Regioselective bromination of 14β-hydroxydihydroquinopimaric acid methyl ester in acetic acid and methanol gave the corresponding 16S-bromo-, 19R-bromo-, and 16S,19R-dibromo-14β,20-epoxy derivatives whose structure was determined by X-ray analysis and NMR spectroscopy. The reactions of 16S-bromo ketones with thiourea afforded 2,1,3-thiadiazoles fused to diterpene fragment.  相似文献

4-Dibromomethyl-4-methyl-2,5-cyclohexadienone and its 2- and 3-methyl-substituted derivatives react with an equimolar amount of molecular bromine in carbon tetrachloride, yielding vinyl bromination products at the -position with respect to the carbonyl group. The reaction of 4-dibromomethyl-3,4-dimethyl-2,5-cyclohexadienone with a large excess of bromine, apart from the vinyl bromination product, gives the corresponding 3-bromomethyl and 3-dibromomethyl derivatives. 4-Dibromomethyl-2,4-dimethyl-2,5-cyclohexadienone takes up bromine molecule at the C 2 ÍC 3 double bond.  相似文献

Vilsmeier-Haak reaction of phlomisoic acid methyl ester gave a mixture of 15- and 16-formyllabdanoids. In addition, methyl 2-formyldodecahydrophenanthro[1,2-b]furan-6-carboxylate was isolated, and its structure was determined by X-ray analysis. Reductive amination of 16-formyllabdanoid with benzylamine or α-amino acid methyl esters led to the formation of labdanoid furfurylamines which reacted with maleic anhydride to produce N-substituted 4-oxo-10-oxa-3-azatricyclo[5.2.1.01,5]dec-8-ene-6-carboxylic acids. Acylation of labdanoid furfurylamines with (E)-but-2-enoyl chloride afforded the corresponding unsaturated amides which were converted into 10-oxa-3-azatricyclo[5.2.1.01,5]dec-8-en-4-ones via intramolecular Diels-Alder reaction. Treatment of the oxa adducts with boron trifluoride-diethyl ether complex gave dihydroisoindol-1-one derivatives containing a diterpene fragment.  相似文献

Diastereoisomeric [(1R,5S)-5-“[(1R)-1-phenylethyl]aminocarbonyl”cyclopent-2-en-1-yl]methyl acetate and [(1S,5R)-5-“[(1R)-1-phenylethyl]aminocarbonyl”cyclopent-2-en-1-yl]methyl acetate reacted with m-chloroperoxybenzoic acid to give the corresponding stereoisomeric α- and β-epoxy derivatives, which were identified on the basis of their spectral parameters.  相似文献

The reactivity of benzo[b]naphtho[2,3-e][1,4]dioxin in the electrophilic aromatic substitution reactions has been studied. Friedel-Crafts acetylation resulted in the formation of three out of the possible five monoacetylated products, with the acetyl group located in positions 8 (major), 7 and 6 (minor) of the heterocycle. In the bromination reaction a higher selectivity was observed with the 6-bromo derivative found as the only monobrominated product and the 6,11-dibromo derivative found as the only polybrominated product. A ratio of unreacted heterocycle:6-bromo:6,11-dibromo derivatives in the bromination reaction has been found to depend strongly on the reaction conditions and on the heterocycle:bromine ratio.  相似文献

The reaction of levulinic acid and its esters with bromine in ionic liquids results in the formation of 3-bromo derivatives as the major products and not the 5-bromo substituted isomers, which are typically formed in organic solvents. The bromination of levulinic acid in ionic liquids in the presence of urea leads to the formation of 5-bromolevulinic acid.  相似文献

Tautomeric transformation of (3,5-dibromo-1-hydroxy-4-oxocyclohexa-2,5-dien-1-yl)acetic acid into previously unknown stereoisomeric γ-lactones, (3aS*,7S*,7aR*)- and (3aS*,7R*,7aR*)-5,7-dibromo-3ahydroxy-3,3a,7,7a-tetrahydro-1-benzofuran-2,6-diones, was revealed by 1H NMR spectroscopy. The concentration of the acid tautomer increases as the solvent polarity rises. The ability of the cycloxexene ring in the lactones to undergo inversion in solution is determined by orientation of the bromine atom on C7.  相似文献

 For a sodium salt of α-sulfonatomyristic acid methyl ester (14SFNa), one of the α-SFMe series surfactants, critical micellization concentration (CMC), solubility and degree of counterion binding (β) were determined by means of electrocon-ductivity measurements at different temperatures (at every 5 °C) ranging from 15 to 50 °C. The phase diagram of 14SFNa in pure water was constructed from the CMC- and solubility-temperature data, in which the Krafft temperature (critical solution temperature) was found around 0 °C. The changes in the Gibbs energy, ΔG 0 m, enthalpy, ΔH 0 m, and entropy, ΔS 0 m, upon micelle formation as a function of temperature were evaluated taking βvalues into calculation. Reaction of the 6-chloro-7-fluoroquinoline 7 with methyl 2-mercaptoacetate, methyl 3-mercaptopropionate, or sodium thiophenolate furnished the quinolone derivatives of 3-carbonylsulfanyl-acetic acid methyl ester 8, the propionoate analogue 10, and 3-carbothioic acid S-phenyl ester 11 respectively. Ester 8 was converted into the 3-carbothioic acid S-carbamoyl derivative 9. Analogously, treatment of the 6,7-diflouroquinolone 12 with amino or mercapto precursors led to the formation of 13 and 14 respectively. Reaction of 14 with aqueous NH3 or H2O2/AcOH afforded the acetamide 15 and the sulfoxide 16 analogues, respectively. The 5′-thioalkyl-acyclic quinolone nucleosides 19 and 20 were obtained from reaction of the mesylate derivative 18, prepared from the free nucleoside 17, with the methanthiolate and thiophenolate anions.  相似文献

Humulene and its derivatives (6R)-hydroxy-α-humulene [(6R)-hydroxy-(1E,4E,8E)-4,8,11,11tetramethylcycloundeca-1,4,8-triene], (6R)-acetoxy-α-humulene [(6R)-acetoxy-(1E,4E,8E)-4,8,11,11tetramethylcycloundeca-1,4,8-triene], a coumaric acid ester, 14-hydroxy-α-humulene [14-coumaroxy(1E,4E,8E)-4,8,11,11-tetramethylcycloundeca-1,4,8-triene], (1E,6R,8E)-4,5-epoxy-6-hydroxy-4,8,11,11tetramethylcycloundeca-1,8-diene, and (6R,9S)-4,11,11-trimethyl-8-methylene-1,4-cycloundecadien-6,9-diol were observed in the hydrocarbon extract of Betula pendula (Betulaceae) buds. It is shown that sodium alkoxides formed from (4R,5R)-2,2-dimethyl-1,3-dioxolane-4,5-bis(diphenylmethanol) ((R,R)-TADDOL) and some of its derivatives can be used as chiral catalysts for enantioselective alkylation of Schiff's bases derived from alanine with reactive alkyl halides. Acid hydrolysis of the reaction products affords (R)-α-methylphenyl-alanine, (R)-α-allylalanine, and (R)-α-methylnaphthylalanine in 61–93% yields and withee 69–94%. When (S,S)-TADDOL is used, the (S)-amino acid is formed. A mechanism explaning the observed features of the reaction is proposed. Translated fromIzvestiya Akademii Nauk. Aiming at preparation of biologically active compounds a bromination of N,N'-substituted malonodiamides in a glacial acetic acid was carried out. The use of one equiv of bromine provided monobromo derivatives, with two equiv of bromine dibromo-substituted products were obtained. Among the N,N'-dibenzylamides of alkylmalonic acids only the methyl homolog underwent bromination. The structure of compounds was proved by IR and 1H NMR spectroscopy. The effect of the compounds synthesized on the central nervous system was investigated.  相似文献

Summary Several bis/tris-salicylaldehyde, salicylaldimine and salicyalethylenediimine chelates of cobalt(III), chromium(III), cobalt(II), nickel(II) and copper(II) readily react with various brominating agents and undergo α-bromo/cyano/succinimido substitution, with or without accompanying bromine substitution of the aryl ring. The selectivity of these reactions on the metal-coordinated salicylaldehyde derivatives allows the preparation in 50–80% yield of hitherto unreported specific α-bromo products. The substituted organic compounds could be isolated by demetallation of the chelate products.  相似文献

Three-component condensation of [(1S,4S)-1,7,7-trimethylbicyclo[2.2.1]hept-2-ylidene]acetaldehyde with naphthalen-1-amine and cyclic β-diketones gave 7-[(1S,4S)-1,7,7-trimethylbicyclo[2.2.1]hept-2-ylidenemethyl]-7,8,9,10,11,12-hexahydro-7H-benzo[c]acridin-8-one derivatives possessing three or more asymmetric carbon atoms. Steric factors were found to be responsible for the predominant formation of the (7R)-isomers (R/S ≈ 7: 5) and orientation of substituents in the cyclohexenone fragment. The same factors determined complete regioselectivity of the reaction with methyl 2,4-dioxocyclohexane-1-carboxylates as dicarbonyl component, which led to exclusive formation of methyl 8-oxobenzoacridine-11-carboxylates. In the reaction of [(1S,4S)-1,7,7-trimethylbicyclo[2.2.1]hept-2-ylidene]acetaldehyde with naphthalen-1-amine and barbituric acid as dicarbonyl component, the only product was that formed by two-component condensation of barbituric acid with bicyclic aldehyde.  相似文献

The heterospirocyclic N‐methyl‐N‐phenyl‐5‐oxa‐1‐azaspiro[2.4]hept‐1‐e n‐2‐amine (6 ) and N‐(5‐oxa‐1‐azaspiro[2.4]hept‐1‐en‐2‐yl)‐(S)‐proline methyl ester ( 7 ) were synthesized from the corresponding heterocyclic thiocarboxamides 12 and 10 , respectively, by consecutive treatment with COCl2, 1,4‐diazabicyclo[2.2.2]octane, and NaN3 (Schemes 1 and 2). The reaction of these 2H‐azirin‐3‐amines with thiobenzoic and benzoic acid gave the racemic benzamides 13 and 14 , and the diastereoisomeric mixtures of the N‐benzoyl dipeptides 15 and 16 , respectively (Scheme 3). The latter were separated chromatographically. The configurations and solid‐state conformations of all six benzamides were determined by X‐ray crystallography. With the aim of examining the use of the new synthons in peptide synthesis, the reactions of 7 with Z‐Leu‐Aib‐OH to yield a tetrapeptide 17 (Scheme 4), and of 6 with Z‐Ala‐OH to give a dipeptide 18 (Scheme 5) were performed. The resulting diastereoisomers were separated by means of MPLC or HPLC. NMR Studies of the solvent dependence of the chemical shifts of the NH resonances indicate the presence of an intramolecular H‐bond in 17 . The dipeptides (S,R)‐ 18 and (S,S)‐ 18 were deprotected at the N‐terminus and were converted to the crystalline derivatives (S,R)‐ 19 and (S,S)‐ 19 , respectively, by reaction with 4‐bromobenzoyl chloride (Scheme 5). Selective hydrolysis of (S,R)‐ 18 and (S,S)‐ 18 gave the dipeptide acids (R,S)‐ 20 and (S,S)‐ 20 , respectively. Coupling of a diastereoisomeric mixture of 20 with H‐Phe‐OtBu led to the tripeptides 21 (Scheme 5). X‐Ray crystal‐structure determinations of (S,R)‐ 19 and (S,S)‐ 19 allowed the determination of the absolute configurations of all diastereoisomers isolated in this series.  相似文献

The bromination of benzo[b]selenophene with an equimolecular amount of bromine and its acetylation with acetic anhydride in the presence of boron trichloride etherate proceed to form a mixture of isomeric 2- and 3-substituted derivatives; 2,3-dibromobenzo[b]selenophene is formed on bromination with two equivalents of bromine. Selectively substituted derivatives of α-cyclodextrin and hexa-O-tert-butyldimethylsilyl-α-cyclodextrin containing a definite number of acylated primary or secondary hydroxy groups were synthesized using benzoic, acetylsalicylic, and 2-(4-isobutylphenyl)propionic acid chlorides in various solvents in the presence of different bases. The acyl residues were assigned to the C2, C3, or C6 atoms in the carbohydrate fragments on the basis of 13C NMR data. Desilylation of the silyl derivatives with ammonium fluoride in methanol gave the corresponding O-acyl derivatives having free primary hydroxy groups in the α-cyclodextrin skeleton.  相似文献

Aiming at the investigation of the mechanism of functioning of steroid estrogens a series of compounds with unnatural rings junction was synthesized. All investigated compounds exhibit a reduced uterotropic activity. It was established applying the NMR spectroscopy that 7α-methyl-3-methoxy-D-homo-6-oxa-8α,14β-estra-1,3,5(10)-trien-17a-one existed in solution in two conformations distinguished by the structure of the rings B, C, and D simultaneously. The reaction of 17-methylidene-3-methoxy-6-oxa-8α-estra-1,3,5(10)-triene with hydrobromic acid in acetic acid promotes a rearrangement with the migration of a methyl group into the position 17 resulting in the formation of 17,17-dimethyl-6-oxa-8α-gona-1,3,5(10),13(14)-tetraene derivatives.  相似文献

Two novel cannabinoid model compounds, (3R, 4R)-Δ1(6)-tetrahydrocannabinol-5″-oic acid (22) and 4″(R, S)-methyl-(3R, 4R)-Δ1(6)-tetrahydrocannabinol-5″-oic acid (23) were synthesized by acid-catalyzed condensation of (+)-trans-p-mentha-2, 8-dien-l-ol (1) with the substituted resorcinols 18 and 19 obtained by a Wittig reaction between 3, 5-bis(benzyloxy)benzaldehyde (7) and methyl 4-bromobutanoate (10) or methyl 4-bromo-2(R, S)-methylbutanoate (11) resp. with subsequent hydrogenation. The stoichiometric protonation constants of some α-amino acid esters (glycine methyl ester, glycine t-butyl ester, l-valine methyl ester, l-valine ethyl ester, l-valine t-butyl ester, l-serine methyl ester, l-serine ethyl ester, l-leucine methyl ester, l-leucine ethyl ester, l-leucine t-butyl ester, l-alanine methyl ester, l-alanine benzyl ester, l-phenylalanine methyl ester, l-phenylalanine ethyl ester, and l-phenylalanine t-butyl ester) in water and 20%, 40%, and 60% (v/v) 1,4-dioxane–water mixtures have been determined at an ionic strength of 0.10 mol⋅L−1 NaCl and at 25.0±0.1 °C under a nitrogen atmosphere. A potentiometric method was used and the calculation of the protonation constants has been carried out using the BEST computer program. The results were discussed in terms of macroscopic properties of the mixed solvent. The stoichiometric protonation constants were influenced by changes in solvent composition and their variations were discussed in terms of preferential solvation. Also, knowledge the protonation constant of α-amino acid esters will be helpful when determining the microscopic equilibrium constants of their corresponding amino acids.  相似文献
