采用超高速液相色谱-质谱(UFLC-MS)研究了吴茱萸醇提取物中入血小分子化合物的体内药代动力学过程. 同时对UFLC-MS 生物样品分析方法进行包括特异性、 线性、 精密度、 准确度、 稳定性、 基质效应和回收率等考察, 结果表明, 该方法稳定可靠, 且醇提取物中10个生物碱类化合物均被胃肠道快速吸收, 并且多数化合物血药浓度在1~2 h左右达到峰值. 将吸收入血的10个生物碱类化合物与乙酰胆碱酯酶和丁酰胆碱脂酶进行柔性分子对接及构效关系分析, 发现其中活性最高的为去氢吴茱萸碱、 吴茱萸碱、 吴茱萸次碱和吴茱萸酰胺Ⅰ 4个吲哚型生物碱, 它们与乙酰胆碱酯酶的对接打分均在-46.02 kJ/mol以下; 与丁酰胆碱脂酶对接打分均在-41.84 kJ/mol以下. 吴茱萸碱、 吴茱萸次碱、 去氢吴茱萸碱和吴茱萸酰胺可能是以乙酰胆碱酯酶和丁酰胆碱脂酶为靶点的胆碱酯酶抑制剂前体化合物.  相似文献

A novel, sensitive and reliable ultra fast liquid chromatography‐tandem mass spectrometry (UFLC‐MS/MS) method has been developed and validated for simultaneous quantitation of eight main active ingredients (evodiamine, rutaecarpine, dehydroevodiamine, limonin, ginsenoside Rb1, Rd, Re and Rg1) in rat plasma after oral administration of Wu‐Zhu‐Yu (WZY) decoction, which is a celebrated and widely used Traditional Chinese Medicine formula for the treatment of headache. The analytes and internal standard (IS) were separated on a SHIM‐PACK XR‐ODS II column, and the detection was performed on a UFLC‐MS/MS system with turbo ion spray source. The lower limits of quantification were 1.5, 0.5, 1.0, 2.0, 2.0, 1.0, 0.5 and 0.2 ng ml?1 for evodiamine, rutaecarpine, dehydroevodiamine, limonin, gensenoside Rb1, Rd, Re and Rg1, respectively. Linearity, accuracy, precision and absolute recoveries of the eight analytes were all within satisfaction. The IS‐normalized matrix factor was adopted for assessing the matrix effect and accompanied with a satisfactory result. The validated method has been successfully applied to compare pharmacokinetic profiles of the eight active ingredients in rat plasma between normal and headache rats after administration. Exact pharmaceutical effect of WZY decoction on headache was demonstrated by the ethological response of headache rats induced by nitric oxide donor after administration. The results indicated that the absorption of evodiamine, rutaecarpine, gensenoside Rb1, Re and Rg1 in headache group were significantly higher than those in normal group with similar concentration–time curves while no significant differences existed in limonin and ginsenoside Rd between the two groups. As increasing numbers of people worldwide are using traditional Chinese medicines, their characterization has become increasingly critical. This study describes the development of a facile and reproducible high-performance liquid chromatography-based method to obtain the fingerprint of the traditional Chinese medicine Yuhuanglian. First, similarity analysis and Principal Component Analysis (PCA) were investigated to interpret differences in the chromatograms of samples from different groups. Seven fingerprint peaks were identified by electrospray ionization tandem mass spectrometry (ESI-MS/MS), including magnolforine, coptisine, berberin, epiberberin, dehydroevodiamine, 12α-hydroxylimonin, and 12α-hydroxyevodol. The established fingerprint method was then applied to the analysis of Evodiae Fructus, Coptis Rhizoma, Yuhuanglian, and analogs of Yuhuanglian, which reflected their chemical constituent properties. The proposed method provides a technical platform for the characterization of Yuhuanglian, which will ensure the safe and effective use of this traditional Chinese medicine.  相似文献

The aim of this study was to evaluate the chromatographic performance of a PEG stationary phase, in comparison with those of C18 columns, for the HPLC analysis of phenethylamine ((+/-)-synephrine) and indoloquinazoline (rutaecarpine and evodiamine) alkaloids in methanolic extracts of fruits of Evodia rutaecarpa (Juss.) Benth. and E. rutaecarpa (Juss.) Benth. var. officinalis (Dode) Huang (i.e., E. officinalis Dode) (Rutaceae family). The method was validated and showed good linearity, precision, accuracy, sensitivity, and specificity. The highest content of both phenethylamine and indoloquinazoline alkaloids was found in methanolic fruit extracts of E. rutaecarpa, and it was closely related to the degree of maturity. E. officinalis fruits displayed low amounts of both types of alkaloids. Furthermore, an enantioselective HPLC method for the enantioseparation of (+/-)-synephrine from Evodia fruits was applied, by using a protein-based chiral stationary phase with cellobiohydrolase (CBH) as the chiral selector (Chiral-CBH). Isolation of synephrine from Evodia aqueous fruit extracts was carried out by strong cation-exchange SPE. The results of the application of the method to the analysis of Evodia samples showed that (-)-synephrine was the main component while (+)-synephrine was present in low concentration.  相似文献

建立了超临界流体色谱快速分析吴茱萸中吲哚类生物碱的方法。以标准品混合物和复杂样品为对象比较4种色谱柱的分离效果,进行色谱柱的筛选;考察了进样体积、改性剂、添加剂、温度和背压对保留行为的影响。结果表明,进样体积对峰形影响显著;添加剂对保留时间和色谱峰形影响有限;改变改性剂能使保留时间显著改变;降低温度,升高背压,保留时间减小。经过优化,确定采用Waters ACQUITY UPC2 BEH色谱柱,以甲醇为改性剂,在35 ℃柱温和2.07×107 Pa背压条件下,15 min内完成复杂样品的分析。同时采用超高效液相色谱完成复杂样品的快速分析。结果表明,超临界流体色谱可用于天然产物的高效快速分析,同时该方法与超高效液相色谱在分离选择上的差异有助于天然产物分析方法的拓展。  相似文献

Evodiamine and rutecarpine are two kinds of indole alkaloids contained in the fruit of Evodiae fructus, which have been shown to exhibit various bioactivities in humans. A liquid chromatography–tandem mass spectrometric method (LC–MS/MS) was developed for the determination of evodiamine and rutecarpine in human serum. The serum was extracted by solid-phase extraction (SPE) and analyzed using a C18 column and a mobile phase consisting of methanol–water (85:15) solution containing 5 mmol/L ammonium formate at a flow rate of 0.5 mL/min. The mass spectrometer was operated in positive mode, employing the extracted ion chromatogram (EIC) for detection and quantitation of evodiamine (m/z 288) and rutecarpine (m/z 304). Good linear relationships between the peak area and the concentration were obtained in the ranges of 5.2–1040 ng/mL and 10.2–1020 ng/mL, with correlation coefficients (r) of 0.999 and 0.998, for evodiamine and rutecarpine, respectively. The repeatabilities (RSD, n=6) of quantitation for evodiamine and rutecarpine were 2.18–4.00% and 2.99–5.67%, respectively, and the recovery ranged from 90.5% to 98.1%. A comparative study of the different ionization and quantitation modes, including ESI–MS, ESI–MS/MS, APCI–MS and APCI–MS/MS, was also accomplished. The MS/MS fragmentation mechanism of the base peak ([M+H]+, m/z 304) of evodiamine was investigated in order to identify the analytes in more complicated body fluid samples.   相似文献

建立了高效液相色谱-电喷雾串联质谱同时检测尿液和胃液中12种有毒生物碱的方法.优化了提取条件及色谱-质谱条件,并考察了基质效应的影响,探讨了质谱碎裂机理.尿液和预先调节至中性的胃液经硼砂-NaOH缓冲液(pH 9.6)碱化,乙酸乙酯液液萃取,采用电喷雾电离(ESI+)、多反应监测(MRM)方式,可同时对黄华碱、倒千里光碱、山莨菪碱、钩吻碱、芦竹碱、哈尔碱、吐根碱、血根碱、吴茱萸碱、吴茱萸次碱、雷公藤吉碱和雷公藤次碱12种有毒生物碱进行定性和定量分析.在优化的条件下,12种成分分别在0.5~200 μg/L、1~200 μg/L和5~200μg/L范围内线性关系良好,尿液中除黄华碱和山莨菪碱外,各生物碱的回收率为61.9%~119.1%,胃液中各生物碱回收率为61,0%~1102%,精密度RSD< 15%.检出限(LOD)为0.1~0 Sμμg/L,定量限(LOQ)为0.5~5.0 μg/L,时回收率不高的生物碱可通过空白基质配制标样校正,满足定量分析的要求.本方法操作简便、快捷、灵敏度高,适用于中毒患者尿液和胃液中有毒生物碱成分的检测.  相似文献

Ginkgo biloba (ginkgo) is one of most frequently used botanical dietary supplements. The bioactive constituents include the terpenoid lactones consisting of bilobalide and the ginkgolides A, B, C and J. A new assay based on high-performance liquid chromatography/electrospray tandem mass spectrometry (LC/MS/MS) was developed for the measurement of the terpenoid lactones in ginkgo products such as leaf powder and extracts. Initially, the MS/MS fragmentation pathways of ginkgolides were investigated to identify abundant fragment ions that might be useful for the sensitive and selective detection of ginkgolides and bilobalide during LC/MS/MS. Then, sample preparation and clean-up procedures were streamlined to maximize throughput by taking advantage of the selectivity of LC/MS/MS detection. Analyte recoveries exceeded 90%, the intra-assay and inter-assay relative standard deviations were <5%, the relative error was <8% and the limits of detection and quantification were 3.6-120 and 11-350 fmol, depending on the analyte that was injected on to the LC column. Therefore, this LC/MS/MS assay facilitated the rapid quantitative analysis of ginkgolides A, B, C and J and bilobalide in ginkgo dietary supplements with excellent recovery, reproducibity, accuracy and sensitivity.  相似文献

Liquid chromatography coupled with electrospray ionization quadrupole time-of-flight tandem mass spectrometry (LC/ESI-QTOF-MS/MS) was performed to study the fragmentation behaviors of steroidal alkaloids from Fritillaria species, the antitussive and expectorant herbs widely used in traditional Chinese medicine. We propose, herein, a strategy that combining key diagnostic fragment ions and the relative abundances and amounts of major fragment ions (the ions exceeding 10% in abundance) to distinguish different sub-classes of Fritillaria alkaloids (FAs). It was found that hydrogen rearrangement and induction effects result in ring cleavage of the basic skeletons occurred in the MS/MS process and produced characteristic fragment ions, which are useful for structural elucidation. This method was finally used to investigate the primary steroidal alkaloids in the extracts of eight major Fritillaria species. As a result, 41 steroidal alkaloids (29 cevanine type, 1 jervine type, 6 veratramine type and 5 secosolanidine type alkaloids) were selectively identified in these Fritillaria species. Twenty-six compounds were unambiguously identified by comparing with the reference compounds and 15 compounds were tentatively identified or deduced according to their MS/MS data. Logical fragmentation pathways for different types of FAs have been proposed and are useful for the identification of these types of steroidal alkaloids in natural products especially when there are no reference compounds available.  相似文献

High-speed counter-current chromatography (HSCCC) with a two-phase solvent system composed of n-hexane-ethyl acetate-methanol-water system (5:5:7:5, v/v) was applied to the isolation and purification of alkaloids from the Chinese medicinal plant Evodia rutaecarpa (Juss.) Benth. Five kinds of alkaloids were obtained and yielded 28 mg of evodiamine (I), 19 mg of rutaecarpine (II), 21 mg of evocarpine (III), 16mg of 1-methy-2-[(6Z,9Z)]-6,9-pentadecadienyl-4-(1H)-quinolone (IV), 12 mg of 1-methyl-2-dodecyl-4-(1H)-quinolone (V) from 180 mg of crude extract in a one-step separation, with the purity of 98.7%, 98.4%, 96.9%, 98.0%, 97.2%, respectively, as determined by high performance liquid chromatography (HPLC). The structures of these compounds were identified by 1H NMR and 13CNMR.  相似文献

A reversed phase liquid chromatographic (LC) method coupled with DAD (250 nm) has been developed and validated for simultaneous quantification of six alkaloids, dehydroevodiamine (1), wuzhuyuamide-I (2), 5-hydroxyrutaecarpine (3), 14-formyldihydrorutaecarpine (4), evodiamine (5) and rutaecarpine (6), in 12 batches evodiae fructus [the dried, unripe fruits of Evodia rutaecarpa (Juss.) Benth. or E. rutaecarpa (Juss.) Benth. var. officinalis (Dode) Huang, E. rutaecarpa (Juss.) Benth. var. bodinieri (Dode) Huang] as a traditional Chinese medicine. The method was carried out by a C18 column (250 × 4.6 mm) with a gradient mobile phase of methanol, acetonitrile, and phosphoric acid–triethylamine–buffer solution. The contents of 1–6 in the evodiae fructus could easily be determined within 70 min. The experimental results were satisfactory for the intra-day and inter-day precision and accuracy of the method for simultaneous determination. The linear calibration ranges of 1–6 were 40–1,000, 20–500, 1–100, 10–500, 40–1,000 and 80–1,000 μg mL−1. The recoveries of 16 were 97.43–103.73% with RSDs from 0.21 to 1.99%. The limits of detection for 16 were 2.0, 2.0, 0.1, 1.0, 5.0 and 5.0 μg mL−1, and the limits of quantification were 6.6, 6.6, 0.3, 3.3, 16.5 and 16.5 μg mL−1. The method was successfully applied to the quantification of six alkaloids in the evodiae fructus.  相似文献

An integrated method combining supercritical fluid extraction (SFE) with liquid chromatography/atmospheric pressure chemical ionization tandem mass spectrometry (LC/APCI-MS/MS) was developed and successfully applied to quantify aflatoxins (AFs) in Zizyphi Fructus (fruits of Zizyphus jujube), a traditional Chinese medicine. To minimize the potential interferences caused by the complex matrix in Zizyphi Fructus, a SFE pretreatment was performed. In addition, electrospray ionization (ESI) and atmospheric pressure chemical ionization (APCI) spectra were also compared. The results showed that the calibration curves of AFB(1), AFB(2), AFG(1), and AFG(2) were all linear over the range of concentration from 1 to 50 ng/g, the squared correlation coefficients (r(2)) were over 0.995, and the detection limits of the method were between 0.17 and 0.32 ng/g. It showed high recovery and good precision in quantitating AFs in Zizyphi Fructus without further clean-up. Further, fragmentation pathways of protonated AFs in APCI-MS/MS were clearly proposed which could predict the existence of AFB or AFG series. To test the empirical validity of the proposed methodology in this paper, eight random samples of Zizyphi Fructus collected from supermarkets and traditional Chinese medicine stores in different geographical areas of Taiwan were analyzed. The results indicated that low levels of AFs were detected in only one of them.  相似文献

In order to develop new drugs from natural products, constituents of natural medicines were examined for their effectiveness in the KCN-induced anoxia model in mice. Methanol extract from a Chinese medicine, evodia (fruits of Evodia rutaecarpa Benth. or E. officinalis Dode), had a significant effect in the KCN-induced anoxia model in mice and therefore the active constituents were further examined. The results indicated that the antianoxic action of evodia was found in the fraction containing evodiamine. Further analysis of the active constituent indicated that evodiamine and rutaecarpine, indole-alkaloids found in large amounts in the Chinese medicine evodia, were mainly responsible for the antianoxic action.  相似文献

This article reviews research on the qualitative and quantitative analysis by high-performance column liquid chromatography/mass spectrometry (LC/MS) and LC/tandem mass spectrometry (LC/MS/MS) of botanical drugs, drug substances or preparations, and finished botanical products. In addition, LC/MS and LC/MS/MS techniques and commercial instruments are described and compared briefly, and prospects for future use of these methods for the analysis of botanicals are suggested. A liquid chromatographic/tandem mass spectrometric (LC/MS/MS) multiresidue method for the simultaneous quantitative determination of oxytetracycline, 4-epi-oxytetracycline, tetracycline, 4-epi-tetracycline, chlortetracycline, 4-epi-chlortetracycline and doxycycline in milk has been developed. An extraction procedure consisting of a liquid extraction of the milk samples with trichloroacetic acid was performed. The extract was centrifuged and the supernatant was filtered. Solid-phase extraction (SPE) with an OASIS HLB SPE column was used to clean up the sample extracts. The samples were analysed by LC/MS/MS. The LC separation was performed on a reversed-phase C18 column using gradient elution with a mobile phase consisting of water and a mixture of methanol/acetonitrile. The tetracycline analytes were detected with a quadrupole mass spectrometer using positive ion electrospray ionisation. The confirmatory method has acceptable detection limits and the different tetracyclines can be detected at a residue concentration between 5 and 20 microg/L. The method is validated according to the European requirements for veterinary drug residues and all determined parameters were found to conform to the criteria. The recovery values ranged from 90.4 to 101.2% with relative standard deviations (RSDs) no larger than 9.7%. The overall or between-day precision of the analytical assay determined as repeatability at several residue concentrations and expressed as RSD ranged from 3.3 to 10%. This analytical assay is a useful tool within the Belgian monitoring programme for confirmation of samples which have been positively screened for residues of tetracyclines in raw farm cow's milk.  相似文献

A simple and rapid qualitative liquid chromatography/tandem mass spectrometry (LC/MS/MS) method was developed and validated for screening cyclo-dopa and diketopiperazine alkaloids in crude extracts of Portulaca oleracea L. at sub-ppm levels. An electrospray ionization orbitrap mass spectrometer, which provides accurate full scan MS and MS/MS data, was used in this study. After simple extraction with ethanol and purification by AB-8 resin, the extracts were subjected to LC/MS/MS analysis. A high mass tolerance (10 ppm) was used in the initial screening to filter the full scan MS data. The cyclo-dopa and diketopiperazine alkaloid standards gave limits of detection (LODs) at or below 5 ng/mL. The results also indicated that the method had an acceptable precision for day-to-day use in the identification of compounds. The alkaloids could be identified based on their MS/MS data, elemental compositions, and retention behavior. This system was used to assay trace amounts of cyclo-dopa and diketopiperazine alkaloids in crude extracts of Portulaca oleracea L., leading to the identification of 5/2 confirmed/unconfirmed cyclo-dopa and 7/6 confirmed/unconfirmed diketopiperazine alkaloids, respectively. The screening method considerably reduces the time and cost involved in the identification of cyclo-dopa and diketopiperazine alkaloids in Portulaca oleracea L., as well as being a simple and convenient approach to the identification of other structural families of natural products.  相似文献

An offline analytical method combining cardiac muscle/cell membrane chromatography (CM/CMC) with liquid chromatography/mass spectrometry (LC/MS) to identify bioactive components from traditional Chinese medicines (TCMs) such as Schisandrae Chinensis Fructus (SCF) and Schisandrae Sphenantherae Fructus (SSF) was developed. The stationary phase of the CM/CMC system employed rat cardiac muscle cell membranes. Fractions retained by the CM/CMC column were collected and analyzed by LC/MS under optimum offline conditions. After evaluating the suitability and reliability of the CM/CMC-offline-LC/MS method using nifedipine and nitrendipine as standards, this method was applied for screening and identification of bioactive components from the extracts of SCF and SSF. The main compounds in both species retained by CM/CMC were deoxyschizandrin (DSD) and schisantherin A (STA) as identified by LC/MS. In vivo pharmacological experiments suggested that DSD and STA could significantly decrease the myocardial-infarct size in rats injured by myocardial ischemia–reperfusion. Our CM/CMC-offline-LC/MS method could be used as an effective alternative for screening multiple receptor-binding bioactive components in TCMs such as FSC and FSS used to remedy cardiac diseases and may be useful for drug discovery using medicinal herbs as lead compounds.  相似文献

By optimizing the extraction, separation and analytical conditions, a reliable and accurate high-performance liquid chromatography (HPLC) method coupled with photodiode array detector (DAD) was developed for simultaneous quantitative determination of six Aconitum alkaloids, i.e., aconitine, mesaconitine, hypaconitine, benzoylaconine, benzoylmesaconine and benzoylhypaconine, in Chinese medicinal herbs, aconite roots, and 12 proprietary Chinese medicines containing processed aconite roots. The separation of these Aconitum alkaloids was achieved on an ODS column with gradient elution using solvents of acetonitrile and ammonium bicarbonate buffer (pH 10.0+/-0.2). Intra-assay and inter-assay precision of the analytes were less than 2.97%, and the average recovery rates obtained were in the range of 90-103% for all with RSDs below 3.28%. Good linear relationships were showed with correlation coefficients for the analytes exceeded 0.999. Quantitative analysis of the six Aconitum alkaloids in the unprocessed and processed aconite roots and in twelve proprietary Chinese medicines containing processed aconite roots showed that the contents of the alkaloids varied significantly. This method and quantitation results can provide a scientific and technical platform to the products manufacturers for setting up a quality control standard as well as to the public for quality and safety assurance of the proprietary Chinese medicines and other herbal preparations containing aconite roots.  相似文献

A dual reactant/catalyst role of glyoxylic acid in the reaction of isatoic anhydride with various amines afforded a novel, robust and rapid synthesis of 3-(un)substituted quinazolin-4(3H)-ones. This metal catalyst-free reaction proceeds via an unusual and unexpected cleavage of C–C bond. A shorter and common route to two alkaloids, that is, rutaecarpine and evodiamine is also accomplished.  相似文献
