New capped silica mesoporous nanoparticles for intracellular controlled cargo release within cathepsin B expressing cells are described. Nanometric mesoporous MCM‐41 supports loaded with safranin O ( S1‐P ) or doxorubicin ( S2‐P ) containing a molecular gate based on a cathepsin B target peptidic sequence were synthesized. Solids were designed to show “zero delivery” and to display cargo release in the presence of cathepsin B enzyme, which selectively hydrolyzed in vitro the capping peptide sequence. Controlled delivery in HeLa, MEFs WT, and MEFs lacking cathepsin B cell lines were also tested. Release of safranin O and doxorubicin in these cells took place when cathepsin B was active or present. Cells treated with S2‐P showed a fall in cell viability due to nanoparticles internalization, cathepsin B hydrolysis of the capping peptide, and cytotoxic agent delivery, proving the possible use of these nanodevices as new therapeutic tools for cancer treatment.  相似文献

Functionalization of monodisperse superparamagnetic magnetite (Fe3O4) nanoparticles for cell specific targeting is crucial for cancer diagnostics and therapeutics. Targeted magnetic nanoparticles can be used to enhance the tissue contrast in magnetic resonance imaging (MRI), to improve the efficiency in anticancer drug delivery, and to eliminate tumor cells by magnetic fluid hyperthermia. Herein we report the nucleus‐targeting Fe3O4 nanoparticles functionalized with protein and nuclear localization signal (NLS) peptide. These NLS‐coated nanoparticles were introduced into the HeLa cell cytoplasm and nucleus, where the particles were monodispersed and non‐aggregated. The success of labeling was examined and identified by fluorescence microscopy and MRI. The work demonstrates that monodisperse magnetic nanoparticles can be readily functionalized and stabilized for potential diagnostic and therapeutic applications.  相似文献

The synthesis and characterisation of new capped silica mesoporous nanoparticles for on‐command delivery applications is reported. Functional capped hybrid systems consist of MCM‐41 nanoparticles functionalised on the external surface with polyesters bearing azobenzene derivatives and rhodamine B inside the mesopores. Two solid materials, Rh‐PAzo8‐S and Rh‐PAzo6‐S, containing two closely related polymers, PAzo8 and PAzo6, in the pore outlets have been prepared. Materials Rh‐PAzo8‐S and Rh‐PAzo6‐S showed an almost zero release in water due to steric hindrance imposed by the presence of anchored bulky polyesters, whereas a large delivery of the cargo was observed in the presence of an esterase enzyme due to the progressive hydrolysis of polyester chains. Moreover, nanoparticles Rh‐PAzo8‐S and Rh‐PAzo6‐S were used to study the controlled release of the dye in intracellular media. Nanoparticles were not toxic for HeLa cells and endocytosis‐mediated cell internalisation was confirmed by confocal microscopy. Furthermore, the possible use of capped materials as a drug‐delivery system was demonstrated by the preparation of a new mesoporous silica nanoparticle functionalised with PAzo6 and loaded with the cytotoxic drug camptothecin (CPT‐PAzo6‐S). Following cell internalisation and lysosome resident enzyme‐dependent gate opening, CPT‐PAzo6‐S induced CPT‐dependent cell death in HeLa cells.  相似文献

Utilizing semirigid lactic acid derivatives (R)‐H2CBA and (S)‐H2CBA as chiral ligands, two pair of homochiral coordination polymers formulated as [Zn((R)‐CBA)(1,4‐DIB)] · H2O ( 1 ‐ D ), [Zn((S)‐CBA)(1,4‐DIB)] · H2O ( 1 ‐ L ), [Co((R)‐CBA)(1,4‐DIB)] · H2O ( 2 ‐ D ) and [Co((S)‐CBA)(1,4‐DIB)] · H2O ( 2 ‐ L ) were prepared under solvothermal reaction condition. Single X‐ray diffraction study reveals that all the complexes are comprised of three kinds of helical chains, which are constructed by corresponding metal ions, CBA2– ligands, and/or 1,4‐DIB ligands. Moreover, some physical characteristics, such as PXRD, thermal stabilities, solid‐state circular dichroism (CD), luminescent and magnetic properties are also investigated.  相似文献

Four chiral polymers P-1, P-2, P-3 and P-4 were synthesized by the polymerization of (S)-2,2'-dioctoxy-1,1'- binaphthyl-6,6'-boronic acid (S-M-3) with (S)-6,6'-dibromo-1,1'-binaphthol (S-M-1), (R)-6,6'-dibromo-1,1'- binaphthol (R-M-1), (S)-3,3'-diiodo-1,1'-binaphthol (S-M-2) and (R)-3,3'-diiodo-1,1'-binaphthol (R-M-2) under Pd-catalyzed Suzuki reaction, respectively. All four polymers can show good solubility in some common solvents due to the nonplanarity of the polymers in the main chain backbone and flexible alkyl groups in the side chain. The analysis results indicate that specific rotation and circular dichroism (CD) spectral signals of the alternative S-S chiral polymers P-1 and P-3 are larger than those of S-R chiral polymers P-2 and P-4, but their UV-Vis and fluorescence spectra are almost similar. The results of asymmetric enantioselectivity of four polymers for diethylzinc addition to benzaldehyde indicate that catalytically active center is (R) or (S)-1, 1'-binaphthol moieties.  相似文献

2′,4′-Dihydroxy-6′-methoxy-3′,5′-dimethylchalcone (DMC), a natural product derived from Syzygium nervosum A. Cunn. ex DC., was investigated for its inhibitory activities against various cancer cell lines. In this work, we investigated the effects of DMC and available anticervical cancer drugs (5-fluorouracil, cisplatin, and doxorubicin) on three human cervical cancer cell lines (C-33A, HeLa, and SiHa). DMC displayed antiproliferative cervical cancer activity in C-33A, HeLa, and SiHa cells, with IC50 values of 15.76 ± 1.49, 10.05 ± 0.22, and 18.31 ± 3.10 µM, respectively. DMC presented higher antiproliferative cancer activity in HeLa cells; therefore, we further investigated DMC-induced apoptosis in this cell line, including DNA damage, cell cycle arrest, and apoptosis assays. As a potential anticancer agent, DMC treatment increased DNA damage in cancer cells, observed through fluorescence inverted microscopy and a comet assay. The cell cycle assay showed an increased number of cells in the G0/G1 phase following DMC treatment. Furthermore, DMC treatment-induced apoptosis cell death was approximately three- to four-fold higher compared to the untreated group. Here, DMC represented a compound-induced apoptosis for cell death in the HeLa cervical cancer cell line. Our findings suggest that DMC, a phytochemical agent, is a potential candidate for antiproliferative cervical cancer drug development.  相似文献

Mesoporous iron‐oxide nanoparticles (mNPs) were prepared by using a modified nanocasting approach with mesoporous carbon as a hard template. mNPs were first loaded with doxorubicin (Dox), an anticancer drug, and then coated with the thermosensitive polymer Pluronic F108 to prevent the leakage of Dox molecules from the pores that would otherwise occur under physiological conditions. The Dox‐loaded, Pluronic F108‐coated system (Dox@F108‐mNPs) was stable at room temperature and physiological pH and released its Dox cargo slowly under acidic conditions or in a sudden burst with magnetic heating. No significant toxicity was observed in vitro when Dox@F108‐mNPs were incubated with noncancerous cells, a result consistent with the minimal internalization of the particles that occurs with normal cells. On the other hand, the drug‐loaded particles significantly reduced the viability of cervical cancer cells (HeLa, IC50=0.70 μm ), wild‐type ovarian cancer cells (A2780, IC50=0.50 μm ) and Dox‐resistant ovarian cancer cells (A2780/AD, IC50=0.53 μm ). In addition, the treatment of HeLa cells with both Dox@F108‐mNPs and subsequent alternating magnetic‐field‐induced hyperthermia was significantly more effective at reducing cell viability than either Dox or Dox@F108‐mNP treatment alone. Thus, Dox@F108‐mNPs constitute a novel soft/hard hybrid nanocarrier system that is highly stable under physiological conditions, temperature‐responsive, and has chemo‐ and thermotherapeutic modes of action.  相似文献

The synthesis, characterization and biological activity of four cyclometalated Ir (III) complexes ( Ir1 ‐ Ir4 ) containing different phosphine‐sulfonate ligands are reported. Most of these complexes showed good activity against A549 cancer cell lines and the human HeLa cervical cell lines. Spectroscopic properties study displays that all four complexes show rich fluorescence with emission maxima in the range of 474–510 nm. Fluorescence property of these complexes provides a tool to investigate the microscopic mechanism by confocal microscopy. Notably, the typical Ir (III) complex Ir4 can specially localize to lysosome, damage it and induce cell death via apoptosis. In addition, Ir4 enters into A549 cancer cells dominantly through energy‐dependent pathway.  相似文献

Water‐soluble phosphorescent polymeric nanoparticles with an average diameter of approximately 100 nm were synthesized by a coordination cross‐linking reaction. The pyridine blocks in poly(4‐vinyl pyridine‐b‐ethylene oxide) (P4VP‐b‐PEO) were cross‐linked by the iridium chloride‐bridged dimer in DMF solution. Owing to the presence of an iridium complex with different ligands in the core of the polymeric nanoparticles, NP‐1, NP‐2, and NP‐3 showed bright green, yellow, and red phosphorescence, respectively. PEG chains in the shell gave the polymeric nanoparticles solubility and biocompatibility, which was confirmed by an MTT assay using HeLa cells as a model cancer cell line. The flow cytometry and laser confocal fluorescence microscopy results revealed NP‐2, as an example, could be effectively uptaken by HeLa cells. Therefore, these polymeric nanoparticles can be used as luminescent probes for living cells. In addition, 1O2 could be effectively generated in the presence of NP‐2 upon irradiation with visible light (λ>400 nm, 300 mW cm?2), which was confirmed by a clear decrease in the fluorescence intensity of 9,10‐dimethylanthracene (DMA). After incubation with NP‐2 at a concentration of 200 μg mL?1 for 6 h, approximately 90 % of HeLa cells were effectively ablated upon irradiation with visible light for only 10 min, indicating the potential for photodynamic therapy with polymeric nanoparticles.  相似文献

A new (S)‐binaphthalene‐based polymer ( P ‐ 1 ) was synthesized by the polymerization of 5,5′‐((2,5‐dibutoxy‐1,4‐phenylene)bis(ethyne‐2,1‐diyl))bis(2‐hydroxy‐3‐(piperidin‐1‐ylmethyl) benzaldehyde ( M ‐ 1 ) with (S)‐2,2′‐dimethoxy‐(1,1′‐binaphthalene)‐3,3′‐diamine ( M ‐ 2 ) through the formation of a Schiff base; the corresponding chiral polymer ( P ‐ 2 ) could be obtained by the reduction of polymer P ‐ 1 with NaBH4. Chiral polymer P ‐ 1 exhibited a remarkable “turn‐on” fluorescence‐enhancement response towards (D )‐phenylalaninol and excellent enantioselective recognition behavior with enantiomeric fluorescence difference ratios (ef) as high as 8.99. More importantly, chiral polymer P ‐ 1 displays a bright blue fluorescence color change upon the addition of (D )‐phenylalaninol under a commercially available UV lamp, which can be clearly observed by the naked eye. On the contrary, chiral polymer P ‐ 2 showed weaker enantioselective fluorescence ability towards the enantiomers of phenylalaninol.  相似文献

The synthesis and characterization of two new capped silica mesoporous nanoparticles for controlled delivery purposes are described. Capped hybrid systems consist of MCM‐41 nanoparticles functionalized on the outer surface with polymer ε‐poly‐L ‐lysine by two different anchoring strategies. In both cases, nanoparticles were loaded with model dye molecule [Ru(bipy)3]2+. An anchoring strategy involved the random formation of urea bonds by the treatment of propyl isocyanate‐functionalized MCM‐41 nanoparticles with the lysine amino groups located on the ε‐poly‐L ‐lysine backbone (solid Ru‐rLys‐S1 ). The second strategy involved a specific attachment through the carboxyl terminus of the polypeptide with azidopropyl‐functionalized MCM‐41 nanoparticles (solid Ru‐tLys‐S1 ). Once synthesized, both nanoparticles showed a nearly zero cargo release in water due to the coverage of the nanoparticle surface by polymer ε‐poly‐L ‐lysine. In contrast, a remarkable payload delivery was observed in the presence of proteases due to the hydrolysis of the polymer’s amide bonds. Once chemically characterized, studies of the viability and the lysosomal enzyme‐controlled release of the dye in intracellular media were carried out. Finally, the possibility of using these materials as drug‐delivery systems was tested by preparing the corresponding ε‐poly‐L ‐lysine capped mesoporous silica nanoparticles loaded with cytotoxic drug camptothecin (CPT), CPT‐rLys‐S1 and CPT‐tLys‐S1 . Cellular uptake and cell‐death induction were studied. The efficiency of both nanoparticles as new potential platforms for cancer treatment was demonstrated.  相似文献

Butelase‐mediated ligation (BML) can be used to modify live bacterial cell surfaces with diverse cargo molecules. Surface‐displayed butelase recognition motif NHV was first introduced at the C‐terminal end of the anchoring protein OmpA on E. coli cells. This then served as a handle of BML for the functionalization of E. coli cell surfaces with fluorescein and biotin tags, a tumor‐associated monoglycosylated peptide, and mCherry protein. The cell‐surface ligation reaction was achieved at low concentrations of butelase and the labeling substrates. Furthermore, the fluorescein‐labeled bacterial cells were used to show the interactions with cultured HeLa cells and with macrophages in live transgenic zebrafish, capturing the latter's powerful phagocytic effect in action. Together these results highlight the usefulness of butelase 1 in live bacterial cell surface engineering for novel applications.  相似文献

Herein, a novel cationic peptide gemini amphiphile containing diacetylene motifs ( DA2P ) is presented, which self‐assembles into novel tadpole‐ and bola‐shaped nanostructures at low concentrations and nanofibers at higher concentrations. Interestingly, the DA2P assemblies can be polymerized into a fluorescent red phase but only during incubation with HeLa cells, most likely owing to the reorganization of the diacetylene chains of DA2P upon interaction with the cell membrane. The red‐fluorescent polymerized DA2P assemblies can serve as a novel cell imaging probe. However, only vesicles, tadpole‐ and bola‐shaped DA2P assemblies can be translocated into HeLa cells, whereas the nanofiber‐like DA2P assemblies are trapped by the cell membranes and do not enter the cells. Hence, morphology‐dependent cell imaging is observed.  相似文献

A 10‐hydroxycamptothecin‐encapsulated magnetic nanovehicle (HEMN) was fabricated by coencapsulating Fe3O4 nanoparticles and 10‐hydroxycamptothecin (HCPT) into a micelle core self‐assembled from the amphiphilic copolymer methoxy‐poly(ethylene glycol)–poly(d,l ‐lactide‐co‐glycolide) through a facile dialysis method. A satisfactory drug‐loading content of (9.03±0.67) % and a relatively high encapsulation efficiency of (53.52±6.46) % were achieved. In vitro drug release was performed by membrane dialysis and a pH‐dependent release behavior was observed. In comparison with free HCPT dissolved in dimethylsulfoxide, HEMNs showed a greatly improved in vitro antitumor efficacy against three different human cancer cell lines—HeLa, A549, and HepG2—and lower IC50 values were measured. The mechanism of cell death was investigated, and it was clearly demonstrated that the apoptosis process was triggered. An in vitro wound‐healing assay and a transwell assay indicated that HEMNs exerted much stronger activity in inhibiting HeLa cell migration. The cellular uptake of HEMNs in a desired area can be significantly enhanced by an external magnetic field. These results demonstrate HCPT‐encapsulated magnetic nanovehicles might have important potential in clinical applications for inhibiting tumor metastasis and for targeted drug delivery.  相似文献

In this report, we describe the characterizations and applications of hybrid nanoparticles. These nanoparticles have been synthesized by combination of organometallic, polymerization process and functionalized with a specific peptide for targeting expressed serpin‐enzyme complex (SEC) receptor of human hepatoma HepG2 cells. By using peptide conjugated hybrid nanoparticles, the specific receptor targeting, collections of cells were successfully achieved. The cell collection results indicated that, the maximum up to 95.32% of HepG2 cell were collected. The 5‐dimethylthiazol‐2‐yl‐2,5‐diphenyltetrazolium bromide (MTT) assay of HepG2 cells incubated with these nanoparticles indicated that, the peptide conjugated hybrid nanoparticles did not possess significant cytotoxicity. The rotating magnetic field induced cell death studies indicated that, the HepG2 cell showed up to 70% of cell death was induced by hybrid nanoparticles under magnetic field. Concluding, these studies demonstrate that the hybrid nanoparticles have the capability of effective separation, imaging, targeting and killing of the human hepatoma cells.  相似文献

Herein, we report the photoinitiated polymerization-induced self-assembly (photo-PISA) of spherical micelles consisting of proapoptotic peptide–polymer amphiphiles. The one-pot synthetic approach yielded micellar nanoparticles at high concentrations and at scale (150 mg mL−1) with tunable peptide loadings up to 48 wt. %. The size of the micellar nanoparticles was tuned by varying the lengths of hydrophobic and hydrophilic building blocks. Critically, the peptide-functionalized nanoparticles imbued the proapoptotic “KLA” peptides (amino acid sequence: KLAKLAKKLAKLAK) with two key properties otherwise not inherent to the sequence: 1) proteolytic resistance compared to the oligopeptide alone; 2) significantly enhanced cell uptake by multivalent display of KLA peptide brushes. The result was demonstrated improved apoptosis efficiency in HeLa cells. These results highlight the potential of photo-PISA in the large-scale synthesis of functional, proteolytically resistant peptide–polymer conjugates for intracellular delivery.  相似文献

Photothermal therapy has attracted much interest for use in cancer treatment in recent years. In this study, Cu2Se nanoparticles as a novel photothermal agent modified by chitosan (CS‐Cu2SeNPs) were successfully synthesized through a facile route at room temperature. The as‐synthesized CS‐Cu2SeNPs exhibited good water solubility and significant stability. CS‐Cu2SeNPs can efficiently convert near‐infrared (NIR) light into heat and exhibit excellent thermostability. In vitro experiments showed that CS‐Cu2SeNPs had selective cellular uptake between cancer and normal cells and expressed clear anticancer activity on A375 and HeLa human cancer cells. In addition, the anticancer activity was increased to about 400 % by combination with a laser at 808 nm, which acted through induction of apoptosis with the involvement of intrinsic and extrinsic pathways. CS‐Cu2SeNPs irradiated with a laser effectively triggered the intracellular reactive oxygen species (ROS) overproduction that promoted cell apoptosis. Therefore, the developed CS‐Cu2SeNPs could be used as a novel phototherapeutic agent for the photothermal therapy of human cancers.  相似文献

Four putative functionalized α‐chloroakyllithiums RCH2CHLiCl, where R=CHCH2 ( 18 a ), CCH ( 18 b ), CH2OBn ( 18 c ), and CH[O(CH2)2O] ( 18 d ), were generated in situ by sulfoxide–lithium exchange from α‐chlorosulfoxides, and investigated for the stereospecific reagent‐controlled homologation (StReCH) of phenethyl and 2‐chloropyrid‐5‐yl ( 17 ) pinacol boronic esters. Deuterium labeling experiments revealed that αchloroalkyllithiums are quenched by proton transfer from their αchlorosulfoxide precursors and it was established that this effect compromises the yield of StReCH reactions. Use of α‐deuterated α‐chlorosulfoxides was discovered to ameliorate the problem by retarding the rate of acid‐base chemistry between the carbenoid and its precursor. Carbenoids 18 a and 18 b showed poor StReCH efficacy, particularly the propargyl group bearing carbenoid 18 b , the instability of which was attributed to a facile 1,2‐hydride shift. By contrast, 18 d , a carbenoid that benefits from a stabilizing interaction between O and Li atoms gave good StReCH yields. Boronate 17 was chain extended by carbenoids 18 a , 18 b , and 18 d in 16, 0, and 68 % yield, respectively; α‐deuterated isotopomers D ‐ 18 a and D ‐ 18 d gave yields of 33 and 79 % for the same reaction. Double StReCH of 17 was pursued to target contiguous stereodiads appropriate for the total synthesis of (?)‐epibatidine ( 15 ). One‐pot double StReCH of boronate 17 by two exposures to (S)‐D ‐ 18 a (≤66 % ee), followed by work‐up with KOOH, gave the expected stereodiad product in 16 % yield (d.r.~67:33). The comparable reaction using two exposures to (S)‐D ‐ 18 d (≤90 % ee) delivered the expected bisacetal containing stereodiad (R,R)‐DD ‐ 48 in 40 % yield (≥98 % ee, d.r.=85:15). Double StReCH of 17 using (S)‐D ‐ 18 d (≤90 % ee) followed by (R)‐D ‐ 18 d (≤90 % ee) likewise gave (R,S)‐DD ‐ 48 in 49 % yield (≥97 % ee, d.r.=79:21). (R,S)‐DD ‐ 48 was converted to a dideuterated isotopomer of a synthetic intermediate in Corey’s synthesis of 15 .  相似文献

Hypoxia is the critical feature of the tumor microenvironment that is known to lead to resistance to many chemotherapeutic drugs. Six novel ruthenium(II) anthraquinone complexes were designed and synthesized; they exhibit similar or superior cytotoxicity compared to cisplatin in hypoxic HeLa, A549, and multidrug‐resistant (A549R) tumor cell lines. Their anticancer activities are related to their lipophilicity and cellular uptake; therefore, these physicochemical properties of the complexes can be changed by modifying the ligands to obtain better anticancer candidates. Complex 1 , the most potent member of the series, is highly active against hypoxic HeLa cancer cells (IC50=0.53 μM ). This complex likely has 46‐fold better activity than cisplatin (IC50=24.62 μM ) in HeLa cells. This complex tends to accumulate in the mitochondria and the nucleus of hypoxic HeLa cells. Further mechanistic studies show that complex 1 induced cell apoptosis during hypoxia through multiple pathways, including those of DNA damage, mitochondrial dysfunction, and the inhibition of DNA replication and HIF‐1α expression, making it an outstanding candidate for further in vivo studies.  相似文献

Catalytic, peptide‐containing metal complexes with a well‐defined peptide structure have the potential to enhance molecular catalysts through an enzyme‐like outer coordination sphere. Here, we report the synthesis and characterization of an active, peptide‐based metal complex built upon the well‐characterized hydrogen production catalyst [Ni(PPh2NPh)2]2+ (PPh2NPh=1,3,6‐triphenyl‐1‐aza‐3,6‐diphosphacycloheptane). The incorporated peptide maintains its β‐hairpin structure when appended to the metal core, and the electrocatalytic activity of the peptide‐based metal complex (≈100,000 s?1) is enhanced compared to the parent complex ([Ni(PPh2NAPPA)2]2+; ≈50,500 s‐1). The combination of an active molecular catalyst with a structured peptide provides a scaffold that permits the incorporation of features of an enzyme‐like outer‐coordination sphere necessary to create molecular electrocatalysts with enhanced functionality.  相似文献
