生物降解聚合物聚三亚甲基环碳酸酯(PTMC)及聚2,2-二甲基三亚甲基环碳酸酯(PDTC)在药物控释载体及其它生物医学技术领域有着良好的应用前景。与脂肪族聚酯不同,PTMC、PDTC降解时,不会产生有害的酸性化合物。PTMC、PDTC主要由三亚甲基环碳酸酯(TMC)及2,2-二甲基三亚甲基环碳酸酯(DTC)开环均聚合制备。本文总结了催化TMC、DTC开环均聚合的不同催化剂及其聚合机理,综述了近年来国内外在TMC、DTC均聚合催化剂开发上的研究进展,并对生物相容性催化剂如稀土催化剂、Ca、Mg、Zn、Fe催化剂以及酶催化剂催化TMC、DTC开环聚合的优缺点进行了比较。  相似文献

研究水了化硅酸三钙(Ca3SiO5,C3S)固化体的体外生物活性行为。结果表明,干C3S固化体中含有23.97wt%的Ca(OH)2,Ca(OH)2的溶解导致模拟体液(Simulated Body Fluids,SBF)的pH值上升;磷灰石颗粒优先诱导沉积于C3S固化体表面,随后碳酸钙和磷灰石颗粒共同沉积于C3S固化体表面;HCO3-是SBF模拟体液内主要的缓冲离子;Ca(OH)2碳化和CaCO3沉积主要导致SBF的pH值下降。由于离子交换作用的减弱,C3S固化体表面最终被磷灰石完全覆盖,磷灰石的沉积促使SBF的pH值进一步降低。因此,对于C3S和其衍生材料的体外生物活性和生物相容性必须考虑到HCO3-在体内的实际含量和行为。  相似文献

Using zirconium(IV) acetylacetonate as an initiator of lactide/trimethylene carbonate copolymerization allowed us to obtain high‐molecular‐weight copolymers with high efficiency. The reactivity ratios of the comonomers were 13.0 for lactide and 0.53 for trimethylene carbonate. Despite the large differences between the values of the reactivity ratios, copolymers with randomized chain structures were obtained. This phenomenon occurred as a result of an intensive intermolecular transesterification process proceeding along with the reaction of copolymer chain growth and modifying its final structure. Conducting the copolymerization at the relatively low temperature of about 110 °C, which minimized the influence of intermolecular transesterification, made it possible to obtain semicrystalline copolymers with multiblock structures. Increasing the temperature of copolymerization up to 180 °C was associated with strong intensification of the transesterification reactions. At this temperature, amorphous copolymers were obtained with identical compositions but highly randomized chain structures. An analysis of the chain microstructures of the obtained copolymers, determining the average length of the blocks, the intermolecular transesterification ratio, and the degree of chain randomization, was conducted by means of NMR spectroscopy. For this purpose, very specific signal assignment in the carbonyl and methylene carbon regions of the 13C NMR spectra to appropriate comonomer sequences of polymeric chains was performed. © 2006 Wiley Periodicals, Inc. Poly(trimethylene carbonate) (PTMC) is a well‐known biodegradable polymer with good biocompatible properties which make it suitable for biomedical applications. Poly(5‐[2‐{2‐(2‐methoxyethoxy)ethyoxy}‐ethoxymethyl]‐5‐methyl‐1,3‐dioxa‐2‐one) (PTMC‐MOE3OM) and copolymers, bearing oligo ethylene glycol (OEG) at the side chain of PTMC backbone, were selected to investigate the cloud point behavior by solvents such as PBS, water, 10% ethanol solution and various ionic strengths. A pH‐responsive copolymer, poly(TMCM‐MOE3OM‐co‐(5‐methyl‐5‐carboxylic‐1,3‐dioxane‐2‐one)) as carboxylic acid carbonate showed a decreased critical temperature at pH 2. Photo‐responsive copolymer, poly(TMCM‐MOE3OM‐co‐coumarin derivatives) bearing 1% and 10% of photo‐induced molecules (7‐[(5‐(5‐methyl‐1,3‐dioxa‐2‐one)methoxy)]‐methoxy coumarin (TMCM‐coumarin)) exhibited a low cloud point because of the hydrophobic moieties. Meanwhile, alternative coumarin polymer including 2% of 4‐methyl‐7‐[(5‐(5‐methyl‐1,3‐dioxa‐2‐one)methoxy)butoxy)]‐methoxy coumarin (TMCM‐4‐methyl‐coumarin) has been successfully synthesized and copolymerized as a novel molecule. The various combinations of monomers were studied and the significant properties were determined via external triggers after copolymerization. This study showed basically synthetic progress toward designs and trivial rationalization of thermoresponsive copolymers close to body temperature. At present, various pendant groups as side part affect to the lower critical solution temperature (LCST) and biodegradable polymer in order to utilize the actual external stimuli application. © 2017 Wiley Periodicals, Inc. The in vitro degradation characteristics of poly(anhydride-imides) containing pyromellitylimidoalanine, with either 1,6-bis(carboxyphenoxy)hexane (CPH) or sebacic acid (SA) were assessed. The copolymers contained up to 50 mol % of the imide monomer, pyromellitylimidoalanine (PMA-ala). Degradation was pH sensitive, being enhanced under basic conditions. Control of degradation times from 2 days to 2 months was achieved by the selection of appropriate monomer units in the polymer backbone. Monomers were chosen based on their solubility in aqueous media, as well as how they influenced the hydrophobicity and crystallinity of the polymer matrices. Polymer degradation was followed by ultraviolet spectroscopy and high-pressure liquid chromatography. Increasing the amount of imide monomer, PMA-ala, and the use of SA (rather than CPH) as the comonomer increased the degradation rate of the polymer matrices. © 1996 John Wiley & Sons, Inc.  相似文献

研究了37℃下吗啉二酮衍生物与L-丙交酯的共聚物在磷酸盐缓冲液(PBS)中的降解,并且与聚(L-丙交酯与乙交酯)(PLGA)的降解进行了比较。通过静滴接触角测量、扫描电镜(SEM)、X衍射(XRD)、凝胶色谱仪(GPC)、核磁共振(1H-NMR)和红外光谱(FT-IR)等方法研究了材料的亲水性、表面形貌、结晶结构、失重率、分子量和结构的变化等。结果表明:与PLGA相比,吗啉二酮衍生物与L-丙交酯的共聚物具有更好的亲水性,接触角达到了74°,降解3个月后数均分子量下降80%以上,该共聚物能够实现两组份的同步降解。  相似文献

The direct enzymatic synthesis of a cyclic trimethylene carbonate (1,3‐dioxane‐2‐one) monomer with/without a methyl substituent was carried out using dimethyl or diethyl carbonate and 1,3‐diol with the objective of producing aliphatic poly(trimethylene carbonate), a typical biodegradable synthetic plastic. The lipase‐catalyzed condensation of dimethyl or diethyl carbonate with aliphatic 1,3‐diols using immobilized Candida antarctica lipase (lipase CA) in an organic solvent at 70 °C afforded the corresponding methyl‐substituted and unsubstituted cyclic trimethylene carbonates. The cyclic trimethylene carbonates obtained by the reaction of dimethyl or diethyl carbonates with 1,3‐propanediol and 2‐methyl‐1,3‐propanediol were polymerized by lipase to produce the corresponding polycarbonates.

In this paper, ring-opening polymerization of trimethylene carbonate (TMC) with rare earth (Nd, Y, La) p-tert-butylcalix[n]arene (n=4, 6, and 8) complexes as catalysts has been studied. Poly(trimethylene carbonate) (PTMC) with Mv of 21,400 was produced by bulk polymerization under the conditions as follows: [TMC]0/[Nd] (molar ratio)=1000,80℃,8h. Mechanism study reveals that the polymerization proceeds via a coordination mechanism.  相似文献

低热-高压法制备PLGA多孔支架及其体外降解研究   总被引:6,自引:1,他引:6  
采用低热-高压法制备了聚(dl-丙交酯/乙交酯)75/25(PLGA75/25)组织工程多孔支架。该方法避免了使用有机溶剂,支架的孔隙率在90%以上,孔径大小分布均匀。多孔支架经过酒精处理后,支架表面产生许多微小的凹陷;用藻酸钙改性处理后,支架形态保持良好。两种处理都使支架的压缩强度有所增大,亲水性增强。虽然孔隙率高的支架降解速率稍慢,但其体外降解规律基本一致:特性粘数争力学强度衰减快,而质量损失较慢,降解6周后,支架的质量损失仅为3%左右;体外降解3周后,支架的形态保持良好,可望在细胞移植争组织修复的早期发挥支撑作用。  相似文献

High‐molecular‐weight (co)polymers of trimethylene carbonate and D ,L ‐lactide are efficiently crosslinked using PETA during gamma irradiation. Form‐stable networks with gel contents of 86 ± 5 to 96 ± 1 are obtained from non‐crystalline (co)polymers. Glass transition temperatures and elastic moduli of the networks can be varied by adjusting the copolymer composition. The PETA‐containing (co)polymer networks are not cytotoxic. Upon incubation in macrophage cultures for 14 d, all (co)polymer films and PETA‐containing network films erode to varying extents, showing that these materials can be degraded by cell‐mediated erosion processes. This method is very useful for the facile preparation of TMC‐ and DLLA‐containing form‐stable networks from high‐molecular‐weight polymers.

The ring‐opening polymerization of trimethylene carbonate (TMC) using homoleptic lanthanide guanidinate complexes [RNC(NR′2)NR]3Ln as single component initiators has been fully investigated for the first time. The substituents on guanidinate ligands and center metals show great effect on the catalytic activities of these complexes, that is, ? N(CH2)5 > ? NiPr2 > ? NPh2 (for R′), ? Cy > ? iPr (for R), and Yb > Sm > Nd. Among them, [Ph2NC(NCy)2]3Yb shows the highest catalytic activity. Some features and kinetic behaviors of the TMC polymerization initiated by [Ph2NC(NCy)2]3Yb were studied in detail. The polymerization rate is first order, with the monomer concentration and Mn of the polymer increasing with the polymer yield increasing linearly. The results indicated the present system having “living character.” A mechanism that the polymerization occurs via acyl‐oxygen bond cleavage rather than alkyl‐oxygen bond cleavage was proposed. Abstract

In this work was evaluated the activity of samarium acetate (III) (Sm(OAc)3) as a possible initiator in the polymerization by ring opening of trimethylene carbonate (TMC). All polymerizations were carried out under solvent-free melt conditions in ampoules-like flasks, equipped with a magnetic stirrer. The effects of different parameters of reaction, such as molar ratio monomer to initiator, temperature and reaction time, on typical variables of polymers, e.g., conversion of TMC to poly(trimethylene carbonate) (PTMC), dispersity and molar mass, were analyzed. The molar ratio of monomer to initiator was varied between 0 and 1000?mol/mol and the temperature among 70 and 150?°C. Nuclear Magnetic Resonance (1H-NMR and HMBC) and Size Exclusion Chromatography (SEC) were used to characterize the polymers. The results indicate that the Sm(OAc)3 induces the polymerization of TMC to high conversion with number-average molecular weights of 3.11?×?103 to 38.40?×?103?Da. Based on the 1H-NMR end-group analysis of low-molecular-weight PTMC, it was proposed a coordination–insertion mechanism for the polymerization, with a breakdown of the acyl-oxygen bond of the TMC. In according to the kinetic study carried out, the polymerization rate is first-order with respect to monomer concentration with apparent rate constants of kap?=?7.02?×?10?4?mol?×?L?1?×?h?1.  相似文献

低左旋度聚丙交酯的分子结构及体外降解行为研究   总被引:1,自引:0,他引:1  
以辛酸亚锡为催化剂,135℃下低左旋度的l-丙交酯([a]_(Na)~(25)=—231°)开环聚合制备了低左旋度聚(l-丙交酯)[l-PLLA]([a]_(CHCl_3)~(25)=-119.7°),并用同核去偶~1H-NMR谱表征其分子链结构。用成纤模压法制备了该聚合物的棒材(φ=3.2mm)试样,研究了其在37℃的模拟体液(SBF)中的降解行为。结果表明,l-PLLA具有良好的力学性能,其初始弯曲强度(σ_b)、剪切强度(σ_s)以及弯曲模量(E_b)虽比聚(l-丙交酯)(PLLA)低,但均比聚(dl-丙交酯)(PDLLA)高得多,且材料表现出很好的韧性,呈非晶态,其分子量下降速率和失重速率都介于PLLA和PDLLA之间,可望是一种良好的骨折内固定材料。  相似文献

合成了2个新的双(三有机锡)2,3-吡啶二甲酸酯2,3-C5H3N(CO2SnR3)2 (R=c-C6H111;C6H5C(CH3)2CH22),利用元素分析、IR和1H NMR表征了其结构。通过X-射线单晶衍射测定了配合物2的晶体结构。该化合物晶体属单斜晶系,空间群P21/na=0.955 3(1),b=1.811 8(2),c=3.533 9(4) nm,β=94.914(2)°,Z=4,V=6.094 1(13) nm3R=0.038 4,wR=0.096 6,S=1.015。2,3-吡啶二甲酸的2个羧基键连2个三有机锡,2个锡原子均为畸变的四面体配位构型。生物活性测试结果表明,化合物1和2对3种人癌细胞HeLa、CoLo205和MCF-7具有好的体外抗癌活性。  相似文献

Tacca leontopetaloides (T. leontopetaloides) contain a number of active compounds such as flavonoids, tannins, phenolics, steroids, and alkaloids. The active compounds from plants have been shown to reduce the risk of cardiovascular disease by lowering cholesterol levels by inhibiting the enzyme 3-hydroxy-3-methylglutaryl-coenzym A (HMG-CoA) reductase activity. This study aims to investigate the potential active compounds in the ethanolic extract of Tacca tubers (T. leontopetaloides) from the Banyak Islands, Aceh Singkil Regency, Aceh Province both in vitro and in silico. Tacca tubers contain secondary metabolites including flavonoids, phenolics, tannins, steroids and saponins, according to phytochemical screening. In vitro investigation of ethanolic extract of Tacca tuber revealed inhibitory activity of HMG Co-A reductase with an IC50 value of 4.92 ppm. Based on the in silico study, active compound from the extract, namely Stigmasterol with the highest binding affinities with HMG Co-A reductase (−7.2 kcal/mol). As a comparison, the inhibition of HMG Co-A reductase activity by simvastatin with an IC50 4.62 ppm and binding affinity −8.0 Kcal/mol. Our findings suggest that the ethanolic extract of Tacca tuber (T. leontopetaloides) from Banyak Islands, Aceh Province has the potential to inhibit the activity of HMG Co-A reductase.  相似文献
