The production of lignin peroxidase fromPhanerochaete chrysosporium was studied using immobilized mycelia in nylon-web cubes in semicontinuous fermentation using glucose pulses or ammonium tartrate pulses. Consistent enzyme production was achieved when glucose pulses were used, leading to an average activity of 253 U/L. The crude enzyme was added to eucalyptus kraft pulp before conventional and ECF bleaching sequences. Optimization of the enzymatic pretreatment led to the following operational conditions: enzyme load of 2 U/g of pulp, hydrogen peroxide addition rate of 10 ppm/h, and reaction time of 60 min. Pulp final characteristics were dependent on the chemical treatment sequence that followed enzymatic pretreatment. The chief advantage of enzymatic pretreatment was pulp viscosity preservation, which was observed in most of the experiments carried out with seven different chemical treatment sequences  相似文献

A study was undertaken to determined if a suitable biosorbent could be found for removal of nickel at low concentrations (< 20 parts per million [ppm]) from a chemically complex wastewater effluent generated by electroplating operations. Algae and cyanobacteria were chosen as candidate biosorbent materials because they are easy to grow and they have the ability to withstand processing into biosorbent materials. Several species were screened for nickel-biosorption capacity initially, and three species of cyanobacteria were selected for further study based on their performance in the scoping tests. When compared to live controls, autoclaving improved the binding capacities of all three species, but usually biosorption data from experiments with live cells were more consistent. None of the three species was able to bind nickel efficiently in actual effluent samples. Further experimentation indicated that sodium ions, which were present in high concentrations in the effluent, were interfering with the ability of the cells to bind nickel. Adsorption isotherm plots for biosorption of nickel by two species ofAnabaena in NiCl2-deionized water solutions were prepared. To determine if compounds added during trichloroethylene (TCE) degradation could reduce the loss of enzyme activity or increase enzyme recovery, different compounds serving as energy and carbon sources, pH buffers, or free radical scavengers were tested. Formate and formic acid (reducing power and a carbon source), as well as ascorbic acid and citric acid (free radical scavengers) were added during TCE degradation at a concentration of 2 mM. A saturated solution of calcium carbonate was also tested to address pH concerns. In the presence of formate and methane, only calcium carbonate and formic acid had a beneficial effect on enzyme recovery. The calcium carbonate and formic acid both reduced the loss of enzyme activity and resulted in the highest levels of enzyme activity after recovery.  相似文献

The use of partially hydrated porous silica particles has been studied as a support for cofactor-dependent enzymatic catalysis in organic solvents. At an optimal pore hydration corresponding to 70% pore volume, horse liver alcohol dehydrogenase catalyzes the oxidation and reduction of alcohols and aldehydes, respectively, with rates sixfold higher than with nonporous glass beads as the enzymatic support and with cofactor recycling numbers in excess of 105. Thus, supported aqueous-phase enzymatic catalysis makes highly effective use of the enzyme and cofactor by coimmobilization and by providing a high interfacial area for reactions in organic media.  相似文献

The degradation of 4-chlorobiphenyl (4CB) was compared in field lysimeters containing 60 Kg of soil contaminated with 5–10 mg/Kg of polychlorinated biphenyls.Alcaligenes A5, a bacterium carrying a plasmid for 4CB degradation, was inoculated into three lysimeters. When compared to an untreated control, soil samples from water, mineral, and yeast extract treated lysimeters with and without a bacterial inoculum exhibited greater than 10-fold increases in the rate of [1-14 C-acetate incorporation into lipids and14CO2 production from [U-14C-4-chlorobiphenyl. Gene probe analyses for the 4CB plasmid and most-probable-number enumerations demonstrated the presence of biodegradative populations in lysimeters and the probable survival of the addedAlcaligenes A5.  相似文献

Aspen wood substrates with varying degrees of deacetylation, xylan, and lignin removal have been prepared and submitted to enzymatic hydrolysis with a cellulase/hemicellulase preparation for an extended constant period of hydrolysis. Controlled deacetylation has been achieved by treating wood with various alkali metal hydroxide solutions, at various alkali/wood ratios. It has been found that samples with the same extent of deacetylation produce the same sugar yields upon enzymatic hydrolysis. Increased degree of deacetylation increases the yield of sugars obtained from enzymatic hydrolysis, all other compositional parameters held constant. The acetyl group removal is proportional to the stoichiometric relation between added base and wood acetyl content, i.e., the same number of milliequivalents of base/weight of wood remove the same extent of acetyl groups, regardless of the concentration of the base solution. No cation effects are found among Li, Na, and K alkali hydroxide solutions, suggesting that swelling is not as important a parameter as is the removal of the acetyl groups from the xylan backbone in determining the extent of hydrolyzability of the resulting sample.  相似文献

Simultaneous saccharification and fermentation (SSF) processes for producing ethanol from lignocellulose are capable of improved hydrolysis rates, yields, and product concentrations compared to separate hydrolysis and fermentation (SHF) systems, because the continuous removal of the sugars by the yeasts reduces the end-product inhibition of the enzyme complex. Recent experiments using Genencor 150L cellulase and mixed yeast cultures have produced yields and concentrations of ethanol from cellulose of 80% and 4.5%, respectively. The mixed culture was employed because B.clausenii has the ability to ferment cellobiose (further reducing end-product inhibition), while the brewing yeastS. cerevisiae provides a robust ability to ferment the monomeric sugars. These experimental results are combined with a process model to evaluate the economics of the process and to investigate the effect of alternative processes, conditions, and organisms.  相似文献

Tributyl phosphate (TBP), a plasticizer and solvent, is used in nuclear fuel reprocessing, generating TBP wastes laden with residual uranium. ACitrobacter sp. accumulated heavy metals via a phosphohydrolase(s) that precipitated metals with inorganic phosphate liberated from an organic phosphate “donor” molecule (TBP). Mutant analysis suggested that TBP hydrolysis was not attributable to a previously documented acid phosphatase (monoesterase). Purified monoesterase had little activity against phospho di- and triesters, had no requirement for Mg2+ or Mn2+, and was EDTA-resistant. Conversely, TBP cleavage by immobilized cells was enhanced by Mg2+, and ininhibited by Mn2+ and EDTA. A separate phosphotri/diesterase was implicated.  相似文献

The formation and decay of lignolytic enzymes, along with the generation of other extracellular metabolites in submerged cultures ofPhanerochaete chrysosporium, were studied under different physiological conditions. Whereas lignin peroxidase (LiP) was detectable only in a narrow range of O2 tension and nitrogen concentration, manganese peroxidase (MnP) reached considerable levels over a broad range. The decay of LiP and MnP activities under lignolytic conditions paralleled that of heme and total proteins. The conditions that decrease or suppress LiP or MnP activities resulted in high levels of extracellular protease activity and/or polysaccharides.  相似文献

Two additional electrophoretically distinct molecular forms, isoforms (iso) 2 and 3, with lectin properties were isolated fromCratylia mollis Mart, seeds (FABACEAE), by extraction with 0.15M NaCl and ammonium sulfate fractionation, followed by chromatography on Sephadex G-75 and Bio-Gel P-200 (iso 2), as well as CM-Cellulose and Sephadex G-75 (iso 3). Both isoforms were human group nonspecific and showed distinct specificity. Polyacrylamide gel electrophoresis resolved iso 2 and 3 in polypeptides of apparent mol wts 60 and 31 kDa, respectively; a distinct isoelectric focusing pattern was obtained for iso 2 and 3, under denaturing and reducing conditions.  相似文献

Xylans are the major components of the hemicellulosic fraction of lignocellulosic biomass and their hydrolysis can be obtained using xylanases fromPenicillium janthinellum. In this work, sugarcane bagasse hemicellulosic hydrolysate was used as the substrate for producing xylanase. The precipitation of these enzymes was studied using ethanol and Na2SO4 as precipitating agents. Ethanol precipitation experiments were performed batchwise in concentrations ranging from 10 to 80%, pH 4.0 to 7.0, at 4áC. The concentrations used in the precipitations with Na2SO4 were from 5 to 60% at pH 5.5 and 25áC. Solubility curves as a function of xylanase activity and total protein for both precipitating agents were made. According to the results, Na2SO4 is not appropriate for precipitating xylanases in this medium since at salt concentrations higher than 25%, the enzyme was denaturated and at this concentration less than 80% of the enzyme and total protein were precipitated. Because of differences in xylanase and total protein solubility, a fractionated precipitation using ethanol can be performed, since with 40% ethanol, 49% of the total protein was precipitated and more than 95% of the enzyme was kept in solution. On the other hand approx 100% of the xylanases were recovered by precipitation after adding 80% ethanol.  相似文献

A 1,4-β-d-glucan cellobiohydrolase (EC 3.2.1.91) and l,4-β-d-glucan glucanohydrolase (EC 3.2.1.4) were purified from the culture filtrates ofPenicillium funiculosum by using preparative isoelectric focusing. Both the enzymes were homogeneous on polyacrylamide gel with and without sodium dodecyl sulphate. The mol wt of the cellobiohydrolase and endoglucanase were 14,400 and 25,000 respectively. The purified enzymes were free of β-glucosidase activity. Acting in isolation, the cellobiohydrolase had little capacity for solubilizing Avicel or Walseth cellulose, but showed increased rates of hydrolysis when combined with endoglucanase. Cellobiose inhibition (50%) was observed in the initial rate of the hydrolysis of Walseth cellulose. Streptomyces setonii 75Vi2 produces an extracellular coal-solubilizing component(s) in the absence of coal. The heat stability, relatively low molecular weight, and insensitivity to proteases of the substance(s) responsible for coal solubilization indicate that the process is nonenzymatic. This report describes factors affecting the production and activity of this substance(s) and the similarity in its action to alkaline buffer solutions in solubilizing coal.  相似文献

Utilization of lipases for synthesis of esters of hydrophilic polyols has been investigated. The choice of a suitable solvent is crucial in this type of reaction. An interesting case is fatty acid esters from neopentylpolyols, such as trimethylolpropane, which are of great interest as high temperature lubricants. Enzymatic synthesis of trimethylolpropane tricaprylate was studied as an alternative to chemical manufacturing. Triester production occurred only if the water produced by esterification was continuously removed from the medium. In these condition, kinetics of appearance and transformation of mono-, di- and triesters were determined in order to define optimal conditions.  相似文献

Multigram quantities (2.5-10 g) of highly purified IgG were obtained within 4 h from serum by using Avid AL packed in a radial-flow column. Avid AL is an affinity gel containing a synthetic, low-mol-wt ligand capable of selectively binding IgG from serum of all animal species tested. By packing the gel in a radial-flow column up to 500 mL, a high flow rate of 50 mL/min can be achieved without adversely affecting the performance of the gel and the purity of the isolated antibody.  相似文献

Urease has been purified from the seeds of Cajanus Cajan. The purification process involves three solvent extraction steps followed by DEAE-cellulose column chromatography. The specific activity of the purified enzyme is found to be 1920 U/mg with the recovery of 8%. The application of the purified enzyme in a biosensor construction is discussed.  相似文献

Several methanotrophic microorganisms, i.e.,Methylococcus capsulatus (Bath),Methylomonas albus (BG-8),Methylosinus trichosporium OB3b, andMethylocystis parvus (OBBP), were evaluated for growth and methane utilization. The effect of temperature was examined in the range of 25 to 45°C for growth and methane utilization. The temperature variations (25–35°C) had minimal effect on growth ofM. albus and M. parvus. Methane consumption varied at different temperatures with a maximum of 0.67 mol%/h and 0.53 mol%/h. at 30 and 35°C, respectively, forM. albus and M. parvus. The growth and methane consumption was slower forM. trichosporium OB3b as a maximum methane consumption of 0.07 mol%/h was obtained at 25°C and growth was inhibited at 35°C.M. capsulatus grew the best at 37°C and growth was affected at higher temperature of 45°C. Of the different cultures examined,M. albus andM. capsulatus grew the best and were further evaluated for the effect of pressure in the range of 10–50 psi. The results obtained usingM. albus demonstrated an enhancement in methane consumption rate by fourfold and final cell concentration by 40% at a pressure of 20 psi by injecting a methane/oxygen mixture, however further increase in the pressure up to 50 psi inhibited the growth. The inhibition was not seen with nitrogen incorporated mixture of oxygen and methane, which suggest that the high partial pressure of methane and/or oxygen are inhibitory for the growth ofM. albus. M. capsulatus was more sensitive to pressure as evidenced by inhibition at the relatively low pressure of 10 psi  相似文献
