A series of 2-aryl-5-alkyl-7-methoxylbenzo[b]furan derivatives have been synthesized by utilizing the coupling of methyl 3-methoxy-4-hydroxy-5-bromocinnamate with cuprous phenylacetylide as the key step.The structures of the new compounds were confirmed by1H NMR,IR and MS.The structure of compound 14 was further confirmed by single crystal X-ray.Compound 17 showed cytotoxic activity against human lung carcinoma A549.  相似文献

An efficient two-step synthesis of p-toluenesulfonic acid salts of optically active 3-amino-3-alkyl-2-oxetanones is reported that involves cyclization of N-Boc-α-alkylserines under Mitsunobu reaction conditions followed by deprotection of the amino group with trifluoroacetic acid in the presence of anhydrous p-toluenesulfonic acid.  相似文献

Derivatives of 2-alkyl-2-mercaptopropionic acid were synthesized based on the substituted 3-cyanopyridin-2(1H)-thiones. The synthesis was carried out by the heating of a mixture of thione, alkyl methyl ketone, and chloroform in the presence of a base. The reaction proceeds readily for acetone, whereas alkyl methyl ketones require a prolonged heating in the presence of a phase-transfer catalyst. Methyl esters were prepared from the acids obtained. 吡喃酮是许多天然产物的结构单元,我们曾由4-异丁酰基庚二酸在过量醋酸酐及乙酰氯存在下回流得到7-氧代-8,8-二甲基-△~9-六氢香豆素.本文由二氰乙基-β-二酮进行酮解水解反应得到4-酰基庚二酸1_(a-c)。 在过量醋酸酐、乙酰氯存在下由1_a、1_c为底物进行反应没有得到双环的香豆素衍生物.其产物和单纯以乙酐为缩合剂时的产物2_a、2_c相同,产率分别为68%、63%。2_c可在硫酸铁催化  相似文献

Reactions between chiral 3-cinnamoyl-4-methyl-5-phenyl-1,3-oxazolidine-2-thiones and aromatic aldehydes in the presence of BF3Et2O diastereoselectively produced tricyclic compounds incorporating a bridgehead carbon bound to four heteroatoms in high yields. Four stereocenters were induced during the reaction. The tricyclic products were transformed into propane-1,3-diols bearing three consecutive stereocenters by acid hydrolysis, S-methylation, and reductive removal of the chiral auxiliary.  相似文献

A stable and storable precursor of 2-SF5-butadiene, 3-SF5-3-sulfolene, has been synthesized and its reactivity studied with several olefinic compounds. When SF5Br is added to sulfolene, 3-bromo-4-SF5-sulfolane is formed and when reacted further with silver tosylate forms 4-SF5-2-sulfolene. The 4-SF5-2-sulfolene undergoes rearrangement with silicic acid to give 3-SF5-3-sulfolene and when heated forms 2-SF5-butadiene; in the absence of a dienophile, dimerization does occur. The new 2-SF5-butadiene is a reactive diene undergoing a Diels-Alder reaction with olefinic sytems such as maleic anhydride, p-naphthoquinone, and methyl acrylate.  相似文献

The reaction of 2-thiophenecarbaldehyde with arene diazonium chlorides in the presence of CuCl2 catalyst gave 5-aryl-2-thiophenecarbaldehydes. Use of 4-nitrobenzene diazonium chloride in the reaction also gave the isomeric 3-(4-nitrophenyl)-2-thiophenecarbaldehyde. Condensation products of the 5-aryl-2-thiophenecarbaldehydes with cyanoacetic acid esters, cyanoacetamide, and with barbituric and dimethylbarbituric acids were prepared. Several 1-alkyl-2-methylazetidin-3-ones were prepared in good yield by the hydride-induced cyclization of the corresponding β-bromo-α,α-dimethoxyketimines, the resulting 3,3-dimethoxyazetidines being hydrolyzed by acid. Imination of these 1,2-disubstituted azetidin-3-ones, followed by alkylation under kinetic control conditions resulted in regioisomeric mixtures of 2,4- and 2,2-dialkylated compounds. Analytical samples of the major 2,4-disubstituted derivatives were obtained after extensive chromatographic separation. The cis stereochemistry of the major 2,4-dialkylated isomer was demonstrated on the basis of NMR data.  相似文献

Several alkyl 2',3'-didehydro-2',3'-dideoxythymidin-5'-yl H-phosphonates were synthesized and analyzed by electrospray ionization multistage tandem mass spectrometry (ESI-MS(n)). Two kinds of novel benzyl rearrangement reactions were observed in ESI - MS(2) of [M + H](+), [M + Na](+) and [M + K](+) of benzyl 2',3'-didehydro-2',3'-dideoxythymidin-5' yl H-phosphonate. Results from tandem mass spectrometry, high-resolution mass spectrometry and control experiments showed that the benzyl migration could undergo a four-membered cyclic rearrangement reaction, and benzyl was essential in the process.  相似文献

The intramolecular cyclization of -ketocarboxylic acids and their esters gives 5-alkylsubstituted 3H-thiolen-2-ones and 3H-furan-2-ones. In order to investigate the biological activity of novel bis-pyrazole compounds,a series of N-(3-alkyl-5-(N-methylcarbamyl)- 1H-pyrazol-4-yl)-3-alkyl-4-substituted-1H-pyrazole-5-carboxamides were designed and synthesized with ethyl 3-alkyl-1H-pyrazole -5-carboxylate 1 as starting materials.N-Methyl-3-alkyl-4-amino-1H-pyrazole-5-carboxamides 6 were obtained from 1 via 5 steps.3-Alkyl-4-substitued-1H-pyrazole-5-carboxyl chlorides 4a,4b,11a,11b,11c or 12 were also obtained from 1 via several steps.Target compounds 7a-7g were obtained after the reaction of 6 with the above 1H-pyrazole-5-carboxyl chlorides.Preliminary bioassay showed some compounds possessing good inactivation effect against TMV(tobacco mosaic virus).Compound 7a showed higher activity superior to ningnanmycin at a concentration of 5.0×10-4 g/mL and equal activity at 1.0×10-4 g/mL;7b and 7c showed equal activity to virazole both at concentrations of 5.0×10-4 g/mL and 1.0×10-4 g/mL.  相似文献

We describe a simple and efficient protocol for accessing some unsaturated heterocyclic compounds in a direct evaluation of allyl bromide as Z-ethyl 3-bromomethyl-4-oxopent-2-enoate. The latter reacts with primary amines via two successive nucleophilic substitutions followed by a 5-exo-trig cyclization to produce selectively 4-acetyl-1-alkyl-1H-pyrrol-2(5H)-ones in good yields.  相似文献

A new isocyanide-based multicomponent reaction between an aromatic aldehyde, malononitrile, an isocyanide, and acetic acid efficiently provides 3,3-dicyano-N-alkyl-2-arylpropanamide derivatives in excellent yields in ethanol at 70 °C. 1,5-Disubstituted 3-trifluoromethylpyrazoles were reacted with N-bromosuccinimide in DMF at room temperature or 70-80 °C for 1-2 h to afford the corresponding 4-bromo-substituted pyrazoles 2 in 95-99% yields. The microwave-assisted Stille coupling reactions of 2 with arylstannanes having a substituent on the benzene ring and allylstannane in refluxing CH3CN in the presence of Pd(PPh3)4 provided the corresponding 1,4,5-trisubstituted 3-trifluoromethylpyrazoles 3 in 75-98% yields.  相似文献

A novel one-pot synthesis of the several bicyclo[2.2.2]oct-5-en-2-ones 19 has been developed in which a cyclic or acyclic ketone 18 is reacted with a cyclic enone 6 in the presence of strong acid to give the bicyclic enone product 19. Alternatively, the intermediate diketone 21 can be prepared separately and subjected to the reaction conditions to give the bicyclic enones 19 in good yields.  相似文献
