In this paper, we review methods for obtaining in vivo optical images of bio-objects with the resolution ranged from several units to tens of micrometers, namely, the methods of spectral-domain optical coherence tomography. We focus on problems that are specific of the spectral-domain optical coherence tomography, namely, the presence of coherent noise and mirror-symmetric phantoms in the image. On the basis of the most important relevant publications, we analyze possibilities of eliminating these drawbacks in modern devices.  相似文献

We describe a novel microscopy technique for quantitative phase-contrast imaging of a transparent specimen. The technique is based on depth-resolved phase information provided by common path spectral-domain optical coherence tomography and can measure minute phase variations caused by changes in refractive index and thickness inside the specimen. We demonstrate subnanometer level path-length sensitivity and present images obtained on reflection from a known phase object and human epithelial cheek cells.  相似文献

The spectroscopic content within optical coherence tomography (OCT) data can provide a wealth of information. Spectroscopic OCT methods are frequently limited by time-frequency trade-offs that limit high spectral and spatial resolution simultaneously. We present spectroscopic spectral-domain optical coherence microscopy performed with a multimodality microscope. Restricting the spatial extent of the signal by using high-numerical-aperture optics makes high-resolution spectroscopic information accessible, facilitated with spectral-domain detection. Simultaneous acquisition of multiphoton microscopy images is used to validate tissue structure and localization of nuclei within individual cells.  相似文献

Broadband interferometry is an attractive technique for the detection of cellular motions because it provides depth-resolved phase information via coherence gating. We present a phase-sensitive technique called spectral-domain phase microscopy (SDPM). SDPM is a functional extension of spectral-domain optical coherence tomography that allows for the detection of nanometer-scale motions in living cells. The sensitivity of the technique is demonstrated, and its calibration is verified. A shot-noise limit to the displacement sensitivity of this technique is derived. Measurement of cellular dynamics was performed on spontaneously beating cardiomyocytes isolated from chick embryos.  相似文献

We report the measurement of the hemoglobin (Hb) oxygen saturation level in human blood with a spectroscopic spectral-domain optical coherence tomography (SSD-OCT) system based on the crossover behavior of Hb and oxyhemoglobin (HbO(2)) absorption coefficients around 800 nm. By calculating the ratio of the exponential decay constant of A-mode scan signal in the long-wavelength range (>800 nm) over that in the short-wavelength range (<800 nm), the relative oxygen saturation level of Hb can be calibrated. Such a relative level can be used for practical diagnosis application after a golden standard is built by comparing the variation of the OCT result with that of a conventional method, such as the use of a blood gas analyzer. In our experiment, the variation curve based on the SSD-OCT measurement, which has the advantages of noninvasive, real-time, and high-resolution measurements, is coincident with that of using a commercial blood gas analyzer.  相似文献

In this paper, a new kind of method is developed to analyse the bit-error rate for a free-space optical communication system, which is based on the three most important parameters of scintillation statistics, i.e. fraction of time of fade and surge, mean frequency of level crossings, and mean duration of fade and surge. By using this method, the relationship between the bit-error rate and scintillation is clearly identified. The tip–tilt error deteriorates the performance of a free-space optical communication system. Fine tracking ability of adaptive optics (through tip–tilt compensation) can overcome this difficulty. The fade and surge statistical parameters with tip–tilt compensation are evaluated for the plane wave model. The bit-error rate as a function of the scintillation statistical parameters (with tip–tilt compensation) is developed. The results show that the bit-error rate is reduced notably through tip–tilt compensation for free-space optical communication.  相似文献

针对通过增加光源谱宽度来提高光学相干层析成像系统分辨率时样品色散特性的限制作用,分析了色散与光学相干层析成像系统纵向光程分辨率的约束关系.根据物质色散特性,采用数值变换方法对光学相干层析成像相干成像信号进行了色散补偿.实验使用了中心波长1 550 nm 自激发辐射光源和光纤迈克尓逊干涉结构,对水和光学快速扫描延迟线引入的二阶和三阶色散进行了数值补偿并通过相位修正因子来改善补偿效果.确定了一种普适的、快速的数值色散补偿方法.在对水中盖玻片和生物组织和的光学相干层析成像图像的色散补偿实验中取得了良好的效果,证明了方法的可行性.  相似文献

提出了基于互相关的多普勒OCT(correlated Doppler optical coherence tomography, CD-OCT)方法, 能够有效的抑制噪声, 实现低信噪比条件下的流速探测. 对CD-OCT算法进行了详细的推导, 分析了噪声的相关性对该算法结果的影响, 最后基于谱域和时域联合探测方法(joint spectral and time domain optical coherence tomography, STD-OCT)以及CD-OCT算法的对比实验证明了该算法能够进一步实现信噪比的提高, 使测量的结果更为稳定.  相似文献

We present a numerical method for compensating dispersion effects in optical coherence tomography that does not require a priori knowledge of dispersive properties of the sample. The method yields results equivalent to recently demonstrated quantum-optical coherence tomography, but without exploiting non-classical states of optical radiation. Dispersion compensation is accomplished by processing phase information present in standard interferograms to calculate the generalized autoconvolution function. The operation of the method can be conveniently visualized using the Wigner distribution function formalism.  相似文献

光学相干层析成像(OCT)系统的纵向分辨力不仅与光源的带宽有关,而且与系统中两干涉臂间的色散匹配有关。如果色散没有得到精确匹配,将使光学相干层析成像系统的纵向分辨力达不到所预期的理论值。色散问题在超高分辨光学相干层析成像系统中尤为突出。提出了一种基于双光栅快速扫描光学延迟线(RSOD),用于光学相干层析成像系统的色散补偿。该方法中新增的光栅引入了光栅间距这一独立变量,其与常规单光栅快速扫描光学延迟线机构中的光栅离焦量一起,可使光学相干层析成像系统中的群速度色散(GVD)和三阶色散(TOD)同时得到补偿。分析了双光栅快速扫描光学延迟线的色散特性和色散调节原则,并提供了一个典型光学相干层析成像系统中的色散补偿实例。  相似文献

We present a moving-scatterer-sensitive optical Doppler tomography (MSS-ODT) technique for in vivo blood flow imaging in real time by using a spectral-domain optical coherence tomography system. In MSS-ODT the influence of stationary scatterers is suppressed by subtracting adjacent complex axial scans before calculating the Doppler frequency shift. We demonstrate that MSS-ODT is a useful technique for accurate determination of blood vessel size by imaging flow in a small capillary tube with a 75 microm inner diameter. The flow profile obtained with MSS-ODT yields a substantially more accurate tube diameter than that obtained with the conventional phase-resolved method, which underestimates the diameter by about 23%. We also demonstrate that MSS-ODT provides improved sensitivity over the conventional phase-resolved method for imaging in vivo blood flow in small vessels in a mouse ear.  相似文献

Photoacoustic ophthalmoscopy(PAOM) is a novel imaging modality, which is capable of non-invasively detecting optical absorption properties in the retina. We visualize the microvasculature in retina and choroid in albino mouse using PAOM guided by spectral-domain optical coherence tomography. Since albino mouse characterizes by lacking melanin in retinal pigment epithelium(RPE), PAOM illumination laser can penetrate through the RPE onto choroid, and consequently provides volumetric visualization of chorioretinal vasculatures as a result of strong hemoglobin optical absorption. The high-quality chorioretinal microvascular imaging acquired by PAOM implies its great potential in understanding pathological mechanisms and developing therapeutic strategies for major chorioretinal diseases that correlate with vascular disorders.  相似文献

Multiangle, fiber-based, spectral-domain Doppler optical coherence tomography with a phase-resolved algorithm is presented to measure three components of an arbitrary velocity vector. A beam divider that divides a probe beam to have five independent viewpoints and path length delays was designed. The divider was inserted into the sampling arm of a Doppler optical coherence tomography system between the collimator and the first galvo mirror of a two-axis galvo scanner. The divider produced five independent D k's (the average difference between the wave vectors of incoming and outgoing beams) after passing through the focusing lens while keeping two-axis scanning capability. After calibration, an unknown velocity vector field inside a microtube was quantified by solving a three-dimensional minimization problem.  相似文献

A high resolution spectral-domain optical coherence tomography (SD-OCT) system based on a thermal light source was presented. A novel normalized method was introduced to remove the background noise and the DC noise of the interference spectrum. A Gaussian spectral calibration procedure was performed to improve axial resolution and image quality before reconstructing OCT image. With the proposed method, the quality of the obtained images was greatly improved. Two-dimensional (2D) cross-sectional images with axial resolution of 1.2?μm were obtained. Furthermore, the film thickness of single-layer film sample was obtained. The experimental result demonstrates the SD-OCT system has potential for film thickness measurement and surface topography.  相似文献

We report in this paper the development of a spectral-domain optical coherence vibration tomography (OCVT) using a broadband CCD-based spectrometer and a short-coherence white light source. We demonstrate that both the vibration amplitude and frequency can be quantified, in the frequency range 0-250 Hz, with an axial resolution of 1 μm. Furthermore, the inner structure (layer thickness) of a vibrating sample can also be quantified simultaneously. The developed OCVT is non-contact and noninvasive in nature, thus is ideal for real time and in situ monitoring of low-frequency micro-vibrations that have critical impacts on many high-precision manufacturing and engineering processes.  相似文献

We propose a new method for authenticating valuable paper sheets such as gift coupons, tickets, and cheques with the use of optical coherence tomography (OCT). This method is based on the detection and evaluation of cross-sectional structural characteristics of valuable paper sheets provided with security features. Commonly used as a security feature is a security thread, which is embedded in such paper sheets. In this paper, the nondestructive detection of the embedded security thread is demonstrated using spectral-domain OCT. The results showed that the security thread was clearly visualized in the OCT image despite the highly scattering nature of papers.  相似文献

We propose and demonstrate a method of real-time dispersion compensation suitable for scanning interferometry and optical coherence tomography. Static grating tilt in a scanning frequency-domain optical delay line is shown to produce dispersion that is linearly proportional to scan position, and we use this property to achieve depth-dependent dispersion compensation during an interferometric scan through a dispersive sample.  相似文献

光学层析成像是一个病态重建问题,为克服重建过程的病态性,提出将多准则优化理论引入到图像重建中。利用了三个用于光学层析图像重建的准则:平方误差函数、图像熵函数和局部平滑函数。采用向量优化方法将多准则优化问题转化为单准则优化问题求解。为了确定各个目标函数间的权重系数,提出一种动态权重系数求解方法。重建过程目标函数关于光学参量的梯度计算是关键,因此提出一种基于梯度树的计算方法。实验过程中对多准则重建结果和基于平方误差函数的单准则重建结果做了比较,证明该方法能够克服传统的偏重单一目标的单准则重建的不足,有效地重建光学层析图像,提高图像重建质量。  相似文献
