Isothiazolinones are used as preservatives, biocides and disinfectants in a variety of industrial and domestic applications. Some of the isothiazolinones are difficult to isolate from water due their high polarity. A sensitive and selective analytical method was developed and optimized for the determination of sub-μg/L levels of three isothiazolinones in water samples. Three isothiazolinones are described in this paper: 2-methyl-3-isothiazolinone, 5-chloro-2-methyl-3-isothiazolinone and 4,5-dichloro-2-methyl-3-isothiazolinone. The analytical method involves a robust large-volume direct on-column loop injection of 2 mL on an Aqua ODS HPLC column and tandem MS detection (HPLC–MS/MS). After filtration, samples are directly injected without further pretreatment. Detection limits of the individual target compounds were between 0.03 and 0.1 μg/L employing Multi-Reaction Monitoring (MRM) mass spectrometry. Based on the constant ratio of two selected product-ions together with the retention time, the identification is very selective and quantification is reliable. The method was successfully applied to real samples of membrane flushings, drinking water, surface waters and waste water. Additional investigations showed the instability of the isothiazolinones in river- and waste water. Preservation of river water and waste water samples with sodium azide (NaN3) promotes the stability of the isothiazolines in solution. In membrane flushings, waste water, surface waters and drinking water, levels of the three isothiazolinones were all below the limit of detection. In effluents of households containing washings from normal shampoo use, isothiazolinones could be detected.  相似文献

A liquid chromatography-tandem mass spectrometry (LC-MS/MS)-based method for the detection of pesticides in tap and treated wastewater was developed and validated according to the ISO/IEC 17025:1999. Key features of this method include direct injection of 100 μL of sample, an 11 min separation by means of a rapid resolution liquid chromatography system with a 4.6 mm × 50 mm, 1.8 μm particle size reverse phase column and detection by electrospray ionization (ESI) MS-MS. The limits of detection were below 15 ng L−1 and correlation coefficients for the calibration curves in the range of 30-2000 ng L−1 were higher than 0.99. Precision was always below 20% and accuracy was confirmed by external evaluation. The main advantages of this method are direct injection of sample without preparative procedures and low limits of detection that fulfill the requirements established by the current European regulations governing pesticide detection.  相似文献

The LC-MS/MS analysis of a group of 14 organic pollutants in wastewater – including pharmaceuticals (analgesics/anti-inflammatories, lipid regulators and diuretics), pesticides (diuron) and disinfectants (chlorophene) – has been carried out using a hybrid triple quadrupole-linear ion trap-mass spectrometer (QqLIT). In order to take advantage of the capabilities of the QqLIT system, two methods have been developed and compared, based on the application of different operation modes. One of them uses selected reaction monitoring (SRM), which is the standard mode for quantitative LC-MS/MS analysis. The other is based on the use of an information dependent acquisition scan function (IDA), which allows the combination of a SRM acting as the survey scan and an enhanced product ion scan (EPI) as dependent scan within the same analysis. Performance of both methods was compared, especially in terms of their limits of detection and identification capability. The advantages and limitations of both techniques are discussed. Finally, the two methodologies developed were applied to real samples for evaluation of effluent wastewater in a treatment plant on the south-eastern Mediterranean coast of Spain. The presence of most of the target compounds was detected at mean concentrations ranging from 50 ng/L (mefenamic acid) to 3373 ng/L (hydrochlorothiazide).  相似文献

Matrix effect profiles can be used to visualize the effect of the sample matrix to the data signals occurring in a chromatogram. In the present study these profiles were generated by postcolumn infusion of a standard pesticide mix with extracts of different food matrices prepared by the QuEChERS method. Complete raw extracts as well as individual clean-up steps were analyzed. This allowed for a detailed comparison of the interferences caused by the matrix effects from various food samples. It also gave an idea about the efficiency of matrix reduction processes. When analyzing the individual clean-up extracts of the QuEChERS method just a slight reduction of matrix effects could be observed from step to step. Matrices causing strong signal effects in the results of the raw extracts also have strong effects after the final clean-up step. Some of the components responsible for the matrix effects show an extremely high retention time. After the injection of extracts from rocket or different types of tea, significant ion suppressions occurred even after rinsing the analytical column for a long time. The experiments have shown that similar matrices can produce different matrix effect profiles. For example, for black teas and green teas significantly different matrix effect profiles were obtained, while the matrix effects of teas within one of these groups were exactly the same. Analogous results could be found for citrus fruits. In order to overcome interfering matrix effects, analytical systems equipped with different electrospray ion sources were tested. Furthermore, profiles of diluted food extracts were generated. Dilution led to a significant decrease in the matrix effects.  相似文献

As a suitable way for routine screening of pesticides and control of other organic contaminants in water, the combination of liquid chromatography triple quadrupole tandem mass spectrometry (LC–QqQ-MS/MS) and liquid chromatography–hybrid quadrupole time-of-flight mass spectrometry (LC–QTOF-MS) has been applied to the analysis of 63 surface and waste water samples after conventional solid-phase extraction (SPE). The extracts were screened for 43 pesticides or degradation products by LC–QqQ-MS/MS achieving limits of detection (LOD) ranged from 0.04 to 2 ng L−1. Of the 43 selected pesticides, 33 were detected in water samples. The ESI–QTOF MS instrument was run using two simultaneous acquisition functions with low and high collision energy (MSE approach) and acquiring the full mass spectra. A home-made database containing more than 1100 organic pollutants was used for substance identification. Around 250 of these compounds were available at the laboratory as reference standards. Five pesticides and 3 of their degradation products, different to those selected in the QqQ method, were detected by QqTOF-MS. Thirteen pharmaceuticals and two drugs of abuse were also identified in the samples. In practice, the sample preparation proved to be suitable for both techniques and for a wide variety of substances with different polarity. Mutual confirmation and evidence of co-occurrence of several other organic contaminants were the main advantages of the combination of both techniques.  相似文献

An efficient, high throughput and cost-effective direct aqueous injection approach for the analysis of neonicotinoid pesticides and a common metabolite in environmental water has been described here. The method determines eight neonicotinoid pesticides (acetamiprid, clothianidin, dinotefuran, flonicamid, imidacloprid, nitenpyram, thiacloprid, thiamethoxam) and 6-chloronicotinic acid (a common metabolite of the first generation neonicotinoids, acetamiprid, imidacloprid, nitenpyram and thiacloprid) without any sample enrichment/cleanup steps. The method detection limits are 2–8 ng/L for the neonicotinoids and 93 ng/L for 6-chloronicotinic acid. The performance of the QTRAP®5500 mass spectrometer was compared against a 4000QTRAP®, and a QTRAP®6500, to provide insights for future method transfer among different generations of instrumentations. Critical mass spectrometric parameters such as collision energy were quite consistent among the three instruments evaluated. However, increased chemical background levels for some target compounds on the more sensitive instruments were observed. The application of differential ion mobility spectrometry combined with tandem mass spectrometry was demonstrated to have great potential in reducing chemical background and/or isobaric interferences inherited in sample matrices. This ISO 17025 accredited method was employed to quantitate neonicotinoids in Ontario stream water samples. Good correlation for analytical results of this direct aqueous injection approach and a previously published solid phase extraction approach warrant high confidence in data quality.  相似文献

A simple and rapid SPE-GC method for the determination of 30 organochlorine, organophosphorus, and organonitrogen pesticides at ng/L levels in washing water of olive processing has been developed. Twenty-nine samples of water were collected directly in the inlet and outlet of the washing machines of three olive mills at various locations in Spain. Preconcentration and clean-up were performed by elution of the pesticides from a C18 SPE cartridge using dichloromethane. The target compounds were determined in the final extract by GC-MS/MS. The most frequently encountered pesticide residues were terbuthylazine, simazine, and diuron. In some of the water samples, coming from the inlet of the washing machines, terbuthylazine and diuron concentrations were above the maximum residue limit.  相似文献

应用高效液相色谱-串联质谱(HPLC-MS-MS)联用技术,建立了检测水体中氨基甲酸酯类农药残留的方法。水样经氨基柱固相萃取,二氯甲烷洗脱,梯度流动相洗脱,电喷雾正离子(ESI+)模式采集、选择反应性监测模式(SRM)对定性、定量离子进行MS/MS测定。5种氨基甲酸酯类农药在5~200ng/L范围内线性关系良好,回收率为80%~92%,相对标准偏差均小于10%,方法检出限分别为:异丙威和仲丁威为1.4ng/L,克百威和甲萘威为1.5ng/L,恶虫威为1.6ng/L。该方法适用于水体中氨基甲酸酯类农药残留的检测。  相似文献

The aim of this study is to propose an analytical method for determining different classes of pesticides in water using LC-ESI-MS/MS. Two techniques of field-sampling and analyte extraction were used: solid phase extraction (SPE) of water samples from active sampling and field exposure of Polar Organic Chemical Integrative Samplers (POCIS). We have worked with thirty-three molecules representing eight pesticide classes: carbamates, chloroacetanilides, dicarboximides, morpholines, organophosphorous, phenylureas, strobilurines and triazines. First, liquid chromatography separation protocols and the optimization of the ESI-MS/MS parameters were developed. Then, the SPE step was optimized to obtain acceptable levels of recovery for the various classes of molecules. The matrix effect that may significantly lower the ionization efficiency with ESI interfaces was evaluated and minimized. The performances (limits of quantification, accuracy and precision) of the SPE and POCIS techniques were evaluated, and a comparison between the active and passive sampling techniques was carried out with a field application.  相似文献

Summary Procedures for the quantitative analysis of industrial effluents which involve concentration by solvent extraction or the purge-and-trap method are time-consuming, labor-intensive, and prone to error. Direct aqueous injection gas chromatography using an electron-capture detector for the analysis of volatile halocarbons at the ppb level is in routine use in many laboratories. We now discuss the development of a similar protocol for the analysis of volatile polar organics such as acetone, methyl ethyl ketone, methyl isobutyl ketone, and tretrahydrofuran using a flame-ionization detector.  相似文献

Reversed-phase liquid chromatography (RPLC) and electrospray (ESI)-linear ion trap (LIT) mass spectrometry was applied to the direct characterization of in vivo S-nitrosylated (SNO) phytochelatins (PCs) expressed in cadmium-stressed Arabidopsis thaliana cells. Cys-nitrosylation is under discussion as in vivo redox-based post-translational modification of proteins and peptides in plants in which the –NO group is involved as signal molecule in different biological functions. The gas-phase ion chemistry of in vivo and in vitro generated SNO-PCs was compared with the aim of evaluating NO binding stability and improving MS knowledge about peptide nitrosation. Using RPLC separation and ESI-LIT-MS, mono-nitrosylated PCs were identified in in vivo cadmium treated A. thaliana cells without derivatization. The in vivo binding of the NO group to PC2, PC3 and PC4 resulted to occur selectively on only one cystein residue. The fragmentation pathway energies of the in vitro GSNO-generated NO-PCs with respect to the in vivo NO-PCs were investigated, suggesting the presence of a different internal stability for these molecules. By carrying out MS2 experiments on these quasi-symmetric peptides, the different stability degree of the NO group was demonstrated to be correlated with the PC chain length. In addition, the data obtained highlight a putative role of the adjacent Glu/Cys motif in the gas-phase stability of the NO-containing molecule.  相似文献

This study describes an SPE coupled with large volume injection (LVI) analytical method for the analysis of organochlorine pesticides, BHC (alpha, beta, delta), aldrin, endosulfan (alpha, beta), endrin, dieldrin, and DDT, from aqueous samples. Determination was carried out by GC with MS. The LODs of organochlorine pesticides were determined at 10 ng/L concentration levels, and the results show that SPE-LVI-GC/MS has the potential to accurately determine organochlorine pesticides in water, as it avoids analyte classes in the various steps of a typical extraction procedure.  相似文献

A sensitive rapid resolution liquid chromatography-tandem mass spectrometry (RRLC-MS/MS) method, combined with solid-phase extraction, ultrasonic extraction and silica gel cartridge cleanup, was developed for 28 steroids including 4 estrogens (estrone (E1), 17β-estradiol (E2), 17α-ethynyl estradiol (EE2), diethylstilbestrol (DES)), 14 androgens (androsta-1,4-diene-3,17-dione (ADD), 17α-trenbolone, 17β-trenbolone, 4-androstene-3,17-dione, 19-nortestoserone, 17β-boldenone, 17α-boldenone, testosterone (T), epi-androsterone (EADR), methyltestosterone (MT), 4-hydroxy-androst-4-ene-17-dione (4-OHA), 5α-dihydrotestosterone (5α-DHT), androsterone (ADR), stanozolol (S)), 5 progestagens (progesterone (P), ethynyl testosterone (ET), 19-norethindrone, norgestrel, medroxyprogesterone (MP)), and 5 glucocorticoids (cortisol, cortisone, prednisone, prednisolone, dexamethasone) in surface water, wastewater and sludge samples. The recoveries of surface water, influents, effluents and sludge samples were 90.6-119.0% (except 5α-DHT was 143%), 44.0-200%, 60.7-123% and 62.6-138%, respectively. The method detection limits for the 28 analytes in surface water, influents, effluents and freeze-dried sludge samples were 0.01-0.24 ng/L, 0.02-1.44 ng/L, 0.01-0.49 ng/L and 0.08-2.06 ng/g, respectively. This method was applied in the determination of the residual steroidal hormones in two surface water of Danshui River, 12 wastewater and 8 sludge samples from two wastewater treatment plants (Meihu and Huiyang WWTPs) in Guangdong (China). Ten analytes were detected in surface water samples with concentrations ranging between 0.4 ng/L (17β-boldenone) and 55.3 ng/L (5α-DHT); twenty analytes in the wastewater samples with concentrations ranging between 0.3 ng/L (P) and 621 ng/L (5α-DHT); and 12 analytes in the sludge samples with concentrations ranging between 1.6 ng/g (E1) and 372 ng/g (EADR).  相似文献

Efficient clean-up is indispensable for preventing matrix effects in multi-residue analysis of pesticides in food by liquid and gas chromatography coupled to mass spectrometry. As a completely new approach, highly automated planar chromatographic tools were applied for powerful clean-up, called high-throughput planar solid phase extraction (HTpSPE). Thin-layer chromatography (TLC) was used to completely separate pesticides from matrix compounds and to focus them into a sharp zone, followed by extraction of the target zone by the TLC-MS interface. HTpSPE resulted in extracts nearly free of interference and free of matrix effects, as shown for seven chemically representative pesticides in four different matrices (apples, cucumbers, red grapes, tomatoes). Regarding the clean-up step, quantification by LC-MS provided mean recovery (against solvent standards) of 90-104% with relative standard deviations of 0.3-4.1% (n=5) for two spiking levels of 0.1 and 0.5 mg/kg. Clean-up of one sample was completed in a manner of minutes, while running numerous samples in parallel at reduced costs, with very low sample and solvent volumes.  相似文献

Summary The determination of selected pesticides and phenols in Portuguese river water samples was carried out from April to September, 1999. The method involved 200 mL samples taken by offline, solid phase extraction by OASIS polymeric cartridges followed by liquid chromatography-atmospheric pressure, chemical ionization-mass spectrometry (LC-APCI-MS). Recoveries of pesticides were 50–96% and 72–120% for the Platform and HP 1100 instruments, respectively. Chlorophenols gave recoveries of 60–91%. Triazines and transformation products like desethylatrazine (DEA) and desisopropylatrazine (DIA) and compounds such as diuron and chlorophenols were positively identified by LC-APCI-MS. The levels detected of the different compounds varied from 0.01–2.61 μg L−1, the most frequently detected compounds being, atrazine, simazine, terbuthylazine, alachlor, metolachlor, Irgarol, diuron, 2,4,6-trichlorophenol, desisopropylatrazine and desethylatrazine.  相似文献

Summary The potential of large-volume PTV injection was studied for the analysis of triazine herbicides in water samples. Direct water injection and in-vial extraction were described and compared. Detection limits were between 0.01–0.02 μg L−1 and relative standard deviations were <9%. Both methods are suitable for the analysis of triazines at ppt-level, although in-vial extraction is favourable for water samples with relatively large amounts of matrix components.  相似文献

An analytical method was developed to quantitatively determine pharmaceuticals in biosolid (treated sewage sludge) from wastewater treatment plants (WWTPs). The collected biosolid samples were initially freeze dried, and grounded to obtain relatively homogenized powders. Pharmaceuticals were extracted using accelerated solvent extraction (ASE) under the optimized conditions. The optimal operation parameters, including extraction solvent, temperature, pressure, extraction time and cycles, were identified to be acetonitrile/water mixture (v/v 7:3) as extraction solvent with 3 extraction cycles (15 min for each cycle) at 100 °C and 100 bars. The extracts were cleaned up using solid-phase extraction followed by determination by liquid chromatography coupled with tandem mass spectrometry. For the 15 target pharmaceuticals commonly found in the environment, the overall method recoveries ranged from 49% to 68% for tetracyclines, 64% to 95% for sulfonamides, and 77% to 88% for other pharmaceuticals (i.e. acetaminophen, caffeine, carbamazepine, erythromycin, lincomycin and tylosin). The developed method was successfully validated and applied to the biosolid samples collected from WWTPs located in six cities in Michigan. Among the 15 target pharmaceuticals, 14 pharmaceuticals were detected in the collected biosolid samples. The average concentrations ranged from 2.6 μg/kg for lincomycin to 743.6 μg/kg for oxytetracycline. These results indicated that pharmaceuticals could survive wastewater treatment processes, and accumulate in sewage sludge and biosolids. Subsequent land application of the contaminated biosolids could lead to the dissemination of pharmaceuticals in soil and water environment, which poses potential threats to at-risk populations in the receiving ecosystems.  相似文献

Summary A new mixed-functional silica material has been developed for direct injection analysis of drugs in serum. The mixed-functional material is synthesized from porous silica by three steps; introduction of 3-glycidoxypropyl groups, introduction of phenyl groups, and hydrolysis of the oxirane ring to diol groups. The prepared column packing can be used for direct injection analysis of hydrophobic and hydrophilic drugs in serum over the eluent pH range employed for ordinary siloxane-bonded silica. The recovery of drugs from serum was almost 100%, regardless of the difference in their protein bindings.  相似文献

A novel and sensitive method was developed for the simultaneous determination of fenbuconazole and its main metabolites enantioselectively using chiral liquid chromatography coupled with tandem mass spectrometry. The separation and determination were performed using reversed-phase chromatography on a cellulose chiral stationary phase, a Chiralcel OD-RH (150 mm×4.6 mm) column, under isocratic conditions at 0.5 mL/min flow rate. The effects of three cellulose-based columns and three amylose-based columns on the separation were also investigated. The elution orders of the eluting enantiomers were identified by an optical rotation detector. The QuEChERS (acronym for Quick, Easy, Cheap, Effective, Rugged and Safe) method and solid-phase extraction (SPE) were used for the extraction and clean-up of the soil and water samples, respectively. Parameters including the matrix effect, linearity, precision, accuracy and stability were evaluated. Under optimal conditions, the mean recoveries for all enantiomers from the soil samples were 82.5-104.1% with 2.7-9.5% intra-day relative standard deviations (RSD) and 5.7-11.2% inter-day RSD at 5, 25 and 50 μg/kg levels; the mean enantiomer recoveries from the water samples were 81.8-104.6% with 2.6-11.4% intra-day RSD and 5.3-10.4% inter-day RSD at 0.25, 0.5 and 2.5 μg/L levels. Coefficients of determination R2≥0.9991 were achieved for each enantiomer in the soil and water matrix calibration curves within the range of 1.0-125 μg/L. The limits of detection (LOD) for all enantiomers in the soil and water were less than 0.8 μg/kg, whereas the limit of quantification (LOQ) did not exceed 2.5 μg/kg. The results of the method validation confirm that this proposed method is convenient and reliable for the enantioselective determination of the enantiomers of fenbuconazole and its main metabolites in soil and water.  相似文献
