A competitive lateral flow assay for detecting the major peanut allergen, Ara h1, has been developed. The detector reagents are Ara h1-tagged liposomes, and the capture reagents are anti-Ara h1 polyclonal antibodies. Two types of rabbit polyclonal antibodies were raised either against the entire Ara h1 molecules (anti-Ara h1 Ab) or against an immunodominant epitope on Ara h1 (anti-peptide Ab). All of them reacted specifically with Ara h1 in Western Blot against crude peanut proteins. Moreover, the anti-Ara h1 Ab was chosen for this assay development because of its highest immunoactivity to Ara h1-tagged liposomes in the lateral flow assay. The calculated limit of detection (LOD) of this assay is 0.45 g mL–1 of Ara h1 with a dynamic range between 0.1 and 10 g mL–1 of Ara h1 in buffer. Additionally, the visually determined detection range is from 1 to 10 g mL–1 of Ara h1 in buffer. Results using this assay can be obtained within 30 min without the need of sophisticated equipment or techniques; therefore, this lateral flow assay has the potential to be a cost-effective, fast, simple, and sensitive method for on-site screening of peanut allergens.  相似文献

Dodecasil 1H, 34SiO2 · 3M12 · 2M12 · 1M20 with M12, M12=N2 and M20=1-aminoadamantane, is hexagonal witha=13.825(2) Å,c=11.189(2) Å and crystallizes in space groupP6/mmm. With 767 unique reflexions the structure has been refined to a weighted reliability factorR w =0.054. The three-dimensional four-connected host framework is built by corner sharing [SiO4] tetrahedra and shows three types of cagelike voids, [512] cages, [435663] cages and [51268] cages, the latter housing the 1-aminoadamantane guest molecule. Difference Fourier synthesis (obs)-(calc(Si,O)) delineate the guest molecules within the different types of cage. Residual electron densities reveal positional disorder of the 1-aminoadamantane guest molecule. Four crystallographically different preferred orientations for the 1-aminoadamantane guest molecule are found.  相似文献

A precise, reliable, sensitive, and selective method for the determination of titanium(IV) is described. Titanium(IV) reacts withN 1-hydroxy-N 1,N 2-diphenylbenzamidine (HDPBA) and thiocyanate to form an orange-coloured mixed-ligand complex of stoichiometry 112 (Ti SCN HDPBA). The complex is quantitatively extractable into toluene from 0.05–0.15M hydrochloric acid. The spectrum of the complex exhibits an absorption maximum at 400 nm with a molar absorptivity of 20000M –1 cm–1 and the coloured system obeys Beer's law in the concentration range 0.20–3.0 gml–1 titanium. The effects of foreign ions and of various experimental parameters have been studied to establish the optimum conditions for the extraction and determination of titanium. The precision of the method has been evaluated and the relative standard deviation has been found to be 0.53%. The method has been successfully applied to the determination of titanium in synthetic matrices corresponding to titanium-containing ores, minerals, and alloys.  相似文献

A selective Spectrophotometric method has been developed for the trace determination of copper after adsorption of its 1-phenyl-4,4,6-trimethyl(1H, 4H)-pyrimidine-2-thiol (PTPT) complex onto microcrystalline naphthalene. The complex is quantitatively adsorbed on naphthalene in the pH range 7.5–11.5, separated by filtration, dissolved in dimethylformamide (DMF) and determined spectrophotometrically at 400 nm. Beer's law is obeyed in the concentration range 2.5–37.5 g of copper in 10 ml of DMF. The molar absorptivity and Sandell's sensitivity are 1.30 × 1041 · mol–1 · cm–1 and 0.0048g cm–2, respectively. Ten replicate analyses of a solution containing 20.0 g of copper gave a mean absorbance of 0.410 with a relative standard deviation of 0.91 %. The interferences of various ions have been studied and the method has been validated by the determination of copper in various standard reference materials, beers, wines, human hair, goat liver and environmental samples.On leave from St. Stephen's College, Delhi 110 007, India  相似文献

The reactions of 2-, 3-, and 4-(1-vinylpyrrol-2-yl)pyridines with methyl iodide afford the corresponding quaternary salts. Analysis of their 1H and 13C NMR spectra showed that the quaternization of the nitrogen atom considerably enhances the -acceptor effect of the pyridine ring on the pyrrole ring and on the vinyl group. 1-Methyl-2-(1-vinylpyrrol-2-yl)pyridinium iodide contains no weak intramolecular C--H...N hydrogen bond present in the starting compound.  相似文献

Heating of 2-benzylidene-1-indanone (1) either in dimethylformamide in the presence of guanidine carbonate or inn-propanol in the presence of thiourea and sodium propylate yields a dimerP with the structure of 1,3-diphenyl-3a,8a-dihydrospiro{cyclopenta[a]indene-2,2(1H,3H)-indene}-1,8(3H)-dione (7); this was deduced from the 400 MHz-1H- and 100MHz-13C-nmr-spectra and the corresponding two-dimensional HH- and CH-correlations. A possible mechanism for the formation of the spirocompound 7 is proposed; the stereo formula of the dimer7 K was established on the basis of a 400 MHz-HH-NOESY experiment.

A new Spectrophotometric method is proposed for the determination of 1-naphthylamine (R), based on its reaction with Mn(CN)5NO2– to form Mn(CN)5NH2R3– and measurement of the absorbance at 472 nm. In aqueous medium the molar absorptivity of the manganese complex is maximum ( = 8.0 × 1031 · mole–1 ·cm–1) in the pH range 5.0–10.0, the colour develops more rapidly at pH 10.0. The absorptivity is increased by a factor of 3.5 if the complex is extracted as an ion-associate into chloroform. The extraction efficiency is 99.2% for a single-step extraction, and a concentration factor of 5 can also be achieved. Linearity of response extends over the range 0.04–1.4 gmg/ml 1-naphthylamine, the coefficient of variation being 1.4% at the 0.29 g/ml level (n = 6). The detection and determination limits are 0.005 and 0.018 g/ml, respectively. The method is selective enough to allow the determination of 1-naphthylamine in the presence of considerable amounts of other amines, such as aminophenols and phenylenediamines. Results obtained in the determination of 1-naphthylamine in human urine were very satisfactory.  相似文献

Two efficient methods for the determination of 1,1-dichloro-1-fluoroethane (Freon F-141b) in formulated polyol and rigid polyurethane foam by dynamic-headspace-gas chromatography-ion trap-mass spectrometry were developed and validated. Rigid polyurethane foam was efficiently dissolved in dimethylformamide by heating at the temperature of 60 °C for 2 h.Validation was carried out in terms of limits of detection (LOD), limits of quantitation (LOQ), linearity, precision and recovery. LOD values of 4.00 g kg–1 for rigid polyurethane foam and 0.73 g kg–1 for formulated polyol were achieved, whereas linearity was statistically verified over one order of magnitude. Precision was evaluated testing two concentration levels. Good results were obtained both in terms of intra-day repeatability and between-day precision: RSD % lower than 4% (n = 6) at the concentration of 15 g kg–1 were calculated for intra-day repeatability. Extraction recoveries up to 92.6±1.6 % (n = 3) were also calculated by the addition of Freon F-141b to the samples analysed. Both the methods were applied for the analysis of a number of formulated polyol and rigid polyurethane foam samples.  相似文献

Pitzer's equation for the activity coefficient has been applied to LiCl, NaCl, KCl, RbCl, CsCl, and HCl in methanol-water mixtures at 25°C. The two parameters, MX (0) and MX (1) , were obtained by the method of least squares. The same values of b and as for aqueous systems could be used for the methanol-water mixtures without greatly affecting the standard deviation of the fit. MX (0) was found to decrease with methanol content for the alkali metal chlorides whereas MX (1) was found to increase. For HCl, however, a slight maximum and a minimum were found in the values of MX (0) while a maximum was observed in the values of MX (1) .  相似文献

Two Kemp's acid diamides were synthesized and applied to chiral amine recognition using 1H NMR analysis. One derivative based on 1-(1-naphthyl)ethylamine had good chiral recognition of six amines and was useful to determine the optical purity for three amines, i.e., methylbenzylamine, 1-(1-naphthyl)ethylamine and 1- henylpropylamine,however, the cyclohexylethylamine derivative showed little discrimination for the amines studied. Together with the results for alkylamines, it was shown that aromatic structure was important for aromatic shielding anisotropy and –– interactions between host and guest. The structure of the 1-(1-naphthyl)ethylamine derivative in solution was also considered based on 1H NMR data and computer simulation.  相似文献

Heats of solution of 13 11 electrolytes in 1-propanol have been determined calorimetrically at various electrolyte concentrations, and extrapolated to zero concentration to give H s o values for these electrolytes. Together with literature data on three additional 11 electrolytes, these measurements yield a self-consistent set of single-ion enthalpies of transfer from water to 1-propanol. Values are tabulated for 10 univalent cations and five univalent anions. It is shown that the H t o (Ph 4 As+)=H t o (Ph 4 B) assumption yields chemically reasonable single-ion values. Using this assumption, it may be deduced that all the univalent ions studied have about the same enthalpy in 1-propanol as in methanol.  相似文献

The crystal structure of 1-(2-pyridiniomethyl)-2,4-bis(phenylsulfonyl)benzene bromide, (C24H20NO4S2)+. Br (I) has been investigated by X-ray diffraction (XRD)analysis. The triclinic structure of I (space group P1, a = 7.863 , b = 8.350 , c = 9.043 , = 94.00°, = 97.81°, = 104.62°, Z = 1) was solved by direct methods and refined by full-matrix least-squares analysis in an anisotropic approximation to R = 0.048 for all 4570 reflections collected (CAD-4 automatic diffractometer, CuK ). The geometrical parameters of the organic cation were determined with a sufficient degree of accuracy. Binding constants for -cyclodextrin ( -CD) complexes with 1- alkanols and 1- alkanoate ions were determined by analyzing changes ( ) in chemical shifts of guest 13C NMR signals with -CD concentration. The guests gave well-separated 13C NMR signals. The curve-fitting analyses of for individual carbons upon assumption of a simple 1:1 complexation gave virtually the same binding constants (K1) in the cases of guests with relatively short alkyl chains, such as 1-propanol, 1-butanol, 1- pentanol , 1- butanoate , 1- pentanoate , and 1-hexanoate. However, thus obtained individual K1 values were significantly different from one another, when the guests had relatively long alkyl chains such as 1- hexanol , 1- heptanol , and 1-octanoate. In these guests, satisfactorily consistent K1 values were obtained by the curve-fitting analyses of for individual carbons upon an assumption that not only 1:1 but also 2:1 (host:guest) complexation occurs.  相似文献

Dispiro[bis(3-aminoindene)-1,4;1,5-(1,3-dithiolanes)] react readily with Cu+, Ag+, Hg+, and Hg2+ salts to give the corresponding bis(1-imino-3-indenyls) and methanedithiol derivatives. Bis(1-oxo-3-indenyls) are formed when the reaction is carried out in the presence of water. The reactivities of the 1,3-dithiolanes were examined with allowance for electronic and steric factors, and the reactivities of the metal cations were examined from the point of view of Pearson's theory. The conductances of eleven 1:1 salts have been measured at 50°C in N,N-dimethylmethanesulfonamide (DMMSA) for electrolyte concentrations of 1.2–55.0×10–4 mol-dm–3. The conductance data were analyzed using the equation of Lee and Wheaton. Calculations for different values of the distance parameter R indicate that all salts studied are only slightly associated in DMMSA. Association was somewhat greater for the trimethylphenylammonium halides than for the tetraalkylammonium salts. Ionic limiting molar conductances were estimated using the tris (isopentyl) butylammonium tetraphenylborate approximation. The markedly smaller value for o (Na+) compared to the values for o (Br) and o (I) indicates that the sodium ion is probably more extensively solvated than the halide ion. In general, it appears that DMMSA (dielectric constant=80.31 at 50°C) is similar in its solvent properties to dipolar aprotic heterocyclic solvents such as 2-cyanopyridine and 3-methyl-2-oxazolidone which have similar dielectric constants.  相似文献

The -orbital structure of the monomeric form of the 2,2,4,6-tetrachloro-2,2-dihydro-1,5,2-diazaphosphorinine has been studied by photoelectron spectroscopy and using quantum-chemical calculations by the semiempirical AM1 method. It has been concluded that the electronic and energy characteristics of four higher -MOs (frontier and three next orbitals) of this compound may be interpreted in terms of semipolar bonds formed by three atoms (C, P, and N). Slow distillation of 5, 5, -dimethyl-2-pyrazoline isomerizes it to 5, 5-dimethyl-1-pyrazoline, and heating the latter with hydrazine hydrochloride reconverts it to the 2 isomer. This is the first experimental evidence that the 2 double bond in a pyrazoline can move to the 1 position, i. e., in a direction the reverse of that previously known. The 5, 5-dimethyl-1 -pyrazoline described is the simplest 1 pyrazoline hitherto isolated.The authors heartily thank A. A. Petrov for making it possible to chart the PMR spectra, and V. B. The structure of 1-(3,5-dimethyl-2-furyl)-3-(p-tolyl)-prop-2-en-1-one, C16H16O2 is determined by single-crystal x-ray diffractometry. Crystals are triclinic, , a=7.381(7), b=8.871(5), c=10.745(13) Å, =93.53(7), =102.25(8),=95.43(7)°, V=682(1) Å3, Z=2, dcalc=1.170 g cm–3, (Cu K)=1.54178 Å,=5.697 cm–1, M=270.30, F(000)=256, R=0.077 for 1601 unique observed reflections. The trans-geometry of the C=C double bond, thes-cis-geometry of the C=C-C=O moiety, and thes-trans arrangement of the carbonyl and furane oxygen atoms are proved.  相似文献
