The 7‐chloro‐3‐(2‐chlorobenzyl)‐ and 7‐chloro‐3‐(2‐fluorobenzyl)‐1,2,3‐triazolo[4,5‐d]pyrimidines ( 1 and 4 ), by nucleophilic replacement with some hydrazides, gave the corresponding 7‐hydrazidoderivatives ( 2a‐e and 5a‐e ). These, by heating in Dowtherm, underwent an intramolecular cyclization to form the new tricyclic 7‐substituted‐3‐(2‐chlorobenzyl)‐ and 3‐(2‐fluorobenzyl)‐1,2,3‐triazolo[4,5‐e]1,2,4‐triazolo[4,3‐c]pyrimidines ( 3a‐d and 6a‐d ). The 7‐hydrazino‐3‐(2‐chlorobenzyl)‐ and 7‐hydrazino‐3‐(2‐fluorobenzyl)‐triazolo‐pyrimidines ( 9a and 9b ) were also prepared via the corresponding mercapto ( 7a and 7b ) and thiomethyl ( 8a and 8b ) derivatives.  相似文献

Reactions of the pyridazine derivatives 1a–c with phenyl isothiocyanate followed by heterocyclization with ethyl chloroacetate gave the thiazolidinone derivatives 6a–c . Derivatives of the new ring system pyrrolo[3,4‐e][1,2,3] triazolo[1,5‐a]pyrimidine 6 were prepared in high yields in one step by reaction of 3‐azidopyrrole 3 and substituted acetonitriles. Compound 6b rearranged, upon heating in dimethyl sulfoxide in the presence of water, to pyrrolo[3,4‐d][1,2,3]triazolo‐[1,5‐a]pyrimidine 7.  相似文献

A series of 1‐substituted 4,5‐diformyl‐[1,2,3]triazole derivatives were prepared by 1,3‐dipolar cyclo‐addition of aryl azides with acetylene dicarboxaldehyde mono‐diethylacetal. The triazoles were readily converted into 1‐substituted [1,2,3]triazolo[4,5‐d]pyridazines in good yields. The 1‐(2‐nitrophenyl)‐[1,2,3]triazolo[4,5‐d]pyridazine was found to be a useful intermediate for the generation of the novel 5H‐benzo[1,2,3]triazolo[1′,2′:1,2]triazolo[4,5‐d]pyridazin‐6‐ium inner salt ring system.  相似文献

Cyclization of 4‐amino‐6‐methyl‐3‐propargylmercapto‐1,2,4‐triazine‐5‐one 3 and 4‐amino‐3‐propargyl mercapto‐1,2,4‐triazole derivatives 6 were afforded 1,2,4‐triazino[3,4‐b][1,3,4]thiadiazines 4 and 1,2,4‐triazolo[3,4‐b][1,3,4]thiadiazines 7 in presence of heteropolyacids, H14[NaP5W29MoO110] and H6P2W18O60 in high yields. Among used heteropoly acids, the yields were higher with H14‐P5Mo, caused to their high acid strengths.  相似文献

A number of new [1,2,4]triazolo[4,3‐b]pyridazines were prepared by either cyclocondesation of substituted hydrazinopyridazines with orthoesters or oxidative cyclization of their hydrazone analogs in nitrobenzene as an oxidizing agent. A host of other new [1,2,4]triazolo[4,3‐b]pyridazine derivatives were synthesized by sequential treatment of the latter compounds with carbon disulfide and alkyl halides.  相似文献

Several new pyrimido[4,5‐e ][1,2,4]triazolo[3,4‐b ][1,3,4]thiadiazine derivatives ( 5a , 5b , 5c , 5d , 5e , 5f , 5g ) were synthesized through the condensation reaction of 5‐bromo‐4,6‐dichloropyrimidine ( 1 ) and 4‐amino‐5‐methyl‐4H‐1,2,4‐triazole‐3‐thiol ( 2 ). The single crystal X‐ray data of one of the derivatives confirmed the occurrence of the S/N type Smiles rearrangement during the course of the reaction.  相似文献

5‐Amino‐4‐formyl pyrazole carboxylate gave facile reactions with malononitrile, hydrazine, and ketones in the presence of piperidine furnished substituted pyrazolo[3,4‐b]pyridines and pyrazolo[3,4‐b]quinolones. The pyridazine sulfonamides were obtained by the reaction of 5‐chloro 4‐formyl pyrazole carboxylate with sulfonamide derivatives.  相似文献

4‐Acetyl‐5‐methyl‐1‐phenyl‐1H‐pyrazole reacts with dimethylformamide dimethylacetal (DMF‐DMA) to afford the corresponding (E)1‐(5‐methyl‐1‐phenyl‐1H‐pyrazol‐4‐yl)‐3‐(N,N‐dimethylamino)‐2‐propen‐1‐one. The latter product undergoes regioselective 1,3‐dipolar cycloaddition with nitrilimines and nitrile oxides to afford the novel 3‐aroyl‐4‐(5‐methyl‐1‐phenyl‐1H‐pyrazol‐4‐yl)carbonyl‐1‐phenylpyrazole and 3‐aroyl‐4‐(5‐methyl‐1‐phenyl‐1H‐pyrazol‐4‐yl)carbonyl isoxazole derivatives, respectively. It reacts also with 1H‐benzimidazole‐2‐acetonitrile, 2‐aminobenzimidazole and 3‐amino‐1,2,4‐triazole to afford the novel pyrido[1,2‐a]benzimidazole, pyrimido[1,2‐a]benzimidazole and the triazolo[4,3‐a]pyrimidine derivatives, respectively. The reaction of 3‐aroyl‐4‐(5‐methyl‐1‐phenyl‐1H‐pyrazol‐4‐yl) carbonyl‐1‐phenylpyrazole derivatives with hydrazine hydrate led to a new pyrazolo[3,4‐d]pyridazine derivatives.  相似文献

A series of novel 6‐2‐methoxy‐5‐[4‐methoxy‐3‐(3‐aryl[1,2,4]triazolo[3,4‐b][1,3,4]oxadiazol‐6‐yl)benzyl]phenyl‐3‐aryl[1,2,4]triazolo[3,4‐b][1,3,4]oxadiazoles 7a , 7b , 7c , 7d , 7e , 7f , 7g , 7h , 7i , 7j has been synthesized and characterized via IR, 1H NMR, 13C NMR, MS, and elemental analyses. Compounds 7a , 7b , 7c , 7d , 7e , 7f , 7g , 7h , 7i , 7j were also screened for their antibacterial activity against Gram‐positive bacteria viz. Bacillus subtilis (MTCC 441), Bacillus sphaericus (MTCC 11), and Staphylococcus aureus (MTCC 96), and Gram‐negative bacteria viz. Pseudomonas aeruginosa (MTCC 741), Klobsinella aerogenes (MTCC 39), and Chromobacterium violaceum (MTCC 2656). The antibacterial screening reveal that the presence of 2,4‐difluorophenyl ( 7e ) or 4‐nitrophenyl ( 7f ) of 2‐pyrazyl ( 7i ), or 2‐furyl ( 7j ) on the triazole moiety exhibited potent inhibitory activity comparable with the standard drug streptomycin, at the tested concentrations, and emerged as potential molecules for further development.  相似文献

Some new 1,2,3-triazolo[4,5-e]-1,2,4-triazolo[3, 4 -c]pyrimidmes were prepared starting from the corresponding 1,2,3-triazolo[4,5-d]pyrimidines via the formation of the 1,2,4-triazole ring. Thus suitable hydrazino derivatives 6 were condensed with triethyl orthoformate, triethyl orthoacetate and triethyl orthobenzoate to give the expected tricyclic derivatives 7 , 8 and 9 . Intramolecular cyclization of the ethoxycarbonylhydrazino derivatives 10 gave the tricyclic compounds 11 bearing an hydroxyl group in the 3 position. The v-triazolo-s-triazolopyrimidine derivatives were tested towards the A1 and A2A adenosine receptors in binding assays, but they did not show any receptor affinity.  相似文献

Ethyl 2‐arylhydrazono‐3‐butyrates 2 reacted with 2‐cyano‐N‐(4‐methylphenyl) acetamide 1a and 2‐cyano‐N‐(thiazol‐2‐yl)acetamide 1b to give the pyridinedione, 4 and pyridazine 5 derivatives in 3:1 ratio. The products 4 and 5 have been transformed into different phthalazine, pyrimido[4,5‐c]‐pyridazine, pyrido[3,4‐c]pyridazine and 1,6‐naphthyridine derivatives.
