Summary Interaction of ethyl 2-acetamido-5-bromothiazole-4-carboxylate (2) with 2-methyl-5-chlorothiophenol (3) afforded the thioether4, which was hydrolysed to the corresponding carboxylic acid5. Attempted cyclization of5 to6 yielded the decarboxylated product7. On the other hand, interaction of 2-acetamido-5-bromothiazole (9) with thiosalicylic acid (10) yielded the thioether11, which was cyclized to compound12. Acid hydrolysis of12 yielded the amino derivative13, which was reacted with certain selected alkyl halides using sodium hydride to afford compounds14–18.

Zusammenfassung Die Reaktion von Ethyl 2-Acetamido-5-bromthiazol-4-carboxylat (2) mit 2-Methyl-5-chlorthiophenol (3) ergab den Thioether4, der zur entsprechenden Carbonsäure5 hydrolysiert wurde. Die versuchte Cyclisierung von5 zu6 ergab das Decarboxylierungsprodukt7. Andererseits ergab die Reaktion von 2-Acetamido-5-bromthiazol (9) mit Thiosalizylsäure (10) den Thioether11, der zu Verbindung12 cyclisiert werden konnte. Saure Hydrolyse von12 ergab das Aminoderivat13, das mit geeigneten Alkylhalogeniden unter Verwendung von Natriumhydrid zu den Verbindungen14–18 führte.

Base-catalyzed intramolecular nucleophilic substitution for the 2-nitro group in 2-hydroxyanilides of 2-nitrobenzoic acids gave dibenzo[b,f][1,4]oxazepin-11(10H)-ones. In particular, 3-nitrodibenzo[b, f][1,4]oxazepin-11(10H)-one was obtained from N-(2-hydroxyphenyl)-2,4-dinitrobenzamide. The nitro group in the product could also be replaced under the action of O- and S-nucleophiles. Published in Russian in Izvestiya Akademii Nauk. A convenient synthesis of a series of pyrido[3,2-e][1,4]-diazepine-2,5-diones 8 and pyrido[2,3-e][1,4]diazepine-2,5-diones 9, is reported using the condensation of α-amino acid methyl ester derivatives with 1H-pyrido[3,2-d][1,3]oxazine-2,4-dione and 1H-pyrido[2,3-d][1,3]oxazine-2,4-dione. Compounds 8 and 9 were also synthesized by peptide coupling of α-amino acid methyl ester derivatives with β-amino acids (2 or 3) followed by the cyclisation in tetrahydrofuran with sodium hydride (NaH).  相似文献

Summary A new series of thiophenic isosters of thioxanthones, namely: 2-substituted-4H-thieno[2,3-b][1]benzothiopyran-4-ones and 5-substituted-2-nitro-8-methyl-4H-thieno[2,3-b][1]benzothiopyran-4-ones were synthesized as potential schistosomicidal agents. The synthesized compounds were characterized by their1H-NMR data.

Die synthetisierten Verbindungen wurden mittels ihrer1H-NMR Daten charakterisiert.

Aminomethylation of 2,6-diamino-4-aryl-4H-thiopyran-3,5-dicarbonitriles in the Mannich reaction with p-toluidine and formaldehyde unexpectedly gave pyrimido[4,3-b][1,3,5]thiadiazine derivatives. The same compounds were obtained in an independent way involving a multicomponent cascade condensation of aromatic aldehydes, cyanothioacetamide, p-toluidine, and formaldehyde. For Part 5, see Izv. Summary 2-Aminothiophenol (1) reacts with 3-chloro-2,4-pentanedione (2) in the presence of pyridine to form 2-acetyl-3-methyl-4H-1,4-benzothiazine (3) in high yields. Reaction of3 with hydrazine gives 4-(2-aminophenylthio)-3,5-dimethylpyrazole (5). Condensation of3 with 4-nitrobenzaldehyde yields the corresponding Schiff base7. Hydroxylamine with benzothiazine3 affords 3,9a-dimethyl-3a, 9a-dihydro-9H-isoxazolo[4,5-b][1,4]benzothiazine (8).

Zusammenfassung 2-Aminothiophenol (1) reagiert mit 3-Chlor-2,4-pentandion (2) in Anwesenheit von Pyridin unter Bildung von 2-Acetyl-3-methyl-4H-1,4-benzothiazin (3) in sehr hoher Ausbeute. Benzothiazin3 kondensiert mit Hydrazin zu 4-(2-Aminophenylthio)-3,5-dimethylpyrazol (5), dessen Aminogruppe reagiert mit 4-Nitrobenzaldehyd zu einer Schiffschen Base (7), die spektroskopisch charakterisiert wurde. Benzothiazin3 mit Hydroxylamin ergibt 3,9a-Dimethyl-3a,9a-dihydro-9H-isoxazolo[4,5-b][1,4]benzothiazin (8). Die Stereochemie der letztgenannten Verbindung wurde ermittelt.

Summary The syntheses of 2-acetyl-, 2-benzyl-, and 2-ethyl-thieno-diltiazem derivatives are described starting from the corresponding 5-substituted 3-nitro-2-thiophenthiolvia reaction with racemic methyltrans-3-(4-methoxyphenyl)-glycidate under different conditions (solvent, catalyst, temperature) to obtain purethreo orerythro products. The nitro groups of these products were reduced and the resulting amino esters cyclized. The thieno[2,3-b][1,4]thiazepin-5(4H)-ones were N-alkylated and acetylated in position 3. The desired 2-substituted 4-(2-dimethylaminoethyl)-4,5,6,7-tetrahydro-7-(4-methoxyphenyl)-5-oxothieno[2,3-b][1,4]thiazepin-6-yl acetates were isolated in good yields.

The products of the reaction of 2-aryl-5-R-5,6-dihydro-7H-[1,2,4]triazolo[5,1-b][1,3]thiazin-7-ones with aryl bromomethyl ketones are 2-aryl-7H-[1,2,4]triazolo[5,1-b][1,3]thiazin-7-ones and aryl methyl ketones or 2,5-diaryl[1,3]thiazolo[3,2-b][1,2,4]triazoles and 3-phenyl-2-propenoic acid, depending on the structure of R. A new synthesis of the indole system has been achieved by chclodehydrogenation of amino phenylmalonate heterocycles. Thus, the 4-amino coumarins1 a,b or the 4-amino-2-quinolones1 c–g are converted to the indoles2 with palladium on charcoal in boiling diphenyl ether. The reaction of the aminocompounds1 with diphenyl carbonate yields the fused polycyclic isoquinolones4.

The interaction has been studied of a series of substituted 3-hetaryl-7-hydroxychromones with amino acids and formaldehyde (reactants ratio 1: 1: 2 respectively). In the case of glycine and Het = 3-isoxazolyl the product of aminomethylation at position 8 of the chromone was obtained, and with other Het (including Het = 4-phenyl-1,2,4-triazol-3-yl) 2-[6-alkyl-3-hetaryl-4-oxo-9,10-dihydro-4H,8H-chromeno[8,7-e][1,3]-oxazin-9-yl]acetic acids were formed. Summary A new approach to the synthesis of 2H-isoindole-4,7-diones is described. Heating -amino acids with carbonyl compounds generates azomethine ylides through the elimination of water and carbon dioxide. The ylides were captured by quinones forming 2H-isoindole-4,7-diones, 2,3,3a,7a-tetrahydro-1H-isoindole-4,7-diones and 2,3-dihydro-1H-pyrrolo[2,1-a]isoindole-6,9-diones. The structures were established on the basis of spectroscopy (NMR, mass).

12H-Benzo[5,6][1,2,4]triazepino[3,4-a]isoindol-5(6H)-one was synthesized by the condensation of anthranilic acid hydrazide with o-phthalaldehyde. The structure of this compound was established by the X-ray diffraction study of its isopropyl derivative. Summary Several substituted crowned phenoselenazin-16-one derivatives were prepared by condensation of substituted zinc 2-amino-benzeneselenolates with 1,4-quinone of benzo[15]crown-5 or its dibromide in alcoholic solution. The dehalogenation of the bromoselenazinones was also investigated.

A reaction of 10-R-substituted tetrahydrobenzo[b][1,6]naphthyridines (R = CN, CONH2, Me) with dimethyl acetylenedicarboxylate, methyl and ethyl propiolates, and acetylacetylene has been studied. It was found that 1-acryloyl-10-cyanotetrahydrobenzo[b][1,6]naphthyridines are the major products of the reaction of alkyl propiolates with 10-cyano-substituted naphthyridines, whereas the reaction with 10-carbamoyl-substituted naphthyridines gives a mixture of 1-acryloyl-substituted naphthyridines and hexahydrobenzo[b]pyrido[3,4,5-d,e][1,6]naphthyridines. The latter are the only products in the reaction of 10-carbamoyl-substituted naphthyridines with acetylacetylene. Published in Russian in Izvestiya Akademii Nauk. Summary Reaction of 2-benzylidene-1-benzocyclanones1 with dithiocarbamic acid afforded open-chain addition products2A–4B. Dehydration of the adducts yielded tricyclic 1,3-thiazine-2-thiones5 and6. Treatment of1 with thiourea under acid conditions gave tricyclic 2-amino-1,3-thiazines7–9. IR and1H NMR spectroscopic investigations showed7–9 to exist predominantly in the amino tautomeric form both in the solid state and in solution.
Die Umsetzung von 2-Benzyliden-benzocyclanonen mit Dithiocarbaminsäure und Thioharnstoff
Zusammenfassung Die Reaktionen der 2-Benzyliden-1-benzocyclanone1 mit Dithiocarbaminsäure liefern kettenförmige Additionsprodukte2A–4B. Die Dehydratisierung der Additionsprodukte führt zu tricyklischen 1,3-Thiazin-2-thionen5 und6. Durch Behandlung von1 mit Thioharnstoff unter sauren Bedingungen wurden tricyclische 2-Amino-1,3-Thiazine7–9 gebildet. IR- und1H-NMR-spektroskopische Untersuchungen zeigten, dass7–9 sowohl in Substanz als auch in Lösungen überwiegend in der amino-tautomeren Form existieren.
