A New Furostanol Saponin from Dioscorea futshauensis   总被引:1,自引:0,他引:1  
A new furostanol saponin presenting moderate bioacitivity of inducting morphological deformation of Pyricularia oryzae mycelia was isolated from Dioscorea futshauensis R.Kunth by bioacitivity-guided fractionation. The structure was established as 26-O-β-D-glucopyranosyl-3β,26-diol-23(S)-methoxyl-(25R)-furost-5,20(22)-diene-3-O-[α-L-rhamnopyranosyl(1→2)-β-D-glucopyranosyl(1→3)]-β-D-glucopyranoside on the basis of chemical evidencesand spectral analysis,especially by 2D-NMR techniques.  相似文献

A New Steroidal Saponin from Dioscorea deltoidea Wall var. orbiculata   总被引:1,自引:0,他引:1  
Bioactivity-guided fractionation led to the isolation of a new steroidal saponin, orbiculatoside B, together with a pair of furostanol saponins, protobioside and methyl protobioside, from the fresh rhizomes of Dioscorea deltoidea Wall var. orbiculate. The new compound was identified to be isonarthogenin 3-O-α-L-thamnopyranosyl(1→2)-[α-L-rhamnopyranosyl-(1→4)]-β-D-glucopyranoside by various NMR techniques in combination with chemical methods. The three saponins showed strong activity against Pyricularia oryzae,and were cytotoxic to cancer cell line K562,HCT-15,A549, and A2780a in vitro.  相似文献

通过正相和反相硅胶柱色以及高效液相色谱,从红葱60%乙醇提取物中分离得到3个新化合物,利用物理化学和光谱学方法鉴定了其结构,分别为9,9'-二羟基-8,8'-二甲氧基-1,1'-二甲基-1H,1H-['4,4']双[萘并(2,3-c)呋喃]-3,3'-二酮(1),6,8-二羟基-3,4-二甲氧基-1-甲基-蒽醌-2-羧酸甲酯(2),2-乙酰基-3,6,8-三羟基-1-甲基蒽醌(3).这3个化合物均能不同程度地诱导稻瘟霉菌丝变形,其最小变形质量浓度(Mini-mummorphologicaldeformationmassconcentration,MMDC)分别为145.8,50.2和124.8μg/mL.利用MTT法测定了化合物抑制人类红白血细胞株K562细胞生长的活性,化合物2的IC50为49.1μg/mL.  相似文献

Gross saponins of Tribulus terrestris(GSTT) have exact effect on cardiovascular and cerebrovascular diseases.But as a mixture,the specific efficient component of GSTT is still unknown.Nine monomers of spirostanol saponins were isolated and idendified as JA―JI(named transitorily) by means of NMR spectrometry.After bio-activity screening on them,we defined that monomers tigogenin 3-O-β-D-xylopyranosyl(1→2)-[β-D-xylopyranosyl(1→3)]-β-D-glucopyranosyl(1→4)-[α-L-rhamnopyranosyl(1→2)]-β-D-galactopyranoside(compound JB) and hecogenin3-O-β-D-glucopyranosyl(1→4)-β-D-galactopyranoside(compound JG) have cytoprotective bio-activity.Compound JB display effective dose in 10-8 and 10-9 mol/L,and JG in 10-6―10-9 mol/L.Survival rate,lactate dehydrogenase(LDH) and apoptosis also show that JB(at dose 10-8,10-9 and 10-10 mol/L) can protect myocardial injury caused by hypoxia/reoxygenation(H/R).While morphology change also shows JG has cytoprotective bio-activity.  相似文献

26-O-β-D-Glucopyranosyl-furost-5(6),20(22)-dien-3β,26-diol 1, a new furostanol saponin, was isolated from the water-extract of Dioscorea nipponica Mak., the raw material of a traditional Chinese herbal medicine Wei Ao Xin. The structure of 1 was determined on the basis of its spectral data especially by NMR spectroscopy. Using high-resolution quadrupole time-of-flight mass spectrometry along with an electrospray ionization source (ESI-QqTOF-MS), accurate molecular weights of 13 steroid saponins extracted from the rhizomes of Dioscorea panthaica were acquired and the corresponding molecular formulae obtained. In order to elucidate the fragmentation pathways of steroid saponins in D. panthaica, 10 authentic samples were investigated using ESI-QqTOF-MS/MS. In addition, atmospheric pressure chemical ionization mass spectrometry combined with ion trap tandem mass spectrometry (APCI-IT-MS/MS) was used to analyze the structures of 13 steroid saponins in D. panthaica. Through the analysis of their tandem mass data, diagnostic fragment ions of the spirostanol and furostanol steroid saponins in D. panthaica were detected as m/z 271.2056 and 253.1951. In addition, four pairs of isomers were detected and the possible structures of four unknown steroid saponins in D. panthaica speculated. ESI-TOF and APCI-MS(n) have proved to be effective tools for research on fragmentation mechanism of steroid saponins and the rapid determination of native steroid saponins in extract mixture, thereby avoiding tedious derivation and separation steps.  相似文献

Ultra high‐performance liquid chromatography (UHPLC) with evaporative light scattering detection was used for the quantification of steroidal saponins and diosgenin from the rhizomes or tubers of various Dioscorea species and dietary supplements that were purported to contain Dioscorea. The analysis was performed on an Acquity UPLC? system with an UPLC? BEH Shield RP18 column using a gradient elution with water and acetonitrile. Owing to their low UV absorption, the steroidal saponins were observed by evaporative light scattering detection. The 12 compounds could be separated within 15 min using the developed UHPLC method with detection limits of 5–12 µg/mL with 2 μL injection volume. The analytical method was validated for linearity, repeatability, accuracy, limits of detection and limits of quantification. The relative standard deviations for intra‐ and inter‐day experiments were <3.1%, and the recovery efficiency was 97–101%. The total content of standard compounds was found to be in the ranges 0.01–14.5% and 0.9–28.6 mg daily intake for dry plant materials and solid commercial preparations, respectively. UHPLC–mass spectrometry with a quadrupole mass analyzer and ESI source was used only for confirmation of the identity of the various saponins. Two new spirostanol saponins, (1β,3β,5β,25S)‐spirostan‐1,3‐diol 1‐(β‐D ‐xylopyranoside) ( 1 ) and (1β,3β,5β,25S)‐spirostan‐1,3‐diol 1‐[α‐L ‐rhamnopyranosyl‐(1→2)‐β‐D ‐fucopyranoside] ( 2 ), along with two known compounds, (1β,3β,5β,25S)‐spirostan‐1,3‐diol 1‐[α‐L ‐rhamnopyranosyl‐(1→2)‐β‐D ‐xylopyranoside] ( 3 ) and (1β,3β,4β,5β,25S)‐spirostan‐1,3,4,5‐tetrol 5‐(β‐D ‐glucopyranoside) ( 4 ) were isolated from the whole plant of Reineckia carnea. The structures of the new steroids were determined by detailed analysis of their 1D‐ and 2D‐NMR spectra and chemical methods, and by comparison with spectral data of known compounds. A new β-resorcylic maerolide, 5'-hydroxyzearalenol (1), was isolated from the culture broth of a marine-derived fungus Fusarium sp. 05ABR26. Three known compounds, zearalenone (2), 8'-hydroxyzearalenone (3) and zearalenol (4) were also isolated. The structure and relative stereochemistry of 1 were elucidated on the basis of spectroscopic data and single-crystal X-ray diffraction data. Compound 2 displayed potent inhibitory activity against Pyricularia oryzae with a MIC value of 6.25 μg/mL, while compound 3 was much less active; however, 1 and 4 showed no obvious activity.  相似文献

Two new spirostanol sapogenins (5β-spirost-25(27)-en-1β,2β,3β,5β-tetrol 3 and its 25,27-dihydro derivative, (25S)-spirostan-1β,2β,3β,5β-tetrol 4) and four new saponins were isolated from the roots and rhizomes of Convallaria majalis L. together with known sapogenins (isolated from Liliaceae): 5β-spirost-25(27)-en-1β,3β-diol 1, (25S)-spirostan-1β,3β-diol 2, 5β-spirost-25(27)-en-1β,3β,4β,5β-tetrol 5, (25S)-spirostan-1β,3β,4β,5β-tetrol 6, 5β-spirost-25(27)-en-1β,2β,3β,4β,5β-pentol 7 and (25S)-spirostan-1β,2β,3β,4β,5β-pentol 8. New steroidal saponins were found to be pentahydroxy 5-O-glycosides; 5β-spirost-25(27)-en-1β,2β,3β,4β,5β-pentol 5-O-β-galactopyranoside 9, 5β-spirost-25(27)-en-1β,2β,3β,4β,5β-pentol 5-O-β-arabinonoside 11, 5β-(25S)-spirostan-1β,2β,3β,4β,5β-pentol 5-O-galactoside 10 and 5β-(25S)-spirostan-1β,2β,3β,4β,5β-pentol 5-O-arabinoside 12 were isolated for the first time. The structures of those compounds were determined by NMR spectroscopy, including 2D COSY, HMBC, HSQC, NOESY, ROESY experiments, theoretical calculations of shielding constants by GIAO DFT, and mass spectrometry (FAB/LSI HR MS). An attempt was made to test biological activity, particularly as potential chemotherapeutic agents, using in silico methods. A set of 12 compounds was docked to the PDB structures of HER2 receptor and tubulin. The results indicated that diols have a higher affinity to the analyzed targets than tetrols and pentols. Two compounds (25S)-spirosten-1β,3β-diol 1 and 5β-spirost-25(27)-en-1β,2β,3β,4β,5β-pentol 5-O-galactoside 9 were selected for further evaluation of biological activity.  相似文献

Two novel 27-norergostanol steroids, panthogenins A (1) and B (2), possessing unusual rings E and F featuring a ketal moiety at C-25 were isolated from rhizomes ofDioscorea panthaica. Their structures and relative configurations were elucidated by extensive spectroscopic methods including 1D and 2D NMR techniques, and the structure of panthogenin A was further confirmed by X-ray crystallography. From the bulbils of Dioscorea bulbifera L. var. sativa, three new clerodane diterpenoids, bafoudiosbulbin C (=methyl (2β,8α,12S)‐17‐oxo‐2,19 : 8,19 : 12,17 : 15,16‐tetraepoxycleroda‐3,13(16), 14‐triene‐18‐carboxylate; 1 ), bafoudiosbulbin D (=methyl (2β,6β,12R)‐17,19‐dioxo‐2,19 : 6,17 : 8,12 : 15,16‐tetraepoxycleroda‐13(16),14‐diene‐18‐carboxylate; 2 ), and bafoudiosbulbin E (=methyl (2β,3α,4α,6β,12R)‐17,19‐dioxo‐2,19 : 3,4 : 6,17 : 8,12 : 15,16‐pentaepoxycleroda‐13(16),14‐diene‐18‐carboxylate; 3 ) were isolated, together with the known compounds bafoudiosbulbins A and B, 3‐Oβ‐D ‐glucopyranosyl‐β‐sitosterol, and 6′‐stearoyl‐3‐Oβ‐D ‐glucopyranosyl‐β‐sitosterol. Their structures were established by high‐field NMR techniques (1H,1H‐COSY, 13C‐DEPT, HSQC, HMBC, and NOESY), MS analyses, as well as by comparison of their spectral data with those of related compounds.  相似文献
