The reaction of n , n '-diphosphonylketones 1 with active methylenenitriles and sulfur in basic conditions led to the new 5-amino-2-(phosphonomethyl)-3-(phosphonoethyl)thiophenes 2 . These compounds were then used in the Horner-Wadsworth-Emmons reaction to give 5-amino-2-alkenyl-3-(phosphonoethyl)thiophenes 3 . The structure of all obtained products was confirmed by NMR ( 1 H, 31 P, 13 C) and IR spectroscopy.  相似文献

3,4-Diamino-2,5-dicarbethoxythieno[2,3-b]thiophene ( 1 ) was allowed to react with NaNO 2 and active methylenes to afford the corresponding azo compounds 2a-c . Hydrazonyl chloride 2a was treated with carbon disulfide, phenyl isothiocyanate, benzonitrile, benzyl cyanide, malononitrile, benzalaniline, ethyl mercaptoacetate, and ethyl glycinate to give 1,3,4-thiadiazolyl-, s-triazolyl-, 1,3,4-thiadiazinyl-, 1,3,4-triazinylthieno[2,3-b]thiophenes 3-6 respectively. The reaction of 2b,c with urea, thiourea, and guanidine afforded pyrimidinyl- and thiazinylazothieno [2,3-b]thiophenes 7-10 respectively. Bithiazolylthieno[2,3-b]thiophenes 11 and 13 were synthesized by treating compound 1 with CS 2 along with halo compounds. The addition of S,S-, N,S-, and N,O-acetals to the Schiff base 14 afforded compounds 15-17 respectively.  相似文献

2-Menthyltrihalothiophene S,S-dioxides are efficient asymmetric dienes in Diels-Alder reactions with dienophiles to give regio- and stereoisomerically pure adducts. Thus styrene, allyl alcohol, methyl acrylate and acrylamide give solely one adduct while flat dienophiles such as indene and acenaphthylene give two diastereomers. Analogous 1- and 3- chirally substituted thiophenes are of little value in generating pure single adducts. Further transformations of the above adducts into useful enantiomerically pure compounds is briefly discussed.  相似文献

The reaction of 2-lithiobenzo[b]thiophenes with ketones produced 2-hydroxyalkylbenzo[b]thiophenes which were cyclized in thionyl chloride to give novel thieno[3,2-b][1]benzothiophenes. Oxidation with peroxyacetic acid gave the S,S-dioxides whose regiochemistry was identified from 1H nmr model studies using substituted benzo[b]thiophenes and their corresponding-1,1-dioxides [1].  相似文献

Pyrazoles were reacted with acylbromoacetylenes in solid Al2O3 at room temperature to afford 2,2-di(pyrazol-1-yl)enones in 22–69% yield. The reaction proceeds via isolable intermediates, (Z)-2-bromo-2-(pyrazol-1-yl)enones. This unexpected 2:1 coupling is in contrast to similar reactions of pyrroles, furans and thiophenes, which give the corresponding acylethynyl derivatives. This reaction opens a one-pot route to inaccessible gem-dipyrazolylenones, which have potential applications as bidentate chelating ligands and building blocks for drug design.  相似文献

A general and versatile route to the 2-alkyl-5-(4'-cyanophenyl)thiophenes was established, involving the condensation of an aryl vinyl ketone with an aldehyde in the presence of a thiazolium catalyst (Stetter procedure) to give a 1,4-diketone which with Lawesson's reagent undergoes ring-closure to give the corresponding arylthiophene. In the final step the exchange of a bromo substituent on the phenyl ring for a cyano group is accomplished by copper(I) cyanide in N,N-dimethylforamamide at reflux. The same sequence afforded 2-alkyl-5-(4'-cyanobiphenylyl)thiophenes. Several of the 1,4-diketones were obtained by conjugate addition of nitroalkanes to aryl vinyl ketones, and treatment of the γ-nitroketones with silica gel-supported potassium permanganate. Three alkyl 5-(4-cyanophenyl)thiophene-2-carboxylates were prepared by condensing β-chlorovinylaldehydes with thioglycolates. The cyanophenylthiophenes exhibited only monotropic phases, but incorporation of an additional phenyl ring provided cyanobiphenylylthiophenes of wide nematic ranges. Transition temperatures of binary mixtures of the cyanophenylthiophenes with 4-n-pentyl-4'-cyanobiphenyl were measured, and extrapolated virtual nematic-isotropic transition temperatures were determined.  相似文献

A facile and efficient one-pot synthesis of highly substituted thiophenes has been developed and employed for the preparation of a small focused library. Treatment of 1,3-dicarbonyl compounds 1 with CS2 in the presence of K2CO3 in DMF at room temperature, followed by stepwise addition of alkyl bromides 2 and methylene active bromides 3, provided via intramolecular cyclization 2,3,4,5-tetrasubstituted thiophenes 4 in yields of 77-94%. This protocol, combining construction and modification of the thiophene ring, increases the structural diversity of final products from readily available materials. A mechanism for the one-pot synthesis of thiophenes of type 4 has been proposed. A small focused library of thiophenes is prepared using the sequential addition of reagents to achieve unique substitution in the 2 and 5 position of the thiophene ring.  相似文献

The reactions of seven thiophenes with benzyne generated from diphenyliodonium-2-carboxylate (DPIC) under a standard set of conditions led among other products to the formation of alpha- and beta-naphthyl phenyl sulfides 2a and 2b from thiophene (1a) and of 2c and 2d from 2-methylthiophene (1b). Dithienyl sulfides 4a-f were produced from the halothiophenes 1c-g. The structures of the naphthyl sulfides were proven by comparison with authentic samples of 2a-f, thus eliminating one of two possible mechanisms of formation. The remaining mechanism involves [4+2]-cycloaddition of benzyne to thiophene or to an S-phenylthiophenium ylide 10 to give the dipolar 2:1 benzyne/thiophene adduct 8 followed by ring-opening. Stevens-like rearrangements of 11, formed from 10 by proton transfer, may also explain the origin of arylated thiophenes such as 12 and 3 found in some reactions of benzynes with thiophene.  相似文献

Electron impact mass spectra of alkyl 4-alkoxy-5-amino-3-methylthiophene-2-carboxylates were studied for the first time. These compounds, except for 4-(1-ethoxyethoxy) and 4-(ferrocenylmethoxy) derivatives, give rise to a stable molecular ion whose decomposition follows three pathways. The main fragmentation pathway of the molecular ion is elimination of alkyl radical from the 4-alkoxy group, the second pathway involves expulsion of alkoxy group from the ester moiety, and the third pathway is decomposition of the thiophene ring. The molecular ions of 4-(1-ethoxyethoxy)thiophenes decompose mainly via elimination of ethyl vinyl ether molecule with formation of [M–VinOEt]+ · odd-electron ion, and fragmentation of the latter follows general pathways. In the mass spectra of 4-(ferrocenylmethoxy)thiophenes the most abundant are ferrocenylmethyl ion with m/z 199 (I rel 100%) and fragment ions derived therefrom.  相似文献

Reaction of 1-(2-chloro-5-nitrophenyl)ethanone via Willgerodt-Kindler route with primary or secondary amines and sulfur allows a simple, efficient one-pot synthesis of 3-aminobenzo[b]thiophenes. Base-catalyzed transformation of 4-(2-chloro-5-nitrophenyl)-1,2,3-thiadiazole in the presence of primary and secondary amines offers a convenient approach towards 2-aminobenzo[b]thiophenes.  相似文献

Treatment of nucleophilic heterocycles like pyrroles and thiophenes, and their tetrahydro derivatives, with S2Cl2 and DABCO in chloroform at room temperature provides a simple one-pot synthesis of fused mono and bispentathiepins. N-Methylpyrrole and its 2-chloro and 2,5-dichloro derivatives and N-methylpyrrolidine all give the same dichloropentathiepin 1a. N-Ethyl, isopropyl and tert-butylpyrrolidine behave similarly; the isopropylpyrrolidine also gives the bispentathiepin 6which undergoes an intriguing rearrangement to the symmetrical monopentathiepin 1c. N-Methyl and ethyl indole give either 2,3-dichloro derivatives 8 or the pentathiepinoindoles 9, depending upon the reaction conditions. Thiophene and tetrahydrothiophene give the pentathiepin 10. X-Ray crystal structures are provided for the pentathiepins 1a and 1d, and possible reaction pathways are suggested for the extensive cascade reactions reported.  相似文献

Iridium-catalyzed borylation has been applied to various substituted thiophenes to synthesize poly-functionalized thiophenes in good to excellent yields. Apart from common functionalities compatible with iridium-catalyzed borylations, additional functional group tolerance to acyl (COMe) and trimethylsilyl (TMS) groups was also observed. High regioselectivities were observed in borylation of 3- and 2,5-di-substituted thiophenes. Electrophilic aromatic C-H/C-Si bromination on thiophene boronate esters is shown to take place without breaking the C-B bond, and one-pot C-H borylation/Suzuki-Miyaura cross-coupling has been accomplished on 2- and 3-borylated thiophenes.  相似文献

N-substituted {2-methyl-benzo[b]thienyl-(3)}-acetamides were oxidized to the corresponding 1,1-dioxides, catalytic hydrogenation of which gave N-substituted {2-methyl-2,3-dihydro-benzo[b]thienyl-(3)}-acetamide-S,S-dioxides. The most concise approach involves sequential coupling of o-bromoiodobenzenes with benzylmercaptan and zinc acetylides to give benzyl o-ethynylphenyl sulfides which react with iodine to give 3-iodobenzo[b]thiophenes in a 5-endo-dig iodocyclization. These iodides can be further elaborated using palladium-mediated coupling and/or metalation techniques. This method has been applied to the synthesis of some novel tubulin binding agents.  相似文献

The behavior of indolo[6,5-d]benzo[b]thiophene in acylation, Mannich, Vilsmeier, and diazo coupling reactions was investigated. Acylation with acetic anhydride proceeds unambiguously at the pyrrole nitrogen atom to give 1-acetylindolo[6,5-d] benzo[b]thiophene. The products of formylation are 2- and 3-formylindolo-[6,5-d] benzo[b]thiophenes. In the case of the Mannich reaction and diazo coupling substitution takes place in the 3 position of the indolobenzo[b]thiophene ring. 2,3-Disubstituted benzo[b]thiophenes have been prepared in excellent yields via coupling of terminal acetylenes with commercially available o-iodothioanisole in the presence of a palladium catalyst and subsequent electrophilic cyclization of the resulting o-(1-alkynyl)thioanisole derivatives. I(2), Br(2), NBS, p-O(2)NC(6)H(4)SCl, and PhSeCl have been utilized as electrophiles. Aryl-, vinyl-, and alkyl-substituted terminal acetylenes undergo this coupling and cyclization to produce excellent yields of benzo[b]thiophenes. (Trimethylsilyl)acetylene also undergoes this coupling/cyclization process with I(2), NBS, and the sulfur and selenium electrophiles to afford the corresponding 2-(trimethylsilyl)benzo[b]thiophenes. However, cyclization of the silyl-containing thioanisole using Br(2) affords 2,3-dibromobenzo[b]thiophene.  相似文献
