N-Benzyl-D-glucamine dithiocarbamate (BGD), N-p-hydroxymethylbenzyl-D-glucamine dithiocarbamate (HBGD), and N-p-carboxybenzyl-D-glucamine dithiocarbamate (CBGD) were compared for their relative efficacies in the distribution and excretion of cadmium in mice exposed to cadmium. Mice were injected intraperitoneally with 109CdCl2 (1 mg of Cd/kg and 2 microCi of 109Cd/one animal). Three days later, they were injected with chelating agents (400 mumol/kg) every other day for 2 weeks. After injections of BGD and HBGD, cadmium was excreted mainly in the feces through the bile, and the fecal excretion of cadmium by HBGD was significantly higher than that by BGD or CBGD. These chelating agents increased the urinary excretion of cadmium to a small extent. The hepatic cadmium content was decreased only after HBGD injection. Also, the injection of HBGD caused a much greater decrease in renal cadmium content than did BGD or CBGD. These chelating agents did not result in the redistribution of cadmium to the brain, testes, or heart. The growth of mice was only slightly retarded by injections of these chelating agents. The results of this study indicate that the injection of HBGD to mice pretreated with cadmium can remove cadmium from the body, mainly through fecal excretion, without redistribution of cadmium to other tissues such as the brain, testes, and heart, more effectively than BGD or CBGD.  相似文献

The effect of N-benzyl-D-glucamine dithiocarbamate (BGD) on the renal toxicity of inorganic mercury in rats was studied. Rats were injected i.v. with saline or HgCl2 (300 micrograms Hg/kg) and 30 min later they were injected i.p. with saline or BGD (2778 mumol/kg, a quarter of an LD50). Urinary excretion of gamma-glutamyl-transpeptidase (gamma-GTP), which is a brush border enzyme, in rats after mercury treatment significantly increased compared to that of the control in the 12-24 h urine specimen and reached a maximum value within 24 h after the treatment. Urinary excretion of N-acetyl-beta-D-glucosaminidase (NAG), which is a lysosomal enzyme, also significantly increased after mercury treatment compared to that of the control in the 12-24 h urine specimen and reached a maximum value within 48 h after the treatment. A change in urinary aspartate aminotransferase (AST) activity after mercury treatment followed a pattern similar to that observed with the urinary NAG. BGD treatment did not increase the urinary excretions of gamma-GTP, NAG, and AST. The uptake of p-aminohippuric acid (PAH) by renal cortical slices significantly decreased 24 h after mercury treatment. BGD injection after mercury treatment did not decrease the uptake of PAH by cortical slices. In addition, the microscopic examination of renal tissue from mercury-treated rats revealed necrosis of the proximal tubular cells. However, a photomicrograph of rat renal cortex after BGD treatment showed little abnormality. These results indicated that the mercury-induced renal damage was protected by the injection of BGD 30 min after mercury treatment.  相似文献

In order to gain further insight into the protective action of Na2MoO4 pretreatment (1.24 mmol/kg, once a day, i.p.) against the acute toxicity of HgCl2 (30 mummol/mmol/kg, once, s.c.), changes of renal function, tissue accumulation of mercury, and urinary excretions of mercury and phenolsulfonphthalein after exposure to HgCl2 were investigated. Lactate content in the kidney and serum calcium were also measured. Na2MoO4 pretreatment enhanced urinary excretion of mercury. Renal function of Na2MoO4-pretreated rats was better maintained as compared to that of the rats given HgCl2 alone at either dose (30 or 15 mumol/kg) although the metal content in the kidney of this group was almost the same as that of the latter HgCl2-alone rats. This pretreatment prevented the rise in lactate content in the kidney and the reduction of urinary excretion of phenolsulfonphthalein caused by HgCl2, Na2MoO4 reduced serum calcium. These results suggest that Na2MoO4 prevented mercury-induced acute renal failure by decreasing tissue accumulation of the metal through urinary excretion of mercury. Better renal hemodynamics attributable to hypocalcemia may be a causative factor in the enhancement of urinary excretion of mercury.  相似文献

The protective effect of N-benzyl-D-glucamine dithiocarbamate (BGD) against gastrointestinal and bone marrow toxicities produced by cis-diamminedichloroplatinum (DDP) injection in rats was studied. Rats were injected i.p. with BGD (2 mmol/kg) immediately after i.v. injection of DDP (20 mumol/kg). A scanning electron micrograph of the jejunum after DDP treatment showed damage in the villi, and that BGD protected the DDP-induced jejunal damage. BGD treatment also had a protective effect against DDP-induced diarrhea. BGD significantly reversed the reduction in maltase and sucrase activities of jejunal mucosa of rats treated with DDP. Platinum (Pt) concentrations in the gastrointestine as well as in the kidney and liver after DDP injection decreased following BGD treatment. The reduction of leukocytes following DDP injection returned to control values after BGD treatment. Biliary and urinary excretions of Pt after DDP injection was remarkably increased by BGD treatment. The results of this study indicated that the injection of BGD to rats treated with DDP can effectively remove Pt from the body through biliary and urinary excretions, resulting in protection of the gastrointestinal and bone marrow toxicities induced by DDP treatment.  相似文献

Mercuric ions (Hg(2+)) can be extracted from solid samples (cellulose matrix) using methanol modified supercritical CO(2) containing the fluorinated chelating agent lithium bis(trifluoroethyl)dithiocarbamate (LiFDDC). Methylmercuric chloride (CH(3)HgCl) and dimethylmercury [(CH(3))(2)Hg] can be extracted by supercritical CO(2) without chelating agent and modifier. The solubility of Hg(FDDC)(2) in supercritical CO(2) has been determined to be 5 x 10(-3)M at 5O degrees C and 150 atm, which is about 3 orders of magnitude greater than that of the non-fluorinated analogue Hg(DDC)(2). Use of methanol (5%)-modified CO(2) further enhances the solubility of Hg(FDDC)(2) by a factor of 2.4. A small amount of water added to the sample matrix tends to facilitate the extraction of Hg(FDDC)(2) and CH(3)HgCl. Potential applications of this in situ chelation-supercritical fluid extraction method for the preconcentration of mercury species and treatment of mercury contaminated wastes are discussed.  相似文献

Comparative tests of net mercury methylation potentials, with cultivated and macrophyte-associated periphyton and using stable ((200)HgCl(2) and CH(3)(199)HgCl) and labeled ((203)HgCl(2)) mercury, have been conducted in the Everglades nutrient removal area (Florida, USA) and in a tropical coastal Brazilian lake (RJ, Brazil). More methylmercury was formed by macrophyte-associated (up to 17% of added (203)Hg(II)) than cultivated (up to 1.6%) periphyton and methylmercury formation was lower in periphyton exposed to light (0.2%). High methylation was also observed for samples incubated with stable mercury isotopes (1.5-7.7% of added (200)Hg(II)), confirming the results obtained with labeled mercury. Simultaneous addition of (200)HgCl(2) and CH(3)(199)HgCl indicated that CH(3)(199)HgCl had no inhibitory effect on Hg methylation. The elevated methylation potentials observed in macrophytes, because of their root-associated periphyton, might contribute significantly to the high levels of methylmercury observed in Everglades biota. Comparative mercury methylation tests were also conducted in the water of a stratified temperate lake (Wisconsin, USA). Similar trends were observed for both stable and radioisotopes, with increasing mercury methylation along the depth profile. The highest levels (0.9% (203)Hg(II) and 0.8% (200)Hg(II)) were obtained below the oxic/anoxic boundary, where sulfide starts to increase, probably as a result of the intense activity of sulfate-reducing bacteria in the anoxic layer.  相似文献

针对湿法脱硫系统中汞还原再释放问题,选用Na_2S、2,4,6-三硫醇基钠代三嗪(TMT-18)、二硫代氨基甲酸盐类(DTCR-2)和Fenton试剂作为添加剂,探究其对脱硫浆液中汞再释放现象的影响。结果表明,四种添加剂均能有效抑制该现象的发生。1.0倍化学计量比的Na_2S足够与浆液中Hg~(2+)结合成HgS,从而将还原再释放的汞含量降低至5.27%。重金属捕集剂TMT-18和DTCR-2的最佳投加量分别为3.0倍和1.0倍化学计量比,通过与Hg~(2+)结合形成有机螯合物将浆液中的汞固定在螯合产物中,从而降低Hg~(2+)的还原,再释放的汞含量分别降低至5.09%和4.62%。具有强氧化性的Fenton试剂,当H_2O_2浓度为0.3 mol/L,[Fe~(2+)]/[H_2O_2]=1∶8时,浆液中仅有2.31%的汞被还原再释放。四种添加剂中Fenton对于浆液中汞还原再释放的抑制效果最好。  相似文献

A rapid method using capillary electrophoresis with laser-induced fluorescence detection (CE-LIF) was developed to determine free and protein-bound glutathione (GSH) in human HepG2 hepatocarcinoma cells. The samples were derivatized with 5-iodoacetamidofluorescein (5-IAF), and analyzed at 22 kV using sodium phosphate buffer (10mM, pH 11.4) and an uncoated 58 cm x 75 microm I.D. fused silica capillary. The analysis time was less than 10 min and N-acetylcysteine was used as internal standard. The derivatization conditions, such as reaction time, 5-IAF concentration, running buffer and cartridge temperature were optimized. Argon gas was used in the study to prevent the oxidization of GSH during sample preparation. The optimized method required only 30-40 nl sample per analysis and was fast and sensitive. The method was applied to the analyses of HepG2 cells treated with the small metal chelating agent, pyrrolidine dithiocarbamate (PDTC). The results demonstrate that the amount of protein-bound GSH, which reflects the amount of protein S-glutathionylation, increased in a time-dependent manner upon cell treatment with PDTC, reaching a maximum of over 50% increase 2h post-PDTC.  相似文献

Mature starfish Leptasterias polaris, collected in the St Lawrence Estuary (eastern Canada), were exposed to two mercury species (HgCI2 and CH3HgCI) via injections into the coelomic fluid. In vivo effects of some complexing agents (glutathione, mercaptoethanol and EDTA) on the distribution of 203Hg-labelled species in starfish organs and tissues and their possible role in mercury transport through membranes were studied over a 24 h period. The excretion of ammonia and mercury was also measured. When injected alone, inorganic mercury and methylmercury [CH3Hg(II)] were distributed in all organs, with a preferential adsorption in gonads, pyloric caeca and stomach. Mercury excretion was very low under all conditions studied. Mercaptoethanol, a small thiol ligand, was very efficient in reducing both mercury species in the coelomic fluid and seems to have promoted translocation towards most organs of the starfish. Its action is attributed to the formation of small and neutral complexes, HgL2 and CH3HgL, which can diffuse through membranes preserving their integrity. Glutathione increased the translocation of CH3Hg(II) towards surrounding organs, but had no apparent effect on inorganic mercury. EDTA promoted the transport of inorganic mercury only. These results highlight (1) the particular interest of starfish to workers studying in vivo chemical complexation of mercury species, and (2) the potential role of complexing molecules in the biotransport of mercury species through living membranes.  相似文献

A preliminary investigation on the introduction of high carbon content solvents into inductively coupled plasma mass spectrometry (ICP-MS) by a direct injection high efficiency nebuliser (DIHEN) is presented. Ethanol, hexane, toluene and a natural gas condensate were introduced using a flow injection system. The performance for determinations of total concentrations of mercury present in organic solvents as different species was evaluated. The most critical operating parameters were the nebuliser gas flow rate and amount of oxygen added to the plasma. For the DIHEN a nebuliser gas flow rate of 0.3 L min(-1) and 50 mL min(-1) of oxygen added to the plasma auxiliary gas flow gave stable conditions and high analyte sensitivity. Species recoveries for HgCl(2), CH(3)HgCl, (CH(3))(2)Hg and Hg(0) in hexane were 99+/-4, 101+/-4, 95+/-4 and 104+/-7%, respectively. Detection limit for mercury in hexane was 85 pg mL(-1) based on 3 sigma of a (201)Hg-spiked blank. A gradual deposition was observed at the nebuliser tip, partly blocking the gas annulus. The rate of deposition seemed to be related to the amount of carbon introduced through the nebuliser. With the optimised conditions used in this work, the nebuliser could be used for approximately 50 hexane samples before cleaning was necessary.  相似文献

A novel method for preconcentration of methylmercury and inorganic mercury from water samples was developed involving the determination of ng l−1 levels of analytes retained on the silica C18 solid sorbent, previous complexation with ammonium pyrrolidine dithiocarbamate (APDC), by slurry sampling cold vapor atomic absorption spectrometry (SS-CVAAS) in a flow injection (FI) system. Several variables were optimized affecting either the retention of both mercury species, such as APDC concentration, silica C18 amount, agitation times, or their determination, including hydrochloric acid concentration in the suspension medium, peristaltic pump speed and argon flow-rate. A Plackett-Burman saturated factorial design permitted to differentiate the influential parameters on the preconcentration efficiency, which were after optimized by the sequential simplex method. The contact time between mercury containing solution and APDC, required to reach an efficient sorption, was decreased from 26 to 3 min by the use of sonication stirring instead of magnetic stirring. The use of 1 mol dm−3 hydrochloric acid suspension medium and 0.75% (m/v) sodium borohydride reducing agent permitted the selective determination of methylmercury. The combination of 5 mol dm−3 hydrochloric acid and 10−4% (m/v) sodium borohydride was used for the selective determination of inorganic mercury. The detection limits achieved for methylmercury and inorganic mercury determination under optimum conditions were 0.96 and 0.25 ng l−1, respectively. The reliability of the proposed method for the determination of both mercury species in waters was checked by the analysis of samples spiked with known concentrations of methylmercury and inorganic mercury; quantitative recoveries were obtained.  相似文献

A graphical method, based on logarithmic concentration diagrams, for construction, without any calculations, of complexometric titration curves is examined. The titration curves obtained for different kinds of unidentate, bidentate and quadridentate ligands clearly show why only chelating ligands are usually used in titrimetric analysis. The method has also been applied to two practical cases where unidentate ligands are used: (a) the complexometric determination of mercury(II) with halides and (b) the determination of cyanide with silver, which involves both a complexation and a precipitation system; for this purpose construction of the diagrams for the HgCl(2)/HgCl(+)/Hg(2+) and Ag(CN)(2)(-)/AgCN/CN(-) systems is considered in detail.  相似文献

An on-line inorganic and organomercury species separation, preconcentration and determination system consisting of cold vapor atomic absorption spectrometry (CV-AAS or CV-ETAAS) coupled to a flow injection (FI) method was studied. The inorganic mercury species was retained on a column (i.d., 3 mm; length 3 cm) packed to a height of 0.7 cm with a chelating resin aminopropyl-controlled pore glass (550 A) functionalized with [1,5-bis (2 pyridyl)-3-sulphophenyl methylene thiocarbonohydrazyde] placed in the injection valve of a simple flow manifold. Methylmercury is not directly determined. Previous oxidation of the organomercurial species permitted the determination of total mercury. The separation of mercury species was obtained by the selective retention of inorganic mercury on the chelating resin. The difference between total and inorganic mercury determined the organomercury content in the sample. The inorganic mercury was removed on-line from the microcolumn with 6% (m/v) thiourea. The mercury cold vapor generation was performed on-line with 0.2% (m/v) sodium tethrahydroborate and 0.05% (m/v) sodium hydroxide as reducing solution. The determination was performed using CV-AAS and CV-ETAAS, both approaches have been used and compared for the speciation of mercury in sea food. A detection limit of 10 and 6 ng l(-1) was achieved for CV-AAS and CV-ETAAS, respectively. The precision for 10 replicate determinations at the 1 microg l(-1) Hg level was 3.5% relative standard deviation (R.S.D.), calculated from the peak heights obtained. Both approaches were validated with the use of two certified reference materials and by spiking experiments. By analyzing the two biological certified materials, it was evident that the difference between the total mercury and inorganic mercury corresponds to methylmercury. The concentrations obtained by both techniques were in agreement with the certified values or with differences of the certified values for total Hg(2+) and CH(3)Hg(+), according to the t-test for a 95% confidence level. It is amazing how this very simple method is able to provide very important information on mercury speciation.  相似文献

Herein, we present a study of the pharmacokinetics and biodistribution of a butadiyne‐linked conjugated porphyrin dimer (Oxdime) designed to have high near‐infrared (NIR) 2‐photon absorption cross‐section for photodynamic therapy (PDT). Changes in biodistribution over time were monitored in mice carrying B16‐F10 melanoma xenografts, following intravenous injection. Using fluorescence imaging of live animals and analyzing isolated organs ex vivo at different time points between 30 min and 24 h after injection, accumulation of Oxdime was measured in several organs (heart, kidney and liver) and in tumor. The concentration in the plasma was about 5–10 times higher than in other tissues. The fluorescence signal peaked at 3–12 h after injection in most tissues, including the tumor and the plasma. The change in the fluorescence emission spectrum of the sensitizer over time was also monitored and a shift in the maximum from 800 to 740 nm was observed over 24 h, showing that the Oxdime is metabolized. Significant quantities accumulated in the tumor, indicating that this PDT sensitizer may be promising for cancer treatment.  相似文献

Starfish, Leptasterias polaris, were exposed between 30 min and 48 h to seawater containing 0.25 nmol dm?3 of radiolabelled methylmercury (Me203HgCI), tributyltin [(C4H9)3113SnCI], and inorganic 203HgCI2 and 113SnCI4, with the objectives of comparing the uptake and distribution kinetics of these metal species in organs and tissues of treated organisms. Some starfish exposed to metals for 48 h were allowed to depurate for 24 h in clean seawater. Whole-body autoradiography was used to locate radiotracers very precisely within starfish tissues. The total amount of methylmercury (MeHg) accumulated in the whole animal after 48 h reached 0.53 nmol compared with 0.09 nmol for inorganic mercury, while tributyltin (TBT) reached 0.72 nmol compared with 0.017 nmol for inorganic tin. No significant reduction of body burdens occurred during the depuration period. The first-order rate constant characterizing the uptake of metals by whole animals, k1, ranged from 0.102 h?1 for MeHg to 3.6 × 10?3h?1 for inorganic mercury(II) and to 8.4 × 10?4 h?1 for inorganic tin(IV). The first-order rate constant characterizing the translocation of metals from seawater-exposed tissues toward internal organs, k3, was available for inorganic Hg and Sn and had values similar to k1. Concentration ratios between external tissues and internal organs after a 48 h exposure were 11.5 and 25.4 for MeHg and TBT, respectively, and 2.1 and 6.1 for inorganic mercury and tin. Furthermore, autoradiograms showed that MeHg and TBT were accumulated only on the external surface of the body wall and podia. This finding indicates a much slower translocation process for organometallic species than inorganic species, a process which seems to be related to the binding mode of MeHg and TBT to the organic matrix of external tissues of starfish.  相似文献

The biodistribution of a new and very potent photosensitizer, benzoporphyrin derivative-monoacid, ring A (BPD-MA), was determined in normal and P815 (mastocytoma) or M1 (rhabdomyosarcoma) tumor-bearing DBA/2J mice. A dose of 80 micrograms of 3H-BPD-MA was determined at 3, 24, 48, 72, 96 and 168 h post injection. The following tissues were tested: blood, brain, heart, intestine, kidney, lung, liver, muscle, skin, stomach, spleen, thymus and tumor. The biodistribution of 3H-BPD-MA in normal and tumor-bearing mice was comparable overall. 3H-BPD-MA localized in tumors better than in other tissues except kidney, liver and spleen. The tumor to tissue ratios were in the range 1.5-3 at 24 h post injection and increased further during the next 72 h. The highest levels of 3H-BPD-MA were observed in all tissues at 3 h post injection and decreased rapidly during the first 24 h. After 24 h the clearance from tissues was rather slow. The preliminary clearance data obtained in a group of five normal mice indicated that the majority of the injected dose (60%) cleared from the body via the bile and feces, while only about 4% cleared via kidneys and urine. Studies in which 3H-BPD-MA was extracted from tumor, kidney and liver 3 and 24 h after injection showed that, at 3 h, all the photosensitizing activity in tumor was retained. At 24 h only 39% of the activity was retained and considerably less active material was present in liver and kidney.  相似文献
