(2S,3R,1'R)-Stegobinone [2,3-dihydro-2,3,5-trimethyl-6-(1'-methyl-2'-oxobutyl)-4H-pyran-4-one], the pheromone of Stegobium paniceum L., was synthesized with stereocontrol at C-2 and C-1' starting from ethyl (R)-3-hydroxybutanoate and methyl (R)-3-hydroxy-2-methylpropanoate. The dihydro-γ-pyrone ring of the pheromone was constructed by an intramolecular acylation followed by acid-catalyzed cyclization.  相似文献

Total synthesis of (±)-sesbanine (1) was carried out using γ-addition of ketene silyl acetal of methyl 3-cyclopentenecarboxylate to quaternized methyl nicotinate. The resulting 1,4-dihydropyridine (7) was oxidized with DDQ to give 4-substituted nicotinate (2) and 2 was converted to alcohol (8) by stereoselective oxymercuration followed by treatment with ammonia to give 1.  相似文献

Asymmetric oxidation of methyl phenyl selenide (3), under anhydrous conditions, by chiral 2-sulfonyloxaziridines, 1–2, gives optically active methyl phenyl selenoxide (8.1–9.3% ee). The stereochemistry of the selenoxide is determined by the configuration of the oxaziridine three membered ring with non-bonded steric interactions responsible for the chiral recognition. Asymmetric oxidation of E-phenyl cinnamyl selenide (7) by 1–2 affords optically active l-phenylallyl alcohol (9). A concerted [2,3] sigmatropic rearrangement via an exo transition state is proposed.  相似文献

An efficient synthesis of C- and N-nucleosides has been developed in an enantioselective and stereocontrolled manner starting from Diels-Alder adduct of furan and dimethyl acetylenedicarboxylate by chemicoenzymatic strategy. The symmetric unsaturated dimethyl esters 2 and 3 were almost quantitatively hydrolysed with pig liver esterase to yield half-esters 4 and 5 with reasonably high optical yields. Decarboxylative ozonolysis of the chiral half-esters 4 followed by chemical transformation afforded methyl L-riboside 12, but after the enantiomer conversion (4 to 13 and 5 to 28) the methyl D-riboside (17), ( + )-showdomycin (22), and ( ? )-6-azapseudouridine (27), were obtained from 13, and ( ? )-cordycepin (32) was obtained from 28.  相似文献

Diethyl bromodifluoromethyl phosphonate reacts readily with cadmium metal to form a stable cadmium complex. Depending on solvent, this functionalized organocadmium reagent exhibits stability for days to months. It reacts with a variety of electrophiles and serves as a synthetically useful source for the introduction of the difluoromethylene phosphonate group into organic compounds.The synthetic utility of a wide variety of fluoromethylene phosphonium ylides has been a major effort in our laboratory over the past several years [1]. The generation and capture of difluoromethylene ylides (1) as a general route to difluoromethylene olefins has been of especial interest to us [2]. In an effort to increase the nucleophilicity of the ylide, we have attempted to prepare the analogous phosphonate ylide (2). Although we have achieved modest success [3] by insitu capture of (2) in the reaction of
sodium dialkyl phosphites with diethyl bromodifluoromethylphosphonate (3), attempts to pregenerate (2), either from diethyl difluoromethylphosphonate (4) or (3), have met with little success. (2) appears to have minimal stability even at low temperatures, and scale up processes of synthetic value would seem to be difficult.  相似文献

Methyl 4-0-benzoyl-6-bromo-6-deoxy-α-D-glucopyranoside (1, Figure 1) was converted, via the corresponding ditosylate 2, into methyl 2,3-di-0-p-toluenesulfonyl-4-0-benzoyl-6-S-acetyl-6-thio-α-D-glucopyranoside (3) by a selective nucleophilic displacement of 6-bromo-group with thioacetate. Unexpectedly, on treating the compound 3 with an excess of sodium methoxide in benzene-methanol (1:1) at room temperature, methyl 2-0-p-toluenesulfonyl-4,6-thioanhydro-α-D-gulopyranoside (4) was obtained in a yield of 84%. In order to determine the structure of the relatively unstable oily product 4, some stable crystalline derivatives (5, 6 and 7) were prepared. Detailed analysis of the 1H-NMR-spectra (200 MHz) of 6 and 7 gave the conclusive evidence for the structure of 4 A self-imposing mechanism of the clean and smooth transformation of 3 to 4 is proposed, involving: a) formation of 9 (Figure 2) as a crucial intermediate and b) a highly regioselective epoxide opening in 9 (at C-4) by an intramolecular nucleophilic attack of the mercaptide anion from C-6.  相似文献

A route towards I-oxygenated eudesmanolides, via a (2+2) photocycloaddition reaction for constructing the decalin framework is described. The following natural substances have been synthesized. (=)-dihydroreynosin (1), (±)-1- oxo-dihydronmagnolialide (2), (±)-maritimin (3), (±)-dihydromagnolialide (4), (±)-magnolialide (5), (±)-dihydrosantamarine (6). The acid catalysed reaction of 4-oxa-homoadamantan-5-one (1) with benzene yielded a mixture of 4a-phenyladamantan-2-one (7), the equatorial isomer' (8) and 2-phenyl-2,4-o- benzenoadamantane (9) A plausible reaction pathway for the occurrence of 9. is put forward. The structure of 9, was deduced from spectroscopic data and reaction of the proposed intermediate 2,4a-diphenyladamantan-2-ol (11) with acid. 2,4-o-Ben-zenoadamantane (16) is prepared likewise.  相似文献

1,3-Diemthyl(5H,7H)imidazo(4,5-dpyrimidine-2,4,6-trithione (1) was synthesized by the reaction of carbon disulfide with 1,3-dimethyl-4-cyano-5-aminoimidazolidine-2-thione (3) which was produced by reacting trimethylsilyl cyanide with methyl isothiocyanate followed by methanolysis. Crystalline N-nitrosothialdine (2) has been prepared in 38% yield by treating thialdine (1) with n-butyl nitrite and acetic acid in hexane. X-Ray crystallography of 2 revealed that its three methyl groups are a11 cis and, rather surprisingly, all equatorial; this finding contrasts sharply with results for other nitrosamine heterocycles which have been investigated, whose bulky alpha substituents are forced into a primarily axial orientation by the large steric requirement of the N-N-0 system. The equatorial methyl group both displaces the nitroso group from the C-N-C plane and twists it somewhat about the N-N bond. N-Nitrosothialdine's unusually low barrier to rotation about the N-N bond (72 kJ/mole) is attributed to this steric crowding, combined with inductive electron withdrawal by the two sulfur atoms.  相似文献

The novel structures assigned by Yunusov et al to the C19-diterpenoid alkaloids, acomonine (1), iliensine (2), 14-dehydroiliensine (3), and 14-bensoyliliensine (4) have been found to be in error. We have demonstrated that these alkaloids are identical with the well-known alkaloids, delsoline (5), delcosine (6), 14-dehydrodeloosine (7), and 14-benzoyldelcosine (8), respectively.  相似文献

Methods were developed to prepare 1 -methyl-, 3-methyl- and 4-0-methyl-ψ-isocytidine by selective methylation.3,?5-?O-Tetraisopropyldisiloxanyl-ψ isocytidine (8) was trimethylsilylated and then treated with MeI and, after deprotection, 1 -methyl-ψ isocytidine (6) was obtained. The 2-?deoxy analog (7) was also prepared in a similar manner from the 2-?deoxy analog (10) of 8. Treatment of 8 with CH2N2 afforded the 3-methyl-ψ-isocytidine derivative (19) as the major product. Methylation with diazomethane also occurred mainly on N3 of the 2-?deoxy analog 10 to form 20. Removal of the 3,? 5-?O-protecting group from 19 and 20 afforded 3-methyl-ψ-isocytidine (14) and its 2-deoxy analog (15), respectively. 2-N-Acetyl- 3,?5-?O-tetraisopropyldisiloxanyl-ψ-isocytidine (24), on the other hand, gave the 4-O-methyl derivative (25) as the major product upon CH2N2 treatment. 6-Oxoprostaglandin E1 methyl ester was synthesized in a single pot from (R)-4-t-butyldimethylsiloxy-2-cyclopentenone by organocopper conjugate addition with an ω side-chain unit, trapping of the resulting enolate with 6-methoxycarbonyl-2-nitrohex-1-ene, and treatment with aqueous titanium(III) trichloride. Hydrolysis of the methyl ester was accomplished by porcine liver esterase. 6-Oxoprostaglandin F, was obtained from 6-nitroprostaglandin E1 methyl ester in four steps.  相似文献
