Nanophase separation has been suggested to influence the biological performance of polyurethane. In a previous work, six different 4,4'-diphenylmethane diisocyanate (MDI)-based poly(carbonate urethane)s (PCUs) that exhibited various degrees of nanophase separation were synthesized and characterized. In the present work, these PCUs were used as a model system to study the effect of nanometric structures on the biocompatibility of polyurethane. Human blood platelet activation, monocyte activation, protein adsorption, and bacterial adhesion on PCU were investigated in vitro. It was found that human blood platelets as well as monocytes were less activated on the PCU surfaces with a greater degree of nanophase separation in general. This phenomenon was closely associated with the lower ratio of human fibrinogen/albumin competitively adsorbed on these surfaces. A series of biomedical polyurethane elastomers (PURs) based on poly(ester‐carbonate)s (PECs) and polycarbonates (PCs) were synthesized and spectrally characterized fully. PEC or PC diols were synthesized by the ring‐opening polymerization of ε‐caprolactone, trimethylene carbonate, and neopentyl carbonate catalyzed by lipase from Candida antarctica. PURs were prepared by free‐metal method from PEC or PC diols and 4,4′‐methylenebis(phenyl isocyanate), with 1,4‐butanediol as a chain extender. The physical and mechanical properties as well as hydrolytic stability of the obtained PURs were determined. The toxicity of the received polymers was evaluated using bacterial luminescence test and two protozoans assays. The presented preliminary studies suggest that PEC or PC diols prepared in this way might be applied for the synthesis of biomedical PURs with improved hydrolytic stability. © 2012 Wiley Periodicals, Inc. Novel enzymatically recyclable poly(carbonate-urethane) consisting of a diurethane moiety as a hard segment and a carbonate linkage as an enzymatically cleavable unit was prepared by the polycondensation of biodegradable diurethanediol and diethyl carbonate using lipase. The produced poly(carbonate-urethane) was readily transformed by lipase into the corresponding cyclic oligomers which were more easily repolymerized by lipase to produce a higher molecular weight poly(carbonate-urethane) than that of the parent poly(carbonate-urethane). 相似文献

Effect of hydrolysis time on molar mass, glass transition temperature, crystallinity, and resistance to thermooxidation at elevated temperatures was analyzed for Estanes 54600, 54610, and 54650. Kinetics of the hydrolysis can be plausibly described in terms of the first-order reaction with an average induction period of about 7 days. Reduction of molar mass induced by hydrolysis brings about an appreciable decrease in glass transition temperature, fraction of crystalline domains of soft segments, and thermooxidative stability. The latter effect is manifested by shortening of the lifetimes (related to 5% mass loss) the temperature dependence of which obeys the Arrhenius plot. The observed differences in hydrolysis resistance of Estanes can be related to their chemical composition. 相似文献

The temperature dependence of thermal, morphological, and rheological properties of amphiphilic polyurethanes was examined with differential scanning calorimetry (DSC), wide‐angle X‐ray scattering (WAXS), small‐angle X‐ray scattering (SAXS), rheological measurements, and Fourier transform infrared spectroscopy. Multiblock (MPU) and triblock (TPU) polyurethanes were synthesized with two crystallizable segments—poly(ethylene oxide) (PEO) as a hydrophilic block and poly(tetramethylene oxide) (PTMO) as a hydrophobic block. DSC and WAXS measurements demonstrated that the microphase of MPUs in the solid state is dominantly affected by the PEO crystalline phase. However, high‐order peaks were not observed in the SAXS measurements because the crystallization of the PEO segments in MPUs was retarded by poor sequence regularity. The microphase in the melt state was induced by the hydrogen bonding between the N? H group of hexamethylene diisocyanate linkers and the ether oxygen of PEO or PTMO blocks. As the temperature increased, the smaller micro‐phase‐separated domains were merged into the larger domains, and the liquidlike ordering was eventually disrupted because of the weakening hydrogen bonding. However, the fully homogeneous state of an MPU with a molar ratio of 5/5 PEO/PTMO (MPU55) was not confirmed even at much higher temperatures with both SAXS and rheological measurements. However, the SAXS patterns of TPU showed weak but broad second‐order peaks below the melting temperature of the PEO block. Compared with MPU55, the ordering of the TPU crystalline lamellar stacks was enhanced because of the high sequence regularity and the low hydrogen‐bonding density. © 2003 Wiley Periodicals, Inc. Segmented poly(ester-siloxane)urethanes containing hard and soft segments were prepared by melt polyaddition using a multistep procedure. The soft segments based on poly(ethylene glycol adipate)diol as ester sequences and α,ω-poly(dimethylsiloxane)diol as silicone sequences were synthesized. The hard segments were based on aromatic or aliphatic diisocyanates and butanediol was used as chain extender. These polyurethane materials were analysed by differential scanning calorimetry and dynamic-mechanical analysis measurements. The shapes of thermal plots are significantly influenced by the soft and hard segment structures and also the compositional parameters. 相似文献

In this work, we synthesized several MDI-based poly(carbonate)urethanes (PCU) by using four different soft segments, including two aliphatic macrodiols (poly(hexyl, ethyl)carbonate diols, MW 2017 and 865, respectively) and two aromatic macrodiols (MW approximately 2000 and 1000, respectively), in different molar ratios to MDI. We demonstrate that these polymers exhibited various degree of micro-phase separation that further influenced their surface protein adsorption, platelet activation as well as cellular attachment and growth. Polyurethanes based on poly(hexyl, ethyl)carbonate diol with MW 2017 in a molar ratio MDI/macrodiol/chain extender of either 3/2/1 or 4/3/1 resulted in greater micro-phase separation as well as superior biocompatibility and biostability. 相似文献

A novel synthesis of poly(ethylene glycol) (PEG)-grafted poly(urethanes) (PURs) is described based on a precursor PUR containing free amino groups in the main chain. Three different poly(urethane) backbones were prepared: a homopoly(urethane) comprised of N-Bocdiethanolamine (BDA) and 4,4′-methylenebis(phenyl isocyanate) (MDI), a copoly(urethane) (COPUR) consisting of BDA, N-benzyldiethanolamine and MDI, and a poly(urethane urea) (PUU) that was prepared from BDA, MDI, and ethylenediamine as the chain extender. The Mn of these poly(urethanes) ranged from 32,000 to 72,000 g/mol. PEG (750, 1,900, and 5,000 g/mol) was grafted onto the boc-deprotected poly(urethanes) via the chloroformate. Films of the polymers were spin cast from dilute solutions, annealed, and the surfaces analyzed by goniometry. Water contact angle data indicates increasing PEG surface coverage of the poly(urethanes) with increasing PEG molecular weight. Reorientation of the polymer films is evidenced by contact angle hysteresis. Polymer thrombogenicity, which was studied using blood perfusion experiments, shows that COPUR-g-PEG5000 and PUU-g-PEG5000 exhibit very little platelet adhesion. © 1999 John Wiley & Sons, Inc. In this article a new technique was developed to fabricate scaffolds with a unique microstructure by solid–liquid separation in combination with particulate-leaching. Firstly, the effects of polymer concentration, quenching temperature on the porous morphology and the mechanical property of obtained scaffolds during solid–liquid separation have been investigated. Then, salt granules as porogen were introduced into the solid–liquid phase separation to produce the unique pore structure of the scaffold. The pore diameter of the scaffold could be controlled with the particulate size and the wall of pores possessed special microstructure, which enhanced the pore interconnectivity. The cell culture results confirmed that a good interconnectivity of the scaffold prepared by the improved solid–liquid separation was useful for nutrition transportation and cell proliferation. Novel poly(ester carbonate)s were synthesized by the ring‐opening polymerization of L ‐lactide and functionalized carbonate monomer 9‐phenyl‐2,4,8,10‐tetraoxaspiro[5,5]undecan‐3‐one derived from pentaerythritol with diethyl zinc as an initiator. 1H NMR analysis revealed that the carbonate content in the copolymer was almost equal to that in the feed. DSC results indicated that Tg of the copolymer increased with increasing carbonate content in the copolymer. Moreover, the protecting benzylidene groups in the copolymer poly(L ‐lactide‐co‐9‐phenyl‐2,4,8,10‐tetraoxaspiro[5,5]undecan‐3‐one) were removed by hydrogenation with palladium hydroxide on activated charcoal as a catalyst to give a functional copolymer, poly(L ‐lactide‐co‐2,2‐dihydroxylmethyl‐propylene carbonate), containing pendant primary hydroxyl groups. Complete deprotection was confirmed by 1H NMR and FTIR spectroscopy. The in vitro degradation rate of the deprotected copolymers was faster than that of the protected copolymers in the presence of proteinase K. The cell morphology and viability on a copolymer film evaluated with ECV‐304 cells showed that poly(ester carbonate)s derived from pentaerythritol are good biocompatible materials suitable for biomedical applications. © 2007 Wiley Periodicals, Inc. Composites of poly(propylene carbonate) (PPC) reinforced with micrometric and nanometric calcium carbonate particles were prepared via melt mixing followed by compression molding. The morphology and mechanical and thermal behaviors of the composites were investigated. Static tensile tests showed that the tensile strength, stiffness, and ductility of the composites tended to increase with increasing contents of micrometric calcium carbonate particles. This improvement in the tensile properties was attributed to good interfacial adhesion between the fillers and matrix, as evidenced by scanning electron microscopy examination. However, because of the agglomeration of calcium carbonate nanoparticles during blending, those composites with nanoparticles exhibited the lowest tensile strength. Thermogravimetric measurements revealed that the incorporation of calcium carbonate into PPC resulted in a slight improvement in its thermooxidative stability. © 2003 Wiley Periodicals, Inc. AB‐type block copolymers with poly(trimethylene carbonate) [poly(TMC); A] and poly(ethylene oxide) [PEO; B; number‐average molecular weight (Mn) = 5000] blocks [poly(TMC)‐b‐PEO] were synthesized via the ring‐opening polymerization of trimethylene carbonate (TMC) in the presence of monohydroxy PEO with stannous octoate as a catalyst. Mn of the resulting copolymers increased with increasing TMC content in the feed at a constant molar ratio of the monomer to the catalyst (monomer/catalyst = 125). The thermal properties of the AB diblock copolymers were investigated with differential scanning calorimetry. The melting temperature of the PEO blocks was lower than that of the homopolymer, and the crystallinity of the PEO block decreased as the length of the poly(TMC) blocks increased. The glass‐transition temperature of the poly(TMC) blocks was dependent on the diblock copolymer composition upon first heating. The static contact angle decreased sharply with increasing PEO content in the diblock copolymers. To investigate by microscopic observation the effects on cell behaviour of immobilized polymers, a micropattern-immobilization technique using a photo-mask was employed. Poly(acrylic acid) or poly(N-isopropylacrylamide-co-acrylic acid) was coupled with azidophenyl groups to form a photo-reactive polymer. The photo-reactive polymer was coated, with or without gelatin, on a cell-culture polystyrene plate and photo-irradiated through a micropatterned photo-mask. Mouse fibroblast STO cells were cultured on the micropattern-immobilized plate. The surface wettability of the immobilized plate was examined by measurement of the contact angle in the cell culture medium. The attachment of cells on the plate was significantly affected by the surface properties. Although the poly(acrylic acid) has the same effect on cell adhesion as a bare polystyrene surface, co-immobilization with gelatin significantly enhanced cell adhesion, while poly(N-isopropylacrylamide) reduced it. However, co-immobilization with gelatin enhanced cell adhesion, and, on the co-immobilized surface, cell detachment was observed by lowering the temperature. Photoresponsive amphiphilic diblock poly(carbonate)s mPEG113‐b‐PMNCn with pendent o‐nitrobenzyl ester group were synthesized through ring‐opening polymerization (ROP) using 1,8‐diazabi‐cyclo[5.4.0]undec‐7‐ene (DBU) as catalyst and monomethoxy poly(ethylene glycol) (mPEG) as macroinitiator. In aqueous solution, the copolymers can self‐assemble to spherical micelles with a PC core and a PEG shell. The critical micelle concentration (CMC), size, and morphology of the micelles were demonstrated by means of fluorescence spectroscopy, transmission electron microscopes (TEM), and dynamic light scattering (DLS). Under UV light irradiation, the amphiphilic copolymer micelles disassembled because of the photocleavage of o‐NB ester, and the light‐controlled release behaviors of payload Nile red were further proved. The composites of biodegradable poly(propylene carbonate) (PPC) reinforced with short Hildegardia populifolia natural fiber were prepared by melt mixing followed by compression molding. The mechanical properties, thermal properties, and morphologies of the composites were studied via static and dynamic mechanical measurements, thermogravimetric analysis, and scanning electron microscopy (SEM) techniques, respectively. Static tensile tests showed that the stiffness and tensile strength of the composites increased with an increasing fiber content. However, the elongation at break and the energy to break decreased dramatically with the addition of short fiber. The relationship between the experimental results and the compatibility or interaction between the PPC matrix and fiber was correlated. SEM observations indicated good interfacial contact between the short fiber and PPC matrix. Thermogravimetric analysis revealed that the introduction of short Hildegardia populifolia fiber led to a slightly improved thermooxidative stability of PPC. © 2004 Wiley Periodicals, Inc. A series of novel poly(ester‐carbonate)s bearing pendant allyl ester groups P(LA‐co‐MAC)s were prepared by ring‐opening copolymerization of L ‐lactide (LA) and 5‐methyl‐5‐allyloxycarbonyl‐1,3‐dioxan‐2‐one (MAC) with diethyl zinc (ZnEt2) as initiator. NMR analysis investigated the microstructure of the copolymer. DSC results indicated that the copolymers displayed a single glass‐transition temperature (Tg), which was indicative of a random copolymer, and the Tg decreased with increasing carbonate content in the copolymer. Then NHS‐activated folic acid (FA) first reacted with 2‐aminoethanethiol to yield FA‐SH; grafting FA‐SH to P(LA‐co‐MAC) in the presence of TEA produced P(LA‐co‐MAC)/FA. The structure of P(LA‐co‐MAC)/FA and its precursor were confirmed by 1H NMR and XPS analysis. Cell experiments showed that FA‐grafted P(LA‐co‐MAC) had improved adhesion and proliferation behavior of vero cells on the polymer films. Therefore, the novel FA‐grafted block copolymer is expected to find application in drug delivery or tissue engineering. © 2008 Wiley Periodicals, Inc. The phase diagram of blends of poly(vinylidene fluoride) and poly (ethyl acrylate) was established by X-ray scattering, optical microscopy and calorimetric techniques. Structure formation, involving phase separation and coarsening was analyzed as a function of temperature variations and annealing times. The nanostructure of a biomaterial surface has strong influence on cell behavior. The migration of cells on nanostructured surfaces, however, has not been investigated so far. In this study, we used PCL/PEG diblock copolymers as model surfaces to examine the effect of nanoislands on migration of different cells, including fibroblasts and endothelial cells. The water sliding angle of the substrates was measured. The cell migration rate was examined under a real-time optical microscope. It was found that a greater cell migration rate correlated with the smaller sliding angle of the substrate. 相似文献
