A new synthesis of the alkaloid (±)-cryptopleurine via anodic oxidation

The alkaoid (±)-cryptopleurine 1 was synthesized by anodic oxidation of the quinolizidinone 3 followed by subsequent transformation of 6a to 1 and 5a to 6a.     

Synthesis of E and Z 1-amino-2-aryl(alkyl)-cyclopropanecarboxylic acids via meldrum derivatives

The reaction of 5-arylidene(alkylidene)-2,2-dimethyl-1,3-dioxane-4,6-diones (1) (Meldrum's acid derivatives) with dimethylsulfoxonium methylide gave 1- aryl(alkyl) - 6,6 - dimethyl - 4,8 - dioxo - 5,7 -dioxaspiro [2.5] octanes (2) which, on treatment with sodium methoxide or ammonium hydroxide, gave exclusively E-1-methoxy-carboyl-2-aryl-cyclopropanecarboxylic acids (4) or Z-1-carbamoyl-2-aryl(alkyl)-cyclopropanecarboxylic acids (7), respectively. Compounds, 4, under conditions of Curtius-type reactions, yielded Z-methyl 1-isocyanate-2-aryl-cyclopropanecarboxylates (5), while derivatives 7 were treated with hypobromite, leading to E-1-methoxy-carbonylamino-2-aryl(alkyl)-cyclopropanecarboxylic acids (8).Reaction of compounds 5 and 8 with hydrochloric acid produced the corresponding Z and E 1-amino-2-aryl (alkyl)-cyclopropanecarboxylic acids hydrochlorides (6). The ¹H-NMR spectral data were analyzed to deduce the stereochemistry of the compounds obtained.     

A new direct synthesis of cinnamic acids from aromatic aldehydes and aliphatic carboxylic acids in the presence of sodium borohydride

Cinnamic acids have been prepared in 59-86% yields by a new direct synthesis from aromatic aldehydes and aliphatic carboxylic acids in the presence of sodium borohydride and N-methyl-2-pyrrolidinone (NMP) as solvent, at reflux (185-190°C), for 9-12 hours. Without sodium borohydride, this reaction is not possible.     

Determination of Na(2)O from sodium aluminate NaAlO(2)

The aim of the work was to find a suitable method and conditions for determining Na(2)O wt.% from NaAlO(2). Problems were encountered while titrating NaAlO(2) with hydrochloric acid. The problematic area was the pH range 4-10 where aluminum precipitates as hydroxides. The different species of the aluminate solution were determined using potentiometric and complexometric titrations. The equivalent point of the potentiometric titration was detected using Gran's plotting method. Precipitation of aluminum hydroxides did not interfere with titrations, because in potentiometric titrations the pH value was over 10 and in complexometric titrations the pH was 4. The results were accurate and determinations were easy to carry out. Sodium was also determined by DCP-AES.     

Transition-metal-free synthesis of (Z)-3-ylidenephthalides from 2-acyl-benzoic acids

We report a highly efficient method for the synthesis of (Z)-3-ylidenephthalides via intramolecular cyclization of readily available 2-acyl-benzoic acids mediated by TSTU at room temperature. Using this method, diversely substituted (Z)-3-ylidenephthalides have been generated in good to excellent yields. The application of the method is highlighted by gram-scale preparation of the antiplatelet drug n-butylphthalide.     

A new method for [c,d]pyridine peri-annelation: synthesis of azapyrenes from phenalenes and their dihydro derivatives

An effective synthesis of various azapyrenes from phenalenes and their dihydro derivatives has been developed using 1,3,5-triazines in polyphosphoric acid (PPA).     

An efficient carbapenem synthesis via an intramolecular Wittig reaction of new trialkoxyphosphorane-thiolesters

New trialkoxyphosphorane-thiolesters 10, obtained by reaction of oxalimides 9 with trialkyl phosphite, were efficiently cyclized by an intramolecular Wittig reaction to give carbapenems 11.     

A new (R)-hydroxynitrile lyase from Prunus mume: asymmetric synthesis of cyanohydrins

A new hydroxynitrile lyase (HNL) was isolated from the seed of Japanese apricot (Prunus mume). The enzyme has similar properties with HNL isolated from other Prunus species and is FAD containing enzyme. It accepts a large number of unnatural substrates (benzaldehyde and its variant) for the addition of HCN to produce the corresponding cyanohydrins in excellent optical and chemical yields. A new HPLC based enantioselective assay technique was developed for the enzyme, which promotes the addition of KCN to benzaldehyde in a buffered solution (pH=4.5).     

A novel synthesis of bis-(α-hydroxyalkyl)phosphinic acids involving microwave irradiation

A simple, efficient, and general method has been developed for the synthesis of bis-(α-hydroxyalkyl)phosphinic acids from hypophosphorous acid using microwave irradiation. Bis-(α-hydroxyalkyl)phosphinic acids were obtained in high yield under mild conditions by reaction of hypophosphorous acid with aldehydes under microwave irradiation.     

Enantioselective synthesis of unsymmetrical benzoins from (S)-mandelic acid enolate and aromatic aldehydes

The reaction of the lithium enolate of the 1,3-dioxolan-4-one derived from optically active (S)-mandelic acid and pivalaldehyde with aromatic aldehydes proceeds readily to give the corresponding aldol products in good yields and diastereoselectivities. Subsequent hydroxyl protection, basic hydrolysis of the dioxolanone, oxidative decarboxylation of the α-hydroxyacid moiety, and hydroxyl deprotection provides chiral unsymmetrical benzoins with high enantiomeric excesses.     

Transition metal free one pot synthesis of aryl carboxylic acids via dehomologative oxidation of styrenes

Iodine/NaOH-catalyzed one-pot dehomologative oxidation of styrenes to aryl carboxylic acids has been reported. A wide range of carboxylic acids are obtained using iodine (I₂) as a catalyst, tert-butyl hydroperoxide (TBHP) as an oxidant and sodium hydroxide (NaOH) as a base. This reliable conversion involves dehomologation of styrene to aromatic aldehyde which on subsequent oxidation affords aryl carboxylic acid. This protocol was used for gram-scale synthesis as it is free from chromatographic purification. This is the first report for the oxidative transformation of styrenes into aryl carboxylic acids under transition metal-free conditions.     

An efficient synthesis of indoles via a CuMgAl-LDH-catalyzed cyclization of 2-alkynylsulfonanilides

A highly efficient method for the synthesis of indoles has been successfully developed via a CuMgAl-LDH-catalyzed intramolecular annulation reaction of 2-alkynylsulfonanilides. This CuMgAl-LDH catalyst features facile preparation, recovery, and reuse at least seven times without a marked loss in the catalytic activity, as well as the unique dual activation. Moreover, the crystal structures and Hirshfeld surface analysis of typical indole compounds were also presented.     

A new method for the conversion of aldehydes (RCH2CHO)to acetylenes (RC&.tbnd;CH) via 1-alkenylstannanes. Application to the synthesis of 9(O)-thia-Δ6-PGI1

A method for the conversion of aldehydes to acetylenes via 1-alkenylstannanes is reported, including its application to the synthesis of 9(O)-thia-Δ⁶-PGI₁.

A practical synthesis of aryl sulfones via cross-coupling of sulfonyl hydrazides with aryltriazenes using copper/ionic liquid combination

A new and efficient approach adopting copper-catalyzed cross-coupling of sulfonyl hydrazides with aryltriazenes has been developed to synthesize aryl sulfones using Brønsted acidic ionic liquid as promoter under ambient conditions. The process employs stable and easy to handle reacting partners, and is endowed with broad substrate scope.     

One-pot synthesis of bis (phenylimino dihydro thiazolyl-2H-chromene) derivatives

A novel series of bis (phenylimino dihydro thiazolyl-2H-chromene) derivatives have been synthesized through an efficient one-pot multicomponent approach involving different substituted of 3-(2-bromoacetyl)-2H-chromen-2-ones, phenyl isothiocyanates and para-phenylenediamine in presence of dimethylformamide solvent. The target compounds were obtained in a shorter reaction time through simple reaction work up with good yields. All the synthesized products were characterized by their spectral and analytical data like IR, ¹H NMR, ¹³C NMR, Roesy, and mass spectral data.     

A novel system for the synthesis of nitriles from aldehydes using aqueous ammonia and sodium dichloroiodate

A simple and mild method for the conversion of varieties of aldehydes to the corresponding nitriles using aqueous ammonia and aqueous sodium dichloroiodate reagent at room temperature is discussed. Advantages of this system are short reaction time, easy work-up and moderate to good yields.     

One-step facile synthesis of deuterium labeled aldehydes from tertiary amides using Cp2Zr(D)Cl

The synthesis of deuterium labeled aldehydes was accomplished in an expedient and facile manner through the reduction of amides using commercially available Cp₂Zr(D)Cl. The reactions proceeded rapidly (ca. 15 min) and in high yields (70-99%).
