Vinylogous β-Cleavage of Enones: UV.-irradiation of 4-(3′,7′,7′-trimethyl-2′-oxabicyclo[3.2.0]hept-3′-ene-1′-yl)but-3-ene-2-on On 1π,π*-excitation (λ = 254 nm) in acetonitrile (E/Z)- 2 is converted into the isomers 4–9 and undergoes fragmentation yielding 10 ; in methanol (E/Z)- 2 gives 7–10 and is transformed into 11 by incorporation of the solvent. On 1π,π*-excitation (λ λ?347 nm; benzene-d6) (E)- 2 is isomerized into (Z)- 2 , which is converted into the isomers 3 and 4 by further irradiation. 1π,π*-Excitation (λ = 254 nm; acetonitrile) of 4 gives 6 and (E)- 9 , whereas UV.-irradiation (λ = 254 nm; acetonitrile-d3) of 5 yields (E)- 7 and 8 . On 1π,π*-excitation (λ = 254 nm; acetonitrile) of (E/Z)- 12 the compounds (E)- 14 and (E)- 15 are obtained.  相似文献

The Photoinduced Cleavage of Conjugated γ, δ-Epoxyenones: UV.-Irradiation of 5,6-Epoxy-3, 4-didehydro-5,6-dihydro-β-ionone On 1n, π*-excitation (λ ≥ 347 nm) in pentane or CClF2CFCl2 (E)- 1 is isomerized to the dihydrofurane (E/Z)- 2 as well to the ethers 3 and 5. Besides these products the isomeric cyclopropane derivative (E)- 4 and the acetal 6 are obtained in methanol. The detection of 6 indicates the formation of an intermediate ketoniumylide a which may give 6 by addition of methanol. ? On 1π, π*-excitation (λ=254 nm) in acetonitrile-d3, CClF2CFCl2 or pentane (E)- 1 yields almost exclusively (E)- 2. In methanol 6 is obtained in addition to (E/Z)- 2 , but no (E)- 4 and 5 is formed.  相似文献

Bis((Z)‐5‐phenyl‐2‐phenylmethylidene‐1, 3‐dithiole‐4‐yl)monosulfane ( 6 ), a molecule consisting of two diphenyldithiafulvene units connected by a sulfur bridge, was synthesized by the selective lithiation of (Z)‐4‐phenyl‐2‐phenylmethylidene‐1, 3‐dithiole ( 7a ) at the endocyclic double bond and by subsequent reaction of the lithiated intermediate with bis(phenylsulfonyl)sulfane. Since this reaction sequence proceeded with retention of configuration, of three possible isomers (E, E, Z, E, and Z, Z) only the Z, Z form was obtained. On the basis of the X‐ray structure analysis and the NMR‐spectroscopic characterization of 6 supplemented by the NMR parameters of (E)‐ and (Z)‐4‐phenyl‐2‐phenylmethylidene‐1, 3‐dithiole, it was demonstrated that two characteristic 5J coupling constants of the proton at the exocyclic double bond indicate the configuration (Z or E) of disubstituted dithiafuvene derivatives.  相似文献

Three novel curcuminoids, curcumaromins A–C ( 1 – 3 , resp.), along with a known compound, longiferone B ( 4 ) were isolated from Curcuma aromatica Salisb . The structures of the new compounds were elucidated as (1E,4Z,6E)‐5‐hydroxy‐7‐{4‐hydroxy‐3‐[(1R*,6R*)‐3‐methyl‐6‐(propan‐2‐yl)cyclohex‐2‐en‐1‐yl)phenyl}‐1‐(4‐hydroxyphenyl)hepta‐1,4,6‐trien‐3‐one ( 1 ), 2,3‐dihydro‐2‐(4‐hydroxyphenyl)‐6‐[(E)‐2‐(4‐hydroxyphenyl)ethenyl]‐5‐[(1R*,6R*)‐3‐methyl‐6‐(propan‐2‐yl)cyclohex‐2‐en‐1‐yl]‐4H‐pyran‐4‐one ( 2 ), and (1E,6E)‐1,7‐bis(4‐hydroxyphenyl)‐4‐[(1R*,6R*)‐3‐methyl‐6‐(propan‐2‐yl)cyclohex‐2‐en‐1‐yl]hepta‐1,6‐diene‐3,5‐dione ( 3 ) on the basis of spectroscopic analysis. Curcumaromins A–C ( 1 – 3 ) represented the first examples of menthane monoterpene‐coupled curcuminoids. The known compound, longiferone B ( 4 ), was the first daucane sesquiterpene isolated from the genus Curcuma.  相似文献

The structures of the main products, i.e. (9Z)‐ ( 2 ), (9′Z)‐ ( 3 ), (13Z)‐ ( 4 ), (13′Z)‐ ( 5 ), and (15Z)‐5,6‐diepikarpoxanthin ( 6 ), were determined by their UV/VIS, CD, 1H‐NMR, and mass spectra. In addition, (9Z,13′Z)‐ or (13Z,9′Z)‐ ( 7 ), (9Z,9′Z)‐ ( 8 ), and (9Z,13Z)‐ or (9′Z,13′Z)‐5,6‐diepikarpoxanthin ( 9 ) were tentatively identified as minor products of the I2‐catalyzed photoisomerization.  相似文献

The reactions of 4,4′‐dimethoxythiobenzophenone ( 1 ) with (S)‐2‐methyloxirane ((S)‐ 2 ) and (R)‐2‐phenyloxirane ((R)‐ 6 ) in the presence of a Lewis acid such as BF3?Et2O, ZnCl2, or SiO2 in dry CH2Cl2 led to the corresponding 1 : 1 adducts, i.e., 1,3‐oxathiolanes (S)‐ 3 with Me at C(5), and (S)‐ 7 and (R)‐ 8 with Ph at C(4) and C(5), respectively. A 1 : 2 adduct, 1,3,6‐dioxathiocane (4S,8S)‐ 4 and 1,3‐dioxolane (S)‐ 9 , respectively, were formed as minor products (Schemes 3 and 5, Tables 1 and 2). Treatment of the 1 : 1 adduct (S)‐ 3 with (S)‐ 2 and BF3?Et2O gave the 1 : 2 adduct (4S,8S)‐ 4 (Scheme 4). In the case of the enolized thioketone 1,3‐diphenylprop‐1‐ene‐2‐thiol ( 10 ) with (S)‐ 2 and (R)‐ 6 in the presence of SiO2, the enesulfanyl alcohols (1′Z,2S)‐ 11 and (1′E,2S)‐ 11 , and (1′Z,2S)‐ 13 , (1′E,2S)‐ 13 , (1′Z,1R)‐ 15 , and (1′E,1R)‐ 15 , respectively, as well as a 1,3‐oxathiolane (S)‐ 14 were formed (Schemes 6 and 8). In the presence of HCl, the enesulfanyl alcohols (1′Z,2S)‐ 11 , (1′Z,2S)‐ 13 , (1′E,2S)‐ 13 , (1′Z,1R)‐ 15 , and (1′E,1R)‐ 15 cyclize to give the corresponding 1,3‐oxathiolanes (S)‐ 12 , (S)‐ 14 , and (R)‐ 16 , respectively (Schemes 7, 9, and 10). The structures of (1′E,2S)‐ 11 , (S)‐ 12 , and (S)‐ 14 were confirmed by X‐ray crystallography (Figs. 13). These results show that 1,3‐oxathiolanes can be prepared directly via the Lewis acid‐catalyzed reactions of oxiranes with non‐enolizable thioketones, and also in two steps with enolized thioketones. The nucleophilic attack of the thiocarbonyl or enesulfanyl S‐atom at the Lewis acid‐complexed oxirane ring proceeds with high regio‐ and stereoselectivity via an Sn 2‐type mechanism.  相似文献

The thermal reaction of homoazulene (=bicyclo[5.3.1]undeca‐1,3,5,7,9‐pentaene; 2 ) with dimethyl acetylenedicarboxylate (ADM) in 1,2‐dichloroethane (ClCH2CH2Cl) results, in contrast to an earlier report [5], in formation of not only dimethyl homoheptalene‐4,5‐dicarboxylate (=bicyclo[5.5.1]trideca‐1,3,5,7,9,11‐hexaene‐4,5‐dicarboxylate; 3 ), but also of a 4 : 1 mixture of 3 and dimethyl homoheptalene‐2,3‐dicarboxylate ( 13 ) in almost quantitative yield (Schemes 1 and 3). The structures of both homoheptalenes have been corroborated by X‐ray crystal‐structure analysis (Fig. 5). The double‐bond‐shifted (DBS) isomers 3 ′ and 13 ′ of 3 and 13 , respectively, could not be detected in their 1H‐NMR spectra (600 MHz threshold of detection ≥0.5%), in agreement with the AM1‐calculated ΔH values of the four isomeric homoheptalene‐dicarboxylates (cf. Table 4). Vilsmeyer formylation of homoazulene ( 2 ) gave homoazulene‐8‐carbaldehyde ( 14 ) in a yield of 67%, which, on treatment with benzylidene‐(triphenyl)‐λ5‐phosphane, gave, in almost quantitative yield, a 1.6 : 1 mixture of (Z)‐ and (E)‐8‐styrylhomoazulene ((Z)‐ 15 and (E)‐ 15 , resp.). Thermal reaction of the latter mixture with ADM in 1,2‐dichloroethane led, in a yield of 42%, to a 5 : 1 mixture of dimethyl (Z)‐ and (E)‐2‐styrylhomoheptalene‐4,5‐dicarboxylate ((Z)‐ 15 and (E)‐ 16 , resp.). Both isomers were separated by column chromatography on silica gel. Again, the DBS isomers of (Z)‐ 16 and (E)‐ 16 , i.e., (Z)‐ 16 ′ and (E)‐ 16 ′, could not be detected in the 1H‐NMR spectra (600 MHz) of pure (Z)‐ 16 and (E)‐ 16 .  相似文献

Methyl (2Z,6Z,10E,14E)‐ ( 3 ) and methyl (2E,6Z,10E,14E)‐geranylfarnesoate ( 4 ) were prepared, and then individually cyclized in the presence of the superacid FSO3H. In the case of substrate 3 , the scalaranic ester 9 (26%) and the cheilanthanic ester 10 (39%) were isolated. Under the same conditions, substrate 4 afforded a mixture of the corresponding stereoisomers 11 (25%) and 12 (63%). The observed product selectivity supports that the internal, (6Z)‐configured C?C bond in these and other biologically relevant substrates plays an essential role in the cyclization process.  相似文献

Caulerpenyne ( 1 ), the most abundant of the ecotoxicologically relevant sesquiterpenoids of the Mediterranean-adapted tropical green seaweed Caulerpa taxifolia, was found to react with Et3N or pyridine in MeOH by initial deprotection of C(1)HO to give oxytoxin 1 ( 2a ), previously isolated from the sacoglossan mollusc Oxynoe olivacea. With BuNH2, without any precaution to exclude light, 1 gave the series of racemic 3 and 4 , and achiral (4E,6E)- 5 , (4E,6Z)- 5 , (4Z,6E)- 5 , and (4Z,6Z)- 5 pyrrole compounds, corresponding to formal C(4) substitution, 4,5-β-elimination, and (E/Z)-isomerization at the C(4)?C(5) and C(6)?C(7) bonds. Changing to CDCl3 as solvent in the dark, 1 gave cleanly, via 2a as an intermediate, 3 and (4E,6E)- 5 . The latter proved to be prone to (E/Z)-photoisomerization. Under standard acetylation conditions, 3 gave (4E,6E)- 5 via acetamide 7 as an intermediate. Particular notice is warranted by selective deprotection of 1 at C(1), mimicking enzyme reactions, and unprecedented formation of pyrrole compounds from freely-rotating, protected 1,4-dialdehyde systems.  相似文献

The new cyrhetrenyl acylhydrazone [(CO)3Re(η5‐C5H4)‐C(O)‐NH‐N = C(CH3)‐(2‐C4H2S‐5‐NO2)] ( E‐CyAH ) has been designed, synthesized and fully characterized to study the effect of having a cyrhetrenyl fragment (sensitizer) covalently bonded to an acylhydrazone moiety (switch), on its photophysical and photochemical properties. The crystal structure reveals that E‐CyAH adopts an E‐configuration around the iminic moiety [‐N = C(CH3)]. The absorption spectrum of E‐CyAH displays two bands at 270 and 380 nm, which are mainly ascribed to π → π* intraligand (IL) and dπ → π* metal‐to‐ligand charge transfer (MLCT) transitions, being consistent with DFT/TD‐DFT calculations. Upon 365 nm irradiation, E‐CyAH photoisomerizes to Z‐CyAH , as evidenced by UV‐Vis and 1H‐NMR spectral changes, with a quantum yield value ΦE‐CyAH →Z‐CyAH of 0.30. Z‐CyAH undergoes a first‐order thermal back‐isomerization process, with a relatively short half‐life τ1/2 of 277 min. Consequently, E‐CyAH was quantitatively recovered after 24 h, making it a fully reversible T‐type molecular photoswitch. This remarkable behavior allows us to measure the individual photophysical properties for both isomers. In addition, E‐CyAH and Z‐CyAH efficiently photosensitize the generation of singlet oxygen (O2 (1Δg)) with good yield (ΦΔ = 0.342).  相似文献

Violaxanthin A (=(all‐E,3S,5S,6R,3′S,5′S,6′R)‐5,6 : 5′,6′‐diepoxy‐5,6,5′,6′‐tetrahydro‐β,β‐carotene‐3,3′‐diol =syn,syn‐violaxanthin; 5 ) and violaxanthin B (=(all‐E,3S,5S,6R,3′S,5′R,6′S)‐5,6 : 5′,6′‐diepoxy‐5,6,5′,6′‐tetrahydro‐β,β‐carotene‐3,3′‐diol=syn,anti‐violaxanthin; 6 ) were prepared by epoxidation of zeaxanthin diacetate ( 1 ) with monoperphthalic acid. Eight novel Schiff bases derived from benzil dihydrazone ( BDH ) or benzil monohydrazone ( BMH ) and four fused‐ring carbonyl compounds (3‐formylindole, FI ; 3‐acetylindole, AI ; 3‐formyl‐1‐methylindole, MFI ; 1‐formylnaphthalene, FN ) were synthesized and characterized by elemental analysis, ESI–QTOF–MS, 1H and 13C NMR spectroscopy, as well as single‐crystal X‐ray diffraction. Moreover, the in vitro cytotoxicity of the eight title compounds was evaluated against two tumour cell lines (A549 human lung cancer and 4T1 mouse breast cancer) and two normal cell lines (MRC‐5 normal lung cells and NIH 3T3 fibroblasts) by MTT assay. The results indicate that four ( BDHMFI , BDHFN , BMHMFI and BMHFN ) are inactive and the other four ( BDHFI , BDHAI , BMHFI and BMHAI ) show severe toxicities against human A549 and mouse 4T1 cells, similar to the standard cisplatin. All the compounds exhibited weaker cytotoxicity against normal cells than cancer cells. The Swiss Target Prediction web server was applied for the prediction of protein targets. After analyzing the differences in frequency hits between these active and inactive Schiff bases, 18 probable targets were selected for reverse docking with the Surflex‐dock function in SYBYL‐X 2.0 software. Three target proteins, i.e. human ether‐á‐go‐go‐related (hERG) potassium channel, the inhibitor of apoptosis protein 3 and serine/threonine‐protein kinase PIM1, were chosen as the targets. Finally, the ligand‐based structure–activity relationships were analyzed based on the putative protein target (hERG) docking results, which will be used to design and synthesize novel hERG ion channel inhibitors.  相似文献

The Z and E isomers of 3‐[4‐(dimethylamino)phenyl]‐2‐(2,4,6‐tribromophenyl)acrylonitrile, C17H13Br3N2, ( 1 ), were obtained simultaneously by a Knoevenagel condensation between 4‐(dimethylamino)benzaldehyde and 2‐(2,4,6‐tribromophenyl)acetonitrile, and were investigated by X‐ray diffraction and density functional theory (DFT) quantum‐chemical calculations. The (Z)‐( 1 ) isomer is monoclinic (space group P21/n, Z′ = 1), whereas the (E)‐( 1 ) isomer is triclinic (space group P, Z′ = 2). The two crystallographically‐independent molecules of (E)‐( 1 ) adopt similar geometries. The corresponding bond lengths and angles in the two isomers of ( 1 ) are very similar. The difference in the calculated total energies of isolated molecules of (Z)‐( 1 ) and (E)‐( 1 ) with DFT‐optimized geometries is ∼4.47 kJ mol−1, with the minimum value corresponding to the Z isomer. The crystal structure of (Z)‐( 1 ) reveals strong intermolecular nonvalent Br…N [3.100 (2) and 3.216 (3) Å] interactions which link the molecules into layers parallel to (10). In contrast, molecules of (E)‐( 1 ) in the crystal are bound to each other by strong nonvalent Br…Br [3.5556 (10) Å] and weak Br…N [3.433 (4) Å] interactions, forming chains propagating along [110]. The crystal packing of (Z)‐( 1 ) is denser than that of (E)‐( 1 ), implying that the crystal structure realized for (Z)‐( 1 ) is more stable than that for (E)‐( 1 ).  相似文献

Reduction of 1,2-Bis[(Z)-(2-nitrophenyl)-NNO-azoxy]benzene1: Synthesis of Cyclotrisazobenzene ( = (5E,6aZ,11E,12aZ,17E,18aZ)-5,6,11,12,17,18-Hexaazatribenzo[aei][1,3,5,7,9,11]cyclododeca-hexaene) Na2S reduction of 1,2-bis[(Z)-(2-nitrophenyl)-NNO-azoxy]benzene ( 2 ) yielded 3 deoxygenated products: the (known) red 2,2′-((E,E)-1,2-phenylenbisazo)dianiline ( 3 , 23%), the orange 2-[2-((E)-2-aminophenylazo)phenyl]-2H-benzotriazol ( 4 , 55%) and the colorless 2,2′-(1,2-phenylene)di-2H-benzotriazol ( 5 , 13%). The constitutions of 3 – 5 and of 6 , the N-acetyl derivative of 4 , were deduced from their 1H-NMR spectra (chemical shifts, couplings, and symmetry properties), and the configurations of 3 , 4 , and 6 at their N,N-double bonds are assumed to be the same as in 2 . Oxidation of 3 with 2 mol-equiv. of Pb(OAc)4 afforded 5 (47%) and a novel, highly symmetrical macrocycle, called cyclotrisazobenzene ( 7 , 24%). The constitution of 7 as a tribenzo-hexaaza[12]annulene and its (E)-configuration at the N,N-bonds was confirmed by X-ray analysis. The molecular symmetry expressed by the 1H-, 13C- and 15N-NMR spectra of 7 reveals a rapid torsional motion around the six N,C bonds. This implies that the N,N-double bonds in the cyclic 12π-electron system (or 24π-electron system if the benzene rings are included) of 7 are highly localized.  相似文献

The conformations of (Z)‐ and (E)‐5‐oxo‐B‐nor‐5,10‐secocholest‐1(10)‐en‐3β‐yl acetates ( 2 and 3 , resp.) were examined by a combination of X‐ray crystallographic analysis and NMR spectroscopy, with emphasis on the geometry of the cyclononenone moiety. The 1H‐ and 13C‐NMR spectra showed that the unsaturated nine‐membered ring of (E)‐isomer 3 in C6D6 and (D6)acetone solution exists in a sole conformation of type B 1 , which is similar to its solid‐state conformation. The (Z)‐isomer 2 in C6D6, CDCl3, and (D6)acetone solution, however, exists in two conformational forms of different families, with different orientation of the carbonyl group, the predominant form (85%) corresponding to the conformation of type A 1 and the minor (15%) to the conformation A 2 present also in the crystalline state. In this solid‐state conformations of the nine‐membered ring of both compounds, the 19‐Me and 5‐oxo groups are ‘β’‐oriented. The NMR analysis suggests that the nine‐membered ring of 4 has a conformation of type C 1 in CDCl3 solution.  相似文献

The crystal structures of the four E,Z,E isomers of 1‐(4‐alk­oxy­phen­yl)‐6‐(4‐nitro­phen­yl)hexa‐1,3,5‐triene, namely (E,Z,E)‐1‐(4‐methoxy­phen­yl)‐6‐(4‐nitro­phen­yl)hexa‐1,3,5‐triene, C19H17NO3, (E,Z,E)‐1‐(4‐ethoxy­phen­yl)‐6‐(4‐nitro­phen­yl)hexa‐1,3,5‐triene, C20H19NO3, (E,Z,E)‐1‐(4‐nitro­phen­yl)‐6‐(4‐n‐propoxyphen­yl)hexa‐1,3,5‐triene, C21H21NO3, and (E,Z,E)‐1‐(4‐n‐butoxy­phen­yl)‐6‐(4‐nitro­phen­yl)hexa‐1,3,5‐triene, C22H23NO3, have been determined. Inter­molecular N⋯O dipole inter­actions between the nitro groups are observed for the meth­oxy derivative, while for the eth­oxy derivative, two adjacent mol­ecules are linked at both ends through N⋯O dipole–dipole inter­actions between the N atom of the nitro group and the O atom of the eth­oxy group to form a supra­molecular ring‐like structure. In the crystal structures of the n‐prop­oxy and n‐but­oxy derivatives, the shortest inter­molecular distances are those between the two O atoms of the alk­oxy groups. Thus, the nearest two mol­ecules form an S‐shaped supra­molecular dimer in these crystal structures.  相似文献

Experiments on the competitive incorporation of farnesol-stereoisomers into cantharidin Farnesol ( 2 ) has been demonstrated to be an efficient precursor for cantharidin ( 1 ), into which it is transformed by elimination of C(1), C(5), C(6), C(7) and C(7′) [1]. The following incorporation experiments with doubly labelled (3H and 14C) stereoisomers of farnesol present strong evidence that (E,E)- farnesol ((E,E)- 2 ) in fact is the precursor for cantharidin, whereas (2E, 6Z)- 2 and (Z,Z)- 2 are not utilized for the biosynthesis of cantharidin. A possible mechanism for the incorporation of (2Z,6E)-farnesol ((2Z,6E)- 2 ) to an extent of 56,8% relative to (E,E)- 2 is discussed.  相似文献

Five new sesquiterpenoids, namely, 8β‐(angeloyloxy)‐4β,6α,15‐trihydroxy‐14‐oxoguaia‐9,11(13)‐dien‐12‐oic acid 12,6‐lactone ( 1 ), 4β,6α,15‐trihydroxy‐8β‐(isobutyryloxy)‐14‐oxoguaia‐9,11(13)‐dien‐12‐oic acid 12,6‐lactone ( 2 ), 11,12,13trinorguai‐6‐ene‐4β,10β‐diol ( 3 ), (1(10)E,4E,8Z)‐8‐(angeloyloxy)‐6α,15‐dihydroxy‐14‐oxogermacra‐(1(10),4,8,11(13)‐tetraen‐12‐oic acid 12,6‐lactone ( 9 ), and (1(10)E,4β)‐8β‐(angeloyloxy)‐6α,14,15‐trihydroxygermacra‐1(10),11(13)‐dien‐12‐oic acid 12,6‐lactone ( 11 ), and three new artifacts, (1(10)E,4Z)‐8β‐(angeloyloxy)‐9α‐ethoxy‐6α,15‐dihydroxy‐14‐oxogermacra‐1(10),4,11(13)‐trien‐12‐oic acid 12,6‐lactone ( 6 ), (1(10)E,4Z)‐8β‐(angeloyloxy)‐9α,13‐diethoxy‐6α,15‐dihydroxy‐14‐oxogermacra‐1(10),4‐dien‐12‐oic acid 12,6‐lactone ( 7 ), and (1(10)E,4Z)‐8β‐(angeloyloxy)‐9α‐ethoxy‐6α,15‐dihydroxy‐13‐methoxy‐14‐oxogermacra‐1(10),4‐dien‐12‐oic acid 12,6‐lactone ( 8 ), together with the three known sesquiterpenoids 4, 5 , and 10 , were isolated from the aerial parts of Siegesbeckia orientalis L.
