The purpose of this pilot study was to establish the dependence or independence of oxalate absorption on the quantity of the test dose of sodium oxalate over a range of test doses corresponding to physiological dietary oxalate intake values. Gastrointestinal oxalate absorption was measured with the [13C2]oxalate absorption test. Six healthy volunteers were always tested under standardized dietary conditions with 63 mg dietary oxalate and 800 mg dietary calcium per day. The volunteers were tested thrice each with sodium oxalate test doses of 25, 50, 200, and 600 mg. Additionally, 1000 mg sodium oxalate was applied once to three of these volunteers. The oxalate absorption of the six volunteers tested under the standardized conditions with 50 mg sodium [13C2]oxalate was 7.2 +/- 2.62 % (mean +/- SD), similar to the 120 volunteers tested previously: 8.0 +/- 4.4 % (mean +/- SD). The tests with sodium [13C2]oxalate doses in the range 25-1000 mg revealed similar percent oxalate absorption values. In conclusion, in healthy volunteers, the amount of oxalate absorbed in the gastrointestinal tract increased proportionally with the higher test doses of oxalate. However, percent oxalate absorption remained unchanged with test doses in the dose range of physiological dietary oxalate intakes.  相似文献

In this work the feasibility of measuring neuronal-glial metabolism in rat brain in vivo using co-infusion of [1,6-13C2]glucose and [1,2-13C2]acetate was investigated. Time courses of 13C spectra were measured in vivo while infusing both 13C-labeled substrates simultaneously. Individual 13C isotopomers (singlets and multiplets observed in 13C spectra) were quantified automatically using LCModel. The distinct 13C spectral pattern observed in glutamate and glutamine directly reflected the fact that glucose was metabolized primarily in the neuronal compartment and acetate in the glial compartment. Time courses of concentration of singly and multiply-labeled isotopomers of glutamate and glutamine were obtained with a temporal resolution of 11 min. Although dynamic metabolic modeling of these 13C isotopomer data will require further work and is not reported here, we expect that these new data will allow more precise determination of metabolic rates as is currently possible when using either glucose or acetate as the sole 13C-labeled substrate.  相似文献

The aim of this study was to investigate the hepatic microsomal and mitochondrial functions by using the 13CO2-breath test in healthy subjects either before or after the consumption of red wine. Fourteen adults received [13C]methacetin and [methyl-13C]methionine together with a standardised dinner. Expired air samples were taken over 6 h. After a wash-out period, the subjects consumed 0.4 ml ethanol/kg/day together with dinner over a 10-day period. Thereafter, 13C-tracer administration was repeated under identical conditions. The 13CO2-enrichments were measured by isotope ratio mass spectrometry. The mean cumulative percentage 13C-dose recovery (CPDR) after administration of [13C]methacetin and [methyl-13C]methionine either without or with red wine consumption amounted to 38.2±6.3 vs. 36.3±6.7% (p=0.363) and 9.5±3.3 vs. 8.8±2.5% (p=0.47), respectively. Moderate alcohol consumption does not induce significant short-term changes of the microsomal and the mitochondrial functions of the human liver in healthy subjects.  相似文献

A newly developed isotope selective nondispersive infrared (NDIR) spectrometer for the measurement of 13CO2 and 12CO2 concentrations in breath samples was applied as a low cost and very simple to operate alternative to isotope ratio mass spectrometry (IRMS). We used this device for several biomedical applications ([13C]urea breath test, [13C]leucine metabolism, [13C]methacetin catabolism of rats) and found that the results agree very well with IRMS.  相似文献

The absorption spectrum of carbon dioxide has been studied between 8800 and 9530 cm−1 by intracavity laser absorption spectroscopy based on a vertical external cavity surface emitting lasers (VeCSEL). Previous laboratory spectra at high resolution were nearly absent in the considered spectral region. Experiments were carried with natural carbon dioxide and with 13C enriched carbon dioxide leading to the determination of the rovibrational parameters of a total of 15 very weak vibrational transitions, including two bands of the 16O13C18O isotopologue. The observed transitions are assigned to components of the 2ν1 + 3ν3 triad and of the much weaker 5ν1 + ν3 hexad. Our measured line positions are found in excellent agreement with the predictions of the effective Hamiltonians developed for 12C16O2 and 13C16O2 but significant deviations were evidenced for the 16O13C18O minor isotopologue. The relative band intensities within each polyad are also discussed on the basis of the effective Hamiltonian model.  相似文献

This work reports the ultraviolet absorption spectrum and cross-sections of ethynyl (C2H) radicals in the wavelength range 235-260 nm, determined at T=298 K and at a total pressure of about 20 kPa (150 Torr). Ethynyl radicals were produced from the single photon 193 nm excimer laser photolysis of dilute mixtures of C2HCF3 or C2H2 in He. Gas chromatographic/mass spectroscopic analysis of the photolyzed samples showed diacetylene (C4H2) as the major stable product. Addition of methane in the photolysis mixtures resulted in reduction of diacetylene and production of methylacetylene and ethane, products of the reaction of ethynyl radicals with methane. Survey of the ultraviolet spectral region, employing time-resolved UV-absorption spectroscopy, resulted in detection of a transient absorption centered at about 243 nm. The spectra obtained from the 193-nm photolysis of both C2HCF3/He and C2H2/He radical precursors were nearly identical, suggesting that the absorption feature can be attributed to the ethynyl radicals. The observed ultraviolet spectrum exhibits a relatively broad absorption feature with some structures and an absorption peak at about 243.5 nm. The absorption cross-sections for ethynyl radicals have been determined in this work for the first time. The C2H cross-section, at the maximum absorption, at 243.5 nm is . The stated uncertainty includes the random and systematic measurement errors. The UV absorption feature, detected in this work, can be assigned to the transitions from the ground electronic state and also possibly from coupled ground and lowest electronic states to the electronic state B (or 32A′) of C2H. This assignment is based on previously reported high level ab initio molecular orbital calculations and results of recent laser-induced fluorescence studies of the ethynyl radical.  相似文献

A resistant starch (RS) mixture (MIX) consisting of fibre of potatoes (FP) and wrinkled pea starch (WPS), and high amylose maize starch (HAMS) were supplemented in adults to evaluate their effects on fat oxidation by means of a 13CO2-breath test. Sixteen subjects received a regular diet either without or with the supplementation of MIX and HAMS in randomised order. After administration of a [U-13C]algal lipid mixture, exhaled air was collected over 14?h in 0.5- and 1-h intervals. The 13C abundances were measured by nondispersive infrared spectroscopy. In comparison to the dry run (DR), supplementation with MIX and with HAMS increased the cumulative percentage dose recovery: (DR: 16.7?%, MIX: 16.9?%, HAMS: 18.0?%), but without statistical significance. The colonic degradation of MIX and HAMS to short-chain fatty acids tends to lower the formation of carbohydrate-derived acetyl-CoA and contributes to a postprandial lipid oxidation increase by using fat-derived acetyl-CoA as a compensatory fuel source.  相似文献

The absorption spectra of H12C13CD and H13C12CD have been observed at high resolution between 6480 and 6610 cm−1 using an external cavity diode laser. The strong 2ν1 band has been observed for each species using a sample enriched in deuterium at natural abundance of 13C. Rotational analyses reveal bands of both species to be essentially unperturbed. Centers of unblended lines are determined with an accuracy of approximately 10 MHz.  相似文献

The absorption spectrum of 13CO2 has been recorded by cw-cavity ringdown spectroscopy with a new set up based on fibered DFB lasers. By using a series of 31 DFB lasers, the spectrum of carbon dioxide could be recorded in the 6130-6750 cm−1 region with a typical sensitivity of 5 × 10−10 cm−1. The spectrum has also been recorded between 4400 and 8500 cm−1 with a Fourier transform spectrometer associated with a multi-pass cell (maximum path length of 105 m). The new observations obtained both by FTS and CRDS represent a significant extension of the available data. For instance, more than 4000 line positions were measured and assigned in the CRDS spectrum while only 232 line positions are listed in the HITRAN database. Altogether, the band by band analysis has led to the determination of the rovibrational parameters of 65, 7, and 24 bands for the 13C16O2, 16O13C17O, and 16O13C18O isotopomers, respectively. As some observed line positions show significant deviations from the predictions of the effective Hamiltonian model, the new observed line positions were gathered with the data available in the literature to refine the set of effective Hamiltonian parameters of the 13C16O2 isotopic species. The refined set of 96 effective Hamiltonian parameters reproduces more than 14 650 line positions of 13C16O2 with an RMS=0.002 cm−1. A detailed comparison with the line positions retrieved from Venus spectra and the line list provided by HITRAN is also presented and discussed.  相似文献

We present a nondispersive infrared spectrometer (NDIRS) for the measurement of the 13CO2/12CO2-ratio in breath samples. A commercial NDIR spectrometer for CO2 concentration measurements in industrial process control was modified using two separate optical channels for the 13CO2 and 12CO2 detection. Cross interference due to overlapping absorption lines of both isotopic gases was successfully eliminated. The sensitivity of this device is ± 0.4‰ of the 13CO2/12CO2-ratio in a range of 2.5 to 5% of total CO2. This is sufficient for biomedical applications. Our spectrometer is small in size, cheap and simple to operate and thus a true alternative to isotope ratio mass spectrometers (IRMS). Several biomedical applications with breath samples were demonstrated and were compared in very good agreement with IRMS.  相似文献

A computer-aided optoacoustic gas analyzer based on a continuous13C16C2 laser for multicomponent pollution of atmospheric air is described. The analyzer has the ability to detect absorption of radiation by detected substances at the level of ∼1·10−9 cm−1 at a time resolution of 30 sec. The absorption spectrum of acetylene-d has been observed at high resolution between 6470 and 6630 cm−1 using an external cavity diode laser. Three cold bands have been observed: the strong 2ν1 band, the weaker ν1 + ν2 + 2ν5 band, and the (ν1 + ν3 + ν5)1 band, which gains its intensity through Coriolis resonance with 2ν1. The usefulness of different ways of water removal in off-line sample preparation of human breath samples for 13CO2 breath tests was examined and compared. Cryogenic water trapping and water removal with common desiccants like silicagel blue, Mg(ClO4)2, and molecular sieves were checked for reliability and reproducibility. With silicagel blue and Mg(ClO4)2 memory effects for 13C content were observed. The use of molecular sieve 4 Å and 5 Å led to tremendous carbon isotope fractionation. Molecular sieve 3 Å was found to be an excellent alternative to the established use of Mg(ClO4)2 and of cryogenic water trapping.  相似文献

The optical absorption spectra and electronic spin resonance parameters (ESR g factors g, g and hyperfine structure constants A, A) for Cu2+ in shattuckite crystal are calculated from the two spin–orbital coupling parameters model, high-order perturbation formulas and complete diagonalization (of energy matrix) method (CDM) of 3d9 ion in tetragonal symmetry. The calculated results are in good agreement with the observed values. Since the ESR parameters are sensitive to the local structure of a paramagnetic impurity center, the defect structure of Cu2+ center in shattuckite crystal is estimated. Abstract

One representative species of each of the three photosynthetic pathways (C3, C4, CAM) were cultivated in growth chambers with high and low nitrogen nutrition level respectively once at 20°C day/13°C night temperature, once at 30°C day/13°C night. Leaf conductances and δ13C values of the leaves of each plant were determined. At 20°C day temperature the C3 species showed higher leaf conductance with low nitrogen nutrition level than with high nitrogen level, which is also reflected in a more negative δ13C value, whereas both C4 and CAM plants did not respond in this manner to nitrogen supply. An increase of day temperature to 30°C diminished the significant response of the C3 plant, while the response of C4 and CAM representatives to nitrogen nutrition did not change markedly.  相似文献

Scanning Infrared Absorption (S.I.R.A.) setup was carried out for investigating and mapping of microdefects distribution in Czochralski silicon wafers. Using CO2 laser, this non destructive, non contact test method allows transmitted beam local attenuation from oxygen precipitates to be determined. Transmitted intensity It maps from peak height of the 1080 cm−1 absorption band were obtained. An inhomogeneous initial oxygen distribution is found in as-grown samples. The samples were then submitted to preannealing at 750°C for 16 hours, followed by annealing at 900°C for 24 hours. Abstract

The [13C]aminopyrine breath test ([13C]ABT) measures the global activity of cytochrome P450 in vivo and is a sensitive indicator of liver metabolic dysfunction. The present study aims to determine whether gender and cigarette smoking influence the results of [13C]ABT as well as to confirm the effect of oral contraceptive steroids (OCS) intake on this metabolic test. Hundred and ten healthy subjects, including men and women, smoker and non-smoker, women taking OCS or not, were phenotyped for CYP1A2 using the [13C]caffeine breath test and underwent a [13C]ABT. Both tests showed large inter-individual variations in accordance with that of CYP450 liver content. [13C]ABT was sensitive enough to point out a significant induction or inhibition related to cigarette smoking habits or OCS. The combined effect of smoking and OCS resulted in an overall unchanged metabolic activity. Consequently, the impact of the studied conditions on the [13C]ABT parameters must be considered by clinicians or clinical investigators.  相似文献

This study evaluated possible actions that can help protect against and reduce radiation exposure by measuring the exposure dose for each type of isotope that is used frequently in nuclear medicine before performing numerical analysis of the effective half-life based on the measurement results. From July to August in 2010, the study targeted 10, 6 and 5 people who underwent an 18F-FDG (fludeoxyglucose) positron emission tomography (PET) scan, 99mTc-HDP bone scan, and 201Tl myocardial single-photon emission computed tomography (SPECT) scan, respectively, in the nuclear medicine department. After injecting the required medicine into the subjects, a survey meter was used to measure the dose depending on the distance from the heart and time elapsed. For the 18F-FDG PET scan, the dose decreased by approximately 66% at 90 min compared to that immediately after the injection and by 78% at a distance of 1 m compared to that at 0.3 m. In the 99mTc-HDP bone scan, the dose decreased by approximately 71% in 200 min compared to that immediately after the injection and by approximately 78% at a distance of 1 m compared to that at 0.3 m. In the 201Tl myocardial SPECT scan, the dose decreased by approximately 30% in 250 min compared to that immediately after the injection and by approximately 55% at a distance of 1 m compared to that at 0.3 m. In conclusion, the dose decreases by a large margin depending on the distance and time. In conclusion, this study measured the exposure doses by isotopes, distance from the heart and exposure time, and found that the doses were reduced significantly according the distance and the time.  相似文献

This study determined the within-subject and between-subject variability of different ways of expressing the results of the 13C-aminopyrine breath test (13C-ABT) and the effect of shortening the test duration. The 13C-ABT was conducted on three separate occasions in 10 healthy volunteers and on a single occasion in 22 patients with established liver cirrhosis. The within-subject variability of cumulative percentage dose recovered (cPDR), using measured CO2 production rate (VCO2), in the reference group over three trials was 15% over 120 min. Higher within-subject variability in cPDR would have been evident if the test was terminated at either 30 or 60 min. Substitution of predicted VCO2 to calculate cPDR yielded comparable values at all time points. Significant differences between cirrhotics and reference group were evident after just 10 min using PDR/h, cPDR or enrichment (all P<0.05). The ABT demonstrates clinically acceptable reproducibility. Shortening of the duration may make the test more acceptable clinically, but it is associated with increasing imprecision.  相似文献
