The use of aliphatic polyesters for biomedical applications is limited by the lack of functionality of their backbones. The aim of the following study was to develop a novel elastic scaffold material containing functional groups to be used for future derivatization to tether peptide ligands to support cell adhesion, migration, and differentiation. The elastomer was based on three‐arm star copolymers composed of ε‐caprolactone and a functionalized ε‐caprolactone, 2‐oxepane‐1,5‐dione, and end‐terminated with acrylate groups. The elastomer thus contains a ketone and two approaches were examined for obtaining a photocrosslinkable elastomer containing functional groups: crosslinking followed by ketone reduction using sodium borohydride to generate pendant hydroxyl groups, and reaction of the ketone with hydrazines. Reduction of the ketone lead to degradation of the elastomer through transesterification and ethanolate mediated cleavage of the polymer backbone. Reaction with hydrazines did not degrade the polymer and resulted in efficient functionalization of the elastomer. © 2008 Wiley Periodicals, Inc. An efficient solid‐phase synthetic method for 2,3,4,5‐Tetrahydro‐1,4‐benzodiazepin‐2,5‐diones, having amine derivatives on the benzene ring, was developed. This method has been successfully applied to the synthesis of several spatially separated drug‐like and information‐rich small‐molecule libraries composed of 400 compounds using ACT‐496 automatic synthesizer and the IRORI radio frequency‐encoded split‐mix synthesis technology.  相似文献

Novel microwave induced method for the synthesis of thiazolidine‐2,4‐dione motif under solvent phase conditions is developed. Further we report an efficient, microwave assisted method for the parallel syntheses of biologically important 5‐benzylidene‐thiazolidine‐2,4‐dione and 5‐benzylidene‐2‐thioxothiazolidine‐4‐one compounds under solid‐phase and solvent‐free conditions. A comparative study between the developed microwave methods and the conventional methods is described. We have also illustrated the possible mechanism behind to address the reason why piperidine, acetic acid and silica gel enhanced the Knoevenagel condensation reaction.  相似文献

2a,4‐Disubstituted 5‐benzoyl‐2‐chloro/2,2‐dichloro‐2a,3,4,5‐tetrahydro‐azeto [1,2‐a] [1,5]benzodiazepin‐1 (2H)‐ones ( 3a–h ) were synthesized by cycloaddition reactions of 2,4‐disubstituted 1‐benzoyl‐2,3‐dihydr o‐1H‐1,5‐benzodiazepines ( 2a–h ) and ketenes, generated from chloroacetyl chloride or dichloroacetyl chloride in the presence of triethylamine, in anhydrous benzene. The relative stability of 1,4‐benzodiazepin‐2‐one tautomers in the gas phase and model solvents was calculated at the M06 and ωB97XD levels of theory. The two density functionals were benchmarked earlier and demonstrated as excellent models to study tautomerism in a vast array of chemical systems. A number of commercially available 1,4‐benzodiazepin‐2‐ones were investigated computationally for the first time. In addition, some biologically active and newly devised benzodiazepines were considered, which may be important in designing structures with desired (bio)chemical features. Special attention was paid to determine substituent effects on the Gibbs free energies of keto, enol, and iminol forms for each respective benzodiazepine. It was demonstrated that (i) the replacement of the benzene ring by the heterocyclic ring in the benzodiazepine system may stabilize the iminol tautomer, and (ii) the electron‐withdrawing substituent at the C3‐position of the respective benzodiazepine may stabilize the enol tautomer relative to the parent keto form. It is concluded that substituent effects may govern the chemical reactivity and biological properties of selected benzodiazepines.  相似文献

In the title potential O,N,N′,O′‐tetradentate Schiff base ligand {systematic name: 2,2′‐[pentane‐1,5‐diylbis(nitrilomethylidyne)]diphenol}, C19H22N2O2, the mutual orientation of the three planar fragments determines the conformation of the molecule. The dihedral angles between the planes of the two salicylidene groups and the plane of the central extended pentane chain are 78.4 (2) and 62.0 (3)°, and the angle between the terminal ring planes is 55.4 (1)°. Strong intramolecular O—H...N hydrogen bonds close almost‐planar six‐membered rings, and the O—H bonds are elongated as a result of hydrogen‐bond formation.  相似文献

The title compounds, 2‐(4‐bromo­phenyl)‐1,2‐di­hydro­pyrimido­[1,2‐a]­benzimidazol‐4‐(3H)‐one, C16H12Br­N3O, (IVa), and 4‐(4‐methylphenyl)‐3,4‐dihydropyrimido[1,2‐a]benzimidazol‐2‐(1H)‐one, C17H15N3O, (Vb), both form R(8) centrosymmetric dimers via N—H?N hydrogen bonds. The N?N distance is 2.943 (3) Å for (IVa) and 2.8481 (16) Å for (Vb), with the corresponding N—H?N angles being 129 and 167°, respectively. However, in other respects, the supra­molecular structures of the two compounds differ. Both compounds contain different C—H?π interactions, in which the C—H?π(centroid) distances are 2.59 and 2.47 Å for (IVa) and (Vb), respectively (the latter being a short distance), with C—H?π(centroid) angles of 158 and 159°, respectively. The supramolecular structures also differ, with a short Br?O distance of 3.117 (2) Å in bromo derivative (IVa), and a C—H?O interaction with a C?O distance of 3.2561 (19) Å and a C—H?O angle of 127° in tolyl system (Vb). The di­hydro­pyrimido part of (Vb) is disordered, with a ratio of the major and minor components of 0.9:0.1. The disorder consists of two non‐interchangeable envelope conformers, each with an equatorial tolyl group and an axial methine H atom.  相似文献

A series of 3‐(thiophen‐2‐yl)‐1,5‐dihydro‐2H‐pyrrol‐2‐one derivatives bearing a carbonic ester group were designed and synthesized by integrating a thiophene nucleus and a pyrroline‐2‐one scaffold in a single molecular architecture. Their structures were confirmed by IR, 1H‐NMR, EI‐MS, and elemental analyses, and their antifungal activities against Fusarium graminearum (Fg), Rhizoctorzia solani (Rs), and Botrytis cinerea (Bc) were evaluated. The antifungal bioassays indicated that some title compounds exhibited desirable antifungal effects against the tested fungi. Strikingly, the title compounds 4i , 4k , 4n , and 4o showed obvious antifungal activities against Rs, with corresponding EC50 values of 35.26, 33.56, 23.90, and 30.48 μg/mL, respectively, which are better than that of hymexazol (37.86 μg/mL). These results indicated that 3‐(thiophen‐2‐yl)‐1,5‐dihydro‐2H‐pyrrol‐2‐one derivatives bearing a carbonic ester group can serve as potential structural templates in the search for novel high‐efficient fungicides.  相似文献

This study represents a facile synthesis of building blocks ( 1–3 ) of crown ethers and amine precoursers ( 4a‐d ). The study also involves synthesis of mono and dibenzo N, N‐disubstituted diaza 18‐crown‐6 derivatives with high yield without chromatographic purification and high vacuum distillation.
