Phytochemical studies on Galanthus elwesii resulted in the isolation of five alkaloids: incartine, hordenine, hippeastrine, 8-O-demethylhomolycorine and lycorine. The NMR data given previously for incartine were revised and completed by two-dimensional 1H-1H and 1H-13C chemical shift correlation experiments. In vitro studies on the bioactivity of incartine were carried out.  相似文献

A liquid chromatographic atmospheric-pressure ionization electrospray mass spectrometry (LC–API–ES–MS) method has been developed for the determination of five bitter orange alkaloids (synephrine, octopamine, n-methyltyramine, tyramine, and hordenine) in bitter orange-containing dietary supplement standard reference materials (SRMs). The materials represent a variety of natural, extracted, and processed sample matrices. Two extraction techniques were evaluated: pressurized-fluid extraction (PFE) and sonication extraction. The influence of different solvents, extraction temperatures, and pH were investigated for a plant material and a processed sample. The LC method uses a new approach for the separation of highly polar alkaloids. A fluorinated, silica-based stationary phase separated the five alkaloids and the internal standard terbutaline in less than 20 min. This method enabled the determination of the dominant alkaloid synephrine and other minor alkaloids in a variety of dietary supplement SRMs.  相似文献

Leaves of Zanthozvlumprocerum (Rutaceae) yielded two major optically inactive alkaloids, culantraramine and culantraraminol, which were assigned bishordeninyl terpene structures based upon spectral evidence and conversions to known alkaloids. Two minor alkaloids, isomeric with culantraraminol, were also found, along with hordenine and N,N-dimethyltryptamine. Although culantraramine could be viewed as a natural self Diels-Alder adduct of dehydropronylhordenine, when this diene was prepared and reacted, it yielded instead an alternate adduct. This room temperature Diels-Alder reaction does, however, represent a model for the biosynthesis of some other known isolates considered as natural Diels-Alder adducts. A total synthesis of culantraramine and culantraminol was instead achieved in high yield from a prenylalcohol precursor under mild acid conditions. The synthetic reaction also yielded the two minor culantraraminol isomers. All four alkaloids were present in similar amounts in both the synthetic mixture and the crude leaf extract. The reaction used in the synthesis of culantraramine and culantraraminol is suggested to be biomimetic for these and certain other dimeric alkaloids.  相似文献

A suite of three dietary supplement standard reference materials (SRMs) containing bitter orange has been developed, and the levels of five alkaloids and caffeine have been measured by multiple analytical methods. Synephrine, octopamine, tyramine, N-methyltyramine, hordenine, total alkaloids, and caffeine were determined by as many as six analytical methods, with measurements performed at the National Institute of Standards and Technology and at two collaborating laboratories. The methods offer substantial independence, with two types of extractions, two separation methods, and four detection methods. Excellent agreement was obtained among the measurements, with data reproducibility for most methods and analytes better than 5% relative standard deviation. The bitter-orange-containing dietary supplement SRMs are intended primarily for use as measurement controls and for use in the development and validation of analytical methods. Citrus aurantium (bitter orange) is a plant belonging to the family Rutaceae, whose fruit extracts have been used recently for the treatment of obesity. The most important biologically active constituents of the C. aurantium fruits are phenethylamine alkaloids (i.e. octopamine, synephrine, tyramine, N-methyltyramine and hordenine). Synephrine is a primary synthesis compound with pharmacological activities such as vasoconstriction, elevation of blood pressure and relaxation of bronchial muscle. Synephrine is present in the peel and the edible part of Citrus fruit. Of the adrenergic amines of natural origin, synephrine has been found to be the main constituent of C. aurantium fruits and extracts; the other alkaloids are either absent or present in only low concentrations. It is known that synephrine and the other amines found in C. aurantium have adverse effects on the cardiovascular system, owing to adrenergic stimulation. In light of the great commercial proliferation of C. aurantium herbal medicines in recent years, this review provides an overview of various extraction, separation and detection techniques employed for the qualitative and quantitative determination of the alkaloids in C. aurantium and related species. The application of chromatographic and electrophoretic methods for the separation and determination of these active components in C. aurantium plant material and derivatives are described. Since synephrine is a chiral compound, enantioselective chromatographic and electrophoretic techniques for the analysis of synephrine enantiomers in natural products are presented. Furthermore, examples of identification of these active compounds in complex matrices by hyphenated methods, such as gas chromatography-mass spectrometry and high-performance liquid chromatography-mass spectrometry, are described. The advantages and limitations of these separation and identification methods are assessed and discussed.  相似文献

A new alkaloid, named alfileramine, has been isolated from Zanthoxylum punctatum. It represents the first alkaloid which has a structure closely related to the tetrahydrocannabinols. The structure was assigned based upon 1H and 13C NMR, UV and mass spectroscopy and by relating alfileramine structure to that of the rearranged dihydrobromide salt. The structure of the dihydrobromide was known from X-ray diffraction. A possible biosynthetic pathway from a dehydrocitral equivalent and two molecules of hordenine is suggested.  相似文献

Hordenine is an active compound found in several foods, herbs and beer. In this work, a novel sorbent was fabricated for selective solid‐phase extraction (SPE) of hordenine in biological samples. The organic polymer sorbent was synthesized in one step in the plastic barrel of a syringe by a pre‐polymerization solution consisting of methacrylic acid (MAA), 4‐vinylphenylboronic acid (VB) and ethylene glycol dimethacrylate (EGDMA). The conditions for preparation were optimized to generate a poly(MAA‐VB‐EGMDA) monolith with good permeability. The monolith exhibited good enrichment efficiency towards hordenine. By using tyramine as the internal standard, a poly(MAA‐VB‐EGMDA)‐based SPE‐HPLC method was established for analysis of hordenine. Conditions for SPE, including volume of eluting solvent, pH of sample solution, sampling rate and sample volume, were optimized. The proposed SPE‐HPLC method presented good linearity (R2 = 0.9992) within 10–2000 ng/mL and the detection limits was 3 ng/mL, which is significantly more sensitive than reported methods. The method was also applied in plasma and urine samples; good capability of removing matrices was observed, while hordenine in low content was well extracted and enriched. The recoveries were from 90.6 to 94.7% and from 89.3 to 91.5% for the spiked plasma and urine samples, respectively, with the relative standard deviations <4.7%. Two new alkaloids (1, 2) were isolated from the whole plants of Crinum asiaticum var. sinicum together with seven known alkaloids. The structures of the new alkaloids were elucidated by spectroscopic analyses and chemical conversions from known alkaloids. New alkaloid 1 was isolated for the first time as a natural product, although it has been prepared as a synthetic product.  相似文献

Three new pavine N-oxide alkaloids, (-)-isocaryachine-N-oxide B, (+)-caryachine-N-oxide, (-)-caryachine-N-oxide, and a new isoquinoline alkaloid, 6,7-methylenedioxy-N-methylisoquinoline together with 11 known alkaloids were isolated and characterized from the stem bark of Cryptocarya chinensis. The structures of the isolated compounds were determined by spectral methods. The stereochemistry of pavine-N-oxide alkaloids is also discussed.  相似文献

A method has been developed to quantify synephrine in bitter orange raw material, extracts, and dietary supplements. Single-laboratory validation has been performed on the method to determine the repeatability, accuracy, selectivity, limit of detection/limit of quantification (LOQ), ruggedness, and linearity for p-synephrine and 5 other biogenic amines: octopamine, phenylephrine (m-synephrine), tyramine, N-methyltyramine, and hordenine, which may be present in bitter orange. p-Synephrine was found to be the primary biogenic amine present in all materials tested, accounting for >80% of the total biogenic amine content in all samples except a finished product. Repeatability precision for synephrine was between 1.48 and 3.55% RSD. Synephrine recovery was between 97.5 and 104%. The minor alkaloids were typically near the LOQ of the method (300-900 microg/g) in the test materials, and between-day precision for the minor compounds was poor because interferences could sometimes be mistakenly identified as one of the minor analytes. Recoveries of the minor components ranged from 99.1 to 103% at approximately 6000 microg/g spike level, to 90.7 to 120% at 300 microg/g spike level.  相似文献

A range of seventeen quinoline alkaloids, involving several types of oxidations during their biosynthetic pathways, have been isolated from leaves of Choisya ternata. In addition to the nine known quinoline alkaloids, eight new members of the furoquinoline family, derived mainly from prenylation at C-5 (including two novel hydroperoxides), have been identified. The absolute configurations and enantiopurity values of all chiral quinoline alkaloids have been determined. One of the isolated alkaloids, 7-isopentenyloxy-gamma-fagarine, has been used as a precursor for the chemical asymmetric synthesis of the enantiopure alkaloids: evoxine, anhydroevoxine and evodine. The possible roles of oxygenase and other oxygen-atom-transfer enzymes, in the biosynthetic pathways of the C. ternata alkaloids, have been discussed.  相似文献

From the leaves and twigs of Tabernaemontana eglandulosa 22 alkaloids, of which 12 are new, have been isolated. Besides tacamine (pseudo-vincamine) 7 other alkaloids of this new class of indole alkaloids were identified. A further three alkaloids of the cleavamine type, namely the known ( + )20R-15, 20-dihydro-cleavamine and (-)20S-15, 20-dihydro-cleavamine and one new alkaloid, which was tentatively identified as 14S, 20R-velbanamine, two new alkaloids of the pseudo-aspidospermidine type, namely 20R-l,2-dehydro-pseudo-aspidospermidine and 20S-hydroxy-l,2-dehydro-pseudoaspidospermidine and an alkaloid of a novel iboga type were isolated. More common alkaloids isolated were coronaridine, 11-hydroxycoronaridine, voaphylline, tubotaiwine, ibogamine, norfluorocurarine and probably a mixture of 20R- and 20S-pseudo-vincadifformine. Only quantitative differences in the alkaloid content of the leaves, twigs and stembark were observed.  相似文献

Two new monoterpenoid indole alkaloids, tabervarines A (1) and B (2), along with seven known monoterpenoid indole alkaloids, were isolated from the methanol extract of the twigs and leaves of Tabernaemontana divaricata. The structures including the absolute configurations of the new alkaloids were elucidated based on MS, NMR, and ECD calculation. The in vitro cytotoxic activities of the isolated alkaloids against several human cancer cell lines were also evaluated.  相似文献

Ten diterpenoid alkaloids, including eight aconitine-type C??-diterpenoid alkaloids and two hetisine-type C??-diterpenoid alkaloids, were isolated from the secondary roots of Aconitum carmichaeli Debx., known as "Fuzi" in Chinese traditional herbal medicine. Their structures were established on the basis of their spectroscopic data and comparison with those of the literature. Among these alkaloids, chasmanine, oxonitine and 15-acetylsongoramine were isolated for the first time from this medicinal plant. The cytotoxic activity of the alkaloids were tested against several cell lines by the MTT method in which aconitine, hypaconitine, mesaconitne and oxonitine were found to strongly inhibit the growth of the HePG2 cell line, which showed that the existence and quantity of the ester groups have a significant influence on the cytotoxicity of the diterpenoid alkaloids.  相似文献

Four new and five known norditerpenoid alkaloids were isolated from the processed tubers of Aconitum carmichaeli. The new alkaloids are 14-O-cinnamoylneoline (3), 14-O-anisoylneoline (4) 14-O-veratroylneoline (5), and lipo-14-O-anisoylbikhaconine (8). The known alkaloids are neoline (1), 14-O-acetylneoline (2), foresaconitine (6), crassicauline A (7), and lipohypaconitine (9). Alkaloids 2, 6, and 7 were isolated from this plant for the first time. The structures of the new alkaloids were established by spectroscopic and chemical methods.  相似文献

Five minor alkaloids were isolated from the seeds of Camptotheca acuminata Decne. Two of them are new indole alkaloids named camptacumotine (1) and camptacumanine (2) respectively.
