The effect of urea on the conformational behavior of poly(N-isopropylacrylamide) (PNIPAM) in dilute aqueous solution has been investigated using fluorescence spectroscopy, fluorescence quenching and fluorescence anisotropy measurements via pyrene (Py) probe and acenaphthylene (ACE) label studies. It was demonstrated that urea promotes the partitioning of the hydrophobic probe, Py, towards the bulk aqueous phase at temperatures above the lower critical solution temperature (LCST) of the polymer due to swelling of the compact coil conformation. However, the compact coil structure of the polymer at temperatures greater than its LCST is not completely destroyed, even for urea concentrations up to 3 M, at which the phase transition is hardly observed. As expected, urea has little effect on the conformational behavior of PNIPAM at temperatures below its LCST. Received: 9 February 2000/Accepted: 13 June 2000  相似文献

Silver nanoparticles (Ag NPs) stabilized by a thermoresponsive polymer, poly(N-isopropylacrylamide) (PNIPAM), have been synthesized by the reduction of silver ions with NaBH(4) in aqueous solutions. The obtained Ag NPs are very stable at room temperature due to the extended coil conformation of the PNIPAM chain at temperatures below its volume phase transition temperature ( approximately 32 degrees C). At higher temperatures (such as 45 degrees C) above the phase transition of PNIPAM, only minute aggregation between Ag NPs was observed, showing that the collapsed PNIPAM chains still retain the ability to stabilize Ag NPs. The PNIPAM-stabilized Ag NPs were then characterized as a function of the thermal phase transition of PNIPAM by UV-vis spectroscopy, dynamic light scattering, transmission electron microscopy, and cyclic voltammeter. Consistent results were obtained showing that the phase transition of PNIPAM has some effect on the optical properties of Ag NPs. Switchable electrochemical response of the PNIPAM-stabilized Ag NPs triggered by temperature change was observed.  相似文献

An imaging method has been developed to examine thermo responsive polymer coagulates by optical and electron microscopy. Poly-N-isopropylacrylamide (PNIPAM), poly-N-dimethylacrylamide (PDMAM) and a 1:1 PNIPAM-PDMAM copolymer were encapsulated in a gelatin matrix as coagulates above the lower critical solution temperature (LCST), and subsequently examined by optical and electron microscopy. The linear macromolecules PNIPAM and PDMAM were synthesized by chain transfer polymerization with mercaptopropionic acid (3-MPA) as chain transfer reagent. The resulting polymers have an average molar mass of ∼1800 g/mol and low polydispersity. The LCST of thermo responsive polymers is defined in pure water but can also be stimulated at lower than the phase transition temperature employing electrolytes containing inorganic salts such as (NH4)2SO4. Under such conditions the polymers show the typical thermo responsive phase transfer property in form of a visible clouding point. Gelatin was used to maintain this biphasic state by slowly adding water-softened gelatin sheets at a temperature above the LCST, followed by cooling to 3 °C in order to induce gelation. Examination of the gelatin-coagulate matrices by optical and electronic microscopy showed that PNIPAM and its copolymer (PNIPAM/PDMAM 1:1) are entrapped as globular spheres and clusters of spheres. In comparison pure PDMAM, even if it shows a clouding point, does not form typical LCST coagulates. With PNIPAM and the copolymer, micro globule formation is also possible with slow gelatin formation, without first provoking an LCST. In this particular case, the phase transition, or entropic demixing of the polymers respectively, are induced in this case by water absorption of the gelatin matrix.  相似文献

Poly(N-isopropylacrylamide) (PNIPAM) has a low critical solution temperature (LCST) at 32°C in water and the hydrophilicity changes through the LCST. The microspheres whose surface was composed of PNIPAM exhibited phase transition behavior around 32°C. Therefore, the interactions between PNIPAM micropheres and granulocytes depended on the temperature. That is, the oxygen consumption and active oxygen production by cells in contact with PNIPAM-containing microspheres and adhesion of the microspheres to the cell surface were more enhanced above the LCST of PNIPAM than below it, whereas no significant temperature dependence of cell–microspheres interaction was observed in nonthermosensitive microsphere systems. It was suggested that the function of cells could be controlled with temperature using the temperature-sensitive microspheres.  相似文献

Poly(N,N‐diethylacrylamide) (PDEA) possesses a lower critical solution temperature (LCST) in aqueous media. The solution properties of PDEA at various temperatures have been characterized with techniques such as rheology and dynamic light scattering. There is a decrease in the coil size before the phase transition due to a coil‐to‐globule transition. At the LCST, rheological and dynamic light scattering studies have also confirmed an aggregation phenomenon. This aggregation modifies the rheological properties of the polymer solutions. Fully atomistic molecular dynamics simulations of poly(2‐[2‐methoxyethoxy]ethyl methacrylate) (PMEO2MA) in water at temperatures below and above its lower critical solution temperature (LCST) were performed to improve the understanding of its LCST behavior. Atomic trajectories were used to calculate various structural and dynamic properties. Simulation results show that PMEO2MA undergo a distinct coil‐to‐globule transition above LCST. Detailed analyses of the number of first hydration shell water molecules around various atomic regions are revealed that the water solubility of PMEO2MA below LCST is mainly provided by the hydrophobic hydration around the side chain carbon atoms. This is achieved by the cage‐like water network formations which are disrupted when the temperature is increased above LCST, accompanied by significant amount of water molecule release and local water‐ordering reduction, which leads to the LCST phase transition. Furthermore, other analyses such as the number of hydrogen bonds and hydrogen bond lifetimes suggest that intermolecular hydrogen bondings between polymer and water molecules have little effect on the phase transition. Our results will contribute to a better understanding on the LCST phase transition of oligo(ethylene glycol) methyl ether methacrylate (OEGMA)‐based homopolymers at atomistic level that will be useful when designing homo‐ and co‐polymers of OEGMAs with desired properties. © 2017 Wiley Periodicals, Inc. 窄分散的聚(N 异丙基丙烯酰胺)水凝胶微球用乳液聚合方法制备,并用动态和静态光散射对其体积相变进行了研究.与水中聚(N 异丙基丙烯酰胺)线性单链比较,水中凝胶微球的体积相变温度较高,对温度的响应比较平缓.相变是连续的,有别于大块凝胶非连续的体积变化.在体积相变过程中,凝胶微球始终是密度均一的热力学稳定球体.从相变过程网络密度的变化可以确定,绝大部分的水在收缩过程被排了出来,但在紧缩的凝胶微球中仍含有约70%的水.  相似文献

The phase transition of poly(N‐isopropylacrylamide) (poly(NIPAM)) solutions was characterized by a dynamic fluid rheometer. Two critical points were observed below the lower critical solution temperature (LCST; ∼︁31.5°C): (1) a critical point resembling sol‐gel transition occurs at ∼︁28.5°C, not far beyond the onset of elasticity (∼︁28.2°C), and (2) a second critical point appears at ∼︁30.4 °C corresponding to the reported Flory temperature (Θ). These findings suggest that intermolecular association and coil‐globule transition of poly(NIPAM) occur below the LCST. The fractal dimension of association of poly(NIPAM) chains which was calculated based upon the rheological method (∼︁1.49) is close to that determined by static light scattering.  相似文献

Collapse of a poly(N-isopropylacrylamide)(PNIPAM) chain upon heating and phase diagrams of aqueous PNIPAM solutions with very flat LCST phase separation line are theoretically studied on the basis of cooperative dehydration(simultaneous dissociation of bound water molecules in a group of correlated sequence),and compared with the experimental observation of temperature-induced coil-globule transition by light scattering methods.The transition becomes sharper with the cooperativity parameterσof hydration.Reentrant coil-globule-coil transition in mixed solvent of water and methanol is also studied from the viewpoint of competitive hydrogen bonds between polymer-water and polymer-methanol. The downward shift of the cloud-point curves(LCST cononsolvency) with the mole fraction of methanol due to the competition is calculated and compared with the experimental data.Aqueous solutions of hydophobically-modified PNIPAM carrying short alkyl chains at both chain ends(telechelic PNIPAM) are theoretically and experimentally studied.The LCST of these solutions is found to shift downward along the sol-gel transition curve as a result of end-chain association (association-induced phase separation),and separate from the coil-globule transition line.Associated structures in the solution,such as flower micelles,mesoglobules and higher fractal assembly,are studied by USANS with theoretical modeling of the scattering function.  相似文献

Protein stability in aqueous solutions is important in numerous fields, particularly biotechnology and food‐science. To shed new light on the protective effect of carbohydrates on proteins, we studied saccharide‐structure effects in aqueous solutions on the coil‐to‐globule transition occurring at the lower critical solution temperature (LCST) of poly‐N‐isopropylacrylamide (PNIPA), an isomer of polyleucine, as a simple model representing certain key behaviors of proteins (e.g., denaturation/renaturation). We systematically selected sugars and polyols to relate structural and physical characteristics of these carbohydrates to their effect on PNIPA solutions. Using isothermal titration‐microcalorimetry, we showed that no significant binding of saccharides to the polymer occurs. Using micro‐DSC, we studied the decreasing polymer LCST temperature with rising carbohydrate concentration. Beyond the expected observation that steric exclusion is important, we observed previously‐unreported significant differences among the effects of isomeric aldohexoses and also among the effects of isomeric diglucoses on PNIPA LCST. We found good correlation between the sugar hydration number and its effect on LCST. We conclude that the larger and denser the hydrated cluster a carbohydrate forms, the worse a cosolvent is for the polymer, and the stronger it's lowering effect of the coil‐to‐globule transition. Such favoring of the compact globule state provides a protective effect against denaturation of globular proteins. © 2008 Wiley Periodicals, Inc. The thermally induced phase transition of the poly(N-isopropylacrylamide) (PNIPAM) brush covalently bound to the surface of the gold nanoparticles was studied using high-sensitivity microcalorimetry. Two types of PNIPAM monolayer protected clusters (MPCs) of gold nanoparticles were employed, denoted as the cumyl- and the cpa-PNIPAM MPCs, bearing either a phenylpropyl end group or a carboxyl end group on each PNIPAM chain, respectively. The PNIPAM chains of both MPCs exhibit two separate transition endotherms; i.e., the first transition with a sharp and narrow endothermic peak occurs at lower temperature, while the second one with a broader peak occurs at higher temperature. With increase of the MPC concentration, the transition temperature corresponding to the first peak only slightly changes but the second transition temperature strongly shifts to lower temperature. The calorimetric enthalpy change in the first transition is much smaller than that in the second transition. The ratio of the calorimetric enthalpy change to the van't Hoff enthalpy change indicates that in the first transition PNIPAM segments show much higher cooperativity than in the second one. The investigation of pH dependence of two-phase transitions further indicates the PNIPAM brush reveals two separate transitions even with a change in interchain/interparticle association. The observations are tentatively rationalized by assuming that the PNIPAM brush can be subdivided into two zones, the inner zone and the outer zone. In the inner zone, the PNIPAM segments are close to the gold surface, densely packed, less hydrated, and undergo the first transition. In the outer zone, on the other hand, the PNIPAM segments are looser and more hydrated, adopt a restricted random coil conformation, and show a phase transition, which is dependent on both concentration of MPC and the chemical nature of the end groups of the PNIPAM chains. Aggregation of the particles, which may also affect the phase transition, is briefly discussed.  相似文献

The interfacial properties of end-grafted temperature-responsive poly(N-isopropylacryamide) (PNIPAM) were quantified by direct force measurements both above and below the lower critical solution temperature (LCST) of 32 degrees C. The forces were measured between identical, opposing PNIPAM films and between a PNIPAM film and a lipid membrane. At the grafting densities and molecular weights investigated, the polymer extension did not change significantly above the LCST, and the polymers did not adhere. Below the LCST, the force-distance profiles suggest a vertical phase separation, which results in a diluter outer layer and a dense surface proximal layer. At large separations, the force profiles agree qualitatively with simple polymer theory but deviate at small separations. Importantly, at these low grafting densities and molecular weights, the end-grafted PNIPAM does not collapse above the LCST. This finding has direct implications for triggering liposomal drug release with end-grafted PNIPAM, but it increases the temperature range where these short PNIPAM chains function as steric stabilizers.  相似文献

Novel biodegradable poly(carbonate‐ether)s (PCEs) with lower critical solution temperature (LCST) at body temperature were synthesized by copolymerization of CO2 and ethylene oxide (EO) under double metal cyanide (DMC) catalyst. The PCEs showed carbonate unit (CU) content of 1.0–42.4 mol % and molecular weight of 2.7–247 kg/mol, which exhibited reversible thermoresponsive feature in deionized water with LCST in a broad window from 21.5 to 84.1 °C. The LCST was highly sensitive to the CU content and the molecular weight of PCEs, and it showed a linear relation with CU content for PCEs with similar molecular weight. In particular, aqueous solution of PCE with a 26.0 mol % of CU showed an LCST around 36.1 °C, which was very close to the body temperature. Interestingly, it was found that the phase transition behavior changed with PCE concentration. For PCE with Mn of 2.7 kg/mol and CU content of 30.0 mol %, the LCST increased from 21.5 to 36.7 °C when the PCE concentration changed from 10 to 1 g/L. Dynamic light scattering indicated that the phase transition was possibly due to a coil‐to‐globule transition. The thermoresponsive biodegradable PCE with LCST at body temperature is promising for biomedical applications, especially for in vivo applications. © 2012 Wiley Periodicals, Inc. Classical molecular dynamics simulations were carried out to investigate the hydrophilic to hydrophobic transition of PNIPAM‐co‐PEGMA close to its lower critical solution temperature (LCST) in 1 M NaCl solution. PNIPAM‐co‐PEGMA is a copolymer of poly(N‐isopropylacrylamide) (PNIPAM) and poly(ethylene glycol) methacrylate (PEGMA). The copolymer consists of 38 monomer units of NIPAM with two PEGMA chains attached to the PNIAPM backbone. The PNIPAM‐co‐PEGMA was observed to go through the hydrophilic?hydrophobic conformational change for simulations at temperature slightly above its LCST. Na+ ions were found to bind strongly and directly with amide O, even more strongly with the O atoms on PEGAMS chains, whereas Cl? ions only exhibit weak interaction with the polymer. Significantly a novel caged stable metal‐organic complex involving a Na+ ion coordinated by six O atoms from the copolymer was observed after the PNIPAM‐co‐PEGMA copolymer went through conformational transition to form a hydrophobic folded structure. © 2011 Wiley Periodicals, Inc. The temperature-induced structural changes of a thermo-responsive poly(N-isopropylacrylamide) (PNIPAM) layer grafted onto a silica substrate were investigated in aqueous solution using an atomic force microscope (AFM) and a quartz crystal microbalance with dissipation (QCM-D). A PNIPAM layer was grafted onto the silicon wafer surface by free radical polymerization of NIPAM to obtain a high molecular weight polymer layer with low-grafting density overall. By AFM imaging, the transition of the grafted PNIPAM chains from a brush-like to a mushroom-like state was clearly visualized: The surface images of the plate were featureless at temperatures below the LCST commensurate with a brush-like layer, whereas above the LCST, a large number of domain structures with a characteristic size of approximately 100 nm were seen on the surface. Both frequency and dissipation data obtained using QCM-D showed a significant change at the LCST. Analysis of these data confirmed that the observed PNIPAM structural transition was caused by a collapse of the brush-like structure as a result of dehydration of the polymer chains.  相似文献

In the present study, we report a method for preparing a fluorescent thermosensitive hybrid material based on monodisperse, thermosensitive poly( N-isopropyl acrylamide) (PNIPAM) microgels covered with CdTe nanocrystals of 3.2 nm diameter. The CdTe nanocrystals were covalently immobilized on the surface of PNIPAM microgels. The chemical environment around the CdTe nanocrystals was modified by changing the temperature and inducing the microgel volume-phase transition. This change provoked a steep variation in the nanocrystal photoluminescence (PL) intensity in such a way that when the temperature was under the low critical solution temperature (LCST) of the polymer (36 degrees C) the PL of the nanocrystals was strongly quenched, whereas above the LCST the PL intensity was restored.  相似文献

The chain end complexation of a functional PNIPAM by a cucurbit[8]uril-viologen complex causes a shift in its lower critical solution temperature (LCST) by over 5 °C. An instantaneous phase change of the thermally responsive polymer beyond its LCST can be induced by addition of the aqueous cucurbituril host-guest complex. Subsequent decomplexation upon addition of a competitive guest releases the PNIPAM terminus and triggers complete reversibility.  相似文献

A new series of cellulose-graft-poly(N-isopropylacrylamide)(cellulose-g-PNIPAM) copolymers were prepared by atom transfer radical polymerization(ATRP) of N-isopropylacrylamide monomers from a cellulose-based macro-initiator, which was homogeneously synthesized in an ionic liquid 1-allyl-3-methylimidazolium chloride(Amim Cl). The composition of cellulose-g-PNIPAM copolymers could be adjusted by altering the feeding ratio and reaction time. The resultant copolymers with relatively high content of PNIPAM segments(molar substitution of PNIPAM ? 18.3) were soluble in water at room temperature. Aqueous solutions of cellulose-g-PNIPAM copolymers exhibited clear temperature-sensitive behavior, and their sol-to-gel phase transition properties were investigated by dynamic light scattering(DLS) and UV measurements. Compared with pure PNIPAM, the cellulose-g-PNIPAM copolymers possessed higher lower critical solution temperatures(LCST) in a range from 36.9 ?C to 40.8 ?C, which are close to normal human body temperature, and could be tuned by adjusting the content of PNIPAM segments in copolymers. Spherical structure of cellulose-g-PNIPAM copolymers formed at temperatures above LCST and its morphology was observed by TEM and SEM. These novel cellulose-g-PNIPAM copolymers may be attractive substrates for some biomedical applications, such as drug release and tissue engineering.  相似文献

Aqueous processes ranging from protein folding and enzyme turnover to colloidal ordering and macromolecular precipitation are sensitive to the nature and concentration of the ions present in solution. Herein, the effect of a series of sodium salts on the lower critical solution temperature (LCST) of poly(N-isopropylacrylamide), PNIPAM, was investigated with a temperature gradient microfluidic device under a dark-field microscope. While the ability of a particular anion to lower the LCST generally followed the Hofmeister series, analysis of solvent isotope effects and of the changes in LCST with ion concentration and identity showed multiple mechanisms were at work. In solutions containing sufficient concentrations of strongly hydrated anions, the phase transition of PNIPAM was directly correlated with the hydration entropy of the anion. On the other hand, weakly hydrated anions were salted-out through surface tension effects and displayed improved hydration by direct ion binding.  相似文献
