2-Substituted [1]benzopyrano[4,3-<Emphasis Type="Italic">d</Emphasis>]pyrimidin-5-ones

A new method is reported for the synthesis of 2-R-substituted [1]benzopyrano[4,3-d]pyrimidin-5-ones (R = p-phenyl, p-chlorophenyl-, p-carbamoylphenyl-, 3-pyridinyl-, 4-pyridinyl-, 2-pyrazinyl-, pyrrolidino-,morpholino-, 3,6-dimethyl-1-pyrazolyl-) by the reaction of 4-chloro-3-formylcoumarin with salts of the corresponding amidines RC(=NH)NH₂HX in DMF in the presence of piperidine or triethylamine. 3-Formyl-4-coumarin and 2-aminobenzimidazole give __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 6, pp. 935–940, June, 2007.     

Oxidative cyclocondensation of cyclic thio-and selenoureas. 5. 2-Thioxothieno-and 2-thioxopyrido[2,3-<Emphasis Type="Italic">d</Emphasis>]pyrimidin-4-ones

A study was carried out on the oxidative cyclocondensation of 2-thioxothieno-and 2-thioxopyrido[2,3-d]pyrimidin-4-ones. The thiophene ring with excess π-electron density facilitates the reaction, while the pyridine ring with diminished π-electron density hinders it. 2-Thioxothieno-[2,3-d]pyridimin-4-ones were converted into previously unreported 7H,13H-[1,2,4]thiadiazolo-[3,2-b:5,4-b′]bis(thieno[2,3-d]pyridimine-7,13-diones). __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 9, pp. 1424–1427, September, 2007.     

Synthesis of pyrido[2,3-<Emphasis Type="Italic">d</Emphasis>]pyrimidin-7(8<Emphasis Type="Italic">H</Emphasis>)-one derivatives from 5-acetyl-4-aminopyrimidines and β-dicarbonyl compounds

Heating of 5-acetyl-4-aminopyrimidine derivatives with ethyl acetoacetate, ethyl benzoylacetate, and diethyl acetone-1, 3-dicarboxylate in the absence of a base gave the corresponding 6-acylpyrido[2, 3-d]pyrimidin-7(8H)-ones. Under analogous conditions, the reaction with ethyl malonate afforded ethyl 7-oxo-7, 8-dihydropyrido[2, 3-d]pyrimidine-6-carboxylates. The pyridone (rather than hydroxypyridine) structures of the pyridopyrimidines obtained were confirmed by IR spectroscopy.__________Published in Russian in Izvestiya Akademii Nauk. New method for the synthesis of thieno[2,3-<Emphasis Type="Italic">d</Emphasis>]pyrimidines

The reaction of 5-formyl-4-thiocyanatopyrimidines with nitromethane was studied under various conditions. The reaction was found unexpectedly to proceed with closure of a thiophene ring to give thieno[2,3-d]pyrimidines. The use of ammonium acetate as the catalyst leads to a side reaction involving closure of an isothiazole ring to give isothiazolo[5,4-d]pyrimidines. X-ray diffraction crystallographic analysis was used to confirm the structure of thieno[2,3-d]pyrimidine.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 10, pp. 1564–1571, October, 2004.     

Fluorine-containing pyrido[1,2-<Emphasis Type="Italic">a</Emphasis>]quinazolin-6-ones

Reactions of 2-aminopyridine and 2-amino-5-methylpyridine with 2,3,4,5-tetrafluorobenzoyl chloride afforded N,N’-diaroylpyridinium salts, which were converted into 6H-pyrido[1,2-a]quinazolin-6-ones by refluxing in toluene in the presence of triethylamine. The angular structure of the tricyclic derivatives obtained was confirmed by ¹⁹F and ¹³C NMR spectroscopy and 2D heteronuclear HetCOR and HMBC experiments.__________Published in Russian in Izvestiya Akademii Nauk. Synthesis of 2,3-disubstituted derivatives of pyrano-, thiopyrano-, and benzoannelated pyrido[2,3-<Emphasis Type="Italic">b</Emphasis>]thieno[3,2-<Emphasis Type="Italic">d</Emphasis>]pyrimidines

A new methods have been developed for the synthesis of condensed pyrido[2,3-b]thieno[3,2-d]pyrimidines based on cyclic derivatives of 4-cyanopyridine-3-thiones. Synthesis and properties of pyrimido[4,5-<Emphasis Type="Italic">a</Emphasis>]- and pyrido[4,3-<Emphasis Type="Italic">a</Emphasis>]carbazole derivatives

Methods for the synthesis of substituted pyrimido [4,5-a]- and pyrido[4,3-a]carbazoles were proposed. Condensation of 2-(dimethylaminomethylene)-6-methyl-2,3,4,9-tetrahydro-1H-carbazol-1-one with guanidine and thiourea afforded 2-amino-8-methyl-6,11-dihydro-5H-pyrimido[4,5-a]carbazole and 8-methyl-3,5,6,11-tetrahydro-2H-pyrimido[4,5-a]carbazole-2-thione, respectively. The reaction of cyano(6-methyl-2,3,4,9-tetrahydro-1H-carbazol-1-ylidene)acetamide with dimethylformamide dimethyl acetal gave N-(dimethylamino-methylene)cyano(6-methyl-2,3,4,9-tetrahydro-1H-carbazol-1-ylidene)acetamide. Cyclization of the latter yielded 1-cyano-8-methyl-3,5,6,11-tetrahydro-2H-pyrido[4,3-a]carbazol-2-one.__________Published in Russian in Izvestiya Akademii Nauk. Ecofriendly synthesis of substituted pyridine and pyrido[2,3-<Emphasis Type="Italic">d</Emphasis>]pyrimidine derivatives

An environmentally benign novel one-pot synthesis of pyridines and pyrido[2,3-d]pyrimidines from chalcones and malononitrile was described. Heterocyclic analogs of 5,12-naphthacenequinone 6. Synthesis of 4,11-dimethoxy derivatives of anthra-[2,3-<Emphasis Type="Italic">b</Emphasis>]thiophene-5,10-dione and anthra[2,3-<Emphasis Type="Italic">d</Emphasis>]isothiazole-5,10-dione

Condensation of 2-formyl-or 2-cyano-3-chloro-1,4-dimethoxyanthraquinone with methyl thioglycolate in the presence of base gave methyl 4,11-dimethoxyanthra[2,3-b]thiophene-5,10-dione-2-carboxylate and its 3-amino derivative respectively. Hydrolysis of the ester group in methyl 4,11-dimethoxyanthra[2,3-b]thiophene-5,10-dione-2-carboxylate and subsequent decarboxylation of the carboxylic acid formed gave 4,11-dimethoxyanthra[2,3-b]thiophene-5,10-dione. Tandem transformations of tetrahydropyrrolo[3,2-<Emphasis Type="Italic">c</Emphasis>]pyridines under the action of dimethyl acetylenedicarboxylate. A novel route to pyrrolo[2,3-<Emphasis Type="Italic">d</Emphasis>]azocines

Reactions of substituted tetrahydropyrrolo[3,2-c]pyridines with dimethyl acetylenedicarboxylate in protic and aprotic solvents were studied. Ionic Liquid Promoted One-Pot Synthesis of Furo[2,3-<Emphasis Type="Italic">d</Emphasis>]pyrimidine-2,4(1<Emphasis Type="Italic">H</Emphasis>,3<Emphasis Type="Italic">H</Emphasis>)-diones

Summary. The three-component condensation of aldehyde, N,N′-dimethylbarbituric acid and alkyl or aryl isocyanide afforded the corresponding furo[2,3-d]pyrimidine-2,4(1H,3H)-diones in 1-butyl-3-methylimidazolium bromide as an ionic liquid in high yields at room temperature within several minutes.     

Pyrano[4,3-<Emphasis Type="Italic">d</Emphasis>]pyrimidinium salts

Construction of a pyrido[4,3-<Emphasis Type="Italic">d</Emphasis>]pyrimidine system on the basis of <Emphasis Type="Italic">N</Emphasis>-cyanobenzamidine and diethyl acetone-1,3-dicarboxylate

Nickel acetate promoted reaction of diethyl acetone-1,3-dicarboxylate with N-cyanobenzamidine led to ethyl 4-amino-6-ethoxycarbonylmethyl-2-phenylpyrimidine-5-carboxylate, upon treatment of which with RNH₂, the corresponding 6-(carbamoylmethyl)pyrimidines were obtained. Cyclization of the latter upon treatment with MeONa afforded 6-R-4-amino-7-hydroxy-2-phenylpyrido[4,3-d]pyrimidin-5(6H)-ones. Published in Russian in Izvestiya Akademii Nauk.
