A major low-energy fragmentation reaction of many protonated dipeptides involves cleavage of the amide bond resulting in formation of either the y(1)" ion or the a(1) ion. For a series of protonated dipeptides H-Val-Xxx-OH it is observed that log(y(1)"/a(1)) is a linear function of the proton affinity of the variable C-terminal amino acid. For the series of protonated dipeptides H-Xxx-Phe-OH log(a(1)/y(1)") gives a poor correlation with the proton affinity or gas-phase basicity of H-Xxx-OH. However, a good limited correlation of log(a(1)/y(1)") with the Taft-Topsom sigma(alpha) for the alkyl group is observed when Xxx is an aliphatic amino acid. It is proposed that fragmentation occurs by initial formation of a proton-bound complex of an aziridinone and an amino acid which may fragment to form either a protonated amino acid (y(1)") or an N-protonated aziridinone with the corresponding neutrals being an aziridinone and an amino acid. Ab initio calculations show that the N-protonated aziridinone is unstable and fragments by loss of CO to form the a(1) immonium ion. However, the proton-bound complex of an aziridinone and an amine base is a stable species which exists in a potential well. Copyright 2000 John Wiley & Sons, Ltd.  相似文献

It was demonstrated that the reaction of epoxide and imine as a latent initiator under highly humid conditions was accelerated by addition of 5‐phenoxymethyl‐1,3‐oxathiolane‐2‐thione ( 1 ). When 1 was added to a mixture of glycidyl phenyl ether and an imine, the reaction of the epoxide with an amine released from the imine became faster than was the case without 1 , that is, 1 worked as a promoter of the reaction. The reaction of 1-methylcyclopentene oxide with alcohols (methanol, ethanol, n-propanol and isopropanol, n-butanol, and n-pentanol) in the presence of Na and of BF3 etherate has been described. The addition of alcohols in an alkaline medium takes place with the formation of secondary ethers of 1-methylcyclopentane-1,2-diol. In an acid medium, tertiary ethers of 1-methylcyclopentane-1,2-diol possibly contaminated with the secondary ethers are formed. In an acid medium, not only does addition take place but the initial oxide isomerizes into a carbonyl compound and to some extent forms trans-1-methylcyclopentane-1,2-diol. With isopropanol, in an alkaline medium the initial oxide is recovered and in an acid medium only the isomerization product is formed.For part III, see [1].  相似文献

Flash photolysis of ethereal solutions of sodium results in the almost complete disappearance of the original absorption band at 15000 cm−1 and in the formation of solvated electrons. After the flash an intermediate with an absorption maximum at 12000 cm−1 is formed. This absorption is assigned to a species which has the stoichiometric composition of a sodium atom. Although it may be described as a metal “monomer” we propose as an alternative model the concept of an “ion pair” of a solvated electron and a sodium cation. In analogy to the 12000 cm−1 transient we propose that the blue species can be described as an associate of a sodium cation with two solvated electrons.  相似文献

The behavior of resazurin (1) as an electron acceptor in glucose oxidase (GOD)-catalyzed oxidation of glucose under anaerobic conditions is described. When a mixture of 1, glucose, and GOD in phosphate buffer (pH 7.4, 0.1 M) was incubated at 25 degrees C, the resulting solution turned purple to fluorescent pink due to the deoxygenated product, resorufin (2). On incubation of 1 with GOD alone or with H2O2 under essentially the same conditions, no color change was seen, indicating that generation of 2 in the enzymatic reaction is brought about through reduction of 1 by the reduced form (GODred) of GOD, which was also supported by the voltammetric behavior of 1. However, it was found that the enzymatic transformation of 1 to 2 is of no practical use as an indicator reaction for glucose determination using only GOD due to a slow reaction of 1 with GODred. Based on a ping-pong type mechanism with a steady-state approximation, KM and kcat for 1 as an electron acceptor from GODred were estimated to be 15+/-1.3 microM and (5.0+/-0.5) x 10(-2) s(-1), respectively.  相似文献

The effect of a pi-electron-donating macrocyclic molecule on the photochromic behavior of viologen derivatives was investigated in a thin polymer film. The intermolecular interactions between the viologens and the macrocyclic molecule were investigated in a solution before photoirradiation. In acetone, benzylviologens, N,N'-dibenzyl-4,4'-bipyridinium hexafluorophosphate (1) and N,N'-dibenzyl-trans-1,2-bis(4-pyridinium)ethylene hexafluorophosphate (2) each derived from 4,4'-bipiyridine and trans-1,2-bis(4-pyridyl)ethylene, respectively, form an inclusion complex with p-benzocrown ether (3) with binding constants of ca 200 M-1, which was driven by a charge transfer interaction. The peak wavelength of the charge transfer absorption band was at 453 and 421 nm for the inclusion complexes of 1 and 2 with 3, respectively. Upon photoirradiation to the polymer film containing 1, the film changed color from colorless to blue, associated with the reduction of 1 from the dication to the radical cation. The original dication was recovered after 120 min. The addition of 3 into the film containing 1 caused not only the color change from colorless to yellow, associated with the charge transfer interaction between 1 and 3 before photoirradiation, but also an acceleration in the bleaching rate of the photoreduced 1. When p-dimethoxybenzene (4) was used as an acyclic analog of 3, a negligible change in the photochromic behavior of 1 was observed. Similar effect of 3 on the photochromic behavior of 2 was observed. These results imply that the pi-electron-donating macrocyclic molecule causes a faster bleaching of photoreduced viologens by forming the inclusion complex.  相似文献

Summary The analysis of spirits with a single stationary phase is still an unsolved problem; the two amyl alcohols, ethyl acetate and acetal are not separated on a polar column, methanol and acetaldehyde coelute from an apolar column. Trials with coupled columns of different polarities showed that optimum results were obtained with a 40 m capillary column, comprising 6 m of Carbowax 20 M, 12 m of 1:1 Carbowax 20M:OV1 mixture and 30 m of PS 264. Best results were achieved when a 1 m retention gap was used, injecting a small amount of sample and using a 7 cm syringe needle.  相似文献

Reaction of an arylacetylene with an azide in hot water gave 1,4-disubstituted 1,2,3-triazoles in high yields, while similar reaction between a terminal aliphatic alkyne and an azide (except m-nitroazidobenzene) afforded a mixture of regioisomers with the ratio of 1,4- to 1,5-isomers ranging from 3:1 to 28.6:1. Reactions of m-nitroazidobenzene with either arylalkynes or aliphatic alkynes formed only 1,4-disubstituted derivatives in excellent yields.  相似文献

An electrochemical immuno-biosensor for detecting glycosylated haemoglobin (HbA1c) is reported based on glassy carbon (GC) electrodes with a mixed layer of an oligo(phenylethynylene) molecular wire (MW) and an oligo(ethylene glycol) (OEG). The mixed layer is formed from in situ-generated aryl diazonium cations. To the distal end of the MW, a redox probe 1,1'-di(aminomethyl)ferrocene (FDMA) was attached followed by the covalent attachment of an epitope N-glycosylated pentapeptide (GPP), an analogon to HbA1c, to which an anti-HbA1c monocolonal antibody IgG can selectively bind. HbA1c was detected by a competitive inhibition assay based on the competition for binding to anti-HbA1c IgG antibodies between the analyte in solution, HbA1c, and the surface bound epitope GPP. Exposure of the GPP modified sensing interface to the mixture of anti-HbA1c IgG antibody and HbA1c results in the attenuation of ferrocene electrochemistry due to free antibody binding to the interface. Higher concentrations of analyte led to higher Faradaic currents as less anti-HbA1c IgG is available to bind to the electrode surface. It was observed that there is a good linear relationship between the relative Faradaic current of FDMA and the concentration of HbA1c from 4.5% to 15.1% of total haemoglobin in serum without the need for washing or rinsing steps.  相似文献

Abstract

A microdetermination method (at the μg1?1 level) for uranium has been developed, based on Solid-Phase Spectrophotometry (SPS). The uranium reacts with pyridylazo-resorcinol in the presence of fluoride to form a 1:1:1 red ternary complex, which is fixed on an anion-exchange resin. The resin absorbance is measured directly, and allows the determination of uranium in the range of 1–10μg1?1, with an RSD of 4%. The method has been applied to the determination of U(VI) in natural waters from wells located near the deposits of industrial wastes from a uranium mineral plant in Andujar (Spain).  相似文献

A triphenylsilyl group is used as an auxiliary in the synthesis of heterodisubstituted p-carboranes via triphenylsilyl-p-carborane (1). The preparation of 1 is statistical, but with recovery of the starting p-carborane, the effective conversion to 1 is about 90%. Carborane 1 has been easily converted to its lithium and copper derivatives, which were reacted with a range of electrophiles including alkyl halides: an aryl iodide, an acetylene bromide, and a sulfenyl chloride. The derivatives of 1 are crystalline and UV active, which facilitates their isolation and purification. The Ph(3)Si group is efficiently removed with fluoride to give monosubstituted p-carboranes I, which upon further nucleophilic substitution yield p-carboranes II. The yields of heterodisubstituted products II are higher than for direct, "statistical" syntheses. For example, 12-pentyl-1, 12-dicarba-closo-dodecaborane-1-carboxylic acid was synthesized from p-carborane via 1 in 62% overall yield, a considerable improvement over the 44% yield obtained by direct methods.  相似文献

Propargyloxycarbonyl chloride, 1, has been used to protect the hydroxyl and amino functionalities of amino alcohols and aminophenols in one pot using triethylamine or pyridine as a base. The increased reactivity of benzyltriethylammonium tetrathiomolybdate, 2, toward propargyl carbonates over propargyl carbamates is studied in detail and has been exploited further to develop an orthogonal protection strategy for the hydroxyl and amino functionalities. For example, 2-amino-1-butanol, 6a, was treated with 1 to get the N,O-diPoc compound 7a in 90% yield, which when treated with 1.1 equiv of 2 at room temperature removes the Poc group attached to oxygen while leaving the one attached to nitrogen intact to yield compound 8a in 85% yield. This particular observation offers a new protecting strategy where an amine and an alcohol group can be protected simultaneously in one pot, and in a later synthetic step, if the alcohol group has to be deprotected selectively, it can be achieved with 1 equiv of 2.  相似文献

The results presented here show that the nature of the axial ligand can alter the distribution of electrons between the metal and the porphyrin in complexes where there is an oxygen atom replacing one of the meso protons. The complexes (1-MeIm)(2)Fe(III)(OEPO) and (2,6-xylylNC)(2)Fe(II)(OEPO(*)) (where OEPO is the trianionic octaethyloxophlorin ligand and OEPO(*) is the dianionic octaethyloxophlorin radical) were prepared by addition of an excess of the appropriate axial ligand to a slurry of [Fe(III)(OEPO)](2) in chloroform under anaerobic conditions. The magnetic moment of (2,6-xylylNC)(2)Fe(II)(OEPO(*)) is temperature invariant and consistent with a simple S = (1)/(2) ground state. This complex with an EPR resonance at g = 2.004 may be considered as a model for the free-radical like EPR signal seen when the meso-hydroxylated heme/heme oxygenase complex is treated with carbon monoxide. In contrast, the magnetic moment of (1-MeIm)(2)Fe(III)(OEPO) drops with temperature and indicates a spin-state change from an S = (5)/(2) or an admixed S = (3)/(2),(5)/(2) state at high temperatures (near room temperature) to an S = (1)/(2) state at temperatures below 100 K. X-ray diffraction studies show that each complex crystallizes in centrosymmetric form with the expected six-coordinate geometry. The structure of (1-MeIm)(2)Fe(III)(OEPO) has been determined at 90, 129, and 296 K and shows a gradual and selective lengthening of the Fe-N(axial bond). This behavior is consistent with population of a higher spin state at elevated temperatures.  相似文献

Medical tablet forming ability of microcrystalline cellulose (MCC) was investigated in relation to the mobility of water molecules in MCC particles. For this purpose, the spin-lattice relaxation time T1 of water in the system was measured by 1H-NMR. Over a wide range of water contents (0.02 H2O/cellulose (g/g) 1.79), two different T1 (T1,l and T1,s) values were observed for water in each MCC sample. Below the equilibrium water content, water having these two different T1 values exchange with each other in an MCC particle reaching an equilibrium state within a given time scale (equilibrium constant K). The T1,l, T1,s and K values for water in MCC, estimated at the equilibrium water content, showed fairly good correlations with the hardness of the tablets made by the MCC samples. Sample with a shorter T1, or larger K tended to have a stronger tablet forming ability. In the spin-spin relaxation time T2 measurements for protons in an MCC/D2O system, two T2 components originating from the glassy cellulose solid (T2,G) and the swelling region (T2,l) were observed. It was found that the mole fraction xL of protons with T2,L in the system exhibits a clear linear correlation with K. From these results, a structural model for the distribution of water in MCC particles was propoed by taking the surface of each microfibril and the disordered region within the microfibril into consideration  相似文献

Time-resolved photoelectron spectroscopy was used to study the energetics and dynamics of solvated electrons in aqueous solution. Solvated electrons are generated by ultrafast photodetachment in a 100 mM aqueous NaI solution. Initially, an ensemble of strongly bound ("cold") solvated electrons and an ensemble of weakly bound ("hot") electrons in an unequilibrated solvent environment are observed. We report an ultrafast recombination channel for the "hot" electrons with a rate of (800 fs)(-1) which is in competition with thermalization occurring with a rate of (1.1 ps)(-1). The thermalized electrons recombine with the iodide radical with a rate of (22 ps)(-1). About 35% of the thermalized electrons escape geminate recombination and form free, solvated electrons. The vertical detachment energy for the solvated electron is determined to be 3.40 eV. No indication for a surface-bound electron at lower binding energies was observed.  相似文献

Purification of A1 adenosine receptor of rat brain membranes was performed using a newly developed affinity gel employing xanthine amine congener (XAC) as an immobilized ligand. The A1 adenosine receptor was solubilized with digitonin-cholate from brain membranes and then purified by a sequential use of affinity chromatography on XAC-agarose, hydroxyapatite chromatography and reaffinity chromatography on XAC-agarose. The A1 adenosine receptor was purified ca. 45,000-fold with a yield of 5%. The final receptor preparation gave a single broad band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis with a Mr approximately 34,000. This band was also shown to be specifically labelled with an affinity labelling reagent for A1 adenosine receptors. This purification method was also applicable for the complete purification of A1 adenosine receptors from rat testis and human brain membranes.  相似文献

Two flow methods for the enzymatic determination of pyrophosphate are described that are used to diminish the consumption of reagents. One method is based on the use of an open-close circuit with manual injection using a syringe. The other is a sequential injection method. The analytical features of both methods are: a linear range of 0.4 - 20 mg L(-1), an LOD of 0.38 mg L(-1), and a CV of 2.0% for the sequential injection method, and a linear range of 0.3 - 15 mg L(-1), an LOD of 0.29 mg L(-1), and CV of 2.2% for the open-close circuit method. The methods were applied to the determination of pyrophosphate in urine. The pyrophosphate concentration determined in urine samples varied from 1.26 to 6.67 mg L(-1).  相似文献

Reactions of DNBF with a series of 2-aminothiazoles (1 a-f) to afford thermodynamically stable C-bonded sigma-adducts have been investigated in acetonitrile. A most significant finding emerged on recording NMR spectra immediately after mixing of equimolar amounts of DNBF and the unsubstituted 2-aminothiazole (1 a) in Me2SO: namely, that the formation of 9 a is preceded by that of a short-lived intermediate species X. From the 1H NMR parameters characterizing this intermediate, as well as the dependence of its lifetime on the experimental conditions-the presence of excess DNBF over 1 a increases the lifetime of X while an excess of base (1 a) accelerates its conversion into 9 a--it is convincingly demonstrated that the structure of X combines the presence of a positively charged Wheland complex moiety (with regard to the thiazole ring) with that of a negatively charged Meisenheimer complex moiety (with regard to the benzofuroxan system). So far, only one intermediate of this type (noted WM) has been successfully characterized, in the reactions of DNBF with 1,3,5-tris(N,N-dialkylamino)benzenes. Among the key features supporting the intermediacy of X along the reaction coordinate leading to 9 a is the fact that the reactions of DNBF with 1 a in the presence of an alcohol (MeOH, EtOH, nPrOH) produce new adducts arising from the addition of an alcohol molecule to the thiazole moiety of WM-1 a. Reflecting the presence of three chiral centres, these species are formed as mixtures of several diastereomers that could all be characterized in their racemic forms in ethanol. These findings generalize the previous report on the formation of Wheland-Meisenheimer carbon-carbon complexes in homocyclic series.  相似文献

A new, robust and simple method is proposed for the chemiluminescent determination of the pesticide Bromoxynil. The empirical procedure is performed with the aid of a flow-injection manifold provided with an on-line photo-reactor to obtain chemiluminescent photofragments. After a period of 12 s of irradiation with an 8 W low-pressure mercury lamp, a chemiluminescent oxidation was performed with the system potassium permanganate in a polyphosphoric acid medium. The photolysis required a basic medium (KOH 0.014 mol l(-1)) with ethanol (1%) as a sensitizer. The method allowed the determination of 134 samples (h-1) of Bromoxynil in a wide interval of concentrations, over the range 5 x 10(-3) - 1 mg l(-1); the detection limit was 5 x 10(-3) mg l(-1). The RSD (n=24) at 0.25 mg l(-1) of the pesticide level was 2.3%. The method was applied to a water sample and to a formulation.  相似文献
