Starch is one of the main carbohydrates in food; it is formed by two polysaccharides: amylose and amylopectin. The granule size of starch varies with different botanical origins and ranges from less than 1 μm to more than 100 μm. Some physicochemical and functional properties vary with the size of the granule, which makes it of great interest to find an efficient and accurate size-based separation method. In this study, the full-feed depletion mode of split-flow thin cell fractionation (FFD-SF) was employed for a size-based fractionation of two types of starch granules (corn and potato) on a large scale. The fractionation efficiency (FE) of fraction-a for corn and potato granules was 98.4 and 99.4%, respectively. The FFD-SF fractions were analyzed using optical microscopy (OM) and gravitational field-flow fractionation (GrFFF). The respective size distribution results were in close agreement for the corn starch fractions, while they were slightly different for the potato starch fractions. The thermal properties of FFD-SF fractions were analyzed, and the results for the potato starch showed that the peak temperature of gelatinization (Tp) slightly decreases as the size of the granules increases. Additionally, the enthalpy of gelatinization (ΔH) increases when the granule size increases and shows negative correlation with the gelatinization range (ΔT).

Centrifugal Partition Chromatography (CPC), a liquid–liquid preparative chromatography using two immiscible solvent systems, benefits from numerous advantages for the separation or purification of synthetic or natural products. This study presents the on-line hyphenation of CPC-Evaporative Light Scattering Detector (CPC-ELSD) with High Performance Liquid Chromatography-UV (HPLC-UV) for the fractionation of flavonols from a solvent-free microwave extract of sea buckthorn (Hippophaë rhamnoides L., Elaeagnaceae) berries. An Arizona G system was used for the fractionation of flavonoids by CPC and a fused core Halo C18 column allowed the on-line analyses of collected fractions by HPLC. The on-line CPC/HPLC procedure allowed the simultaneous fractionation step at preparative scale combined with the HPLC analyses which provide direct fingerprint of collected fractions. Thus the crude extract was simplified and immediate information on the composition of fractions could be obtained. Furthermore, this methodology reduced the time of post-fractionation steps and facilitated identification of main molecules by Mass Spectrometry (MS). Rutin, isorhamnetin-3-O-rutinoside, isorhamnetin-3-O-glucoside, quercetin-3-O-glucoside, isorhamnetin-rhamnoside, quercetin and isorhamnetin were identified. CPC-ELSD/HPLC-UV could be considered as a high-throughput technique for the guided fractionation of bioactive natural products from complex crude extracts.  相似文献

A combination of gravitational split-flow thin (SPLITT) fractionation and sedimentation/steric field-flow fractionation (Sd/StFFF) has been used for continuous size-sorting of a sediment sample and for size analysis of the collected fractions. An IAEA (International Atomic Energy Agency) sediment material was separated into four size fractions (with theoretical size ranges <1.0, 1.0–3.0, 3.0–5.0, and >5.0 m in diameter) by means of a three-step gravitational SPLITT fractionation (GSF) for which the same GSF channel was used throughout. The GSF fractions were collected and examined by optical microscopy (OM) and by Sd/St FFF. The mean diameters of the GSF fractions measured by OM were within the size interval predicted by GSF theory, despite the theory assuming that all particles are spherical, which is not true for the sediment particles. The Sd/St FFF results showed that retention shifted toward shorter elution time (or larger size) than expected, probably because of the shape effect. The results from GSF, OM, and Sd/StFFF are discussed in detail.  相似文献

Trace metals such as Cadmium (Cd) and Zinc (Zn) are known to exhibit adverse effects on many aquatic organisms including early life stages of fish. In contact with contaminated sediment, fish eggs and embryos may be exposed to metals via the water phase as well as via direct contact with contaminated particles. This may result in body burdens that are difficult to predict and may vary according to individual micro scale exposure conditions. The highly sensitive total reflection X-ray fluorescence spectrometry (TXRF) may provide a tool to analyse individual embryos for internal contaminant concentrations and thus helps to develop a better understanding of dose–response relationships. To test this hypothesis, embryos of Danio rerio were exposed to Cd and Zn spiked sediment in different treatments applying an ion exchange resin for modification of bioavailable concentrations. The TXRF analysis indicated individual embryos with dramatically enhanced exposure compared to other individuals despite uniform exposure conditions on a macro scale. Ion exchanger reduced embryo Zn concentrations to values close to control value with a comparably low standard deviation. Cadmium concentrations in embryos were in the range of 4000 to 7000 µg/g with a median of 5740 µg/g. A commercial ion exchanger reduced individual body burdens by a factor 50 to 100. Individual peak body burdens of up to 3160 µg/g were accompanied by reduced weight of the fish eggs due to early death i.e. coagulation. The investigation of exposure and effects on an individual-based scale may significantly help to reduce uncertainty and inconsistencies occurring in conventional analysis of pooled fish embryo samples.  相似文献

This study was designed to measure the distribution of pesticides within the mobile phase of simulated irrigation run-off water, using centrifugal split-flow thin-channel (SPLITT) fractionation, a novel technique providing a gentle separation of natural sediment and suspended particles. Particular attention is paid to the extraction of pesticide residues for enzyme-linked immunosorbent assay (ELISA) analysis; ELISA was used because of the limited sample size.Centrifugal SPLITT fractionation combined laminar flow hydrodynamics and centrifugal sedimentation to obtain a continuous binary separation of suspended particles. The non-destructive technique allowed an accurate separation of particles into fractions with divisions at 0.5, 2 and 10 μm, with those above 25 μm being performed by wet sieving. ELISA was used to analyse the concentration of endosulfan and diuron for each fraction generated by the SPLITT technique.This data can be used to determine the role that particulate fines and colloidal fractions play in the transport of bound organic pollutants within the environment and to examine prospects for remediation on farms.  相似文献

The availability of robust and highly efficient separation methods represents a major requirement for proteome analysis. This study investigated the characteristics of two different gel-free proteomic approaches to the fractionation of proteolytic peptides and intact proteins, respectively, in a first separation dimension. Separation and mass spectrometric detection by matrix-assisted laser desorption/ionization tandem mass spectrometry (MALDI-MS/MS) were performed at the peptide level in both methods. Bottom-up analysis (BU) was carried out employing well established peptide fractionation in the first separation dimension by strong cation-exchange chromatography (SCX), followed by ion-pair reversed-phase chromatography (IP-RPC) in the second dimension. In the semi-top-down approach (STD), which involved intact protein fractionation in the first dimension, the separation mode in both dimensions was IP-RPC utilizing monolithic columns. Application of the two approaches to the proteome analysis of proteins extracted from a tumor tissue revealed that the BU method identified more proteins (1245 in BU versus 920 in STD) while STD analysis offered higher sequence coverage (14.8% in BU versus 17.5% in STD on average). The identification of more basic and larger proteins was slightly favored in the BU approach, most probably due to higher losses of these proteins during intact protein handling and separation in the STD method. A significant degree of complementarity was revealed by an approximately 33% overlap between one BU and STD replicate, while 33% each of the protein identifications were unique to both methods. In the STD method, peptides obtained upon digestion of the proteins contained in fractions of the first separation dimension covered a broad elution window in the second-dimension separation, which demonstrates a high degree of “pseudo-orthogonality” of protein and peptide separation by IP-RPC in both separation dimensions.  相似文献

In situ fusion on the boat-type graphite platform has been used as a sample pretreatment for the direct determination of Co, Cr and Mn in Portland cement by solid sampling graphite furnace atomic absorption spectrometry (SS-GF AAS). The 3-field Zeeman technique was adopted for background correction to decrease the sensitivity during measurements. This strategy allowed working with up to 200 µg of sample. The in situ fusion was accomplished using 10 µL of a flux mixture 4.0% m/v Na2CO3 + 4.0% m/v ZnO + 0.1% m/v Triton® X-100 added over the cement sample and heated at 800 °C for 20 s. The resulting mould was completely dissolved with 10 µL of 0.1% m/v HNO3. Limits of detection were 0.11 µg g− 1 for Co, 1.1 µg g− 1 for Cr and 1.9 µg g− 1 for Mn. The accuracy of the proposed method has been evaluated by the analysis of certified reference materials. The values found presented no statistically significant differences compared to the certified values (Student's t-test, p < 0.05). In general, the relative standard deviation was lower than 12% (n = 5).  相似文献

The purpose of the present study is the association of natural uranium in seawater with colloidal and suspended-particulate matter was determined. The separation of suspended particulate material (>0.45 µm) and colloidal fraction (as dissolved fractions) in seawater were done by suction and ultra filtration techniques. Seawater samples were collected at 1 km away from the shore and subjected to sequential fractionation in nine stages ranging from 2.7 µm to 1.1 nm. Suspended particulate matter were separated in three different size groups namely >2.7 µm, <2.7–>0.45 µm and <0.45–>0.22 µm by suction filtration using cellulose acetate and nitrate membranes filters. To concentrate the solution with colloidal particles <0.22 µm–1.1 nm (0.5 k Nominal Molecular Weight cut-off Limit {NMWL}), the solution obtained from filtration through <0.22 µm was passed through stirred ultra-filtration cell. The pH and conductivity at different stages of fractionation (dissolved) showed minor variations. The concentration of uranium was measured in suspended and dissolved fractions by using a pulsed nitrogen laser at 337.1 nm. In order to evaluate the role of mineral colloids in various stages of filtration, concentration of calcium, magnesium, potassium were measured by using ion chromatography and atomic absorption spectrometry. The clay mineral at seawater pH (approximately 8) behave as negative ions and provides binding site for the positively charge species of uranium. Among the dissolved fraction, the maximum concentrations of colloidal uranium was observed about 4 times higher than that compared to average concentration of 6.93±3.10 ppb in other fractions. In the case of suspended particulate matter, the concentration of uranium was below detection limits (<1 ppb). The maximum concentration of Ca, Mg and K in the dissolved fraction were in the <1.1 nm fraction, while for suspended particulate matter, the concentration of Ca, Mg and K decreased with the decrease in size and it is highest in the fraction of 0.22 –0.45 µm.This revised version was published online in November 2005 with corrections to the Cover Date.  相似文献

Pinched inlet gravitational split-flow thin fractionation (PI-GSF) has been applied to the continuous size fractionation of marine sediments in order to study the difference in sediment size distribution and the concentration of PCDD/Fs contained in different particle sizes. A PI-GSF channel, known to improve the separation efficiency by reducing the sample inlet thickness, was utilized to fractionate sediments collected from three different bay areas (Geoje, Ulsan, and Pohang) in Korea into 5 different sub-populations (<2.0, 2.0-5.0, 5.0-10, 10-20, 20-63 microm in diameter). The sorted sediment fractions from PI-GSF were examined using electron microscopy to obtain size distribution and the results showed a variation in particle size distribution between bay areas. When the collected particle fractions were examined for size dependent levels of PCDD/Fs, the concentrations of total PCDD/Fs were shown to be much greater for samples collected close to heavy industry complexes than sediments from bay areas without major industry.  相似文献

Nanofibrous scaffolds of silk fibroin (SF) and poly(l-lactic acid-co-?-caprolactone) (P(LLA-CL)) blends fabricated via electrospinning possessed good mechanical property and biocompatibility, as demonstrated by a previous study in vitro. However, the degradation behavior of the scaffolds, which may significantly influence tissue repair and regeneration, needs further exploration. In this study, in vitro degradation of pure SF, P(LLA-CL) and SF/P(LLA-CL) blended nanofibrous scaffolds were performed in phosphate-buffered saline (PBS, pH 7.4 ± 0.1) at 37 °C for 6 months. A series of analyses and characterizations (including morphologic changes, loss weight, pH changes of PBS solutions, DSC, XRD and FTIR-ATR) were conducted to the nanofibrous scaffolds after degradation and the results showed that the pure SF nanofibrous scaffolds were not completely degradable in PBS while pure P(LLA-CL) nanofibrous scaffolds had the fastest degradation rate. Moreover, the addition of SF reduced the degradation rate of P(LLA-CL) in SF/P(LLA-CL) blended nanofibrous scaffolds. This was probably caused by the intermolecular interactions between SF and P(LLA-CL), which hindered the movement of P(LLA-CL) molecular chains.  相似文献

A column chromatography procedure was developed for the clean-up of solvent-extracted sediment samples for the fractionation of polybrominated diphenyl ethers (PBDEs) and polybrominated dibenzo-p-dioxins and dibenzofurans (PBDD/Fs). The procedure included multiple column chromatography steps for clean-up for the separation of PBDEs from PBDD/Fs. The separation of the two chemical groups overcame the mutual interfering problem during the GC-ion trap MS analysis. The method was validated with the analysis of quality control samples. The method accuracy represented with relative error was less than 16% for all targeted PBDEs and PBDD/Fs congeners. Recoveries of the 13C-labeled standards ranged from 64% to 117% with relative standard deviation from 7.3% to 15%. Results from the analysis of environmental sediment samples collected in the vicinity of a recycling site for electronic wastes showed high levels of PBDEs (1.5-12 ng/g, dry weight), trace levels of PBDFs (0.025-0.92 ng/g, dry weight) and non-detectable PBDDs.  相似文献

The potential of 3D selective laser melting (SLM) technology to produce compact, temperature and pressure stable titanium alloy chromatographic columns is explored. A micro bore channel (0.9 mm I.D. × 600 mm long) was produced within a 5 × 30 × 30 mm titanium alloy (Ti–6Al–4V) cuboid, in form of a double handed spiral. A poly(butyl methacrylate-co-ethyleneglycoldimethacrylate) (BuMA-co-EDMA) monolithic stationary phase was thermally polymerised within the channel for application in reversed-phase high-performance liquid chromatography. The prepared monolithic column was applied to the liquid chromatographic separation of intact proteins and peptides. Peak capacities of 69–76 (for 6–8 proteins respectively) were observed during isothermal separation of proteins at 44 °C which were further increased to 73–77 using a thermal step gradient with programmed temperature from 60 °C to 35 °C using an in-house built direct-contact heater/cooler platform based upon matching sized Peltier thermoelectric modules. Rapid temperature gradients were possible due to direct-contact between the planar metal column and the Peltier module, and the high thermal conductivity of the titanium column as compared to a similar stainless steel printed column. The separation of peptides released from a digestion of E.coli was also achieved in less than 35 min with ca. 40 distinguishable peaks at 210 nm.  相似文献

The portable capillary electrophoresis instrument is automated and features three independent channels with different background electrolytes to allow the concurrent optimized determination of three different categories of charged analytes. The fluidic system is based on a miniature manifold which is based on mechanically milled channels for injection of samples and buffers. The planar manifold pattern was designed to minimize the number of electronic valves required for each channel. The system utilizes pneumatic pressurization to transport solutions at the grounded as well as the high voltage side of the separation capillaries. The instrument has a compact design, with all components arranged in a briefcase with dimensions of 45 (w) × 35 (d) × 15 cm (h) and a weight of about 15 kg. It can operate continuously for 8 h in the battery-powered mode if only one electrophoresis channel is in use, or for about 2.5 h in the case of simultaneous employment of all three channels. The different operations, i.e. capillary flushing, rinsing of the interfaces at both capillary ends, sample injection and electrophoretic separation, are activated automatically with a control program featuring a graphical user interface. For demonstration, the system was employed successfully for the concurrent separation of different inorganic cations and anions, organic preservatives, additives and artificial sweeteners in various beverage and food matrices.  相似文献

A convenient and scalable method for the preparation of (3RS,3aSR,8aSR)-phenyloctahydrocyclohepta[b]pyrrol-4(1H)-one based on the aza-Cope-Mannich rearrangement is described. A new confocal three-dimensional micro X-ray fluorescence (3D micro-XRF) facility based on polycapillary X-ray optics in the detection channel and Kirkpatrick–Baez (KB) mirrors in the excitation channel is designed. The lateral resolution (lx, ly) of this confocal three-dimensional micro-X-ray fluorescence facility is 76.3(lx) and 53.4(ly) µm respectively, and its depth resolution dz is 77.1 μm at θ = 90°. We present a fast and versatile method to produce functional micro free-flow electrophoresis chips. Microfluidic structures were generated between two glass slides applying multistep liquid-phase lithography, omitting troublesome bonding steps or cost-intensive master structures. Utilizing a novel spacer-less approach with the photodefinable polymer polyethyleneglycol dimethacrylate (PEG-DA), microfluidic devices with hydrophilic channels of only 25 μm in height were generated. The microfluidic chips feature ion-permeable segregation walls between the electrode channels and the separation bed and hydrophilic surfaces. The performance of the chip is demonstrated by free-flow electrophoretic separation of fluorescent xanthene dyes and fluorescently labeled amino acids.  相似文献

Gravitational field-flow fractionation is used for the separation of particles according to their sizes in the range 1-100 microns: larger particles elute before smaller ones. This phenomenon can be explained as a result of the steric exclusion of the particles from the vicinity of the channel walls, and/or hydrodynamic effects supposedly associated with the inertia of the liquid. The method was used for the investigation of red blood cells. The dependence of the retention ratio on the flow-rate, sample volume, concentration of blood and relaxation time was studied. Analysis of fifteen individual fractions by Coulter counter and reinjection of three other fractions were studied in order to verify fractionation of red blood cells.  相似文献

Apple pomace, a by-product in the processing of apple juice, was investigated as a potential source of polyphenols. Two methods of separation and purification of polyphenols from apple pomace extract were established by combination of gel chromatography with high-speed counter-current chromatography (HSCCC) and solvent extraction with HSCCC, respectively. The optimal separation was performed on a Sephadex LH-20 column using gradient aqueous ethanol as eluting solvent from 0% to 100% in increments of 10%. HPLC analysis indicated that main polyphenols existed in fractions eluted between 40% and 50% aqueous ethanol. The fractions of interest from column were separated by HSCCC with the solvent system hexane–ethyl acetate–1% aqueous acetic acid (0.5:9.5:10, v/v/v). Ethyl acetate fractionation of the apple pomace extract followed by direct HSCCC separation by the same solvent system in the volume ratio of 1:9:10 also produced a good separation of the main polyphenols of interest. Six high-purity polyphenols were achieved tentatively and identified by HPLC/MS: chlorogenic acid (1, m/z 354), quercetin-3-glucoside/quercetin-3-glacaside (2, m/z 464), quercetin-3-xyloside (3, m/z 434), phloridzin (4, m/z 436), quercetin-3-arabinoside (5, m/z 434), and quercetin-3-rhamnoside (6, m/z 448). These results provided a preliminary foundation for further development and exploration of apple pomace.  相似文献

The separation and manipulation of microparticles in lab on a chip devices have importance in point of care diagnostic tools and analytical applications. The separation and sorting of particles from biological and clinical samples can be performed using active and passive techniques. In passive techniques, no external force is applied while in active techniques by applying external force (e.g. electrical), higher separation efficiency is obtained. In this article, passive (pinched flow fractionation) and active (insulator‐based dielectrophoresis) methods were combined to increase the separation efficiency at lower voltages. First by simulation, appropriate values of geometry and applied voltages for better focusing, separation, and lower Joule heating were obtained. Separation of 1.5 and 6 μm polystyrene microparticles was experimentally obtained at optimized geometry and low total applied voltage (25 V). Also, the trajectory of 1.5 μm microparticles was controlled by adjusting the total applied voltage.  相似文献

In this study, preparative ion-pair high-speed countercurrent chromatography was directly coupled to an electrospray ionization mass-spectrometry device (IP-HSCCC/ESI-MS-MS) for target-guided fractionation of high molecular weight acyl-oligosaccharide linked betacyanins from purple bracts of Bougainvillea glabra (Nyctaginaceae). The direct identification of six principal acyl-oligosaccharide linked betacyanins in the mass range between m/z 859 and m/z 1359 was achieved by positive ESI-MS ionization and gave access to the genuine pigment profile already during the proceeding of the preparative separation. Inclusively, all MS/MS-fragmentation data were provided during the chromatographic run for a complete analysis of substitution pattern. On-line purity evaluation of the recovered fractions is of high value in target-guided screening procedures and for immediate decisions about suitable fractions used for further structural analysis. The applied preparative hyphenation was shown to be a versatile screening method for on-line monitoring of countercurrent chromatographic separations of polar crude pigment extracts and also traced some minor concentrated compounds. For the separation of 760 mg crude pigment extract the biphasic solvent system tert.-butylmethylether/n-butanol/acetonitrile/water 2:2:1:5 (v/v/v/v) was used with addition of ion-pair forming reagent trifluoroacetic acid. The preparative HSCCC-eluate had to be modified by post-column addition of a make-up solvent stream containing formic acid to reduce ion-suppression caused by trifluoroacetic acid and later significantly maximized response of ESI-MS/MS detection of target substances. A variable low-pressure split-unit guided a micro-eluate to the ESI-MS-interface for sensitive and direct on-line detection, and the major volume of the effluent stream was directed to the fraction collector for preparative sample recovery. The applied make-up solvent mixture significantly improved smoothness of the continuously measured IP-HSCCC-ESI-MS base peak ion trace in the experimental range of m/z 50–2200 by masking stationary phase bleeding and generating a stable single solvent phase for ESI-MS/MS detection. Immediate structural data were retrieved throughout the countercurrent chromatography run containing complete MS/MS-fragmentation pattern of the separated acyl-substituted betanidin oligoglycosides. Single ion monitoring indicated clearly the base-line separation of higher concentrated acylated betacyanin components.  相似文献
