The reactions of [Ag(NH=CMe2)2]ClO4 with cis-[PtCl2L2] in a 1:1 molar ratio give cis-[PtCl(NH=CMe2)(PPh3)2]ClO4 (1cis) or cis-[PtCl(NH=CMe2)2(dmso)]ClO4 (2), and in 2:1 molar ratio, they produce [Pt(NH=CMe2)2L2](ClO4)2 [L = PPh3 (3), L2= tbbpy = 4,4'-di-tert-butyl-2,2'-dipyridyl (4)]. Complex 2 reacts with PPh3 (1:2) to give trans-[PtCl(NH=CMe2)(PPh3)2]ClO(4) (1trans). The two-step reaction of cis-[PtCl2(dmso)2], [Au(NH=CMe2)(PPh3)]ClO4, and PPh3 (1:1:1) gives [SP-4-3]-[PtCl(NH=CMe2)(dmso)(PPh3)]ClO4 (5). The reactions of complexes 2 and 4 with PhICl2 give the Pt(IV) derivatives [OC-6-13]-[PtCl3(NH=CMe2)(2)(dmso)]ClO4 (6) and [OC-6-13]-[PtCl2(NH=CMe2)2(dtbbpy)](ClO4)2 (7), respectively. Complexes 1cis and 1trans react with NaH and [AuCl(PPh3)] (1:10:1.2) to give cis- and trans-[PtCl{mu-N(AuPPh3)=CMe2}(PPh3)2]ClO4 (8cis and 8trans), respectively. The crystal structures of 4.0.5Et2O.0.5Me2CO and 6 have been determined; both exhibit pseudosymmetry.  相似文献

Platinum bisphosphine complexes bearing dichalcogen-derivatised naphthalene, acenaphthene or phenanthrene ligands have been prepared by either oxidative addition to zero-valent platinum species or from [PtCl(2)(PPhR(2))] (R=Ph or Me) and the disodium or dilithium salts of the parent disulfur, diselenide or mixed S/Se species. The parent naphthalene, acenaphthene and phenanthrene chalcogen compounds were treated with either [Pt(PPh(3))(4)] or [Pt(C(2)H(4))(PMe(3))(2)] (prepared in situ from [PtCl(2)(PMe(3))(2)], ethene and sodium naphthalide or super hydride [LiBEt(3)H]) to give the appropriate platinum(II) species. The dilithium salts of 1,8-E(2)-naphthalene (E=S or Se) prepared in situ by reduction of the E-E bond with [LiBEt(3)H] were treated with [PtCl(2)(PPh(3))(2)] to give [Pt(1,8-E(2)-nap)(PPh(3))(2)]. The tetraoxides [Pt(1,8-(S(O)(2))(2)-nap)(PR(3))(2)] (PR(3)=PPh(3) or PMe(2)Ph) were prepared in a similar metathetical manner from the appropriate [PtCl(2)(PR(3))] complexes and the disodium salt of naphthalene 1,8-disulfinic acid (1,8-(S(O)ONa)(2)-nap). The X-ray structures of selected examples reveal bidentate coordination with the naphthalene-E(2) unit hinged (111-137 degrees) with respect to the coordination plane. The naphthalene ring suffers significant distortion from planarity.  相似文献

The monofunctional substitution reactions between trans-[PtCl(H2O)(NH3)(pip)]+,trans-[Pt(H2O)2(NH3)(pip)]2+,trans-[PtCl(H2O)(pip)2]+,trans-[Pt(H2O)2(pip)2]2+ (pip = piperidine) and adenine/guanine nucleotides are explored by using B3LYP hybrid functional and IEF-PCM salvation models. For the trans-[Pt(H2O)2(NH3)(pip)]2+ and trans-[PtCl(H2O)(NH3)(pip)]+ complexes, the computed barrier heights in aqueous solution are 13.5/13.5 and 11.6/11.6 kcal/mol from trans-Pt-chloroaqua complex to trans/cis-monoadduct for adenine and guanine, and the corresponding values are 20.7/20.7 and 18.8/18.8 kcal/mol from trans-Pt-diaqua complex to trans/cis-monoadduct for adenine and guanine, respectively. For trans-[PtCl(H2O)(pip)2]+ and trans-[Pt(H2O)2(pip)2]2+,the corresponding values are 21.5/21.3 and 19.4/19.4 kcal/mol,and 26.0/26.0 and 20.7/20.8 kal/mol for adenine and guanine, respectively. Our calculations demonstrate that the barrier heights of chloroaqua are lower than the corresponding values of diaqua for adenine and guanine. In addition, the free energies of activation for guanine in aqueous solution are all smaller than that for adenine, which predicts a preference of 1.9 kcal/mol when trans-[PtCl(H2O)(NH3)(pip)]+ and trans-[Pt(H2O)2(NH3)(pip)]2+ are the active agents and ~1.9 and ~ 5.3 kcal/mol when trans-[PtCl(H2O)(pip)2]+ and trans-[Pt(H2O)2(pip)2]2+ are the active agents, respectively. For the reaction of trans-Pt-chloroaqua (or diaqua) to cis-monoadduct,we obtain the same transition-state structure as from the reaction of trans-Pt-chloroaqua (or diaqua) to trans-monoadduct,which seems that the trans-Pt-chloroaqua (or diaqua) complex can generate trans-or cis-monoadduct via the same transition-state.  相似文献

The complex cis,trans,cis-[PtCl(2)(OAc)(2)NH(3)(c-C(6)H(11)NH(2))] (JM-216) is currently undergoing clinical evaluation as an antitumor agent. In support of characterization and analysis of this complex a study of its isomers and other complexes [PtCl(m)()(OAc)((4)(-)(m)()())NH(3)(c-C(6)H(11)NH(2))] (m = 0-4) has been undertaken. The complexes have been obtained by a variety of synthetic routes which now extend the scope for the preparation of platinum(IV) antitumor complexes. As platinum(IV) complexes are very stable to substitution in the absence of catalysis, use has been made of both light and base catalysis to promote substitution. Oxidative addition to platinum(II) using hypervalent iodine reagents has also been used. The stereochemistry of the complexes has been confirmed by spectroscopic studies, primarily NMR including natural abundance (15)N NMR spectroscopy.  相似文献

Treatment of trans-[PtCl(4)(RCN)(2)] (R = Me, Et) with ethanol allowed the isolation of trans-[PtCl(4)[E-NH[double bond]C(R)OEt](2)]. The latter were reduced selectively, by the ylide Ph(3)P[double bond]CHCO(2)Me, to trans-[PtCl(2)[E-NH[double bond]C(R)OEt](2)]. The complexed imino esters NH[double bond]C(R)OEt were liberated from the platinum(II) complexes by reaction with 2 equiv of 1,2-bis(diphenylphosphino)ethane (dppe) in chloroform; the cationic complex [Pt(dppe)(2)]Cl(2) precipitates almost quantitatively from the reaction mixture and can be easily separated by filtration to give a solution of NH[double bond]C(R)OEt with a known concentration of the imino ester. The imino esters efficiently couple with the coordinated nitriles in trans-[PtCl(4)(EtCN)(2)] to give, as the dominant product, [PtCl(4)[NH[double bond]C(Et)N[double bond]C(R)OEt](2)] containing a previously unknown linkage, i.e., ligated N-(1-imino-propyl)-alkylimidic acid ethyl esters. In addition to [PtCl(4)[NH[double bond]C(Et)N[double bond]C(Et)OEt](2)], another compound was generated as the minor product, i.e., [PtCl(4)(EtCN)[NH[double bond]C(Et)N[double bond]C(Et)OEt]], which was reduced to [PtCl(2)(EtCN)[NH[double bond]C(Et)N[double bond]C(Et)OEt]], and this complex was characterized by X-ray single-crystal diffraction. The platinum(IV) complexes [PtCl(4)[NH[double bond]C(Et)N[double bond]C(R)OEt](2)] are unstable toward hydrolysis and give EtOH and the acylamidine complexes trans-[PtCl(4)[Z-NH[double bond]C(Et)NHC(R)[double bond]O](2)], where the coordination to the Pt center results in the predominant stabilization of the imino tautomer NH[double bond]C(Et)NHC(R)[double bond]O over the other form, i.e., NH(2)C(Et)[double bond]NC(R)[double bond]O, which is the major one for free acylamidines. The structures of trans-[PtCl(4)[Z-NH[double bond]C(Et)NHC(R)[double bond]O](2)] (R = Me, Et) were determined by X-ray studies. The complexes [PtCl(4)[NH[double bond]C(Et)N[double bond]C(R)OEt](2)] were reduced to the appropriate platinum(II) compounds [PtCl(2)[NH[double bond]C(Et)N[double bond]C(R)OEt](2)], which, similarly to the appropriate Pt(IV) compounds, rapidly hydrolyze to yield the acylamidine complexes [PtCl(2)[NH[double bond]C(Et)NHC(R)[double bond]O](2)] and EtOH. The latter acylamidine compounds were also prepared by an alternative route upon reduction of the corresponding platinum(IV) complexes. Besides the first observation of the platinum(IV)-mediated nitrile-imine ester integration, this work demonstrates that the application of metal complexes gives new opportunities for the generation of a great variety of imines (sometimes unreachable in pure organic chemistry) in metal-mediated conversions of organonitriles, the "storage" of imino species in the complexed form, and their synthetic utilization after liberation.  相似文献

A new family of heterometallic compounds 3-6 containing ferrocenyl and platinum(II) centers has been synthesized by reaction of 1-β-aminoethylferrocene (1) and 1,1'-bis(β-aminoethyl)ferrocene (2) with Pt(II) precursors. Using K(2)[PtCl(4)] as the Pt(II) source, the cis-square-planar neutral compounds [Fe{η(5)-C(5)H(4)(CH(2))(2)NH(2)}(2)PtCl(2)] (3) and [{Fe(η(5)-C(5)H(4)(CH(2))(2)NH(2))(η(5)-C(5)H(5))}(2)PtCl(2)] (5) were obtained. Reaction of cis-[PtCl(2)(dmso)(2)] with 1 and 2 resulted in the displacement of dmso and chloride ligands from the platinum coordination sphere, affording the cationic and neutral compounds [Fe{η(5)-C(5)H(4)(CH(2))(2)NH(2)}(2)Pt(dmso)Cl]Cl (4) and [Fe(η(5)-C(5)H(4)(CH(2))(2)NH(2))(η(5)-C(5)H(5))Pt(dmso)Cl(2)] (6). Compounds 3-6 were thoroughly characterized using multinuclear ((1)H, (13)C, (195)Pt) NMR, IR spectroscopy, ESI mass spectrometry and elemental analysis. Single-crystal X-ray analysis of heterometallic 6 confirmed the cis geometry of the molecule and revealed that the platinum atom is held in a perfect square-planar geometry. The electrochemical behaviour of the heterometallic compounds 3-6, which has been examined by cyclic (CV) and square wave (SWV) voltammetries in dichloromethane and dmso solution, is characterized by the reversible one-electron oxidation of the ferrocene moieties. The results of the biological activity studies revealed that the organometallic complex 5 is active against all cell lines with GI(50) values in the range 1.7-2.3 μM. When compared to the standard anticancer drug cisplatin, heterotrimetallic 5, possessing two aminoethylferrocenyl units coordinated to the Pt(II) center, showed a greater activity profile in the colon cancer cell line. Cell cycle studies revealed that the new mixed compound exhibits a mechanism of action different to cisplatin.  相似文献

Capillary electrophoresis-electrospray ionization-mass spectrometry (CE-ESI-MS) has been used for investigating the influence of the sulfur containing amino acid L-methionine (L-Met) on the binding behavior of oxaliplatin (trans-R,R-diaminocyclohexane-(oxalato)platinum(II)) to 5'-GMP. L-Methionine competes with 5'-GMP for the platinum binding site and forms as well as 5'-GMP adducts with oxaliplatin. The formation of the prognosed complexes [Pt(DACH)(L-Met-S,N)]+ and [Pt(DACH)(5'-GMP)2]2- (DACH = 1,2-diaminocyclohexane) could be proved directly by using CE-ESI-MS. Furthermore, we could now bring forward proofs, that the coordination of 5'-GMP with oxaliplatin is inhibited by L-methionine and could show, that the 5'-GMP ligands of the [Pt(DACH) (5'-GMP)2]2- complex can be replaced slowly by L-methionine whereas methionine can not be replaced by GMP.  相似文献

Treatment of the bridging bidentate 1,Z-bis(aminopropyl)-1,Z-dicarba-closo-dodecaborane(12)(1,Z-bis(aminopropyl)-1,Z-carborane) ligands of the type 1,Z-[H(2)N(CH(2))(3)](2)-1,Z-C(2)B(10)H(10)(L(1), Z= 7, 5) or (L(2), Z= 12, 6) with two equivalents of trans-[PtClI(2)(NH(3))](-), followed by halogen ligand metathesis with AgOTf and HCl((aq)) afforded the novel diplatinum(II)-amine species cis-[[PtCl(2)(NH(3))](2)L(n)](7(n= 1) or 8(n= 2), respectively). Similarly, the reaction of L(1) or L(2) with the labile trans-[PtCl(dmf)(NH(3))(2)](+) afforded trans-[[PtCl(NH(3))(2)](2)L(n)](OTf)(2)(9(n= 1) or 10(n= 2), respectively) in good yield and purity. However, isolation of the analogous 1,2-carborane complexes was not possible owing to decomposition reactions that led to extensive degradation of the carborane cage and reduction of the metal centre. The mixed dinuclear complex [cis-[PtCl(2)(NH(3))]-L(1)-trans-[PtCl(NH(3))(2)]]OTf (19) was prepared by treatment of the Boc-protected amine ligand 1-[(Boc)(2)N(CH(2))(3)]-7-[H(2)N(CH(2))(3)]-1,7-C(2)B(10)H(10)(L(3), 15) with trans-[PtCl(dmf)(NH(3))(2)](+) to yield trans-[PtCl(NH(3))(2)L(3)]OTf (16), followed by acid deprotection of the pendant amine group, complexation with trans-[PtClI(2)(NH(3))](-), and halogen ligand metathesis using AgOTf and HCl((aq)). A novel trinuclear species containing 5 was prepared by the addition of two equivalents of 15 to the labile precursor cis-[Pt(dmf)(2)(NH(3))(2)](2+) followed by acid deprotection of the pendant amine groups. Further complexation with two equivalents of trans-[PtClI(2)(NH(3))](-) followed by halogen ligand metathesis using AgOTf and HCl((aq)) afforded the triplatinum(II)-amine species [cis-[Pt(NH(3))(2)(L(1))(2)]-cis-[PtCl(2)(NH(3))](2)](OTf)(2)(23). Complexes 7-10, 19 and 23 represent the first examples of multinuclear platinum(ii)-amine derivatives containing carborane cages. Preliminary in vitro cytotoxicity studies for selected complexes are also reported.  相似文献

Treatment of trans-[PtCl(4)(RCN)(2)](R = Me, Et) with the hydrazone oximes MeC(=NOH)C(R')=NNH(2)(R' = Me, Ph) at 45 degrees C in CH(2)Cl(2) led to the formation of trans-[PtCl(4)(NH=C(R)ON=C(Me)C(R')=NNH(2))(2)](R/R' = Me/Ph 1, Et/Me 2, Et/Ph 3) due to the regioselective OH-addition of the bifunctional MeC(=NOH)C(R')=NNH(2) to the nitrile group. The reaction of 3 and Ph(3)P=CHCO(2)Me allows the formation of the Pt(II) complex trans-[PtCl(2)(NH=C(Et)ON=C(Me)C(Ph)=NNH(2))2](4). In 4, the imine ligand was liberated by substitution with 2 equivalents of bis(1,2-diphenylphosphino)ethane (dppe) in CDCl(3) to give, along with the free ligand, the solid [Pt(dppe)(2)]Cl(2). The free iminoacyl hydrazone, having a restricted life-time, decomposes at 20-25 degrees C in about 20 h to the parent organonitrile and the hydrazone oxime. The Schiff condensation of the free NH(2) groups of 4 with aromatic aldehydes, i.e. 2-OH-5-NO(2)-benzaldehyde and 4-NO(2)-benzaldehyde, brings about the formation of the platinum(II) complexes trans-[PtCl(2)(NH=C(Et)ON=C(Me)C(Ph)=NN=CH(C(6)H(3)-2-OH-5-NO(2))2](5) and trans-[PtCl(2)(NH=C(Et)ON=C(Me)C(Ph)=NN=CH(C(6)H(4)-4-NO(2))2](6), respectively, containing functionalized remote peripherical groups. Metallization of 5, which can be considered as a novel type of metallaligand, was achieved by its reaction with M(OAc)(2).nH(2)O (M = Cu, n= 2; M = Co, n= 4) in a 1:1 molar ratio furnishing solid heteronuclear compounds with composition [Pt]:[M]= 1:1. The complexes were characterized by C, H, N elemental analyses, FAB+ mass-spectrometry, IR, 1H, 13C[1H] and (195)Pt NMR spectroscopies; X-ray structures were determined for 3, 4 and 5.  相似文献

The reaction of platinum(IV) complex trans-[PtCl4(EtCN)2] with pyrazoles 3,5-RR'pzH (R/R' = H/H, Me/H, Me/Me) leads to the formation of the trans-[PtCl4{NH=C(Et)(3,5-RR'pz)}2] (1-3) species due to the metal-mediated nitrile-pyrazole coupling. Pyrazolylimino complexes 1-3 (i) completely convert to pyrazole complexes cis-[PtCl4(3,5-RR'pzH)2] by elimination of EtCN upon reflux in a CH2Cl2 solution or upon heating in the solid state; (ii) undergo exchange at the imino C atom with another pyrazole different from that contained in the pyrazolylimino ligand. The reaction of trans-[PtIICl2(EtCN)2] and 3,5-RR'pzH, conducted under conditions similar to those for trans-[PtIVCl4(EtCN)2], is much less selective, and the composition of the products strongly depends on the pyrazole employed: (a) with pzH, the reaction gives a mixture of three products, i.e., [PtCl2NH=C(Et)pz-kappa2N,N}] (4), [PtCl(pzH){NH=C(Et)pz-kappa2N,N}]Cl (5), and [Pt(pzH)2{NH=C(Et)pz-kappa2N,N}]Cl2 (6) (complexes 5 and 6 are rather unstable and gradually transform to trans-[PtCl2(pzH2] and [Pt(pzH)(4)]Cl(2) and free EtCN); (b) with 3,5-Me(2)pzH, the reaction leads to the formation of [PtCl2NH=C(Et)(3,5-Me2pz)-kappa2N,N}] (7) and [PtCl(3,5-Me2pzH)3]Cl (8); (c) in the case of asymmetric pyrazole 3(5)-MepzH, which can be added to EtCN and/or bind metal centers by any of the two nonequivalent nitrogen sites, a broad mixture of currently unidentified products is formed. The reduction of 1-3 with Ph3P=CHCO2Me in CHCl3 allows for the formation of corresponding platinum(II) compounds trans-[PtCl2{NH=C(Et)(3,5-RR'pz)}2] (9-11). Ligands NH=C(Et)(3,5-RR'pz) (12-14) were almost quantitatively liberated from 9-11 with 2 equiv of 1,2-bis-(diphenylphosphino)ethane in CDCl3, giving free imines 12-14 in solution and the precipitate of trans-[Pt(dppe)2](Cl)2. Pyrazolylimines 12-14 undergo splitting in CDCl3 solution at 20-25 degrees C for ca. 20 h to furnish the parent propiononitrile and the pyrazole 3,5-RR'pzH, but they can be synthetically utilized immediately after the liberation.  相似文献

A possible explanation for the low bioavailability of platinum antitumor compounds is their high reactivity with the sulfur-containing tripeptide glutathione (GSH; deprotonated GSH = SG). GSH is located in the intracellular matrix of the cell with a normal concentration of 5-10 mM. In vivo, only a small fraction of the administered drug will migrate into the cell, resulting in relatively high concentrations of GSH compared to that of the drug. The products of the reactions of [[trans-PtCl(NH(3))(2)](2)-mu-[trans-Pt(NH(3))(2)(NH(2)(CH(2))(6)NH(2))(2)]](NO(3))(4) (BBR3464; 1,0,1/t,t,t, n = 6), [[trans-PtCl(NH(3))(2)](2)-mu-(H(2)N(CH(2))(6)NH(2))](NO(3))(2) (BBR3005; 1,1/t,t, n = 6), [[trans-PtCl(NH(3))(2)](2)-mu-(H(2)N(CH(2))(3)NH(2)(CH(2))(4)NH(2))]Cl(3) (BBR3571; 1,1/t,t-spermidine, n = 3, 4), and trans-[PtCl(2)(NH(3))(2)] (t-DDP) with reduced GSH in phosphate-buffered saline (pH 7.35) have been characterized by (1)H, (195)Pt, and (1)H(-)(15)N gradient heteronuclear single-quantum coherence NMR spectroscopy and high-performance liquid chromatography (HPLC) coupled with electrospray ionization time-of-flight mass spectrometry to determine likely metabolites of the complexes with GSH. Chemical shifts (NMR) and retention times (HPLC) established via analysis of the t-DDP profile served as a fingerprint to compare results obtained for the products afforded by the degradation of the polynuclear compounds by GSH. Identical kinetic profiles and chemical shifts between the metabolites and the t-DDP/GSH products allowed identification of the final product for the 1:2 Pt:GSH reaction as a dinuclear species [[trans-Pt(SG)(NH(3))(2)](2)-mu-SG], in which glutathione bridges the two platinum centers via only the sulfur atom.  相似文献

Three 2,2'-bipyridine (2,2'-bpy) complexes of Pt(IV) have been synthesized, characterized by X-ray crystallography, and their solution behavior in D(2)O studied by (1)H NMR spectroscopic analysis: mer-[PtCl(3)(2,2'-bpy)(MeNH(2))]ClH(2)O (4), trans-[PtCl(2)(2,2'-bpy)(MeNH(2))(2)]Cl(2) (5), and trans-[Pt (2,2'-bpy)(MeNH(2))(2)(OH)(2)]Cl(2) (6; MeNH(2)=methylamine). Complexes 4 and 5 undergo hydrolysis of the Cl(-) ions, both in the dark and daylight, as evident from a drop in the pH value. Two solvolysis products were detected in the case of 4, which is indicative of species with equatorial and axial OH(-) groups. The hydrolysis reaction of 5 implies that an axial Cl(-) group is replaced by an OH(-) moiety; in contrast, 6 remains virtually unaffected. Ordinary daylight, in particular irradiation with a 50-W halogen lamp, initially causes ligand-isomerization processes, which are followed by the reduction of 4 and 5 to Pt(II) species. This reduction of 4 and 5 is accompanied by the formation of hypochlorous acid, as demonstrated qualitatively in the decoloration test of indigo, and loss of MeNH(2), which is particularly pronounced in the case of 5. The formation of Pt(II) compounds is established on the basis of the J coupling constants of (195)Pt with selected (1)H NMR resonances. The results obtained herein are possibly also relevant to the chemistry of Cl-containing Pt(IV) antitumor agents and their reactions with DNA.  相似文献

Trans complexes such as trans-[PtCl(2)(NH(3))(2)] have historically been considered therapeutically inactive. The use of planar ligands such as pyridine greatly enhances the cytotoxicity of the trans geometry. The complexes trans-[PtCl(R'R'SO)(A)(2)]NO(3) (R'R'SO = substituted sulfoxides such as dimethyl (Me(2)SO), methyl benzyl (MeBzSO), and methyl phenyl sulfoxide (MePhSO) and A = NH(3), pyridine (py) and 4-methylpyridine or picoline (pic)) were prepared for comparison of the chemical reactivity between ammine and pyridine ligands. The X-ray crystal structure determination for trans-[PtCl(Me(2)SO)(py)(2)]NO(3) confirmed the geometry with S-bound Me(2)SO. The crystals are orthorhombic, space group P2(1)2(1)2(1), with cell dimensions a = 7.888(2) A, b = 14.740(3) A, c =15.626(5) A, and Z = 4. The geometry around the platinum atom is square planar with l(Pt-Cl) = 2.304(4) A, l(Pt-S) = 2.218(5) A, and l(Pt-N) = 2.03(1) and 2.02(1) A. Bond angles are normal with Cl-Pt-S = 177.9(2) degrees, Cl-Pt-N(1) = 88.0(4) degrees, Cl-Pt-N(2) = 89.3(5) degrees, S-Pt-N(1) = 93.8(4) degrees, S-Pt-N(2) = 88.9(4) degrees, and N(1)-Pt-N(2) = 177.2(6) degrees. The intensity data were collected with Mo Kalpha radiation with lambda = 0.710 69 A. Refinement was by full-matrix least-squares methods to a final R value of 3.80%. Unlike trans-[PtCl(2)(NH(3))(2)], trans-[PtCl(2)(A)(2)] (A = py or pic) complexes do not react with Me(2)SO. The solvolytic products of cis-[PtCl(2)(A)(2)] (A = py or pic) were characterized. Studies of displacement of the sulfoxide by chloride were performed using HPLC. The sulfoxide was displaced faster for the pyridine complex relative to the ammine complex. Chemical studies comparing the reactivity of trans-[PtCl(R'R'SO)(amine)(2)]NO(3) with a model nucleotide, guanosine 5'-monophosphate (GMP), showed that the reaction gave two principal products: the species [Pt(R'R'SO)(amine)(2)(N7-GMP)], which reacts with a second equivalent of GMP, forming [Pt(amine)(2)(N7-GMP)(2)]. The reaction pathways were different, however, for the pyridine complexes in comparison to the NH(3) species, with sulfoxide displacement again being significantly faster for the pyridine case.  相似文献

The hexaphosphapentaprismane P(6)C(4)(t)Bu(4) undergoes specific insertion of the zerovalent platinum fragment [Pt(PPh(3))(2)] into the unique P-P bond between the 5-membered rings to afford [Pt(PPh(3))(2)P(6)C(4)(t)Bu(4)]. A similar reaction with the Pt(ii) complexes [{PtCl(2)(PMe(3))}(2)] and [PtCl(2)(eta(4)-COD)] results in both insertion and chlorine migration reactions. The complexes [Pt(PPh(3))(2)P(6)C(4)(t)Bu(4)], trans-[PtCl(PMe(3))P(6)C(4)(t)Bu(4)Cl], cis-,trans-[{PtCl(2)(PMe(3))}micro-{P(6)C(4)(t)Bu(4)}{PtCl(2)(PMe(3))}], [{PtClP(6)C(4)(t)Bu(4)Cl}(2)] and cis-[PtClP(6)C(4)(t)Bu(4)Cl(P(6)C(4)(t)Bu(4))] have been structurally characterized by single crystal X-ray diffraction and multinuclear NMR studies.  相似文献

The hairpin-stabilized double-stranded oligonucleotides d(TATGGTATT4ATACCATA) (I) and d(TATAGTATT4ATACTATA) (II) were allowed to react with the three aquated forms of the antitumor drug cisplatin (cis-[PtCl2(NH3)2], 1) which are likely candidates for DNA binding, that is, cis-[PtC1(NH3)2(H2O)]+ (2), cis-[Pt(NH3)2(H2O)2]2+ (3), and its conjugate base cis-[Pt(OH)(NH3)2(H2O)]+ (4). The reaction between I and [Pt(NH3)3(H2O)]2+ (5) was also studied for comparison. All reactions were monitored by HPLC. The platination reactions of I and II were carried out in NaClO4 (0.1M) at 293 K and at a constant pH of 4.5 +/- 0.1 for 2, 3, and 5. The data relative to the platination by 4 were obtained from measurements in unbuffered NaClO4 solutions (0.1M) at a starting pH close to neutrality, where 3 and 4 are present in equilibrium. In this case, a fit function describing the pH-time curve allowed the determination of the actual concentrations of 3, 4, and the dihydroxo complex. The platination rate constants characterizing the bimolecular reactions between either I or II and 2, 3, and 4 were individually determined along with the rate constants for hydrolysis of the chloro-monoadducts and for the chelation reactions of the aqua-monoadducts. The reactivity of compounds 2-5, which have the general formula cis-[Pt(NH3)2(H2O)(Y)]2+/-, decreases in the order 3>4>5>2, that is, Y= H2O > OH- >NH3 > Cl-, which is the order of decreasing hydrogen-bond donating ability of Y. Deprotonation of 3 to 4 reduces the reactivity of the platinum complex only by a factor of approximately equals 2, and both complexes discriminate between the different purines of I and II in the same manner. Whereas 3 and 4 react approximately three times faster with the GG sequence of I than with the AG sequence of II, 2 shows a similar reactivity towards both sequences. In view of the well-established preferential binding of cisplatin to GG sequences of DNA in vivo and in vitro, this result suggests that the actual DNA platination species are derived from double hydrolysis of cisplatin.  相似文献

The (benzophenone imine)platinum(II) compounds trans-[PtCl2(Ph2C=NH)(RR'SO)] [R, R'=Me, Me (2); n-Pr, n-Pr (3); (CH2)4 (4); Me, Ph (5); Me, p-MeC6H4 (6)] were prepared by the reaction of Ph2C=NH with K[PtCl3(RR'SO)], obtained in situ from K2[PtCl4] and the corresponding sulfoxide, giving 2-6 as well as cis-[PtCl2(Ph2C=NH)2] (1) as a minor product. The complexes were characterized by 1H, 13C, and 195Pt NMR and IR spectroscopy, electrospray ionization mass spectrometry, and C, H, and N elemental analysis. The X-ray crystallography of 1 enables confirmation of the cis configuration of the complex, while in 2 and 4.1/2CHCl3, the imine and sulfoxide ligands are mutually trans. The solid-state structure of 4.1/2CHCl3 consists of two dimeric Pt moieties representing a rather weak Pt...Pt interaction. The dimeric architecture of 4.1/2CHCl3 is enhanced by the hydrogen bonding between imine H atoms and O atoms. The orthometalation of 1 and 2-6 proceeds both in the solid phase and in a toluene suspension, leading to the formation of [PtCl{Ph(C6H4)C=NH}(Ph2C=NH)] (7) and [PtCl{Ph(C6H4)C=NH}(RR'SO)] (8-12), respectively, isolated in nearly quantitative yields. Complexes 8-12 are emissive at room temperature both in solution (lambdaemmax approximately 535 nm) and in the solid state (lambdaemmax 560-610 nm), with excited-state lifetimes of ca. 300-600 ns, representing a new family of PtII-based luminescent complexes. Compounds 8 and 10 have been characterized by X-ray analysis, confirming the square-planar coordination geometry of the metal center with the almost planar platinacycles. In 8, the asymmetric unit contains two independent Pt molecules, while in 10, it includes four Pt molecules linked by the intermolecular hydrogen-bonding network between the NH group and Cl atoms.  相似文献

The platinum mixed-phosphine complexes (SP-4,2)-[PtCl(8-MTT)(PPh3)(PTA)] (2) and cis-[Pt(8-MTT)2(PPh3)(PTA)] (3) (MTTH2 = 8-(methylthio)theophylline, PTA = 1,3,5-triaza-7-phosphaadamantane) have been prepared from the precursor cis-[PtCl2(PPh3)(PTA)] (1), which has been fully characterized by X-ray diffraction determination. Antiproliferative activity tests indicated that the presence of one lipophilic PPh3 and one hydrophilic PTA makes 1-3 more active than the analogues bearing two PPh3 or two PTA. The reactivity of cis-[PtCl2(PPh3)2], cis-[PtCl2(PTA)2], and cis-[PtCl2(PPh3)(PTA)] with the bis(thiopurines) bis(S-8-thiotheophylline)methane (MBTTH2), 1,2-bis(S-8-thiotheophylline)ethane (EBTTH2), and 1,3-bis(S-8-thiotheophylline)propane (PBTTH2) has also been investigated. New binuclear complexes have been prepared and identified by spectroscopic techniques and their antiproliferative activities on T2 and SKOV3 cell lines evaluated.  相似文献

Addition of excess R(2)NCN to an aqueous solution of K(2)[PtCl(4)] led to the precipitation of [PtCl(2)(NCNR(2))(2)] (R(2) = Me(2) 1; Et(2) 2; C(5)H(10) 3; C(4)H(8)O, 4) in a cis/trans isomeric ratio which depends on temperature. Pure isomers cis-1-3 and trans-1-3 were separated by column chromatography on SiO(2), while trans-4 was obtained by recrystallization. Complexes cis-1-3 isomerize to trans-1-3 on heating in the solid phase at 110 degrees C; trans-1 has been characterized by X-ray crystallography. Chlorination of the platinum(II) complexes cis-1-3 and trans-1-4 gives the appropriate platinum(IV) complexes [PtCl(4)(NCNR(2))(2)] (cis-5-7 and trans-5-8). The compound cis-6 was also obtained by treatment of [PtCl(4)(NCMe)(2)] with neat Et(2)NCN. The platinum(IV) complex trans-[PtCl(4)(NCNMe(2))(2)] (trans-5) in a mixture of undried Et(2)O and CH(2)Cl(2) undergoes facile hydrolysis to give trans-[PtCl(4)[(H)=C(NMe(2))OH](2)] (9; X-ray structure has been determined). The hydrolysis went to another direction with the cis-[PtCl(4)(NCNR(2))(2)] (cis-5-7) which were converted to the metallacycles [PtCl(4)[NH=C(NR(2))OC(NR(2))=NH]] (11-13) due to the unprecedented hydrolytic coupling of the two adjacent dialkylcyanamide ligands giving a novel (for both coordination and organic chemistry) diimino linkage. Compounds 11-13 and also 14 (R(2) = C(4)H(8)O) were alternatively obtained by the reaction between cis-[PtCl(4)(MeCN)(2)] and neat undried NCNR(2). The structures of complexes 11, 13, and 14 were determined by X-ray single-crystal diffraction. All the platinum compounds were additionally characterized by elemental analyses, FAB mass-spectrometry, and IR and (1)H and (13)C[(1)H] NMR spectroscopies.  相似文献

The novel dinuclear Pt(II) complexes [{trans-Pt(NH(3))(2)Cl}(2)(μ-pyrazine)](ClO(4))(2) (Pt1), [{trans-Pt(NH(3))(2)Cl}(2)(μ-4,4'-bipyridyl)](ClO(4))(2)·DMF (Pt2), and [{trans-Pt(NH(3))(2)Cl}(2)(μ-1,2-bis(4-pyridyl)ethane)](ClO(4))(2) (Pt3), were synthesized. Acid-base titrations, and temperature and concentration dependent kinetic measurements of the reactions with biologically relevant ligands such as thiourea (Tu), glutathione (GSH) and guanosine-5'-monophosphate (5'-GMP) were studied at pH 2.5 and 7.2. The reactions were followed under pseudo-first-order conditions by stopped-flow and UV-vis spectrophotometry. (1)H NMR spectroscopy was used to follow the substitution of chloride in the complex [{trans-Pt(NH(3))(2)Cl}(2)(μ-4,4'-bipyridyl)](ClO(4))(2)·DMF by guanosine-5'-monophosphate (5'-GMP) under second-order conditions. The results indicate that the bridging ligand has an influence on the reactivity of the complexes towards nucleophiles. The order of reactivity of the investigated complexes is Pt1 > Pt2 > Pt3.  相似文献

The ion flotation of rhodium(III) and palladium(II) with some anionic surfactants has been investigated. Two flotation procedures are proposed for the separation of some platinum metals, based on differences in the kinetic properties of the chloro-complexes of rhodium(III), palladium(II) and platinum(IV). The first involves the selective flotation of Rh(H(2)O)(3+)(6) from PdCl(2-)(4) and PtCl(2-)(6) in dilute hydrochloric acid with sodium dodecylbenzenesulfonate (SDBS). After precipitation of the hydroxide and redissolution in dilute acid, the Rh(III) is converted into Rh(H(2)O)(3+)(6), Pd(II) and Pt(IV) remaining as PdCl(2-)(4) and PtCl(2-)(6) respectively, and separation is achieved by floating the Rh(H(2)O)(3+)(6) with SDBS. The second is for separation of Pd(II). Prior to flotation, the solution of PdCl(2-)(4) and PtCl(2-)(6) is heated with ammonium acetate to convert PdCl(2-)(4) into Pd(NH(3))(2+)(4). The chloro-complex of Pt(IV) is unaffected. The complex cation, Pd(NH(3))(2+)(4), is then selectively floated with SDBS. The procedures are fast, simple and do not require expensive reagents and apparatus.  相似文献
