A new version of TLC with a forced flow of the mobile phase has been suggested. The variant involves the use of a TLC plate, the sorption layer on which is covered with a polymer film, and evacuation as an additional driving force for the mobile phase. The advantages of the new method were analyzed; the velocity coefficient of the movement of the mobile phase front was found to be linearly dependent on the rarefaction at the end of the plate.Translated fromIzvestiya Akademii Nauk. Aqueous ethylene glycol (ethane 1,2 diol) as a green mobile phase has been used for thin layer chromatographic (TLC) studies of cationic surfactants on alumina layers. Nineteen solvent systems were used to examine the mobility of the surfactants and to discover the best TLC system for the selective separation of dodecyl trimethylammonium bromide (DTAB) from multi-component mixture of other surfactants. Among the TLC systems studied, M3 (ethylene glycol: water, 8:2) was best for achieving the selective separation of DTAB from multi-component mixture of other surfactants because in this mobile phase mobility of all surfactants except DTAB were insignificant. Effect of organic additives in aqueous ethylene glycol mobile phase on the mobility of surfactants was examined. The results obtained on laboratory made alumina TLC plates and commercially available precoated alumina HPTLC plates were compared. The lower limits of detection of DTAB, CPC, CTAB, HDTAC, and TTAB were 0.02, 0.05, 0.04, 0.06, or 0.08 µg per zone respectively. The resolution of mixture of cationic surfactants was also examined in the presence metal cations as an impurity in the analyzed sample.  相似文献

Linear solvation energy relationships (LSERs) were used to delineate which specific intermolecular interactions are responsible for changes in retention for a variety of well characterized analytes when acidic and basic additives were used in reversed phase HPLC. The effects of trifluoroacetic acid, triethylamine and a combination of trifluoroacetic acid and triethylamine on the LSERs were compared to those observed in the absence of additives. These effects were examined using four different mobile phase modifiers and five different stationary phases. Trifluoroacetic acid alone and in combination with triethylamine produced LSER regression coefficients nearly identical to those obtained with no additive present in the mobile phase. Triethylamine alone produced different LSER regression coefficients from the other systems unless the mobile phase contained trifluoroethanol as the mobile phase modifier, or the stationary phase consisted of a polymeric support.  相似文献

 提出了以醋酸 水作为流动相的体系中 ,在ODS柱上分离生物大分子的反相高效液相色谱 (RPLC)方法。实验结果表明 ,醋酸 水的洗脱能力强于甲醇 水 三氟醋酸体系 ,在一定程度上克服了色谱分离中一些蛋白质的不可逆吸附且具有便于冷冻干燥的优点。用参数Z(1mol溶剂化溶质被溶剂化固定相吸附时从两者接触表面释放出置换剂的摩尔总数 ) ,logI(与 1mol溶质对固定相亲和势有关的常数 )和 j(与 1mol溶剂对固定相亲和势有关的常数 )对 9种蛋白质在此流动相体系中的保留进行了表征。  相似文献

Abstract

A method for the thin layer chromatographic (TLC) separation of enantiomers and diastereomers involving the use of chiral ion interaction agents is described. Several aromatic amino alcohols were resolved by TLC on diol and/or high performance silica gel plates using a mobile phase containing (1R)-(-)- ammonium-10-camphorsulfonate or N-benzoxycarbonyl-glycyl-L-proline (ZGP). Many of these chiral aromatic amino alcohols are of pharmacological importance as α- and β-adrenergic blockers, adrenergic compounds, and anti-glaucoma agents. A comparison was made between various N-CBZ-amino acid derivatives as chiral counter ions/chiral mobile phase additives (CMAs). These separations could not be achieved on other normal phase TLC stationary phases including microcrystalline cellulose, alumina and ordinary silica gel plates.  相似文献

Summary The chiral discrimination of phenoxypropionic acid herbicides by reversed-phase chromatography on a teicoplanin phase has been re-examined using the perturbation method to calculate the solute distribution isotherms. The effects of both temperature and the methanol (organic modifier) content of the mobile phase on the chiral discrimination mechanism were well described by the bi-Langmuir model. The method confirmed a change in the mechanism of enantiomer retention at a critical temperature, T*, and showed that the mechanism was independent of (i) herbicide molecular structure, i.e. the position of the chloro group on the phenol ring, and (ii) the absolute configuration of the carbon atom. Enantioselectivity was enhanced by increasing the mobile phase methanol content. Use of this approach also revealed that secondary sites on the teicoplanin surface were involved in the processes determining both retention and selectivity. It was clearly demonstrated that these secondary sites of low affinity were not affected by the temperature change and were not involved in the chiral recognition mechanism.  相似文献

测定了乙醇在不同温度下的粘度,比较了乙醇与甲醇的理化性质和作反相高效液相色谱(RP HPLC)溶剂的特点。用乙醇 水作RP HPLC流动相测定中药有效成分,并将测定结果与甲醇 水或乙腈 水作流动相的测定结果进行比较。研究结果表明,选择合适的柱温等色谱条件,乙醇一般可以代替甲醇或乙腈用作RP HPLC流动相。  相似文献

This paper reports the development and validation of an assay for the determination of acetonitrile in the recycled mobile phase using high performance liquid chromatography(HPLC). The method is based on that the retention in reversed-phase liquid chromatography increases with decreasing concentration of organic phase in the mobile phase. The natural logarithm of the capacity ratio for a given solute is linearly related to the volume fraction of the organic modifier in the mobile phase. For dimethylphthalate and diethylphthalate, the linearity range is 30%--60%, and for biphenyl and terphenyl, the range is 60%--95%. Precision values(RSD) were both 〈1% and the accuracy(RE) was in the range of ±1%. The assay was successfully applied to the determination of acetonitrile concentration of recycled mobile phase after the distillation of the column eluent in our laboratory.  相似文献

Summary An equation describing RM values in TLC with a binary mobile phase has been derived and examined by using TLC data. Two different methods have been proposed to determine the chromatographic parameters characterizing energetic heterogeneity of the adsorption system and solute-solvent interactions. These methods differ in the estimation of the mole fractions of solvents in the surface phase. In method I an analytical equation for evaluating these mole fractions is assumeda priori.  在硅胶薄层板上用氯仿-乙酸乙酯-甲醇-水(体积比为15∶40∶22∶10)作展开剂,测定了常见人参皂甙的Rf值。为了研究它们的结构与保留值之间的关系,对它们的17种结构参数进行了计算。除了拓扑指数和理化参数外,引入了代表构成人参皂甙的母体化合物种类并反映它们分子极性特征的新参数“E”。通过相关分析优化选出范德华分子表面积AW、拓扑指数0B和参数E,建立了多参数线性回归方程,较好地描述了在正相薄层色谱中常见人参皂甙结构与保留值之间的关系,并与人工神经网络方法进行了比较。  相似文献

The retention behaviour of fifteen closely related coumarins in normal-phase overpressured layer chromatography (OPLC) was studied with the aim of comparing the retentions with those in normal-phase thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC) when optimization of the mobile phase was carried out according to the PRISMA system. The mobile phase optimization was carried out on TLC plates in unsaturated chambers. The resulting mobile phases were transposed to off-line, non-equilibrated OPLC and further to HPLC. The retention in TLC was measured at 37 selectivity points and in OPLC and HPLC at 13 points. Capacity factors (k′) and separation factors () were calculated in order to study the retention behaviour in the different systems. Two- and three-dimensional evaluations of k′ against selectivity points showed similar retention behaviours for the coumarins in TLC, OPLC and HPLC. The values for TLC, OPLC and HPLC showed similar patterns in the three-dimensional evaluations. The retention behaviour at different solvent strengths was also examined. According to quadratic regression, k′ showed a dependence on the change in solvent strength. OPLC, which can be considered as a “planar column” technique, and TLC are closely related methods, whereas HPLC shows a different behaviour in the elution process with regard to solvent strength.  相似文献

The adsorption isotherms of methanol in water solutions which were used as mobile phase in RP HPLC on adsorbents modified with dimethyloctadecylchlorosilane were measured. On the basis of these isotherms the chemical composition of the stationary phase as a function of the mobile phase concentration was determined. The influence of both those phases on retention factors k′ of alkyl derivatives of benzene was discussed. It was stated that k′ of benzene and its derivatives decreases with the various rate together with the increase of the methanol concentration in both phases. Quotient of volume coefficient corresponding to a unit of adsorbed methanol (logk′/ns Me) decreases linearly.
