Pueraria lobata is a rich source of isoflavonoids. The detection and identification of isoflavonoid components from root, stem, leaf, callus and cell samples, is very important for the best, safest and most efficacious use of kudzu as a medicinal plant, and for the studies on quantitative analysis in the secondary metabolism of isoflavonoids. In this paper, a simple, rapid and precise high-performance capillary electrophoresis (HPCE) method with diode array detection (DAD) has been developed for separation and determination of isoflavonoids in several kudzu samples. The isoflavonoids could be well separated within 15 min in a 40 cm length capillary at a separation voltage of 15kV in a 30 mmol L(-1) borax buffer (pH9.29), and this proposed method demonstrated excellent reproducibility and accuracy with relative standard deviations of less than 5% for isoflavonoid content (n = 5) of different kudzu samples. The relationship between peak areas and isoflavone concentrations, in a specified working range with linear response, was determined by first-order polynomial regression over the range 0.05-0.5 mg mL(-1) for puerarin and 2.5-50 microg mL(-1) for 3'-methoxypuerarin, daidzin and daidzein, respectively, and quantitative evaluation of those four main isoflavonoid components was determined by ultraviolet absorption at lambda = 192 nm. The differences were also illustrated by comparison of the determination of isoflavonoid components from kudzu root, stem, leaf samples and plant tissue cultures in vitro.  相似文献

A new, rapid analytical method using near-infrared spectroscopy (NIRS) was developed to differentiate two species of Radix puerariae (GG), Pueraria lobata (YG) and Pueraria thomsonii (FG), and to determine the contents of puerarin, daidzin and total isoflavonoid in the samples. Five isoflavonoids, puerarin, daidzin, daidzein, genistin and genistein were analyzed simultaneously by high-performance liquid chromatography-diode array detection (HPLC-DAD). The total isoflavonoid content was exploited as critical parameter for successful discrimination of the two species. Scattering effect and baseline shift in the NIR spectra were corrected and the spectral features were enhanced by several pre-processing methods. By using linear discriminant analysis (LDA) and soft independent modeling class analogy (SIMCA), samples were separated successfully into two different clusters corresponding to the two GG species. Furthermore, sensitivity and specificity of the classification models were determined to evaluate the performance. Finally, partial least squares (PLS) regression was used to build the correlation models. The results showed that the correlation coefficients of the prediction models are R = 0.970 for the puerarin, R = 0.939 for daidzin and R = 0.969 for total isoflavonoid. The outcome showed that NIRS can serve as routine screening in the quality control of Chinese herbal medicine (CHM).  相似文献

The isoflavonoid puerarin in extracts of the well-known traditional Chinese drug Radix puerariae (root of the plant Pueraria lobata) can be separated from other isoflavonoids by adsorption chromatography on the cross-linked 12% agarose gel Superose 12 equilibrated in distilled water. The adsorption is totally quenched by the addition of 50% acetic acid. The separation of the isoflavonoids is tentatively ascribed to interaction with the residues of the cross-linking reagents used in the manufacturing process of Superose 12. Thus, no useful separation can be achieved with non-cross-linked 12% agarose gel media. Symmetric elution profiles at high sample loadings (16 mg on a 24 ml column) suggest linear adsorption isotherms for the isoflavonoids.  相似文献

采用反相C18毛细管液相色谱柱,以乙腈(含0.1%(体积分数,下同)三氟乙酸)和水(含0.1%三氟乙酸)为流动相梯度洗脱,在26 min内分离了葛根异黄酮提取物以及愈风宁心片中的主要成分。采用毛细管液相色谱/四极杆飞行时间串联质谱仪对葛根提取物以及片剂中的几种主要异黄酮类化合物做了结构分析,发现葛根素是主成分(提取物中其平均质量分数是13.32%;片剂中每片含量19.28~24.34 mg)。对微量未知化合物,用它们的子离子谱图与已知化合物的谱图比较,推测其成分为3′-甲氧基葛根素和3′-甲氧基大豆苷。  相似文献

For thousands of years Pueraria thomsonii Benth has been used to treat a number of diseases in traditional Chinese pharmacopeia. Despite these uses, there is still insufficient information on its biological activity and chemical composition. In this respect, the in vitro callus culture of P. thomsonii was subjected to identify anticancer and antibacterial compounds. Based on significant preliminary cytotoxicity and antibacterial activities; the chemical investigation led to the isolation of isoflavonoids, coumaric acid derivative and dihydroxyflavanone-type of compounds viz., daidzin (1), puerarin (2), biochanin A (3), daidzein (4), p-coumaric acid ethyl ester (5) and liquiritigenin (6), respectively. These compounds were tested for their cytotoxicity and antibacterial activities. Among them, p-coumaric acid ethyl ester (5) exhibited significant cytotoxicity with GI50 values of 14.73, 15.64 and 20.88 μM/mL against 4T1, NC1-H1975 and A549, respectively; the other isoflavones and aflavonoid showed moderate to weak activities. Moreover, p-coumaric acid ethyl ester (5) inhibited the growth of K. pneumonia, MRSE and MRSA at very low MIC values of 6.01, 12.01 µg/mL 24.02, respectively. On the other hand compounds biochanin A (3) and liquiritigenin (6) showed moderate antibacterial activity. Because of the potential anticancer and antibacterial activities of bioactive compounds from P. thomsonii, they can be used to treat various cancer and emerging bacterial infections.  相似文献

To evaluate the bioavailability of puerarin from Pueraria lobata isoflavone self-microemulsifying drug delivery systems (SMEDDS) and Yufengningxin tablets, a rapid and specific liquid chromatography--mass spectrometric method was developed and validated to determine puerarin in rabbit serum. The analyte was extracted from serum samples by precipitating the serum proteins, separated on a Diamonsil C(18) column and detected by mass spectrometry with an electrospray ionization interface. 4-Hydroxybenzaldehyde was used as the internal standard. The method has a limit of quantitation of 10 ng/mL using 200 microL serum. The intra-day relative standard deviations (RSDs) ranged from 3.7 to 6.9% and inter-day RSDs were within 6.5%. After administration of SMEDDS and tablets to rabbits, a significant difference was observed in main pharmacokinetic parameters of t(max), C(max) and AUC(0--infinity) between SMEDDS and tablets, and a 2.2-fold increase in the relative bioavailability of puerarin was observed with the SMEDDS compared with Yufengningxin tablets. It was concluded that the absorption of puerarin from Pueraria lobata isoflavone SMEDDS was enhanced.  相似文献

A method based on capillary electrophoresis with electrochemical detection was developed for the determination of puerarin, daidzein and rutin. Effects of several important factors such as the acidity and concentration of running buffer, separation voltage, injection time, and detection potential were investigated to acquire the optimum conditions. The working electrode was a 300-microm diameter carbon disc electrode positioned opposite the outlet of capillary. The three analytes could be well separated within 12 min in a 40 cm length capillary at a separation voltage of 9 kV in a 50 mmol/l borate buffer (pH 9.0). The relationship between peak currents and analyte concentrations was linear over about three orders of magnitude with detection limits (SIN=3) ranging from 0.241 x 10(-6) to 0.511 x 10(-6) mol/l for all compounds. This proposed method demonstrated long-term stability and reproducibility with relative standard deviations of less than 5% for both migration time and peak current (n=7). It has been successfully applied for the determination of puerarin, daidzein and rutin in Chinese traditional drugs, the vines of Pueraria lobata (Wild.) Ohwi and Puerariae Radix.  相似文献

The levels of 6 kinds of isoflavonoids found in 11 domestic and imported soybeans, and 12 kinds of soybean-based processed foods in Japan were systematically analyzed, and the Japanese daily intake of isoflavonoids from those foods was estimated. The total isoflavonoids (daidzein, glycitein, and genistein) were analyzed with acid hydrolysis and the intact isoflavonoids (daidzein, glycitein, genistein, daidzin, glycitin, and genistin) were analyzed without hydrolysis. This was followed by cleanup with an ODS cartridge column and determined by liquid chromatography with a diode array detector. The highest content of isoflavonoids was found in kinako (a roasted soybean powder) and the lowest was found in soy sauce. The contents and composition of the isoflavonoids in the 11 soybeans varied by species and country of origin. The level of isoflavonoids found in the processed foods varied by manufacturing method or ingredients. The percentage of aglycone tended to be higher in miso (fermented soybean paste) and soy sauce, which are heated and fermented during the manufacturing process. Japanese daily intake of isoflavonoids from soybeans and soybean-based processed foods was estimated as 27.80 mg per day (daidzein 12.02 mg, glycitein 2.30 mg, and genistein 13.48 mg).  相似文献

The optimization of a high-performance liquid chromatographic method to determine three isoflavonoids (daidzein, genistein, and biochanin A) in the fruit of Psoralea corylifolia is developed and validated. Dried psoralea fruit powder is extracted with aqueous methanol followed by the hydrolysis of the analytes' conjugated glycosides with hydrochloric acid. The HPLC assay is performed on a reverse-phase C18 column with gradient elution using acetonitrile and 10% acetic acid as the mobile phase at a flow rate of 0.8 mL/min. Flavone is used as the internal standard and the substances are detected at 260 nm. Calibrations are linear (correlation coefficient > or = 0.995) for all three analytes. The limits of detection are 0.01 microg/mL for daidzein and genistein and 0.1 microg/mL for biochanin A. The overall intra- and interassay precision range from 2.5% to 4.9% and from 0.5% to 4.7%, respectively. The method proved to be sensitive, specific, accurate, and precise for the determination of daidzein, genistein, and biochanin A in Psoralea corylifolia.  相似文献

Isoflavones are phytochemicals found in many plants. Because of their structural similarity to beta-estradiol, health benefits of isoflavones have been evaluated in age-related and hormone-dependent diseases. Dietary supplement preparations contain extracts from soy, Red Clover and kudzu. Soy products contain primarily genistein, daidzein, and glycitein, while Red Clover products contain primarily formononetin and biochanin A. Kudzu extracts contain puerarin and daidzein among other components. Previous methods of analysis focused on the determination of isoflavones from a single botanical source, while dietary supplements are often a blend of extracts from different plants. We developed a method for the analysis of isoflavones in dietary supplements regardless of their botanical composition, using HPLC-PDA because of its applicability to routine analysis. Isoflavones are found as free compounds, glucoside derivatives, 6'-O-malonyl-beta-d-glucoside and 6'-O-acetyl-beta-d-glucoside derivatives. In this study, the samples were extracted at room temperature with 50:50 (v/v) MeCN/water, and then analyzed before and after hydrolyzing the isoflavones by acid or basic digestion. 2'-Methoxy-flavone and 6-methoxy-flavone were used as internal standards and were added together to every sample. Daidzein, glycitein, genistein, puerarin, calycosin, pratensein, pseudobaptigenin, formononetin, biochanin A and prunetin were among the isoflavones determined.  相似文献

In order to differentiate two species of Radix Puerariae (Radix Puerariae lobatae and Radix Puerariae thomsonii) and to determine major isoflavonoids (puerarin, daidzin, daidzein and genistein) in the samples, a simple high performance liquid chromatography (HPLC) method with isocratic elution employing cyclodextrins (CDs) as mobile phase additives was developed. Various factors affecting the retention of isoflavonoids in the C18 reversed-phase column, such as the nature of CDs, the concentration of hydroxypropyl-β-cyclodextrin (HP-β-CD) and the methanol percentage in the mobile phase, were studied. Experimental results confirmed that HP-β-CD, as a very effective mobile phase additive, could markedly reduce the retention of isoflavonoids, especially daidzein and genistein. The elution of four isoflavonoids could be achieved on a Kromasil® C18 column within 56 min by using the methanol–water contained 5 mM HP-β-CD (25/75, v/v) mixture as the mobile phase. The formation of the inclusion complexes between isoflavonoids and HP-β-CD explained the modification of the retention of analytes. The apparent formation constants determined by HPLC confirmed that the stoichiometry of HP-β-CD-isoflavonoid complexes was 1:1, and the stability of the complexes depended on the size and property of isoflavonoids. The optimized method was successfully applied for the simultaneous determination of major isoflavonoids in P. lobatae and P. thomsonii samples. This work provides a useful method for the analysis of traditional Chinese herbs.  相似文献

The roots of Pueraria lobata (Wild.) Ohwi and Pueraria thomsonii Benth have been officially recorded in all editions of Chinese Pharmacopoeia under the same monograph 'Gegen' (Radix Puerariae, RP). However, in its 2005 edition, the two species were separated into both individual monographs, namely 'Gegen' (Radix Puerariae Lobatae, RPL) and 'Fenge' (Radix Puerariae Thomsonii, RPT), respectively, due to their obvious content discrepancy of puerarin, the major active constituent. In present paper, the fingerprint of high-performance thin-layer chromatography (HPTLC) combining digital scanning profiling was developed to identify and distinguish the both species in detail. The unique properties of the HPTLC fingerprints were validated by analyzing ten batches of Pueraria lobata and P. thomsonii samples, respectively. The common pattern of the HPTLC images of the roots of Pueraria spp. and the respective different ratios of the chemical distribution can directly discern the two species. The corresponding digital scanning profiles provided an easy way for quantifiable comparison among the samples. Obvious difference in ingredient content and HPTLC patterns of the two species questioned their bio-equivalence and explained that recording both species separately in the current edition of Chinese Pharmacopoeia (2005 edition) is reasonable due to not only the content of major constituent, puerarin, but also the peak-to-peak distribution in the fingerprint and integration value of the total components. Furthermore, the HPTLC fingerprint is also suitable for rapid and simple authentication and comparison of the subtle difference among samples with identical plant resource but different geographic locations.  相似文献

A very fast chromatographic separation of isoflavonoids genistein, daidzein, formononetin and biochanin A was developed on a C18 high-speed column under isocratic conditions. The method was validated in terms of detection limits, quantitation limits (LOQs), linearity and precision. LOQs in 0.04-0.2 microg/g range were calculated, making feasible the determination of these compounds of nutritional concern at trace levels. Good linearity was demonstrated over three concentration orders of magnitude for each analyte (r2 0.990-1.000). The intra-day and inter-day repeatability was evaluated in terms of relative standard deviation (RSD%) at two concentration levels for each analyte (RSD% <9%). An optimization strategy was adopted to find the best conditions for the extraction of isoflavonoid aglycones from yellow soybeans using microwave-assisted extraction. The most relevant parameters resulted to be the microwave power, the extraction time and the acid concentration, optimal values being 600 W, 1 min and 12 M, respectively. When performing sample treatment on a fortified soybean sample, high recovery percentage was obtained for both compounds (94+/-8% for daidzein and 97+/-5% (n = 4) for genistein). The concentration level at which daidzein and genistein were found in the soybean sample were 1.21+/-0.15 mg/g and 2.38+/-0.09 mg/g (n=4), respectively.  相似文献

Tong-Mai oral liquid, containing three herbs (Pueraria radix, radix Salviae miltiorrhizae and rhizoma Chuanxiong), is a formula created under the theory of traditional Chinese medicine (TCM) to invigorate the ‘blood’ and dispel ‘blood stasis’, which arises from poor blood circulation. In Tong-Mai formula, Pueraria radix is used for improved blood circulation and to treat cardiovascular diseases and arrhythmia. Radix Salviae miltiorrhizae was widely used in TCMs for treatment of coronary heart disease, blood circulation diseases, and other cardiovascular diseases; rhizoma Chuanxiong is used to treat thrombosis and platelet aggregation. Clinically, combining the use of these herbs was therapeutically valuable, which suggests that they might synergistically prevent thrombosis. In this study, the pharmacokinetic behaviors of puerarin and daidzein in rat plasma were studied after orally administrated to four kinds of extracts. Puerarin and daidzein were set as index components. In the Tong-Mai oral liquid group, puerarin and daidzein showed the maximum plasma concentration and the lowest clearance rate of plasma. The results indicated that the herbal constituents in Tong-Mai formula showed synergism, and it may be helpful to certificate the rationality of the matching in Tong-Mai formula.  相似文献

The isoflavonoid puerarin, a well-known traditional Chinese drug, has been purified in one step from an extract of Radix puerariae (root of the plant Pueralria lobata) by adsorption chromatography on an epichlorohydrin polymerized beta-cyclodextrin ligand coupled to brominated allyl-group substituted Sepharose HP. Acetic acid (10%) was used as the mobile phase and the optimum loading capacity was around 1.2 mg crude extract/ml packed gel. The purity of the collected puerarin was about 98% with a recovery of about 62%.  相似文献

High-speed counter-current chromatography (HSCCC) was applied to the semipreparative separation and purification of puerarin and related isoflavones from a crude extract of Pueraria lobata. Analytical HSCCC was used for the preliminary selection of a suitable solvent system composed of ethyl acetate-n-butanol-water (2:1:3, v/v/v). Using the above solvent system the preparative HSCCC was successfully performed yielding six relatively pure isoflavones including puerarin from 80 mg of the crude extract in one-step separation.  相似文献

A new isoflavonoid glycoside, gehuain, together with three flavonoids and eight isoflavonoids, was isolated from the flowers of Pueraria lobata. Three of them, apigenin, apigenin 4′‐O‐β‐D‐glucoside, and wistin, were reported the first time from this plant. Their structures were established on the basis of spectral evidence.  相似文献

Poly(vinyl acetate-triallyl isocyanurate-divinylbenzene) matrix coupled with oligo-β-cyclodextrin was synthesized to separate puerarin. The separation of three isoflavone homologues, puerarin, daidzin and daidzein, was achieved by increasing the proportion of methanol from 40 to 85%. The interaction mechanism between the crosslinkers and puerarin was investigated through molecular dynamics simulation and the mixed-mode character was demonstrated by computational and conformational methods. The research confirms that in order to enhance the recognition, the addition of triallyl isocyanurate is feasible. Meanwhile, an alternative matrix for the separation of puerarin was provided, with the advantages of moderate mechanical rigidity, ease of modification and low cost.  相似文献
