater samples previously analysed only for parent compounds using LC-QTOF under MS(E) mode (simultaneous recording of two acquisition functions, at low and high collision energy). The retrospective analysis was applied to search for pharmaceutical metabolites in parent-positive effluent wastewaters from the Spanish Mediterranean region. Five metabolites, such as clopidogrel carboxylic acid or N-desmethyl clarithromycin, were identified in the samples.  相似文献

Pesticides are widely applied and they can produce a variety of transformation products (TPs), through different pathways and mechanisms. Nowadays there is a growing interest related to the determination of pesticide TPs in several matrices (environmental, food and biological samples), due to these compounds can be more toxic and persistent than parent compounds, and some of them can be used as markers of exposure to different pesticides. Although solid-phase extraction (SPE) is mainly used for the extraction of TPs, alternative techniques such as solid-phase microextraction (SPME) and liquid-phase extraction (LPE) can be used. These TPs are mainly determined by liquid chromatography (LC) due to the recent developments in this technique, especially when it is coupled to mass spectrometry (MS) detectors, allowing the determination of known and/or unknown TPs. Furthermore, MS is a very valuable tool for the structural elucidation of unknown TPs. This review discusses all phases of analytical procedure, including sample treatment and analysis, indicating the main problems related to the extraction of TPs from several matrices due to their high polarity, as well as the different alternatives found for the simultaneous determination of parent compounds and TPs, using chromatographic techniques coupled to MS detection.  相似文献

A new multiresidue method has been validated in cucumber matrix for the routine analysis of 130 multiclass pesticide residues by gas chromatography/triple quadrupole mass spectrometry. The pesticides were extracted with ethyl acetate. A first identification of the pesticides was based on a tandem mass spectrometric (MS/MS) screening method, which monitors a single transition for each target compound, in less than 12 min. After that, potentially non-negative samples were analyzed again by the MS/MS confirmation/quantification method, which monitors two or three MS/MS transitions for each compound, also in less than 12 min. Performance characteristics, such as trueness, precision, linear range, detection limit (LOD) and quantification limit (LOQ), for each pesticide were calculated. The average recoveries obtained ranged between 70 and 120% at three different fortification levels (25, 200 and 500 microg/kg) with precision, expressed as relative standard deviation (RSD), values lower than 15%. The calculated LOD and LOQ were typically <3.2 and 9.6 microg/kg, respectively. Such limits were much lower than the maximum residue levels (MRLs) established by European legislation. The proposed methodology was applied to the determination of pesticides in real vegetable samples from Almería (Spain).  相似文献

ABSTRACT

An on-line automated method for phototransformation studies of alachlor, aldicarb, and methiocarb by means of liquid chromatography/mass spectrometry (LC/MS) with particle beam (PB) interface is described. Surface water samples were first spiked with 50 μg/1 of each pesticide and then exposed to the radiation of a medium-pressure mercury lamp. Next, in regular intervals of 60 min, aliquots of 50-ml sample were enriched on a solid-phase extraction (SPE) cartridge and on-line eluted by a gradient of LC eluent onto analytical column for separation, followed by MS detection. A phototransformation experiment with each pesticide was performed twice, with MS operated in electron ionisation (EI) and positive chemical ionisation (PCI) modes. Many transformation products (TPs) of parent pesticides were detected and in numerous instances, tentatively identified. Appearance of several transformation products is reported for the first time.  相似文献

The widely-used pesticide propanil is a selective post-emergent general-use acetanilide herbicide registered for control of broadleaf and grass weeds in rice, small grain, and turf. Because broad application and quite heavy use of this herbicide lead to contaminated sites and, consequently, contaminated water, immunoanalytical methods with very low limits of detection (LOD) and low limits of quantification (LOQ) are becoming increasingly important for environmental analysis and, especially, for monitoring drinking-water quality. Environmental monitoring of pesticides, hormones, endocrine-disrupting chemicals, and antibiotics in aqueous samples (e.g. surface, ground, waste, or drinking water) with quite difficult matrices places large demands on chemical analysis. Biosensors have suitable characteristics such as efficiency in enabling very fast, sensitive, and cost-effective detection. Here we describe the steps of progress toward sub-nanogram per liter detection of propanil with a fully automated immunoassay. In contrast with common analytical methods such as GC–MS or HPLC–MS the biosensor used requires no sample pre-treatment and pre-concentration. The basis of our sensitive assay is an antibody with a high affinity constant toward propanil. During the optimization process, we compared different surface modifications (four different immobilized derivatives) and reduced the amount of antibody per sample. In fact, optimization of the assay resulted in an LOD of 0.6 ng L–1 and an LOQ of 4.5 ng L–1 without any sample pre-treatment and without pre-concentration. These results for propanil with the RIANA instrument, and its improved sensitivity for detection of a single pesticide at the low nanogram per liter range, show that biosensors can compete with common analytical methods in the field of water analysis.  相似文献

In this work, we have developed a sensitive method for detection and quantification of eight N-nitrosamines, N-nitrosodimethylamine (NDMA), N-nitrosomorpholine (NMor), N-nitrosomethylethylamine (NMEA), N-nitrosopirrolidine (NPyr), N-nitrosodiethylamine (NDEA), N-nitrosopiperidine (NPip), N-nitroso-n-dipropylamine (NDPA) and N-nitrosodi-n-butylamine (NDBA) in drinking water. The method is based on liquid chromatography coupled to tandem mass spectrometry, using atmospheric pressure chemical ionization (APCI) in positive mode with a triple quadrupole analyzer (QqQ). The simultaneous acquisition of two MS/MS transitions in selected reaction monitoring mode (SRM) for each compound, together with the evaluation of their relative intensity, allowed the simultaneous quantification and reliable identification in water at ppt levels. Empirical formula of the product ions selected was confirmed by UHPLC-(Q)TOF MS accurate mass measurements from reference standards.Prior to LC–MS/MS QqQ analysis, a preconcentration step by off-line SPE using coconut charcoal EPA 521 cartridges (by passing 500 mL of water sample) was necessary to improve the sensitivity and to meet regulation requirements. For accurate quantification, two isotope labelled nitrosamines (NDMA-d6 and NDPA-d14) were added as surrogate internal standards to the samples. The optimized method was validated at two concentration levels (10 and 100 ng L−1) in drinking water samples, obtaining satisfactory recoveries (between 90 and 120%) and precision (RSD < 20%). Limits of detection were found to be in the range of 1–8 ng L−1. The described methodology has been applied to different types of water samples: chlorinated from drinking water and wastewater treatment plants (DWTP and WWTP, respectively), wastewaters subjected to ozonation and tap waters.  相似文献

The herbicide bromacil has been extensively used in the Spanish Mediterranean region, and although plant protection products containing bromacil have been withdrawn by the European Union, this compound is still frequently detected in surface and ground water of this area. However, the fast and complete disappearance of this compound has been observed in water intended for human consumption, after it has been subjected to chlorination. There is a concern about the possible degradation products formed, since they might be present in drinking water and might be hazardous. In this work, the sensitive full‐spectrum acquisition, high resolution and exact mass capabilities of hybrid quadrupole time‐of‐flight (QTOF) mass spectrometry have allowed the discovery and proposal of structures of transformation products (TPs) of bromacil in water subjected to chlorination. Different ground water samples spiked at 0.5 µg/mL were subjected to the conventional chlorination procedure applied to drinking waters, sampling 2‐mL aliquots at different time intervals (1, 10 and 30 min). The corresponding non‐spiked water was used as control sample in each experiment. Afterwards, 50 μL of the water was directly injected into an ultra‐high‐pressure liquid chromatography (UHPLC)/electrospray ionization (ESI)‐(Q)TOF system. The QTOF instrument enabled the simultaneous recording of two acquisition functions at different collision energies (MSE approach): the low‐energy (LE) function, fixed at 4 eV, and the high‐energy (HE) function, with a collision energy ramp from 15 to 40 eV. This approach enables the simultaneous acquisition of both parent (deprotonated and protonated molecules) and fragment ions in a single injection. The low mass errors observed for the deprotonated and protonated molecules (detected in LE function) allowed the assignment of a highly probable molecular formula. Fragment ions and neutral losses were investigated in both LE and HE spectra to elucidate the structures of the TPs found. For those compounds that displayed poor fragmentation, product ion scan (MS/MS) experiments were also performed. On processing the data with specialized software (MetaboLynx), four bromacil TPs were detected and their structures were elucidated. To our knowledge, two of them had not previously been reported. A multiresidue method using gas chromatography coupled to ion-trap tandem mass spectrometry (MS/MS) was developed for the analysis of 27 pesticides, commonly used in Alsace, in atmospheric samples (particle and gas phases). As pesticides are expected to be present at very low concentrations and in a complex matrix, the analytical method used was both highly selective and sensitive. These two properties were obtained by associating chromatography with ion-trap MS/MS. To develop this method, analysis of electron impact in single MS was first conducted to choose the parent ions of the pesticides studied. Among the 27 pesticides analysed, seven of them require a derivatisation step. This was the case of some ureas (chlorotoluron, diuron and isoproturon), phenoxy acids (2,4-dichlorophenoxyacetic acid, 4-chloro-2-methylphenoxyacetic acid and mecoprop) and of bromoxynil. The derivatisation was performed with success with pentafluorobenzylbromide. Then, a MS/MS method was optimised by parameters such as the radio frequency storage level and the collision-induced dissociation excitation voltage. Finally, a last step enabled the development of two calibrating programs based on the quantification of daughter ions for the 20 pesticides analysed directly (run 1) and for the seven pesticides which needed derivatisation (run 2). With this analytical procedure, the detection limits varied between 2.5 and 1,250 pg m–3 depending on the compounds studied. This method was tested with success for atmospheric samples collected in Strasbourg (France) during intensive pesticide treatment in 2002.  相似文献

Until now, time-of-flight (TOF) mass analysers have only been very rarely used in pesticide residue analysis (PRA) of water samples. However, the inherent characteristics of TOF MS make these analysers well-suited to this field, mainly for qualitative purposes. Thus, the high sensitivity obtained from full-scan acquisition in comparison to other MS analysers and the high resolution of TOF MS suggest its suitability for screening purposes; it also increases the multiresidue capabilities of methods based on it and decreases the chance of recording false positives. Although these characteristics can also be helpful for quantification, confirmation and elucidation, some limitations on the use of TOF for these purposes have been observed. These limitations are more noticeable when dealing with samples containing very low analyte concentrations, which is the normal situation for PRA in water. The use of hybrid quadrupole–time-of-flight instruments (QTOF) minimises the limitations of TOF, facilitating the simultaneous detection and unequivocal confirmation of pesticides found in the sample. Additionally, the acquisition of accurate product ion full-scan mass spectra can help to elucidate the structures of unknown compounds. In this paper, the potential of TOF and QTOF hyphenated to liquid chromatography for PRA in water is explored, emphasizing both the advantages and limitations of this approach for screening, quantification, confirmation and elucidation purposes. Emphasis is placed on the determination of polar pesticides and transformation products—the analytes that fit well with LC–API–(Q)TOF MS technology.  相似文献

As a suitable way for routine screening of pesticides and control of other organic contaminants in water, the combination of liquid chromatography triple quadrupole tandem mass spectrometry (LC–QqQ-MS/MS) and liquid chromatography–hybrid quadrupole time-of-flight mass spectrometry (LC–QTOF-MS) has been applied to the analysis of 63 surface and waste water samples after conventional solid-phase extraction (SPE). The extracts were screened for 43 pesticides or degradation products by LC–QqQ-MS/MS achieving limits of detection (LOD) ranged from 0.04 to 2 ng L−1. Of the 43 selected pesticides, 33 were detected in water samples. The ESI–QTOF MS instrument was run using two simultaneous acquisition functions with low and high collision energy (MSE approach) and acquiring the full mass spectra. A home-made database containing more than 1100 organic pollutants was used for substance identification. Around 250 of these compounds were available at the laboratory as reference standards. Five pesticides and 3 of their degradation products, different to those selected in the QqQ method, were detected by QqTOF-MS. Thirteen pharmaceuticals and two drugs of abuse were also identified in the samples. In practice, the sample preparation proved to be suitable for both techniques and for a wide variety of substances with different polarity. Mutual confirmation and evidence of co-occurrence of several other organic contaminants were the main advantages of the combination of both techniques.  相似文献

An ultra-high-pressure liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) method for the determination of 37 pesticides (herbicides, insecticides and fungicides) in environmental and wastewater has been developed. To efficiently combine UHPLC with MS/MS, a fast-acquisition triple quadrupole mass analyzer was used. This analyzer (minimum dwell time, 5 ms) allows acquiring up to three simultaneous transitions in the selected reaction monitoring mode for each compound assuring a reliable identification without resolution or sensitivity losses. A pre-concentration step based on solid-phase extraction using Waters Oasis HLB cartridges (0.2 g) was applied with a 100-fold pre-concentration factor along the whole analytical procedure. The method was validated based on European SANCO guidelines using surface, ground, drinking and treated water (from an urban solid residues treatment plant) spiked at two concentration levels (0.025 and 0.1 μg/L), the lowest having been established as the limit of quantification objective. The method showed excellent sensitivity, with instrumental limits of detection ranging from 0.1 to 7 pg. It was applied to environmental water samples (ground and surface water) as well as to samples of urban solid waste leachates (raw leachate and treated leachate after applying reversed osmosis) collected from a municipal treatment plant. Matrix effects have been studied in the different types of water samples analyzed, and several isotope-labelled internal standards have been evaluated as a way to compensate the signal suppression observed for most of the compounds studied, especially in wastewater samples. As a general remark, only those pesticides which response was corrected using their own isotope-labelled molecule, could be satisfactorily corrected in all type of samples, assuring in this way the accurate quantification in all matrix samples.  相似文献

Gas chromatography coupled to tandem mass spectrometry (GC-MS/MS) is one of the most powerful techniques in pesticide residue analysis. MS/MS can be conceived in two ways: tandem in space (e.g. triple quadrupole, QqQ) or in time (e.g. ion trap, IT). QqQ and IT are commonly interfaced to GC; however, there has not been any direct comparison between them in pesticide residue analysis so far. In the present work, the performance of GC coupled to these two analyzers (GC-QqQ-MS/MS and GC-IT-MS/MS) was studied and compared for pesticide residue analysis as well as its application in food analysis. The large volume injection (LVI) technique together with programmed-temperature vaporization (PTV) was applied. For this purpose, 19 pesticides, including organochlorine and organophophorus pesticides and pyrethroids, were analyzed in both systems. Mass spectrometric data, performance characteristics (linearity, intra-day and inter-day precision) and the influence of the matrix nature on the analysis of low concentrations were compared. The target compounds were analyzed in solvent and in two representative food matrices such as cucumber (high water content) and egg (high fat content). MS data and intra-day precision were similar in QqQ and IT, whereas inter-day precision was significantly worse in QqQ. Linearity (expressed as determination coefficient, R(2)) in the range 10-150mugL(-1) was adequate in both systems; however, better R(2) values were obtained with the QqQ analyzer in high and low concentration ranges (1-50 and 1-750mugL(-1), respectively). The influence of the matrix nature on the analysis of low concentrations of each analyzer was also evaluated. The QqQ and IT performance was similar in cucumber and solvent. However, QqQ provided better sensitivity in egg working in selected reaction monitoring (SRM).  相似文献
