Summary The Claisen rearrangement of 7-(3-phenyl-2-propenyloxy)-3-phenyl-(4H)-1-benzopyran-4-one (2 a) gave 7-hydroxy-8-(1-phenyl-2-propenyl)-3-phenyl-(4H)-1-benzopyran-4-one (3 a) and 2,3-dihydro-2,6-diphenyl-3-methyl-(7H)furo[2,3-h]-1-benzopyran-7-one (7 a). 2-Methyl-7-(3-phenyl-2-propenyloxy)-3-phenyl-(4H)-1-benzopyran-4-one (2 b) afforded4 b and7 b. 8-Methyl-7-(3-phenyl-2-propenyloxy)-3-phenyl-(4H)-1-benzopyran-4-one (12) gave only the alkali soluble product 7-hydroxy-8-methyl-6-(1-phenyl-2-propenyl)-3-phenyl-(4H)-1-benzopyran-4-one (13).3 a,4 b, and13 were further cyclized in acidic medium to9 a,10 b, and14 followed by dehydrogenation.This paper is dedicated to Dr. F. The synthesis of some substituted 4-hydroxy-2,5,6,7-tetrahydro-pyrano[2,3-c]pyrrole-2,5-diones (5) and 4-hydroxy-1,2,6,7-tetrahydro-5H-pyrrolo[3,4-b]pyridine-2,5-diones (6) by reacting 1,5-diaryl-pyrrolidine-2,4-diones (1) and 1,5-diaryl-1,5-dihydro-4-amino-2H-pyrrol-2-ones (3) with bis-2,4,6-trichlorophenyl malonates (4) is described.Dedicated to Prof. Dr. Dr. h. c.O. The reactions of 1-substituted 2-nitro-3-phenylaminoprop-2-en-1-ones with cyanothioacetamide afforded the corresponding 6-substituted 3-cyano-5-nitropyridine-2(1H)-thiones, which were used for the synthesis of 6-substituted 3-cyano-2-methylthio-5-nitropyridines and 7-substituted 4-hydroxy-8-nitropyrido[2",3":4,5]thieno[2,3-b]pyridin-2(1H)-ones.  相似文献

3-(N",N",S-trialkylisothioureido)quinazolin-4(3H)-ones obtained by the reactions of 3-(N",N"-dialkylthioureido)quinazolin-4(3H)-ones with alkyl halides undergo unusual recyclization into 5-(2-aminophenyl)-2-dialkylamino-1,3,4-oxadiazoles under the action of aqueous solutions of alkali, hydrazine, and primary aliphatic amines. A plausible mechanism of the recyclization was proposed.  相似文献

A new method for the synthesis of N-substituted 2-(4-oxo-4,5,6,7-tetrahydroindol-3-yl)acetic acids was developed. Alkylation of cyclic 1,3-diketones with 3,5,5,5-tetrachloropentan-2-one affords 1,4-diketones, which undergo cyclization with different primary amines into N-substituted 3-(2,2,2-trichloroethyl)-4,5,6,7-tetrahydroindol-4-ones. Acid hydrolysis of the latter gives the corresponding indol-3-ylacetic acids. The structures of the compounds obtained were confirmed by 1H and 13C NMR data.  相似文献

Summary By condensation of 2,2-difluoro-4-methyl-benzo[d]-1,3,2-2H-dioxaborines (12) with cyano acetic acid derivatives in presence of weak bases, 3-cyano-4-methyl-benzo[b]pyran-2-ones (13) or their 3-cyano-4-methyl-benzo[b]pyran-2-imine precursors (14) are available in satisfactory yields.

The reaction of 5-aryl-2,3-dihydrofuran-2,3-diones with cyanoacetylhydrazide, o-(hydrazinocarbonyl)phenylthiourea, and the diphenylhydrazone of diaminoglyoxal leads to the synthesis of the corresponding N-cyanoacetylhydrazides of aroylpyruvic acids, 2-(N-aroylpyruvoylhydrazinocarbonyl)-phenylthioureas, and 5,6-bis(phenylhydrazono)-3-aroylmethylenepiperazin-2-ones. The results of the primary investigation of the biological activity of N-cyanoacetylhydrazides of aroylpyruvic acids are given.  相似文献

The reactions of the monosubstituted guanidines2 b-h with methyl acrylate in dimethylformamide or ethanol as solvent preferentially afford 1-substituted 2-amino-5,6-dihydro-4(1H)-pyrimidinones6 b-h. The structures of 1-hexyl- and 1-benzyl-4-pyrimidinones6 c, e and of the picrate of 1-phenylpyrimidinone6 g were proved by comparison with authentic samples, which were prepared from N-substituted ethyl 3-amino-propionates14 c, e andg and cyanamide. Accordingly,6 g is not identical with authentic 2-phenylaminopyrimidinone7 g (prepared from 2-methylthio-4-pyrimidinone10 and 2-thioxo-4-pyrimidinone12 respectively, compare10).N,N-Disubstituted guanidines2 i-m react with methyl acrylate in dimethylformamide as solvent to afford N2,N2-disubstituted 2-amino-5,6-dihydro-4-(1H)-pyrimidinones7 i-m. Action of morpholine-4-carboxamidine (21) on methyl acrylate in ethanol yields 2-morpholinopyrimidinone71 as byproduct and 3-ethoxy-N-[morpholino(amino)methylene]propionamide (91) as mainproduct.

Substituted 2-amino-4-aryl-3-cyano-5-oxo-5,6-dihydro-4H-pyrano[2,3-d]pyrido[3",2":4,5]thieno[3,2-b]pyridines were synthesized by the reactions of 4-hydroxy-1H-thieno[2,3-b;4,5-b]dipyridin-2-ones with arylidenemalononitriles or by the three-component reactions of hydroxythienodipyridinones with aldehydes and malononitrile in DMF in the presence of triethylamine. Methods for syntheses of substituted 3-alkoxycarbonyl-6-amino-4-aryl-2-(3-cyanopyridin-2-ylthiomethyl)-4H-pyrans were developed on the basis of the reactions of 4-(3-cyanopyridin-2-ylthio)acetoacetates and arylidenemalononitriles or aldehydes and malononitrile. Ethyl 4-(3-cyanopyridin-2-ylthio)acetoacetate and 4-methoxybenzylidenecyanothioacetamide were used for the synthesis of 6-(pyridin-2-ylthiomethyl)-3-cyanopyridine-2(1H)-thione.  相似文献

An efficient and novel synthesis of 2,3-disubstituted 2,3-dihydroquinazolin-4(1H)-ones via one-pot, three-component reaction of isatoic anhydride, primary amines and aromatic aldehydes catalyzed by Bi(NO3)3·5H2O under solvent-free conditions is described. Oxidation of these 2,3-dihydroquinazolin-4(1H)-ones to their quinazolin-4(3H)-ones was also successfully performed in the presence of Bi(NO3)3·5H2O. This new method has the advantages of convenient manipulation, short reaction times, excellent yields, very easy work-up, and the use of commercially available, low cost and relatively non-toxic catalyst. The role of Bi(NO3)3·5H2O was also investigated in these transformations.  相似文献

The reaction of 3-phenyl-1,2,4-triazin-5(4H)-one (1) with l-menthol in the presence of aliphatic acid anhydrides results in (6S)- and (6R)-1-acyl-6-(l-menth-3-yl)-1,6-dyhydro-3-phenyl-1,2,4-triazin-5(4H)-ones. The reaction is diastereoselective with predominant formation of (6S)-isomers. The reaction diastereoselectivity increases with enhancement of the steric hindrance in the vicinity of the reaction center of the azine.  相似文献

Summary 6-Phenylimino-3,6-dihydro-2H-thiopyran-4-amines (1) were converted to 1-phenyl-5,6-dihydropyridine-2(1H)-thiones (3). Those were akylated and hydrolyzed, thus yielding 6-methylthio-1-phenyl-2,3-dihydro-4(1H)-pyridinones (5). Finally, the methylthio group was removed withRaney nickel giving the title compounds6. The relative configurations of the formed diastereoisomeric dihydropyridinones have been investigated by NOE measurements.

Zusammenfassung 6-Phenylimino-3,6-dihydro-2H-thiopyran-4-amine (1) wurden in 1-Phenyl-5,6-dihydropyridin-2(1H)-thione (3) umgewandelt. Diese wurden alkyliert und zu 6-Methylthio-1-phenyl-2,3-dihydro-4(1H)-pyridinonen (5) hydrolysiert. Zuletzt gelangte man durch selektives Entfernen der Methylthiogruppe mitRaney-Nickel zu den Titelverbindungen6. Die relativen Konfigurationen der gebildeten diastereomeren Dihydropyridinone wurden durch NOE-Messungen aufgeklärt.

Summary 4-Phosphoranylidene-5(4H)-oxazolones (2), a hardly known class of phosphorus ylides, were readily prepared from 4-unsubstituted-5-(4H)-oxazolones (1) by treatment with Ph3P-Br2, Bu3P-Br2, Ph3P-CCl4, or Ph3P-CBr4 adducts in the presence of triethylamine in CH2Cl2 at room temperature in a novel, efficient one-pot procedure. The spectroscopic properties of the ylides are reported and discussed.
4-Phosphoranyliden-5(4H)-oxazolone — Eine neue Synthese und Eigenschaften
Zusammenfassung 4-Phosphoranyliden-5(4H)-oxazolone, eine sehr wenig bekannte Gruppe der Phosphor-ylide, wurden auf einfache Weise nach einem neuen Eintopfverfahren mit guten Ausbeuten hergestellt. Als Ausgangsverbindungen wurden 4-unsubtituierte-5-(4H)-Oxazolone (1) eingesetzt, die unter der Einwirkung von Addukten wie Ph3P-Br2, Bu3P-Br2, Ph3P-CCl4 oder Ph3P-CBr4 in Anwesenheit von Triethylamin in CH2Cl2 bei Zimmertemperatur die Titelverbindungen liefern. Die spektroskopischen Eigenschaften der Ylide werden berichtet und diskutiert.

5-Alkyl(aryl)-3-arylidene-3H-pyrrolin-2-ones were synthesized by ammonolysis of their O-heteroanalogs or by the reactions of 5-alkyl(aryl)-3H-pyrrolin-2-ones with aromatic aldehydes. The structures of the compounds obtained were confirmed by 1H NMR spectra.  相似文献

Summary 3(Bis(alkylthio)methylene)-3H-isobenzofuran-1-ones2a–e and 2-(bis(alkylthio)methylene)-3(2H)-benzofuranone derivatives4a–c are obtained by reaction of phthalides1a–d or 3(2H)-benzofuranone (coumaranone3), respectively, with carbon disulfide under basic conditions followed by alkylation. The reaction behaviour of the new compounds2 and4 is investigated. 2-((2-Dimethylthio-1-oxo)ethyl)benzoic acid N,N-dimethylamide (7a) and 2-((2-dimethylthio-1-oxo)ethyl)-benzoic acid 2-methylpiperidide (7b) are surprisingly formed instead of the methylthio substitution products by treatment of2a with the corresponding amine in the presence of aluminum chloride.

4-Alkylamino-2(1H)-pyridinethiones (1) react with bis-trichlorphenylethylmalonate (3) to give four isomeric products: 5-thioxo-1,6-naphthyridine-2(1H)-ones (4), 7,7-dimethyl-4H-thiopyrano[2,3-b]pyridine-4-ones (6), 2,2-dimethyl-2H-thiopyrano[2,3-b]pyridine-5,7-dioles (7) and 2,2-dimethyl-2H-thiopyrano[2,3-b]pyridine-5(3H)-ones (8). On treatment with alkali the thioxogroup of4 is hydrolyzed and 1,6-naphthyridinediones (5) are formed. Compound5 can also be synthesized by heating the alkylaminopyridones (2) together with3.6 can be hydrolyzed to 2-thioxo-3-pyridylpropylketones (12). On treatment with diluted alkali or conc. acid the aminogroup of7 and8 is hydrolyzed and10 is formed. By heating in 20% NaOH10 is transformed to the dihydroxymercapto-3-pyridylmethylketone (11).

Summary (±)-4-Thiacarnegin (3) was synthesized by reaction of 6,7-dimethoxy-3-methyl-2H-1,3-benzothiazinium iodide (2) with methyl magnesium iodide, thereby opening a new synthetic route to 4-substituted dihydro-2H-1,3-benzothiazines. Compound3 was also obtained by reduction of 6,7-dimethoxy-3,4-dimethyl-2H-1,3-benzothiazinium iodide (5). In a similar way, reduction of the quaternary salts9 a–c afforded the (±)-4-thia analogues of cryptostilin-I, -II and -III (10 a–c). The isomers of the former compounds (12 a–c) were also synthesized by reduction of the 4H-1,3-benzothiazinium salts11 a–c.

Zusammenfassung Aus 6,7-Dimethoxy-3-methyl-2H-1,3-benzothiaziniumjodid (2) wurde mit Methylmagnesiumjodid (±)-Thiacarnegin (3) dargestellt. Diese Reaktion bietet ein neues Verfahren für die Synthese von 4-substituierten Dihydro-2H-1,3-benzothiazinen.3 wurde auch durch Reduktion von 6,7-Dimethoxy-3,4-dimethyl-2H-1,3-benzothiaziniumjodid (5) erhalten. Die Reduktion der quartären Ammoniumsalze9 a–c ergab ebenfalls die Cryptostillin I-, II-, III-(±)-4-thiaanalogen Verbindungen (10 a–c). Reduktion der 4H-1,3-Benzothiazinium-Salze11 a–c lieferte die entsprechenden Isomeren12 a–c der obengenannten Verbindungen.

The C=O stretching frequencies of 32 5,6-polymethylenepyrimidin-4(3H)-one (1a–1w) andcis-5,6-polymethylene-5,6-dihydropyrimidin-4(3H)-one (2a–2h) derivatives were measured in tetrachlormethane and in chloroform and correlated with the substituent constantsX + (R) in the sense of the modified and extendedSeth-Paul-Van Duyse equation. It was found that the C=O stretching frequency and the transmission of substituent effects through the heterocyclic ring are significantly influenced by the size of the fused hydrocarbon ring. Evidence was obtained that in molecules linked by intermolecular hydrogen bonds the substituent effects are transmitted to the C=O group predominantly via the C=N–C=C part of the pyrimidinone ring. In the free molecules the transmission of substituent effects takes place mainly through the NH group.

Zusammenfassung Es wurden die C=O-Streck-Frequenzen von 32 5,6-Polymethylenpyrimidin-4(3H)-onen (1a–1w) undcis-5,6-polymethylen-5,6-dihydropyrimidin-4(3H)-onen (2a–2h) in CCl4 und CHCl3 gemessen und mit den SubstituentenkonstantenX + (R) im Sinn der modifizierten und erweitertenSeth-Paul-Van Duyse-Gleichung korreliert. Es wurde festgestellt, daß die C=O-Streck-Frequenzen und die Übertragung von Substituenteneffekten durch den heterocyclischen Ring von der Größe des kondensierten Kohlenwasserstoffringes signifikant beeinflußt werden. Es konnte belegt werden, daß in Molekülen, die mit Wasserstoffbrücken verbunden sind, die Substituenteneffekte vorwiegend über den C=N–C=C-Teil des Pyrimidinonringes zur C=O-Gruppe übertragen werden. In den freien Molekülen erfolgt die Übertragung der Substituenteneffekte hauptsächlich über die NH-Gruppe.
