In the present study, we found that plasmid DNA could induce single-chained cationic surfactants cetyltrimethylammonium bromide (CTAB), dodecyltrimethylammonium bromide (DTAB), and dodecyltriethyl ammonium bromide (DEAB) to form vesicles once its concentration reached a critical value. Moreover, the gene for follicle-stimulating hormone was delivered into cells with these single-chained cationic surfactant/DNA vesicles and the transfection efficiency was comparable to that with lipofectamine? 2000, a famous and widely used commercial transfection reagent, and also to that using electroporation method, although it was generally thought conventional single-chained cationic surfactant was not suitable for gene transfer. The conventional single-chained cationic surfactant is very cheap and stable and the vesicles are very easy to be prepared. Thereby, this study may suggest that the vesicles formed between plasmid DNA and surfactant should be prospective to transfer DNA.  相似文献

In the recent genomic era, a novel gene silencing approach has been introduced based on the use of small synthetic oligonucleotides, such as antisense RNAs, siRNAs, to inhibit the expression of a specific target gene. Successful implementation of this methodology calls for the development of efficient systems to deliver small oligonucleotides into the cells using various natural and synthetic cationic agents. While extensive studies have focused on the interaction of various natural and synthetic cationic surfactants with long DNA, less attention has been paid to surfactant interaction with small oligonucleotides. In this study, the interaction between 14mer double stranded DNA and alkyltrimethylammonium bromides of C16 (cetyl, CTAB), C14 (tetradecyl, TTAB), and C12 (dodecyl, DTAB) chain lengths was investigated at different charge ratios by gel electrophoresis, ethidium bromide exclusion, circular dichroism, and UV melting. Our gel studies at 1 microM oligonucleotide concentration showed that CTAB, TTAB, and DTAB neutralize the oligonucleotides at a charge ratio (Z+/-) of 1, 14, and 50, respectively. At lower charge ratios, CTAB and TTAB interact with oligonucleotides, and the complexes show electrophoretic mobility shifts in the gel, while such mobility shifts were completely absent in the case of DTAB. UV melting experiments revealed that interaction with all three surfactants increased the thermostability of the oligonucleotide. The extent of thermal stabilization was highest in the case of CTAB, moderate in the case of TTAB, and extremely low in the case of DTAB. Oligonucleotides within fully neutralized complexes denatured at further higher temperatures, and again, stabilization was the highest in the case of CTAB followed by TTAB and DTAB, hence revealing that the oligonucleotides interacted more strongly with CTAB than with the other two surfactants. Ethidium bromide exclusion studies also supported our UV melting studies, confirming that CTAB binds most strongly to the oligonucleotide. CD titrations of oligonucleotides with increasing amounts of surfactants revealed common spectral patterns consisting of the progressive loss of CD signals for native helical DNA conformations. Overall, our results demonstrate that interaction between oligonucleotides and cationic surfactants, although qualitatively similar to long double stranded DNA, shows subtle differences that need to be understood to improve small oligonucleotide delivery into the cells by using common delivery agents that have been used to deliver long pieces of DNA.  相似文献

The interaction between silver nanoparticles (Ag NPs) of different surface charge and surfactants relevant to the laundry cycle has been investigated to understand changes in speciation, both in and during transport from the washing machine. Ag NPs were synthesized to exhibit either a positive or a negative surface charge in solution conditions relevant for the laundry cycle (pH 10 and pH 7). These particles were characterized in terms of size and surface charge and compared to commercially laser ablated Ag NPs. The surfactants included anionic sodium dodecylbenzenesulfonate (LAS), cationic dodecyltrimethylammoniumchloride (DTAC) and nonionic Berol 266 (Berol). Surfactant-Ag NP interactions were studied by means of dynamic light scattering, Raman spectroscopy, zeta potential, and Quartz Crystal Microbalance. Mixed bilayers of CTAB and LAS were formed through a co-operative adsorption process on positively charged Ag NPs with pre-adsorbed CTAB, resulting in charge reversal from positive to negative zeta potentials. Adsorption of DTAC on negatively charged synthesized Ag NPs and negatively charged commercial Ag NPs resulted in bilayer formation and charge reversal. Weak interactions were observed for nonionic Berol with all Ag NPs via hydrophobic interactions, which resulted in decreased zeta potentials for Berol concentrations above its critical micelle concentration. Differences in particle size were essentially not affected by surfactant adsorption, as the surfactant layer thicknesses did not exceed more than a few nanometers. The surfactant interaction with the Ag NP surface was shown to be reversible, an observation of particular importance for hazard and environmental risk assessments.  相似文献

We have investigated dilute aqueous solutions of an anionic polymer (carboxymethylcellulose) mixed with cationic surfactants of different chain lengths (dodecyl to octadecyl trimethylammonium bromides: DTAB, TTAB, CTAB and OTAB). The structures of the concentrated phases formed above the precipitation threshold were studied by X-ray diffraction. Different body-centred cubic structures with space groups Pm3n were observed in the presence of surfactant with a short aliphatic chain (DTAB), despite the fact that the polymer persistence length is comparable to the repeat distance of the structure (5 nm). For larger surfactant chain lengths (TTAB and CTAB), the structure of the precipitates can be either cubic (Pm3n) or 2D hexagonal depending on the initial surfactant and polymer concentrations. For still larger chain length (OTAB), the structure becomes lamellar. This structural evolution from micellar cubic towards 2D hexagonal and lamellar is attributed to the decrease of the local curvature of the surfactant aggregates, as observed for flexible synthetic polymers and short DNA fragments under similar conditions. Furthermore, the structure of the bulk complexes formed just below the precipitation threshold anticipates the structure seen in the precipitated phases.  相似文献

The compaction of long duplex DNA by cationic nanoparticles (NP) used as a primary model of histone core particles has been investigated. We have systematically studied the effect of salt concentration, particle size, and particle charge by means of single-molecule observations-fluorescence microscopy (FM) and transmission electron microscopy (TEM)-and molecular dynamics (MD) simulations. We have found that the large-scale DNA compaction is progressive and proceeds through the formation of beads-on-a-string structures of various morphologies. The DNA adsorbed amount per particle depends weakly on NP concentration but increases significantly with an increase in particle size and is optimal at an intermediate salt concentration. Three different complexation mechanisms have been identified depending on the correlation between DNA and NPs in terms of geometry, chain rigidity, and electrostatic interactions: free DNA adsorption onto NP surface, DNA wrapping around NP, and NP collection on DNA chain.  相似文献

The interaction between cationic surfactants (hexadecyl and dodecyl trimethyl ammonium bromide) and gelatin was characterized by measuring the circular dichroism. The interaction between the cationic surfactants and gelatin is weak in comparison to that of anionic surfactants. When the concentration of cationic surfactants is sufficiently low, refolding of the gelatin-strands to the triple helical structure by rechilling the solution from 298 K to 283 K is complete. The triple helical content of the solution is affected more strongly by the cationic surfactants in acidic solution than at pHs 7 or 10. The interaction depends on the apolar group of the surfactant and is found to be stronger for DTAB than for CTAB at 298 K. Coagulation of the hydrophobic gelatin-cationic surfactant complexes does not comprise that pan of gelatin which is able to refold the triple helical structure. Therefore, the gelatin-strands of lower molecular weights are thought to react favorably with the surfactant ions.  相似文献

The interactions between oppositely charged surfactant-polymer systems have been studied using surface tension and conductivity measurements and the dependence of aggregation phenomenon over the polyelectrolyte concentration and chain length of cationic ATAB surfactants, cetyltrimethyl ammonium bromide (CTAB), tetradecyltrimethyl ammonium bromide (TTAB), and dodecyltrimethyl ammonium bromide (DTAB) have been investigated. It was observed that cationic surfactants induce cooperative binding with anionic polyelectrolyte at critical aggregation concentration (cac). The cac values of ATAB surfactants in the presence of anionic polyelectrolyte, sodium carboxy methyl cellulose (NaCMC), are considerably lower than their critical micelle concentration (cmc). After the complete complexation, free micelles are formed at the apparent critical micelle concentration (acmc), which is slightly higher in polyelectrolyte aqueous solution than in pure water. Among the cationic surfactants (i.e., CTAB, TTAB, and DTAB), DTAB was found to have least interaction with NaCMC. Surfactants with longer tail size strongly favor the interaction, indicating the dependence of aggregation phenomenon on the structure, morphology, and tail length of the surfactant.   相似文献

Ionic surfactants are known to aggregate around the surface of a nanoparticle as a single layer in premicellar and a double layer in micellar concentrations. This motif of arrangement indicates the development of a layer of confined water of lower polarity than bulk water around the surface of the nanoparticle. We have demonstrated the behavior of a twisted intramolecular charge transfer (TICT) probe, trans-2-[4-(dimethylamino)styryl]benzothiazole (DMASBT), in the confined aqueous layer developed at the surface of spherical silver nanoparticles (Ag NPs) at and above the critical micellar concentrations (CMC) of a cationic and an anionic surfactant, namely, cetyltrimethylammonium bromide (CTAB) and sodium dodecyl sulfate (SDS). It is observed that the presence of charged surfactant head groups affects DMASBT differentially in the ground and the excited states. In presence of CTAB, DMASBT turns over in the excited state and interacts with the Ag NP surface, whereas in SDS the probe remains in its original orientation during the interaction. Steady-state and time-resolved fluorescence spectral studies provide enough evidence for orientation of the TICT probe in the peripheral water of Ag NP created by the surfactants. The results were confirmed by steady-state anisotropy measurements. The data show the difference between the properties of the confined peripheral water and the bulk aqueous environment. The TICT probe, DMASBT, is proved to be an excellent marker for the phenomenon.  相似文献

The addition of cationic surfactants to an aqueous solution of an anionic polymer, carboxymethylcellulose (carboxyMC), causes the spontaneous formation of aggregates in a certain range of concentrations. Here we studied two surfactants, dodecyl and hexadecyl trimethylammonium bromide (DTAB and CTAB, respectively). Using different techniques (light scattering, potentiometry, viscosimetry, and zetametry), we found that a simple lengthening of the surfactant tail length by four CH2 groups drastically changes the aggregate morphology, size, and charge. We explored in detail how the surfactant and polymer concentrations act on these systems.  相似文献

The alkaline hydrolysis of curcumin was studied in three types of micelles composed of the cationic surfactants cetyl trimethylammonium bromide (CTAB) and dodecyl trimethylammonium bromide (DTAB) and the anionic surfactant sodium dodecyl sulfate (SDS). At pH 13, curcumin undergoes rapid degradation by alkaline hydrolysis in the SDS micellar solution. In contrast, alkaline hydrolysis of curcumin is greatly suppressed in the presence of either CTAB or DTAB micelles, with a yield of suppression close to 90%. The results from fluorescence spectroscopic studies reveal that while curcumin remains encapsulated in CTAB and DTAB micelles at pH 13, curcumin is dissociated from the SDS micelles to the aqueous phase at this pH. The absence of encapsulation and stabilization in the SDS micellar solution results in rapid hydrolysis of curcumin.  相似文献

Recent investigations of the DNA interactions with cationic surfactants and catanionic mixtures are reviewed. Several techniques have been used such as fluorescence microscopy, dynamic light scattering, electron microscopy, and Monte Carlo simulations.

The conformational behaviour of large DNA molecules in the presence of cationic surfactant was followed by fluorescence microscopy and also by dynamic light scattering. These techniques were in good agreement and it was possible to observe a discrete transition from extended coils to collapsed globules and their coexistence for intermediate amphiphile concentrations. The dependence on the surfactant alkyl chain was also monitored by fluorescence microscopy and, as expected, lower concentrations of the more hydrophobic surfactant were required to induce DNA compaction, although an excess of positive charges was still required.

Monte Carlo simulations on the compaction of a medium size polyanion with shorter polycations were performed. The polyanion chain suffers a sudden collapse as a function of the concentration of condensing agent, and of the number of charges on the polycation molecules. Further increase in the concentration increases the degree of compaction. The compaction was found to be associated with the polycations promoting bridging between different sites of the polyanion. When the total charge of the polycations was lower than that of the polyanion, a significant translational motion of the compacting agent along the polyanion was observed, producing only a small-degree of intrachain segregation, which can explain the excess of positive charges necessary to compact DNA.

Dissociation of the DNA–cationic surfactant complexes and a concomitant release of DNA was achieved by addition of anionic surfactants. The unfolding of DNA molecules, previously compacted with cationic surfactant, was shown to be strongly dependent on the anionic surfactant chain length; lower amounts of a longer chain surfactant were needed to release DNA into solution. On the other hand, no dependence on the hydrophobicity of the compacting agent was observed. The structures of the aggregates formed by the two surfactants, after the interaction with DNA, were imaged by cryogenic transmission electron microscopy. It is possible to predict the structure of the aggregates formed by the surfactants, like vesicles, from the phase behaviour of the mixed surfactant systems.

Studies on the interactions between DNA and catanionic mixtures were also performed. It was observed that DNA does not interact with negatively charged vesicles, even though they carry positive amphiphiles; however, in the presence of positively charged vesicles, DNA molecules compact and adsorb on their surface.

Finally Monte Carlo simulations were performed on the adsorption of a polyelectrolyte on catanionic surfaces. It was observed that the mobile charges in the surface react to the presence of the polyelectrolyte enabling a strong degree of adsorption even though the membrane was globally neutral. Our observations indicate that the adsorption behaviour of the polyelectrolyte is influenced by the response given by the membrane to its presence and that the number of adsorbed beads increases drastically with the increase of flexibility of the polymer. Calculations involving polymers with three different intrinsic stiffnesses showed that the variation is non-monotonic. It was observed also that a smaller polyanion typically adsorbs more completely than the larger one, which indicates that the polarisation of the membrane becomes less facilitated as the degree of disruption increases.  相似文献

The interactions between double-stranded DNA (dsDNA) and three different kinds of surfactants, i.e., cationic, anionic, and nonionic surfactants, were investigated by cyclic voltammetry, electrochemical impedance spectroscopy and UV-vis spectroscopy. Multilayer films composed of DNA and surfactants were prepared at gold electrode by electrostatic or hydrophobic interactions. It was found that the cationic surfactant, CTAB, can bind to DNA by electrostatic interaction, and the electron transfer resistance of CTAB-DNA complex film increases first and then decreases with CTAB concentration. The anionic surfactant, LAS, can bind to DNA but by hydrophobic interaction, and the electron transfer resistance of the complex film keeps decreasing with LAS concentration. Nonionic surfactants can also directly bind to DNA by hydrophobic interaction. All the three different kinds of surfactants can form multilayer films with DNA on the electrode surface. The chemical structure of DNA keeps unchanged during interacting with these surfactants. The binding modes of DNA with these three different kinds of surfactants were also deduced.  相似文献

The size-dependent interaction of anionic silica nanoparticles with ionic (anionic and cationic) and nonionic surfactants has been studied using small-angle neutron scattering (SANS). The surfactants used are anionic sodium dodecyl sulfate (SDS), cationic dodecyltrimethyl ammonium bromide (DTAB), and nonionic decaoxyethylene n-dodecylether (C(12)E(10)). The measurements have been carried out for three different sizes of silica nanoparticles (8, 16, and 26 nm) at fixed concentrations (1 wt % each) of nanoparticles and surfactants. It is found that irrespective of the size of the nanoparticles there is no significant interaction evolved between like-charged nanoparticles and the SDS micelles leading to any structural changes. However, the strong attraction of oppositely charged DTAB micelles with silica nanoparticles results in the aggregation of nanoparticles. The number of micelles mediating the nanoparticle aggregation increases with the size of the nanoparticle. The aggregates are characterized by fractal structure where the fractal dimension is found to be constant (D ≈ 2.3) independent of the size of the nanoparticles and consistent with diffusion-limited-aggregation-type fractal morphology in these systems. In the case of nonionic surfactant C(12)E(10), micelles interact with the individual silica nanoparticles. The number of adsorbed micelles per nanoparticle increases drastically whereas the percentage of adsorbed micelles on nanoparticles decreases with the increase in the size of the nanoparticles.  相似文献

The dependence of the electrochemiluminescence of Ru(bpy)3(2+) (bpy = 2,2'-bipyridine) with tri-n-propylamine as co-reactant on the anionic surfactant SDS (sodium dodecyl sulfate) and the cationic surfactants CTAX (CTA = cetyltrimethylammonium cation, X = bromide, chloride and hydrogensulfate) was studied. Both SDS and CTAX, at low surfactant concentrations below the critical micelle concentrations, enhance the electrochemiluminescence at a platinum working electrode. A further enhancement of the light emission intensity by bromide ions was observed when CTAB (B = bromide) was used-an overall 30-fold increase in electrochemiluminescence efficiency was obtained at a CTAB concentration of 0.08 mM. Voltammetric data support adsorption of surfactant molecules on the electrode surface as the cause of the enhancement of electrochemiluminescence by ionic surfactants.  相似文献

通过UV-Vis吸收光谱、同步荧光光谱、圆二色(CD)光谱等方法对阴离子型表面活性剂——琥珀酸二辛酯磺酸钠(AOT)和十二烷基苯磺酸钠(SDBS)、阳离子型表面活性剂——十六烷基三甲基溴化铵(CTAB)和十二烷基三甲基溴化铵(DTAB)、两性离子型表面活性剂——3-[(3-胆固醇氨丙基)二甲基氨基]-1-丙磺酸(CHAPS)与马心高铁肌红蛋白(metMb)的不同作用机理进行了探讨.结果显示:阴、阳离子型表面活性剂可以与蛋白发生较强烈的作用,且相互作用与表面活性剂的浓度密切相关.AOT和SDBS浓度的升高使得metMb的Soret带发生红移且出现两个新的Q带,伴随着配体金属电荷转移(LMCT)带的消失,蛋白从水合的六配位高自旋复合物(6-cHs)转化成六配位低自旋高铁血红素复合物(6-cLs),低浓度的AOT和SDBS对Tyr和Trp微环境均有影响,能使metMb的二级结构发生变化;而CTAB和DTAB在低浓度时对metMb的血红素中心影响不大,但是对Trp和Tyr的微环境影响很大,高浓度时主要通过静电吸引作用以聚合体形式直接作用于血红素中心,使Soret带发生蓝移,metMb形成五配位高自旋(5-cHs)复合物,血红素从疏水腔中释放出来,metMb的α螺旋含量减少.DTAB由于自身结构的特点,与CTAB作用于蛋白的过程有些区别,形成了一个中间态,但最终也导致血红素的暴露.两性离子型表面活性剂在测定浓度范围内不与metMb发生作用,原因是CHAPS整体呈电中性,其与metMb的阴离子性或者阳离子性位点作用的能力很弱,同时也说明metMb表面带相反电荷的位点相距较远.结果充分证明表面活性剂与蛋白相互作用的方式与表面活性剂的种类、结构及其浓度有关.  相似文献

The polymerization of acrylamide in mixed micellar solutions of surfactants, initiated by NaHSO3 has been studied at 20 and 3Q° C with time variable method of thermokinetics for 1. 5-order reaction. The results indicate that the mixed micellar systems of cationic or anionic with zwitterionic surfactants (SLS/ CTAB, SLS/ TTAB, SLS/ SDS) and cationic with nonionic surfactants (Brij 357sol; CTAB, Bri-J35/TTAB, Brij35/ DTAB) have catalytic effect on the polymerization in the order, at 20° C. SLS/ SDS SLS/ TTAB SLS/ CTAB Brij35/ CTAB at 30° C SLS/ SDS SLS/ TTAB≈ / CTAB Bri-j35/ DTAB= sBrij35/ TTAB as Brij35/ CTAB, while Brij35/ SDS mixed micellar system has inhibition. These effects are attributed to the effect of the Stern layer of mixed micelles on the step of initiator (HSOT) to form free radical.  相似文献

In this article, we discuss the structure and composition of mixed DNA-cationic surfactant adsorption layers on both hydrophobic and hydrophilic solid surfaces. We have focused on the effects of the bulk concentrations, the surfactant chain length, and the type of solid surface on the interfacial layer structure (the location, coverage, and conformation of the DNA and surfactant molecules). Neutron reflectometry is the technique of choice for revealing the surface layer structure by means of selective deuteration. We start by studying the interfacial complexation of DNA with dodecyltrimethylammonium bromide (DTAB) and hexadecyltrimethylammonium bromide (CTAB) on hydrophobic surfaces, where we show that DNA molecules are located on top of a self-assembled surfactant monolayer, with the thickness of the DNA layer and the surfactant-DNA ratio determined by the surface coverage of the underlying cationic layer. The surface coverages of surfactant and DNA are determined by the bulk concentration of the surfactant relative to its critical micelle concentration (cmc). The structure of the interfacial layer is not affected by the choice of cationic surfactant studied. However, to obtain similar interfacial structures, a higher concentration in relation to its cmc is required for the more soluble DTAB surfactant with a shorter alkyl chain than for CTAB. Our results suggest that the DNA molecules will spontaneously form a relatively dense, thin layer on top of a surfactant monolayer (hydrophobic surface) or a layer of admicelles (hydrophilic surface) as long as the surface concentration of surfactant is great enough to ensure a high interfacial charge density. These findings have implications for bioanalytical and nanotechnology applications, which require the deposition of DNA layers with well-controlled structure and composition.  相似文献

Self-assembly in mixtures of two single-chain cationic surfactants, with different tail lengths (CTAB and DTAB) as well as of a single-chain (DTAB) and a double-chain (DDAB) cationic surfactant, with identical tail lengths, have been investigated with small-angle neutron scattering (SANS) and rationalised in terms of bending elasticity properties. The growth behaviour of micelles with respect to surfactant composition appears completely different in the two surfactant mixtures. DTAB form small oblate spheroidal micelles in presence of [NaBr] = 0.1 M that transform into prolate spheroidal mixed CTAB/DTAB micelles upon adding moderate amounts of CTAB, so as to give a mole fraction y = 0.20 in solution. Most unexpectedly, upon further addition of CTAB the mixed CTAB/DTAB micelles grow with an almost equal rate in both length and width directions to form tablets. In contrast to this behaviour, mixed DDAB/DTAB micelles grow virtually exclusively in the length direction, in presence of [NaBr] = 0.1 M, to form elongated ellipsoidal (tablet-shaped) and subsequently long wormlike micelles as the fraction of DDAB in the micelles increases. Mixed DDAB/DTAB micelles grow to become as long as 2000 Å before an abrupt transition to large bilayer structures occurs. This means that the micelles are much longer at the micelle-to-bilayer transition as compared to the same mixture in absence of added salt. It is found that the point of transition from micelles to bilayers is significantly shifted towards higher fractions of aggregated DTAB as an appreciable amount of salt is added to DDAB/DTAB mixtures, indicating a considerable reduction of the spontaneous curvature with an increasing [NaBr]. By means of deducing the various bending elasticity constants from our experimental results, according to a novel approach by ours, we are able to conclude that the different growth behaviours appear as a consequence of a considerably lower bending rigidity, as well as higher saddle-splay constant, for DDAB/DTAB surfactant mixtures in presence of [NaBr] = 0.1 M, as compared to mixtures of CTAB/DTAB in [NaBr] = 0.1 M and DDAB/DTAB in absence of added salt.  相似文献

The interactions between oppositely charged surfactant/polymer mixtures have been studied using conductivity and turbidity measurements. The dependence of aggregation phenomenon on the chain length and head group modifications of conventional cationic surfactants, i.e., hexadecyl- (HTAB), tetradecyl- (TTAB), and dodecyltrimethylammonium bromides (DTAB) and dimeric cationic surfactants, i.e., decyl- (DeDGB) and dodecyldimethylgemini bromides (DDGB), is investigated. It was observed that cationic surfactants induce cooperative binding with anionic polyelectrolytes at critical aggregation concentration (cac). The cac values are considerably lower than the critical micelle concentration (cmc) values for the same surfactant. After the complete complexation, free micelles are formed at the apparent critical micelle concentration (acmc), which is slightly higher in aqueous polyelectrolyte than in pure water. Among the conventional and dimeric cationic surfactants, DTAB and DeDGB, respectively, have been found to have least interactions with oppositely charged polyelectrolytes.  相似文献
