In the crystal structure of 6‐phenyl‐3‐thioxo‐2,3,4,5‐tetrahydro‐1,2,4‐triazin‐5‐one, C9H7N3OS, (I), the 1,2,4‐triazine moieties are connected by face‐to‐face contacts through two kinds of double hydrogen bonds (N—H...O and N—H...S), which form planar ribbons along the a axis. The ribbons are crosslinked through C—H...π interactions between the phenyl rings. The molecular structures of two regioisomeric compounds, namely 6‐phenyl‐2,3‐dihydro‐7H‐1,3‐thiazolo[3,2‐b][1,2,4]triazin‐7‐one, C11H9N3OS, (II), and 3‐phenyl‐6,7‐dihydro‐4H‐1,3‐thiazolo[2,3‐c][1,2,4]triazin‐4‐one, C11H9N3OS, (III), which were prepared by the condensation reaction of (I) with 1,2‐dibromoethane, have been characterized by X‐ray crystallography and spectroscopic studies. The crystal structures of (II) and (III) both show two crystallographically independent molecules. While the two compounds are isomers, the unit‐cell parameters and crystal packing are quite different and (II) has a chiral crystal structure.  相似文献

The synthesis of 5,6,7,8‐tetrahydro‐4H‐oxazolo[4,5‐c]azepin‐4‐ones 5a,b and 1,3‐benzoxazol‐4‐amines 4a,b are described starting from 4,5,6,7‐tetrahydro‐1,3‐benzoxazol‐4‐ones. Thionation of 5a,b followed by alkylation with ethyl bromoacetate led to the corresponding S‐alkyl azepines 7a,b .  相似文献

N‐(2‐Amino‐5,6,7,8‐tetrahydro‐6‐quinazolinyl)acetamide ( 9 ) and N‐(2,4‐diamino‐5,6,7,8‐tetrahydro‐6‐quinazolinyl)acetamide ( 6 ) were synthesized from N‐(4‐oxocyclohexyl)acetamide ( 5 ) as novel peptidomimetic building blocks. With similar purpose, N‐(6‐oxo‐5,6,7,8‐tetrahydro‐2‐quinazolinyl)acetamide ( 18 ) and N‐[2‐(acetylamino)‐6‐oxo‐5,6,7,8‐tetrahydro‐4‐quinazolinyl]acetamide ( 14 ) were prepared from cyclohexane‐1,4‐dione monoethylene ketal ( 11 ).  相似文献

Anhydrous zinc bromide catalysed reactions of arylidine‐3‐acetyl coumarins ( 1a‐c ) and 5,6‐benzoanalogs of arylidine 3‐acetyl coumarins ( 4a,4b ) with 1,3‐cyclohexanedione gives ‐(4‐aryl‐5‐oxo‐5,6,7,8‐tetrahydro‐4H‐chromen‐2yl)‐2H‐chromen‐2‐ones ( 3a, 3c ) and 5,6‐benzoanalogs of 3‐(4‐aryl‐5‐oxo‐5,6,7,8‐tetrahydro‐4H‐chromen‐2yl)‐2H‐chromen‐2‐one ( 5a,5b ). Under similar conditions arylidine‐3‐acetylcoumarins ( 1a, 1b,1d, 1e, 1f ) and 5,6‐benzoanalog of arylidine 3‐acetyl coumarin ( 4b ) react with 5,5‐dimethyl‐1,3‐cyclohexanedione (dimedone) yielding 3‐(4‐aryl‐7,7‐dimethyl‐5‐oxo‐5,6,7,8‐tetrahydro‐4H‐chromen‐2‐yl)‐2H‐chromen‐2‐ones ( 3d‐3h ) and the 5,6‐benzoanalog of 3.(4‐aryl‐7,7‐dimethyl‐5‐oxo‐5,6,7,8‐tetrahydro‐4H‐chromen‐2‐yl)‐2H‐chromen‐2‐one ( 5c ).  相似文献

The molecules of (I) and (II) are linked into centrosymmetric dimers and simple chains, respectively, by C—H...O hydrogen bonds, augmented only in (I) by a C—H...π hydrogen bond. The molecules of (III) are linked by a combination of C—H...O and C—H...π hydrogen bonds into a chain of edge‐fused centrosymmetric rings, further linked by weak hydrogen bonds into supramolecular arrays in two or three dimensions. The heterocyclic molecules in (IV) are linked by two independent C—H...O hydrogen bonds into sheets, from which the partial‐occupancy ethanol molecules are pendent. The significance of this study lies in its finding of a very wide range of supramolecular aggregation modes dependent on rather modest changes in the peripheral substituents remote from the main hydrogen‐bond acceptor sites.  相似文献

rac‐5‐Diphenylacetyl‐2,2,4‐trimethyl‐2,3,4,5‐tetrahydro‐1,5‐benzothiazepine, C26H27NOS, (I), and rac‐5‐formyl‐2,2,4‐trimethyl‐2,3,4,5‐tetrahydro‐1,5‐benzothiazepine, C13H17NOS, (II), are both characterized by a planar configuration around the heterocyclic N atom. In contrast with the chair conformation of the parent benzothiazepine, which has no substituents at the heterocyclic N atom, the seven‐membered ring adopts a boat conformation in (I) and a conformation intermediate between boat and twist‐boat in (II). The molecules lack a symmetry plane, indicating distortions from the perfect boat or twist‐boat conformations. The supramolecular architectures are significantly different, depending in (I) on C—H...O interactions and intermolecular S...S contacts, and in (II) on a single aromatic π–π stacking interaction.  相似文献

The Bigenelli acid catalyzed condensation of 2‐pyridylcarboxaldehyde ( 1 ), urea ( 2 ) and an alkyl acetoacetate ( 3 ) afforded the respective alkyl (Me, Et, i‐Pr, i‐Bu, t‐Bu) 6‐methyl‐4‐(2‐pyridyl)‐1,2,3,4‐tetrahydro‐2H‐pyrimidine‐2‐one‐5‐carboxylates ( 4a‐e ). The most potent calcium channel antagonist ethyl 6‐methyl‐4‐(2‐pyridyl)‐1,2,3,4‐tetrahydro‐2H‐pyrimidine‐2‐one‐5‐carboxylate ( 4b , IC50 = 1.67 × 10?5 M) wasa much weaker calcium channel antagonist than the reference drug nifedipine (Adalat®, IC50 = 1.40 × 10?8 M) on guinea pig ileal longitudinal smooth muscle (GPILSM). The alkyl 6‐methyl‐4‐(2‐pyridyl)‐1,2,3,4‐tetrahydro‐2H‐pyrimidine‐2‐one‐5‐carboxylates did not show any inotropic effect on heart since no increase, or decrease, in the contractile force of guinea pig left atrium was observed. These structure activity studies show that the alkyl 6‐methyl‐4‐(2‐pyridyl)‐1,2,3,4‐tetrahydro‐2H‐pyrimidine‐2‐one‐5‐carboxylates ( 4a‐e ) are partial bioisosteres of nifedipine with respect to calcium channel antagonist activity on guinea pig ileal longitudinal smooth muscle (GPILSM).  相似文献

5‐Benzoyl‐4‐(substituted phenyl)‐6‐phenyl‐1,2,3,4‐tetrahydro‐2‐thioxopyrimidines ( 4a‐d ) were synthesized using the Biginelli three component cyclocondensation reaction of an appropriate β‐diketone, arylaldehyde, and thiourea in acetic acid under reflux condition in approximately 52‐65% yields. The acetylation of compounds 4a‐d gave 3‐acetyl thioxopyrimidine derivatives 5a‐d . Also, pyrimidothiazine compounds 6a‐d were prepared by a simple one‐pot condensation reaction of starting pyrimidine derivatives 4a‐d and 3‐bromopropionic acid. The structures of compounds were characterized on the basis of elemental analyses, IR, 1H and 13C‐NMR spectra.  相似文献

The 1H‐pyrazole‐3‐carboxylic acid 1 was converted via reactions of its acid chloride 3 with various asymmetrical disubstituted urea and alcohol derivatives into the corresponding novel 4‐benzoyl‐N‐(N′,N′‐dialkylcarbamyl)‐1‐(4‐methoxyphenyl)‐5‐phenyl‐1H‐pyrazole‐3‐carboxamide 4a , b and alkyl 4‐benzoyl‐1‐(4‐methoxyphenyl)‐5‐phenyl‐1H‐pyrazole‐3‐carboxylate 7a‐c , respectively, in good yields (57%‐78%). Friedel‐Crafts reactions of 3 with aromatic compouns for 15 min.‐2 h led to the formation of the 4‐3‐diaroyl‐1‐(4‐hydroxyphenyl)‐5‐phenyl‐1H‐pyrazoles 9a‐c , 4‐benzoyl‐1‐(4‐methoxyphenyl)‐3‐aroyl‐5‐phenyl‐1H‐pyrazoles 10a , b and than from the acylation reactions of 9a‐c were obtained the 3,4‐diaroyl‐1‐(4‐acyloxyphenyl)‐5‐phenyl‐1H‐pyrazoles 13a‐d . The structures of all new synthesized compounds were established by NMR experiments such as 1H, and 13C, as well as 2D COSY and IR spectroscopic data, and elemental analyses. All the compounds were evaluated for their antimicrobial activities (agar diffusion method) against eight bacteria and two yeasts.  相似文献

The treatment of 3,3a,4,5‐tetrahydro‐3‐aryl‐2‐phenyl‐2H‐benzo[g]indazoles 4 with I2/DMSO led to the oxidation of the five‐member rings ( 5 ) as well as the iodination of N‐phenyl moieties along with oxidation of the five‐member rings ( 6 ). However, the reactions of 4 with CuCl2/DMSO gave only compounds 5 . The reaction of N‐bromosuccinimide (NBS) with compounds 4 resulted in fully aromatization along with bromination at C‐5 of the indazole rings ( 7 ). The indazole six‐member rings in compounds 5 and 6 also underwent aromatization along with bromination by using NBS ( 7 and 8 ).  相似文献

In this study, 10 different substituted aromatic bis‐benzaldehydes were synthesized by treating hydroxy benzaldehydes with various dihaloalkanes. Bis aldehydes 5a , 5b , 5c , 5d , 5e , 5f , 5g , 5h , 5i , 5j were treated with 2‐(5‐phenyl‐1H‐tetrazole‐1‐yl)acetohydrazide ( 3 ) in acidic medium and in the presence of ammonium acetate to yield a series of new isomeric bis(2‐(5‐((5‐phenyl‐1H‐tetrazol‐1‐yl)methyl)‐4H‐1,2,4‐triazol‐3‐yl)phenoxy)alkanes ( 6a , 6b , 6c , 6d , 6e , 6f , 6g , 6h , 6i , 6j ) in excellent to good yield. The newly synthesized compounds were characterized by the available spectroscopic analysis.  相似文献

(2SR,4RS)‐2‐exo‐Phenyl‐2,3,4,5‐tetrahydro‐1H‐1,4‐epoxy‐1‐benzazepine, C16H15NO, (I), (2SR,4RS)‐2‐exo‐(4‐chlorophenyl)‐2,3,4,5‐tetrahydro‐1H‐1,4‐epoxy‐1‐benzazepine, C16H14ClNO, (II), and (2SR,4RS)‐2‐exo‐(3‐methylphenyl)‐2,3,4,5‐tetrahydro‐1H‐1,4‐epoxy‐1‐benzazepine, C17H17NO, (III), all crystallize with Z′ = 2, in the space groups Cc, P21/n and P21/c, respectively. In each of (II) and (III), the conformations of the two independent molecules are significantly different. The molecules in (I) are linked by C—H...π(arene) hydrogen bonds to form two independent chains, each containing only one type of molecule. The molecules in (II) are linked into sheets by a combination of C—H...O, C—H...(N,O) and C—H...π(arene) hydrogen bonds, all of which link pairs of molecules related by inversion, while in (III), the molecules are linked into sheets by a combination of C—H...N, C—H...O and C—H...π(arene) hydrogen bonds. There are no direction‐specific intermolecular interactions of any kind in the structure of (2SR,4RS)‐7‐bromo‐2‐exo‐phenyl‐2,3,4,5‐tetrahydro‐1H‐1,4‐epoxy‐1‐benzazepine, C16H14BrNO, (IV), but in the structure of (2SR,4RS)‐2‐exo‐(4‐bromophenyl)‐7‐chloro‐2,3,4,5‐tetrahydro‐1H‐1,4‐epoxy‐1‐benzazepine, C16H13BrClNO, (V), a combination of one C—H...N hydrogen bond and one C—H...O hydrogen bond links the molecules into sheets of alternating centrosymmetric R22(14) and R66(22) rings. 3‐Alkyl/aryl‐3‐ureido‐1H,3H‐quinoline‐2,4‐diones ( 2 ) and 3a‐alkyl/aryl‐9b‐hydroxy‐3,3a,5,9b‐tetrahydro‐1H‐imidazo[4,5‐c]quinoline‐2,4‐diones ( 3 ) react in boiling concentrated HCl to give 5‐alkyl/aryl‐4‐(2‐aminophenyl)‐1,3‐dihydro‐2H‐imidazol‐2‐ones ( 6 ). The same compounds were prepared by the same procedure from 2‐alkyl/aryl‐3‐ureido‐1H‐indoles ( 4 ), which were obtained from the reaction of 3‐alkyl/aryl‐3‐aminoquinoline‐2,4(1H,3H)‐diones ( 1 ) with 1,3‐diphenylurea or by the transformation of 3a‐alkyl/aryl‐9b‐hydroxy‐3,3a,5,9b‐tetrahydro‐1H‐imidazo[4,5‐c]quinoline‐2,4‐diones ( 3 ) and 5‐alkyl/aryl‐4‐(2‐aminophenyl)‐1,3‐dihydro‐2H‐imidazol‐2‐ones ( 6 ) in boiling AcOH. The latter were converted into 1,3‐bis[2‐(2‐oxo‐2,3‐dihydro‐1H‐imidazol‐4‐yl)phenyl]ureas ( 5 ) by treatment with triphosgene. All compounds were characterized by 1H‐ and 13C‐NMR and IR spectroscopy, as well as atmospheric pressure chemical‐ionisation mass spectra.  相似文献

Short pathways are described for the synthesis of a representative example of each of the 7,8‐dihydro‐and 1,2,3,4‐tetrahydro‐1,6‐naphthyridine‐5(6H)‐one ring systems from simple pyridine precursors. An attempted synthesis of the related 4,6‐dihydro‐1,6‐naphthyridin‐5(1H)‐one ring system from a common intermediate was unsuccessful.  相似文献

A series of 1‐acetyl(or phenyl)‐3‐aryl‐5‐(1‐phenyl‐3‐methyl‐5‐aryloxyl‐pyrazol)‐4,5‐dihydropyrazole derivatives have been efficiently synthesized under refluxing in glacial acetic acid with two kinds of hydrazines and five kinds of chalcones of 1‐phenyl‐methyl‐5‐phenoxyl‐pyrazol‐4‐aldehyde. The structures were confirmed on the basis of 1H NMR, IR, MS and element analysis, and the crystal structure of the compound 4c was determined by single crystal X‐ray diffraction. The compound 4c belongs to monoclinic system with space group P2(1)/n, a = 11.8779(3) nm, b = 12.0901(2) nm, c = 17.7004(4) nm, α = 90°, β = 100.05(10)°, γ = 90°, Formula weight: 549.46, Triclinic V = 2502.89(9) nm3, Dc = 1.458 Mg/m3, Z = 4, F (000) = 1128.  相似文献

Each such dimer makes rather short Br...Br contacts with two other dimers. In (2RS,4SR)‐2‐exo‐(5‐methyl‐2‐thienyl)‐2,3,4,5‐tetrahydro‐1H‐1,4‐epoxy‐1‐benzazepine, C15H15NOS, (II), a combination of C—H...O and C—H...π(thienyl) hydrogen bonds links the molecules into chains of rings. A more complex chain of rings is formed in (2RS,4SR)‐7‐chloro‐2‐exo‐(5‐methyl‐2‐thienyl)‐2,3,4,5‐tetrahydro‐1H‐1,4‐epoxy‐1‐benzazepine, C15H14ClNOS, (III), built from a combination of two independent C—H...O hydrogen bonds, one C—H...π(arene) hydrogen bond and one C—H...π(thienyl) hydrogen bond.  相似文献

This paper communicates the (regio) synthesis and a convenient NMR structural assignment method for N‐methyl‐tetrahydro‐5H‐indazol‐5‐one isomers. The cyclization reaction of 7‐(hydroxymethylene)‐1,4‐dioxaspiro[4,5]decan‐8‐one ( 3 ) with methylhydrazine yields, after de‐protection predominately the N‐2 methyl isomer 2 . Analysis of the product ratio and structural assignments are based on NMR data including NOE difference experiments and subsequently confirmed with X‐ray crystallography. These findings are in sharp contrast with the literature. The experimental conditions used to optimize the synthesis of the individual isomers are discussed.  相似文献

A number of 1‐substituted 4H,5H,6H‐[1,3]thiazolo[3,2‐a][1,5]benzodiazepinium‐11‐bromides and S‐(2‐oxo‐2‐phenyl‐X‐(p)‐ethyl)‐3‐(2‐methyl‐1H‐benzimidazol‐1‐yl) propane (or butane) thioate hydrobromides were obtained by direct reaction of the 5‐acetyl(or formyl, or anilinocarbonyl)‐substituted tetrahydro‐1,5‐benzodiazepine‐2‐thiones with aromatic α‐bromoketones. 2‐[(1‐Acetyl‐2(or 3)‐methyl‐2,3‐dihydro‐1H‐1,5‐benzodiazepin‐4‐yl) sulfanyl]‐1‐phenylethanones as intermediates of the formation of thiazolo [3,2‐a][1,5]benzodiazepine and N‐substituted 2‐methyl‐1H‐benzimidazole derivatives have been synthesized. Semiempirical AM1 calculations of a mechanism and energetic parameters for the heptatomic nucleus rearrangement to benzimidazole ring are presented. © 2008 Wiley Periodicals, Inc. The isomeric 2‐substituted‐7(5)‐methyl‐2,3‐dihydro‐5(7)H‐oxazolo[3,2‐a]pyrimidin‐5‐ones 3a‐b and 7‐ones 2a‐b,7a were synthesized by cyclocondensation from the 5‐substituted‐2‐amino‐2‐oxazolines 1a‐b with biselectrophiles. In boiling ethanol, the reaction of 1a‐b with acetylenic esters led to a mixture of 2a‐b,7a with a small amount of (E)‐2‐N‐(2‐ethoxycarbonylethylene)‐5‐substituted‐2‐iminooxazolines 5a‐b . The ring annulation between 1a‐b and diketene gave the 2‐substituted‐7‐hydroxy‐7‐methyl‐2,3,6,7‐tetrahydro‐5H‐oxazolo[3,2‐ a ]pyrimidin‐5‐ones 4a‐b which can be easily dehydrated to provide the 2‐substituted‐7‐methyl‐2,3‐dihydro‐5H‐oxazolo[3,2‐a]pyrimidin‐5‐ones 3a‐b .  相似文献

The bifunctional 3/4‐[acetyl]phenylsydnones 1a, 1b were subjected to a one‐pot ring conversion to 3‐[3/4‐acetyl]phenyl‐5‐methyl‐3H‐[1,3,4]‐oxadiazol‐2‐ones 2a, 2b , which on further bromination yielded the 3‐[3/4‐bromoacyl]phenyl‐5‐methyl‐3H‐[1,3,4]‐oxadiazol‐2‐ones 3a, 3b . Reaction of these compounds with thiourea yielded the 3‐[3/4‐(2‐aminothiazol‐4‐yl)]phenyl‐5‐methyl‐3H‐[1,3,4]‐oxadiazol‐2‐ones 4a, 4b . The other thiazole derivatives 5a, 5b–7a, 7b were prepared by using thiosemicarbazide, thioacetamide, and thiobenzamide, respectively. In another reaction of the bromoacetyl compounds ( 3a, 3b ) with 2‐aminopyridine and 2‐aminothiazole, the fused biheterocyclic compounds 3‐[3/4‐imidazo‐[1,2‐a]pyridine‐2‐yl]phenyl‐5‐methyl‐3H‐[1,3,4]‐oxadiazol‐2‐ones 8a, 8b and 3‐[3/4‐imidazo‐[2,1‐b]‐thiazol‐6‐yl]phenyl‐5‐methyl‐3H‐[1,3,4]‐oxadiazol‐2‐ones 9a, 9b were obtained. The 3‐[3/4‐(benzofuran‐2‐carbonyl)]phenyl‐5‐methyl‐3H‐[1,3,4]‐oxadiazol‐2‐ones 10a, 10b were obtained by treatment of compounds 3a, 3b with o‐hydroxy benzaldehyde. Most of these compounds exhibited antifungal activity greater than the reference drugs used. © 2007 Wiley Periodicals, Inc.
