Abstract

Residual carboxyl groups in gel permeation column materials (hydrophilic polymer gels) (Asahipak GS-520) were methylated with boron trifluoride methanol complex to minimize the interaction of metals between the ligands in substrates and in gel materials. Although zinc ions were eluted very slowly as an extremely broad peak on the original column, the metal ions were eluted faster as a lesser broad peak on the methylated column. Cadmium ions were eluted faster as a relatively sharp peak on the methylated column than the original column. Alkali earth metal ions were eluted also as sharper peaks on the methylated column. Zinc in rat serum was eluted more with globulins and less with albumin on the methylated column than on the original column. Globulins and albumin in rat and human sera were separated more efficiently on the methylated column. These results suggest that methylation of residual carboxyl groups in gel materials decreased the interaction of metals with gel materials and increased hydrophobicity of the gel materials.  相似文献

A novel open‐tubular capillary electrochromatography column coated with β‐cyclodextrin was prepared using the sol‐gel technique. In the sol‐gel approach, owing to the three‐dimensional network of sol‐gel and the strong chemical bond between the stationary phase and the surface of capillary columns, good chromatographic characteristics and unique selectivity in separating enantiomers were shown. The influences of capillary inner diameter, coating time, organic modifier, buffer pH, and buffer concentration on separation were investigated. The sol‐gel‐coated β‐cyclodextrin column has shown improved enantioseparation efficiency of chlorphenamine, brompheniramine, pheniramine, zopiclone in comparison with the sol‐gel matrix capillary column. The migration time relative standard deviation of the separation of the enantiomers was less than 0.89% over five runs and 2.9% from column to column. This work confirmed that gold nanoparticles are promising electrochromatographic support to enhance the phase ratio of open‐tubular capillary electrochromatography column in capillary electrochromatography.  相似文献

Summary β-Cyclodextrin sulfate having heparin-mimicking activity was immobilized on a hydrophilic vinyl-polymer gel, TSKgel AF-Epoxy, TOYOPEARL 650M. A column packed with this material could be used for high-performance affinity chromatographic separation of heparin-binding substances such as growth factors, enzymes, coagulation factors and lipoproteins. The substances retained on the column were recovered more easily than those on a heparin-immobilized column. Furthermore, the β-Cyclodextin sulfate-immobilized gel was superior in stability to the heparin-immobilized gel when exposed to acidic and basic solutions.  相似文献

As one of approaches of developing novel HPLC stationary phases, we prepared Cu-octabromotetrakis(4-carboxyphenyl)porphine derivative-immobilized silica gels (Cu-OBTCPP(D)), and evaluated the availability of the resultant Cu-OBTCPP(D) as a stationary phase for separation of poly-aromatic-hydrocarbons (PAHs) and their related compounds. A Cu-OBTCPP(D) column was revealed to have an ability to separate simple PAHs and be useful as a stationary phase in both polar and non-polar eluents. The retention property of the Cu-OBTCPP(D) column was evaluated in various comparative experiments using commercially available columns. In comparison with 2-(1-pyrenyl)ethyl dimetylsilyl silica gel column (PYE column) regarding the retention behavior for PAHs etc., the Cu-OBTCPP(D) column showed stronger interactions involving pi electron in non-polar eluent than PYE column. In comparison with a pentabromobenzyloxy propylsilyl silica gel column (PBB column) regarding the influence of bromination, the Cu-OBTCPP(D) column was affected differently from the PBB column. In comparison with nitrophenylethyl silica gel column (NPE column) regarding the retention behavior for compounds having a dipole in a non-polar eluent, the Cu-OBTCPP(D) column showed electrostatic interactions such as dipole-dipole interaction equivalent to or larger than the NPE column.  相似文献

Heparin cofactor II (HCII) is an inhibitor of thrombin in human plasma whose activity is enhanced by heparin and dermatan sulphate. HCII was purified to homogeneity from normal human plasma with an overall yield of 7.5%. After treatment with barium chloride, precipitation with 50% saturated ammonium sulphate and dialysis of the resuspended precipitate against 0.02 M Tris-HCl (pH 7.4), the sample was chromatographed on a heparin-Sepharose CL 6B affinity column, DEAE-Sepharose CL 6B ion-exchange gel and an AcA 34 gel permeation column. For the final steps, a high-performance liquid chromatographic system was used which included ion-exchange chromatography on a Mono-Q column and gel permeation using a Superose column. The purified protein was homogeneous by sodium dodecyl sulphate-polyacrylamide gel electrophoresis. The specific activity of purified HCII was 12.2 U/mg. The HCII activity was evaluated as antithrombin dermatan sulphate cofactor activity. A specific antiserum against HCII was raised in the rabbit.  相似文献

A column packed with red blood cells (RBCs) was prepared for electrochromatography as a separation and reaction column. RBCs were kept inside a piece of fused silica capillary tubing with 2% agarose gel. In the column, RBCs were uniformly distributed in the agarose gel matrix and their electrophoretic movements due to an applied voltage were suppressed well. The durability of the biological function of the column under applied voltage was about 1 h, although it could remain for 2-3 days without applied voltage. The column could not be used when hemolysis of the RBCs was observed in the column. When the developed "RBC-gel column" was used, both pyridoxamine and serotonin were converted to other compounds through their direct contact with RBCs.  相似文献

In the genital tract of the male rat two different forms of the enzyme transglutaminase (TGase) could be identified and characterized. The coagulating gland and the dorsal prostate secrete a glycosylated and acylated TGase with a molecular weight of 65,000 dalton and pI value of 8.7. This secretory form was purified to homogeneity using preparative isoelectric focusing and gel filtration on a Superdex 200 column. Running fast protein liquid chromatographic gel filtration on a Superose 12 column in the presence of calcium ions, high-molecular-weight aggregates were physically formed which could only be eluted using drastic conditions (0.1 M sodium hydroxide). In the presence of 10 mM EDTA this tendency to aggregate was greatly diminished. Utilizing a Superdex 200 column for gel filtration, the secretory TGase was even eluted as a monomeric protein. Testicular TGase was isolated by ion-exchange fast protein liquid chromatography on a Mono Q and by gel filtration on a Superdex 200 column. This enzyme represents a tissue-type TGase with a molecular weight of 82,000 dalton and pI value of 5.25. Hydrophobic interaction chromatography on a phenyl-Superose column showed no further enrichment of the GTP-binding form of transglutaminase.  相似文献

Plant membrane lipids were separated by multiple solid-phase extraction (SPE) in a single run. Elution was performed continuously through the modulated stationary phase employing only non-aqueous solvent systems. At the different stages of the glycerolipid separation the SPE manifold combined arninopropyl, arninopropyl/silica gel and silica gel/aminopropyl weak anion exchanger columns. The glycerolipid extract of pigment-containing plant tissues was cleared from the pigments onto the aminopropyl column. The aminopropyl column with the glycerolipid extract was then connected to a silica gel column from which monogalactosyldiacylglycerol, phosphatidylethanolamine, phosphatidylglycerol and digalactosyldiacylglycerol were eluted as individual fractions. The elution was performed under polarity, pH and temperature gradient conditions. To continue the separation, the aminopropyl column was discarded and the silica gel column containing the remaining glycerolipid extract was connected to an aminopropyl anion exchanger column. Individual fractions of sulfoquinovosyldiacylglycerol, phosphatidylcholine and phosphatidylinositol were now eluted. The separation process was supported by ammonium counter ions and by the polarity gradient of the elution systems used. The membrane lipids were isolated from pigment-containing (rice and maize leaves and rice leafy stems) and pigment-free (rice roots) tissues. The repeatability for a standard glycerolipid mixture was 2-6% (n=7), and for rice leaf lipid extracts, 3-7% (n=5). Glycerolipid recovery was 87-95%.  相似文献

纤维素三(3,5-二甲基苯基氨基甲酸酯)是液相色谱中使用最广泛的手性柱。该文详细地研究了不同程度衍生的纤维素(3,5-二甲基苯基氨基甲酸酯)以及不同硅胶(粗制硅胶、氨丙基粗制硅胶、精制硅胶、氨丙基精制硅胶、大孔硅胶、氨丙基大孔硅胶)作为支撑体对该柱手性分离能力的影响。自制了13根手性色谱柱,分别考察了其对16种外消旋体的拆分,分离结果显示:三取代纤维素柱 > 二取代纤维素柱 > 纤维素柱;精制硅胶和大孔硅胶优于粗制硅胶,大孔硅胶的柱压更低;硅胶的氨丙基化对手性选择性有一定的影响;这些手性柱之间具有一定的互补性,尤其是纤维素柱。该文有助于人们更深刻地理解和更好地把握高效液相色谱手性柱的制备。  相似文献

A new chiral stationary phase (CSP) was developed for the direct optical resolution of enantiomeric amino acid derivatives. The CSP was readily prepared by a three-step reaction carried out in a pre-packed aminopropylsilyl silica gel column. In the first step, a solution of disuccinimido carbonate (DSC) was delivered through the pre-packed column to give a succinimido carbamyl aminopropylsilyl-bonded, activated-carbamate type silica gel (ACsil) column. Through the column was then delivered a solution of pentaethylenehexamine to afford a polyamine-bonded column. Finally, a solution of optically active succinimido (S)- or (R)-naphthylethyl carbamate was delivered through the polyamine column, to give a naphthylethylurea multiple-bonded CSP. p-Bromophenylcarbamyl derivatives of enantiomeric protein amino acids were resolved on these CSPs by elution with an aqueous mobile phase. Simultaneous analysis of these amino acid enantiomers by means of gradient elution was also accomplished.  相似文献

Fritless packed silica gel columns were prepared using sol‐gel technology. A part of a 75 μm i.d. fused silica capillary was filled with a mixture of tetramethoxysilane and poly(ethylene glycol). After gelling at 40°C and heating at 300°C, the resultant silica gel was derivatized with dimethyloctadecylchlorosilane. A scanning electron micrograph of a cross‐section of the capillary column showed that the gel took the form of a spherical particle aggregate and adhered to the column inner wall. The column performance was evaluated for electrochromatography using acetonitrile–50 mM HEPES buffer (pH 6.6) (60/40 or 40/60, v/v) as the mobile phase. An electroosmotic flow of 1.0 mm/s was generated with (60/40, v/v) acetonitrile/HEPES buffer at a field strength of 546 V/cm. Using a sol‐gel‐derived packed column at an electroosmotic flow of 0.5 mm/s, efficiencies of up to 1.1×105 plates/m were obtained for retained solutes.  相似文献

Summary This contribution to the problems of carbohydrate (sugar) column liquid chromatography, evaluates several currently used systems (reversed phase octadecylated silica gel, amino modified silica gel, cation exchangers on polystyrene and on silica gel basis, polyol derivatised silica gel and anion exchange systems). The elution pattern, analysis time, column efficiency and column life time expectancy are considered to be the important points for this comparison. The application of silica gel-based cation exchanger and of polyol derivatised silica gel is new to this field. The comparison and/or evaluation tries to be critical and to come to a conclusion for the choice of a recommended system.  相似文献

以rProtein A-琼脂糖凝胶为载体,同时偶联抗黄曲霉素B1( AFB1)、玉米赤霉烯酮( ZEN)和脱氧雪腐镰刀菌烯醇( DON)单抗,制备了AFB1-ZEN-DON三合一免疫亲和柱,并对非特异性吸附、柱空白、柱容量、柱效及样品加标回收率等指标进行评价。结果表明,0.25 mL胶对应的柱容量分别为:AFB1295 ng,ZEN 905 ng,DON 2342 ng;柱空白为0;rProtein A-琼脂糖凝胶(0.25 mL胶)对3种毒素的非特异吸附率均低于8%,3种毒素不同浓度的平均柱回收分别为97.4%、98.0%和98.4%。通过优化条件,选择80%甲醇-水(80:20, V/V)为提取溶剂,PBST稀释;FAPAS质控样本经不同批次三合一亲和柱净化后测定结果接近靶心值。制备的三合一免疫亲和柱能满足食品及饲料样品的前处理,可替代常规单一亲和柱,为多种毒素的一步富集、净化、检测奠定基础。  相似文献

以溶胶-凝胶(sol-gel)技术制备开管毛细管电色谱(OTCEC)柱,考察了制柱过程中盐酸pH值和凝胶溶液在毛细管内反应时间对柱性能的影响。对制得的电色谱柱进行了评价,在最佳制柱条件下,硫际、苯乙酸、萘、联苯、2,6-二甲基萘的平均柱效分别为46.6×104,52.4×104,50.2×104,47.7×104,35.8×104塔板数/m。  相似文献

Summary The influence of the alcohol content of the mobile phase and water, acetic acid and aniline as mobile phase additives on the generation and shape of two additional changes of the streaming current, generated inside the liquid chromatography column by injection of any sample and recorded before the responses of retained solutes, was studied in a normal-phase system using silica gel as the stationary phase. The mobile phases were based on a n-heptane-1-propanol mixtures. Under the same conditions the relationships between the column interparticle volume, the column void volume and the total liquid volume in the column and the retention volumes of these two streaming current responses, having the form of chromatographic peaks, were studied. The column void volume was identified with the retention volume of n-octane. The total liquid volume in the column (column hold-up) was calculated from the weight loss of the column wetted with water at first and then dried in nitrogen stream. The retention volume of the first streaming current response equals the column interparticle volume disregarding the mobile phase composition. If the 95∶5 n-heptane-1-propanol mobile phase contains water up to 80% of its saturated concentration (up to 0.114% by vol.), the retention volume of the second response agrees with the total volume of the liquid in the silica gel column, with a precision better than 2%. At a higher relative water saturation the retention volume of the second response increases, while the column void volume decreases. Both changes are explained by the spontaneous formation of a highly polar stagnant liquid in the pores of the silica gel.  相似文献

A novel open‐tubular CEC column coated with chitosan‐graft‐(β‐CD) (CDCS) was prepared using sol‐gel technique. In the sol‐gel approach, owing to the 3D network of sol‐gel and the strong chemical bond between the stationary phase and the surface of capillary columns, good chromatographic characteristics and unique selectivity in separating isomers were shown. The column efficiencies of 55 000~163 000 plates/m for the isomeric xanthopterin and phenoxy acid herbicides using the sol‐gel‐derived CDCS columns were achieved. Good stabilities were demonstrated that the RSD values for the retention time of thiourea and isoxanthopterin were 1.3 and 1.4% (run to run, n = 5), 1.6 and 2.0% (day to day, n = 3), 2.9 and 3.1% (column to column, n = 3), respectively. The sol‐gel‐coated CDCS columns have shown improved separations of isomeric xanthopterin in comparison with CDCS‐bonded capillary column.  相似文献

采用固液相表面连续反应法,先将偶联剂γ-[(2,3)-环氧丙氧]丙基三甲氧基硅烷(KH-560)键合到球形硅胶表面,然后再将植物有效成分丁香酚与硅胶上的KH-560活性基团反应,合成了丁香酚键合硅胶液相色谱固定相(EGSP)。采用元素分析、热重分析和红外光谱对该固定相的结构进行了表征。以萘作为溶质探针,乙腈-水(35:65, v/v)为流动相,流速为0.8 mL/min,测得EGSP柱的柱效。以一系列的中性、碱性和酸性化合物为溶质探针,C18柱和苯基柱作参比,对该固定相的色谱性能及保留机理进行了研究。结果表明,硅胶表面成功键合上了丁香酚配体,键合量为0.28 mmol/g, EGSP柱理论塔板数约为24707 N/m。该固定相不仅具有良好的反相色谱性能,同时由于配体结构中含有芳环、烯基和甲氧基,还能与溶质发生π-π电荷转移、偶极-偶极和氢键作用。与传统的反相C18柱和苯基柱相比,EGSP在极性芳香族化合物的快速、简便分离中占优势。  相似文献

建立了一种测定烟草中拟除虫菊酯类农药和有机磷类农药残留量的气相色谱法.用普通的聚苯乙烯凝胶柱代替昂贵的凝胶渗透色谱仪;层析柱填料用弗罗里硅土代替硅胶;用工作标准溶液过柱曲线作为标准曲线;有机磷类农药气相检测器用PFPD代替FPD.方法的检测量为0.01μg/g~0.04 μg/g,平均加标回收率为70.18%~95.86%,RSD为4.36﹪~9.96﹪,对样品进行了拟除虫菊酯类农药和有机磷类农药残留量的测定,结果满意.  相似文献

The radioisotope99Mo generated through the fission of neutron-irradiated uranium targets was separated in an extremely radiochemically pure state. The target dissolution mixture was run on a silica gel column whereby the95Zr−95Nb activity, the serious radiocontaminants of99Mo, was adsorbed on the gel. The effluent from the gel column was processed and treated with sodium dihydrogen phosphate. The processed solution was run on an activated alumina column whereby99Mo was adsorbed as phospho-molybdate complex ion, then desorbed from the column as the molybdate ion by eluting with 10% NaOH solution. By recycling the adsorption and desorption steps on alumnia,99Mo was finally obtained totally free from all other radiocontaminants.  相似文献
