The review focuses on the analysis of small phosphorus-containing compounds by capillary electrophoresis (CE) with different detection modes including UV absorption, laser-induced fluorescence, conductometry, amperometry, atomic spectrometry, and mass spectrometry. Determinations of phosphates, organophosphate, and chemical warfare agents in environmental samples such as water, soil and grains are emphasized. To achieve better sensitivity, high-resolving power, and reproducibility, the optimum separation conditions for various analytes (samples) are different. We compare the merits and demerits of the different detection modes for the detection of the analytes. Although the present methods are successful in many cases, there is still a need to develop high-throughput CE techniques for screening numerous environmental samples and sample stacking techniques in CE for the analysis of trace analytes.  相似文献

In the first part of this work, the use of capillary electrophoresis (CE) for the separation of two groups of pharmaceuticals, namely a metabolite of tamoxifen and a basic drug substance, DS1, was investigated. The effects of pH and types of modifiers, e.g. surfactant, bile salt, γ-cyclodextrin and hydroxypropyl-β-cyclodextrin on selectivity, separation and peak shape were studied. Besides achieving complete separation of the compounds, the CE system was capable of providing separation with significant improvements in overall peak shape of the compounds compared with HPLC. In the case of the basic drug substance DS1, validation of the CE system developed in terms of linearity, selectivity, sensitivity and reproducibility was satisfactorily performed. At the same time, a study of the sample solvent matrix effects on the separation of this group of compounds was examined. The system was successfully applied to the analysis of laboratory-synthesized samples. Good correlation was observed between CE and HPLC, although higher efficiency and faster speed of separation were obtained using the CE system developed. For the tamoxifen metabolite, special emphasis was placed on the use of CE for the separation of the pair of isomers. This was readily achieved through the introduction of γ-cyclodextrin in the electrolyte. Resolution of at least 1.5 was obtained for the isomers using the CE method.  相似文献

A capillary electrophoretic assay for determining aloe-emodin (AE), methyl p-coumarate (MC), and 3,4-dihydro-6,8-dihydroxyl-[(3s)-2'-acetyl-3'-hydroxyl-5'-methoxy-benzyl]-isocoumarin (DDI) in Aloe vera has been developed. Baseline separation was achieved within 15 min using a running buffer of 20 mm borax containing 10% (v/v) acetonitrile at pH 10.5. A linear relationship between the peak area and the concentration of the analytes was found in the ranges 5-500, 10-1000 and 2-1000 micro g/mL for AE, MC and DDI, respectively, with correlation coefficients of 0.9992-0.9998. The relative standard deviations of migration time and peak area were within 0.17-0.19 and 1.52-3.37%, respectively. The recoveries of AE, MC and DDI were 105, 102 and 96.4%. The contents of AE, MC and DDI in Aloe vera were measured to be 5.13, 0.768 and 1.30 mg/g, respectively.  相似文献

Summary Thin layer chromatography on silica gel high performance layers and automated multiple development was used to separate the polar aromatic flavor compounds vanillin, ethyl vanillin, 4-hydroxybenzaldehyde, 4-hydroxybenzoic acid, 4-hydroxybenzyl alcohol, vanillic acid, coumarin, piperonal, anisic acid, and anisaldehyde commonly found in extracts of natural and artificial vanilla flavors. The ratio of 4-hydroxybenzoic acid, 4-hydroxybenzaldehyde and vanillic acid to vanillin in natural vanilla extracts was used to confirm the authenticity of extracts purchased in the United States of America and the United Kingdom. Natural vanilla extracts purchased in Mexico and Puerto Rico were identified as counterfeit products based on changes in the above ratio and the presence of synthetic flavor compounds such as ethyl vanillin and coumarin. It is also demonstrated that the proposed method is suitable for the determination of natural and synthetic vanilla flavors in solvent extracts from food, beverage and confectionery products. The main advantage of thin layer chromatography for the analysis of vanilla extracts and food stuffs flavored with vanilla is its high sample throughput since sample preparation requirements are minimal and multiple samples can be separated simultaneously.  相似文献

In this paper, the dissociation constants (pKa1, pKa2) of five anthraquinones were determined from the relation between the effective mobility at different pH values and the buffer pH value, which was derived from the basic electrophoresis theory and the dissociation equilibrium of a binary acid. In addition, the changes of pKa values of the five compounds were also investigated when organic modifiers were added to the buffer system.  相似文献

Summary A new method for analysing carbonyls (aldehydes and ketones) by capillary electrophoresis (CE) is described. The carbonyls are derivatized with DNSH [5-(dimethylamino)naphthalene-1-sulphonehydrazide, Dansylhydrazine] to form detectable compounds and also to form negative charged molecules (buffer pH 7.1). Detection is carried out by UV or, alternatively, by fluorescence. Eight carbonyls are separated in less than 8 min. Electrokinetic injection is made by using a new experimental set-up. The electroendosmatic flow (EOF) is reversed during sample injection by pressure. Research on the structure–activity relationships of molecules with acidic carbon atoms led us to undertake a feasibility study on the determination of their acidity constants by capillary electrophoresis (CE). The studied molecules had diverse structures and were tetronic acid, acetylacetone, diethylmalonate, Meldrums acid, 3-methylrhodanine, nitroacetic acid ethyl ester, pyrimidine-2,4,6-trione, 3-oxo-3-phenylpropionic acid ethyl ester, 1-phenylbutan-1,3-dione, 5,5-dimethylcyclohexan-1,3-dione and homophthalic anhydride. The pKa range explored by CE was therefore very large (from 3 to 12) and pKa values near 12 were evaluated by mathematical extrapolations. The analyses were carried out in CZE mode using a fused silica capillary grafted (or not) with hexadimethrine. Owing to the electrophoretic behaviour of these compounds according to the pH, their acidity constants could be evaluated and appeared in perfect agreement with the literature data obtained, a few decades ago, by means of potentiometry, spectrometry or conductimetry. The pKa of homophthalic anhydride and 3-methylrhodanine were evaluated for the first time.  相似文献

CZE has been applied for determination of acid-base dissociation constants (pKa) of ionogenic groups of newly synthesized amino- and (amino)guanidinopurine nucleotide analogs, such as acyclic nucleoside phosphonate, acyclic nucleoside phosphonate diesters and other related compounds. These compounds bear characteristic pharmacophores contained in various important biologically active substances, such as cytostatics and antivirals. The pKa values of ionogenic groups of the above compounds were determined by nonlinear regression analysis of the experimentally measured pH dependence of their effective electrophoretic mobilities. The effective mobilities were measured by CZE performed in series of BGEs in a broad pH range (3.50-11.25), at constant ionic strength (25 mM) and temperature (25 degrees C). pKa values were determined for the protonated guanidinyl group in (amino)guanidino 9-alkylpurines and in (amino)guanidinopurine nucleotide analogs, such as acyclic nucleoside phosphonates and acyclic nucleoside phosphonate diesters, for phosphonic acid to the second dissociation degree (-2) in acyclic nucleoside phosphonates of amino and (amino)guanidino 9-alkylpurines, and for protonated nitrogen in position 1 (N1) of purine moiety in acyclic nucleoside phosphonates of amino 9-alkylpurines. Thermodynamic pKa of protonated guanidinyl group was estimated to be in the range of 7.75-10.32, pKa of phosphonic acid to the second dissociation degree achieved values of 6.64-7.46, and pKa of protonated nitrogen in position 1 of purine was in the range of 4.13-4.89, depending on the structure of the analyzed compounds.  相似文献

建立了同时测定香兰素和其异构体邻位香兰素的毛细管区带电泳法(CZE)。考察了缓冲溶液的种类、浓度和pH以及分离电压等因素对分离结果的影响。在缓冲溶液为50 mmol/L硼砂-150 mmol/L磷酸氢二钠(pH 7.5)、分离电压15 kV的优化条件下,6 min内即可实现分离。香兰素和邻位香兰素在10~240 mg/L范围内线性关系良好,相关系数分别为0.9999和0.9997;方法的检出限均为1.0 mg/L (信噪比为3);样品的加标回收率为99.4%~101.2%,相对标准偏差为0.19%~0.73%。该方法操作简单、快速,已应用于实际样品的分析,并获得了令人满意的结果。  相似文献

Capillary electrophoresis has been increasingly used during the past few years for the separation and determination of surfactants. These substances are applied in many household and industrial products such as laundry detergents, cosmetics and pharmaceuticals, often as homologous and isomeric mixtures. Product development and control as well as toxicological and environmental analyses require selective and sensitive analytical methods. This review presents capillary electrophoretic techniques to determine important representatives of cationic, anionic, and neutral surfactants. The application of different buffer additives such as organic solvents, cyclodextrins or micelles to enhance the resolution of complex mixtures is discussed. Besides direct and indirect UV and fluorescence detection, examples for conductivity and mass spectrometric detection are also given. Derivatization procedures to improve the detectability and implement charge in neutral analytes are described. The successful use of capillary electrophoresis for surfactant determinations has proven that it can serve as a routine technique in many real-world applications. Robust, validated methods for the quantitation of single compounds, such as alkylbenzene sulfonates, sodium dodecyl sulfate and benzalkonium salts, are now available. Characteristic peak patterns (fingerprint analysis) can be used for the identification of surfactants in multicomponent formulations (e.g. ethoxylates and phosphonates).  相似文献

The electrophoretic behavior of polyhexamethyleneguanidine hydrochloride (PHMG HC), hexamethylenediamine (HMDA), and guanidine hydrochloride (GHC) was studied. It was shown that PHMG HC and the initial toxic components of its synthesis, GMDA and GHC, can be separated. A procedure was developed for determining PHMG HC oligomers, GMDA, and GHC in aqueous solutions in concentrations from 0.007 to 0.05 mg/mL by capillary electrophoresis. The procedure was applied to the analysis of model mixtures of these compounds.  相似文献

A capillary zone electrophoresis method is presented to separate sulfadiazine, sulfamethoxazole, trimethoprim, bromhexine and guaiacol by using a fused-silica capillary (60.2 cm x 75 microm I.D.). The separation was carried out at 30 kV and 25 degrees C in a 15 mM phosphate buffer adjusted to pH 6.2 as electrolyte. Under these conditions, the run time was 6 min and the limits of quantification were about 1 mg/l for every component. The method was applied to pharmaceutical preparations and the results provided recoveries close to 100%.  相似文献

The determination of quaternary ammonium ions by capillary electrophoresis (CE) is reviewed. The analytes include tetraalkylammonium and alkylbenzyldimethylammonium compounds frequently used as antiseptic and antibacterial agents as well as in various household products, several plant growth regulators and herbicides, by-products in bile acid sequestrants, and a range of anticholinergic drugs. Besides direct and indirect UV detection, hyphenation with electrospray mass spectrometry is particularly suited for quaternary ammonium ions and may lower the detection limits by two orders of magnitude. In comparison with established liquid chromatographic techniques, CE may exhibits superior separation efficiency. Applications in routine analysis have demonstrated that CE is reliable and robust enough to represent a real alternative to chromatography.  相似文献

建立了毛细管区带电泳法同时测定注射用头孢哌酮钠他唑巴坦钠中目标成分含量的方法。采用石英毛细管(75 cm×75 μm i.d.,有效长度63 cm),以40 mmol/L硼砂溶液为背景电解质(BGE),以氢氯噻嗪为内标,分离电压12.0 kV,检测波长220 nm,重力进样10 s(高度10 cm)。实验结果表明,头孢哌酮钠与他唑巴坦钠分别在0.25~3.96 g/L和0.062~0.99 g/L范围内呈良好的线性关系,相关系数(r)分别为0.9995和0.9996。精密度和重复性均良好,相对峰面积的相对标准偏差(RSD)均小于3%,制剂在208 min内稳定,回收试验结果符合方法学要求。该方法简单、准确、耗费少、污染小,可用于注射用粉针剂头孢哌酮钠他唑巴坦钠中头孢哌酮钠和他唑巴坦钠的定量分析。  相似文献

Doripenem, the latest carbapenem antibiotic licensed in the United States (15 October 2007) and the European Union (25 July 2008), has been implemented into therapeutic use along with imipenem, meropenem and ertapenem. The described method of zone electrophoresis in a low pH buffer for the separation of doripenem from its impurities has been successfully performed using field‐amplified sample stacking (FASS), followed by UV absorption detection at 214 nm. The best results were obtained with phosphate buffer (100 mM) pH 2.9 containing 10% (v/v) of methanol, as the background electrolyte. Uncoated fused‐silica capillary (60/52 cm; 75 μm id) with normal polarity, and voltage values of 25 kV, was used throughout the investigation. The optimised method of doripenem determination was validated in terms of linearity, accuracy and precision, and provides a detection limit of 3.0 μg/mL of doripenem. The repeatability, expressed by relative standard deviation (RSD) of the migration time, for doripenem and its degradation products varied from 1.37 to 2.51%, whereas the corrected peak areas were about 0.91–9.87%. Satisfactory separation was achieved within 20 min of electrophoresis; moreover, all carbapenems (imipenem, meropenem, ertapenem and doripenem) were well separated from each other during this time. The evaluated CZE method was applied in the analysis of a medicinal product containing doripenem Doribax® powder for solution for infusion.  相似文献

Additives included in the review are preservatives, antioxidants, sweeteners, colourings, caffeine, niacin, choline, nitrate, and nitrite. An improved capillary electrophoresis assay for histamine in crude extracts was developed and used to determine histamine levels in a selection of tomato fruits and pastes. Performance in terms of reproducibility and sensitivity was optimized by use of a high sensitivity detector flow cell, sodium hydroxide rinses, and a voltage gradient. The method was linear down to 0.2 microgram/mL (signal-to-noise ratio = 4:1), which was below the endogenous level in all samples.  相似文献
