Well‐defined diblock and triblock copolymers composed of poly(N‐isopropylacrylamide) (PNIPAM) and poly(ethylene oxide) (PEO) were successfully synthesized through the reversible addition–fragmentation chain transfer polymerization of N‐isopropylacrylamide (NIPAM) with PEO capped with one or two dithiobenzoyl groups as a macrotransfer agent. 1H NMR, Fourier transform infrared, and gel permeation chromatography instruments were used to characterize the block copolymers obtained. The results showed that the diblock and triblock copolymers had well‐defined structures and narrow molecular weight distributions (weight‐average molecular weight/number‐average molecular weight < 1.2), and the molecular weight of the PNIPAM block in the diblock and triblock copolymers could be controlled by the initial molar ratio of NIPAM to dithiobenzoate‐terminated PEO and the NIPAM conversion. © 2004 Wiley Periodicals, Inc. Functional poly(N-isopropylacrylamide) (PNIPAM) hydrogels were prepared by reversible addition fragmentation chain transfer (RAFT) polymerization of NIPAM in the presence of four-arm poly(ethylene glycol) (4A-PEG) as backbone and 4-cyanopentanoic acid dithiobenzoate functional α -cyclodextrin threaded onto the PEG as chain transfer reagent (CTA).The structure of the hydrogels was characterized in detail with FTIR techniques. The analytical results demonstrated that α -cyclodextrin remains in as-obtained hydrogels. The swelling behavior was investigated and the functional hydrogels (functional gels) showed accelerated shrinking kinetics and higher swelling ratio comparing with conventional hydrogel (CG). It could be attributed to the presence of dangling chains. The hydrogel exhibited rapid swelling and deswelling kinetics. In principle, the hydrogel might find a number of applications including an on-off system and drug delivery systems.  相似文献

Poly(methyl methacrylate)/poly(N-isopropylacrylamide) (PMMA/PNIPAM) core-shell particles were synthesized by seeded precipitation polymerization of N-isopropylacrylamide (NIPAM) in the presence of PMMA seed particles. The anionic potassium persulfate was used as initiator, and acrylic acid as functional comonomer. It was shown that the weight ratio of the PNIPAM shell to the PMMA core can be greatly increased through continuous addition of NIPAM monomer at a relatively slow rate. PMMA/PNIPAM particles with different shell thickness were obtained by varying the amount of charged NIPAM monomers. These particles exhibited unique nonspherical core-shell morphology. PMMA core was partially coated by dense hair-like or antler-like PNIPAM shell depending on the shell thickness. The measurement of these particles' zeta potential at different temperatures showed that the absolute value of zeta potential unusually decreased as the particle size decreased with temperature.  相似文献

With the ever-increasing demands for personalized drugs, disease-specific and condition-dependent drug delivery systems, four-dimensional (4D) printing can be used as a new approach to develop drug capsules that display unique advantages of self-changing drug release behavior according to the actual physiological circumstances. Herein, a plant stomata-inspired smart hydrogel capsule was developed using an extrusion-based 4D printing method, which featured with UV cross-linked poly(N-isopropylacrylamide) (PNIPAM) hydrogel as the capsule shell. The lower critical solution temperature (LCST) of the PNIPAM hydrogels was approximately 34.9 °C and macroporous PNIPAM hydrogels were prepared with higher molecular weight polyethylene glycols (PEGs) as the pore-forming agents. Owing to the LCST-induced shrinking/swelling properties, the prepared PNIPAM hydrogel capsules exhibited temperature-responsive drug release along with the microstructure changes in the PNIPAM hydrogels. The in vitro drug release test confirmed that the PNIPAM hydrogel capsules can autonomously control their drug release behaviors on the basis of ambient temperature changes. Moreover, the increased PEG molecular weights in the macroporous PNIPAM hydrogel capsules caused an obvious improvement of drug release rate, distinctly indicating that the drug release profiles can be well programmed by adjusting the internal pore size of the hydrogel capsules. In vitro biocompatibility studies confirmed that the PNIPAM hydrogel capsules have great potential for biomedical applications. The bioinspired 4D printed hydrogel capsules pioneer the paradigm of smart controlled drug release.  相似文献

Aminopropylisobutyl polyhedral oligomeric silsesquioxane (POSS) was used to prepare a POSS‐containing reversible addition‐fragmentation transfer (RAFT) agent. The POSS‐containing RAFT agent was used in the RAFT polymerization of N‐isopropylacrylamide (NIPAM) to produce tadpole‐shaped organic/inorganic hybrid Poly(N‐isopropylacrylamide) (PNIPAM). The results show that the POSS‐containing RAFT agent was an effective chain transfer agent in the RAFT polymerization of NIPAM, and the polymerization kinetics were found to be pseudo‐first‐order behavior. The thermal properties of the organic/inorganic hybrid PNIPAM were also characterized by differential scanning calorimetry. The glass transition temperature (Tg) of the tadpole‐shaped inorganic/organic hybrid PNIPAM was enhanced by POSS molecule. The self‐assembly behavior of the tadpole‐shaped inorganic/organic hybrid PNIPAM was investigated by atomic force microscopy and dynamic light scattering. The results show the core‐shell nanostructured micelles with a uniform diameter. The diameter of the micelle increases with the molecular weight of the hybrid PNIPAM. Surprisingly, the micelle of the tadpole‐shaped inorganic/organic hybrid PNIPAM with low molecular weight has a much bigger and more compact core than that with high molecular weight. © Wiley Periodicals, Inc. DNA:Au nanosphere bioconjugates have applications in biosensing and in the bottom-up assembly of materials. These bioconjugates can be selectively assembled into three-dimensional aggregates upon addition of complementary DNA oligonucleotides and can be dissociated by heating above a melting transition temperature at which the DNA duplexes are denatured. Herein we describe the impact of polymeric solutes on the thermal denaturation behavior of DNA:Au nanoparticle bioconjugate assemblies. Polymeric solutes can dramatically impact biochemical reactions via macromolecular crowding. Poly(ethylene glycol)s (PEGs) and dextrans of varying molecular weights were used as crowding reagents. While both PEG and dextran increased the stability of DNA:Au aggregates, melting transition temperatures in the presence of PEG were impacted more significantly. Polymer molecular weight was less important than polymer chemistry and weight percent in solution. For a high (15%) weight percent of PEG, aggregation was observed even in the absence of complementary oligonucleotides. These results underscore the importance of polymer chemistry in addition to physical volume exclusion in macromolecular crowding and point to the importance of understanding these effects when designing biorecognition-based nanoparticle assembly schemes in complex matrixes (i.e., any involving polymeric solutes).  相似文献

结合大分子自组装和原位自由基聚合方法,采用油溶性引发剂偶氮二异丁腈(AIBN),在聚(ε-已内酯)(PCL)纳米粒子表面引发聚合单体N-异丙基丙烯酰胺(NIPAM)和交联剂亚甲基双(丙烯酰胺)(MBA),制备得到了核-壳结构的PCL/PNIPAM聚合物纳米微球.系统研究了单体和交联剂用量、壳层目标交联度、初始PCL/DMF溶液的浓度及引发剂AIBN含量4个反应参数对核-壳结构PCL/PNIPAM纳米微球的PNIPAM壳层得率、微球尺寸、温敏性能及电镜形貌的影响.结果表明,在制备核-壳结构PCL/PNIPAM纳米微球的反应过程中,PCL粒子表面的聚合和水中的聚合二者之间相互竞争.适当增加引发剂AIBN的添加量,有利于制备得到核/壳比例可控的PCL/PNIPAM纳米微球;交联剂MBA较高的反应活性导致形成了非均匀交联的PNIPAM壳层.  相似文献

A polypseudorotaxane (PPR) comprising γ‐cyclodextrin (γ‐CD) as host molecules and poly(N‐isopropylacrylamide) (PNIPAM) as a guest polymer is prepared via self‐assembly in aqueous solution. Due to the bulky pendant isopropylamide group, PNIPAM exhibits size‐selectivity toward self‐assembly with α‐, β‐, and γ‐CDs. It can fit into the cavity of γ‐CD to give rise to a PPR, but cannot pass through α‐CD and β‐CD under the same conditions. The ratio of the number of γ‐CD molecules to entrapped NIPAM repeat units is kept at 1:2.2 or 1:2.4, determined by 1H NMR spectroscopy and TGA analysis, respectively, indicating that there are more than 2 but less than 3 NIPAM repeat units included by one γ‐CD molecule. This finding opens new avenues to PPR‐based supramolecular polymers to be used as solid, stimuli‐responsive materials.  相似文献

Macroporous, temperature‐sensitive poly(N‐isopropylacrylamide) (PNIPAAm) hydrogels were synthesized with poly(ethylene glycol)s (PEGs; molecular weight = 2000–6000) as the pore‐forming agents. The influence of the molecular weight and PEG content on the responsive kinetics of these macroporous hydrogels was investigated. The PEG‐modified PNIPAAm hydrogels were characterized by the swelling ratio, deswelling–reswelling kinetics, Fourier transform infrared, and differential scanning calorimetry. The morphology of these hydrogels was analyzed with scanning electron microscopy. The prepared macroporous hydrogels exhibited some unique properties in comparison with the gels with low molecular weight PEGs (molecular weight < 2000) as the pore‐forming agents. In addition, a preliminary study on the controlled release of bovine serum albumin from these macroporous hydrogels was carried out. © 2002 Wiley Periodicals, Inc. Hybrid nanogels consisting of cholesteryl-modified pullulan (CHP) and poly(N-isopropylacrylamide) (PNIPAM) were synthesized by graft free-radical copolymerization of N-isopropylacrylamide (NIPAM) onto methacryloyl-substituted CHP nanogels (CHPMA) in water at 50 degrees C in the presence of a water-soluble free radical initiator. Depending on the initial NIPAM/CHPMA ratio, CHP-PNIPAM (CN) nanogels containing 30.8-84.8 wt % PNIPAM were obtained in the form of self-assembled nanoparticles with a hydrodynamic radius (Rh) of 69.0-116.0 nm in water kept at 20 degrees C. Hybrid nanogels of sufficiently high NIPAM content, such as the sample CN90, which contains 79.6 wt % NIPAM, exhibited a two-step response to changes in solution (3 mg/mL) temperature: a decrease in Rh from 93 to 57 nm as the temperature increased from 20 to 35 degrees C, followed by a sharp increase in Rh from 57 nm to 90 nm at 55 degrees C. Both steps in this temperature response were reversible. The multistep response to temperature of the CN nanogels was attributed to the morphology of the nanogels, which are seen as consisting of grape-like (botryoidal) clusters of associated native nanogels held together via cholesteryl cross-linking points and held together by the grafted PNIPAM chains.  相似文献

Well‐defined H‐shaped pentablock copolymers composed of poly(N‐isopropylacrylamide) (PNIPAM), poly(N,N‐dimethylaminoethylacrylamide) (PDMAEMA), and poly(ethylene glycol) (PEG) with the chain architecture of (A/B)‐b‐C‐b‐(A/B) were synthesized by the combination of single‐electron‐transfer living radical polymerization, atom‐transfer radical polymerization, and click chemistry. Single‐electron‐transfer living radical polymerization of NIPAM using α,ω azide‐capped PEG macroinitiator resulted in PNIPAM‐b‐PEG‐b‐PNIPAM with azide groups at the block joints. Atom‐transfer radical polymerization of DMAEMA initiated by propargyl 2‐chloropropionate gave out α‐capped alkyne‐PDMAEMA. The H‐shaped copolymers were finally obtained by the click reaction between PNIPAM‐b‐PEG‐b‐PNIPAM and alkyne‐PDMAEMA. These copolymers were used to prepare stable colloidal gold nanoparticles (GNPs) in aqueous solution without any external reducing agent. The formation of GNPs was affected by the length of PDMAEMA block, the feed ratio of the copolymer to HAuCl4, and the pH value. The surface plasmon absorbance of these obtained GNPs also exhibited pH and thermal dependence because of the existence of PNIAPM and PDAMEMA blocks. © 2010 Wiley Periodicals, Inc. Polyethylene glycols (PEGs) are non-ionic, water-soluble synthetic polymers which have been widely used for many applications. Since they are of very low toxicity and are readily excreted in urine, PEGs in the molecular weight range 400-6000 have been used extensively in the study of intestinal physiology in man. A high-performance liquid chromatographic (HPLC) method has been developed for the determination of PEG 600 in human urine, which includes a pre-column derivatisation step. The dibenzoate derivatives of PEG 600 can be quantitatively prepared, and this, coupled with ultraviolet detection at 230 nm, has greatly improved the limit of detection for the determination of PEGs by HPLC. A suitable extraction procedure has also been developed which enabled PEG levels in urine to be monitored with much greater sensitivity than any previously reported method.  相似文献

Polymeric micelles with a polystyrene core, poly(acrylic acid)/poly(4-vinyl pyridine) (PAA/P4VP) complex shell and poly(ethylene glycol) & poly(N-isopropylacrylamide) (PEG & PNIPAM) mixed corona were synthesized and used as the supporter for the gold nanoparticles (GNs). It was concluded from the result of 1H NMR characterization that hydrophilic channels formed around PEG chains when PNIPAM collapsed above its lower critical solution temperature. The density of the channels in the corona can be tuned by changing the weight ratios of PEG chains to PNIPAM chains. The GNs were set in the PAA/P4VP complex layer and the catalytic activity of the GNs can be modulated by the channels. The catalytic activity increased with increasing the density of the channels in the corona. Meanwhile, the whole Au/micelle nanoparticles were stabilized by the extended PEG chains.  相似文献

Thermoresponsive colloidal particles were prepared by seeded precipitation polymerization of N-isopropylacrylamide (NIPAM) in the presence of a crosslinking monomer, N,N-methylenebisacrylamide (MBA), using polystyrene latex particles (ca. 50 nm in diameter) as seeds in aqueous dispersion. Phase transitions of the prepared poly(N-isopropylacrylamide), PNIPAM, shells on polystyrene cores were studied in comparison to colloidal PNIPAM microgel particles, in H2O and/or in D2O by dynamic light scattering, microcalorimetry and by 1H NMR spectroscopy including the measurements of spin–lattice (T1) and spin–spin (T2) relaxation times for the protons of PNIPAM. As expected, the seed particles grew in hydrodynamic size during the crosslinking polymerization of NIPAM, and a larger NIPAM to seed mass ratio in the polymerization batch led to a larger increase of particle size indicating a product coated with a thicker PNIPAM shell. Broader microcalorimetric endotherms of dehydration were observed for crosslinked PNIPAM on the solid cores compared to the PNIPAM microgels and also an increase of the transition temperature was observed. The calorimetric results were complemented by the NMR spectroscopy data of the 1H-signal intensities upon heating in D2O, showing that the phase transition of crosslinked PNIPAM on polystyrene core shifts towards higher temperatures when compared to the microgels, and also that the temperature range of the transition is broader.  相似文献

Novel AB2‐type amphiphilic block copolymers of poly(ethylene glycol) and poly(N‐isopropylacrylamide), PEG‐b‐(PNIPAM)2, were successfully synthesized through single‐electron transfer living radical polymerization (SET‐LRP). A difunctional macroinitiator was prepared by esterification of 2,2‐dichloroacetyl chloride with poly(ethylene glycol) monomethyl ether (PEG). The copolymers were obtained via the SET‐LRP of N‐isopropylacrylamide (NIPAM) with CuCl/tris(2‐(dimethylamino)ethyl)amine (Me6TREN) as catalytic system and DMF/H2O (v/v = 3:1) mixture as solvent. The resulting copolymers were characterized by gel permeation chromatography and 1H NMR. These block copolymers show controllable molecular weights and narrow molecular weight distributions (PDI < 1.15). Their phase transition temperatures and the corresponding enthalpy changes in aqueous solution were measured by differential scanning calorimetry. As a result, the phase transition temperature of PEG44b‐(PNIPAM55)2 is similar to that in the case of PEG44b‐PNIPAM110; however, the corresponding enthalpy change is much lower, indicating the significant influence of the macromolecular architecture on the phase transition. This is the first study into the effect of macromolecular architecture on the phase transition using AB2‐type amphiphilic block copolymer composed of PEG and PNIPAM. © 2009 Wiley Periodicals, Inc. A new method, adopting inorganic clay (synthetic hectorite) as a physical cross-linker, was used to prepare poly(N-isopropylacrylamide) (PNIPAM) microgels via surfactant-free emulsion polymerization. The effect of hectorite content on the temperature-sensitivity of PNIPAM microgels was investigated by means of DLS, UV/Vis and DSC. It was found that, in the absence of surfactant, with increasing hectorite content, the particle size tends to decrease to 300 nm at room temperature, while increases as weight ratio (WR) of hectorite and N-isopropylacrylamide (NIPAM) exceeds 21%. Furthermore, with increasing WR from 7% to 21%, the volume phase transition temperature of PNIPAM microgels has little shift, while decreases slightly when WR increases up to 28%.  相似文献

A reliable and efficient route for preparing thermoresponsive hollow microgels based on cross-linked poly(N-isopropyl acrylamide) (PNIPAM) was developed. Firstly, monodisperse thermoresponsive core–shell microspheres composed of a P(styrene (St)-co-NIPAM) core and a cross-linked PNIPAM shell were prepared by seeded emulsion polymerization using P(St-co-NIPAM) particles as seeds. The size of the P(St-co-NIPAM) core can be conveniently tuned by different dosages of sodium dodecyl sulfate. The thickness of the cross-linked PNIPAM shell can be controlled by varying the dosage of NIPAM in the preparation of PNIAPM shell. Then, hollow PNIPAM microgels were obtained by simply dissolving the P(St-co-NIPAM) core with tetrahydrofuran. The core–shell microspheres and the hollow microgels were characterized by transmission electron microscopy, dynamic light scattering, atomic force microscopy, and Fourier-transform infrared spectroscopy.  相似文献

BackgroundPolyethylene glycol (PEG) is a polyether compound which is an excellent candidate to study possible specific interactions of protein-polymer systems at the atomic level. Such studies would be beneficial in obtaining a more thorough understanding of PEG-protein interactions and might help to explain PEG's effects on protein behavior when used as a crowder.Scope of reviewPEGs has been called double edged sword showing assorted effects on the protein due its hydrophilic, hydrophobic and amphiphatic nature. It has been observed that the stabilization of the protein due to PEGs is because of exclusion volume effect (hard core interactions) shown by the higher molecular sizes of the crowder, however there are soft (chemical) interactions also which leads to destabilization of proteins. Most of low molecular size PEGs are noticed to destabilize proteins due to soft interactions.General significanceHere, the core characteristics of the polyethylene glycols that are responsible for the various interactions with proteins and trying to delineate the behaviour of PEGs being ambiguous, are discussed. To conclude its paradox nature being a factual inert molecule or not, many different reports of PEG interactions with various proteins are clubbed together. In addition, owing to low toxicity, high polarity, high water solubility, typically unhydrolysing or undeteriorating characteristics, it has a practical significance in pharmaceutical and cosmetic industries, and in the research field on which this review shall enlighten from earlier to now.  相似文献

The melting temperature (T(m)) of DNA is affected not only by salt but also by the presence of high molecular weight (MW) solutes, such as polyethylene glycol (PEG), acting as a crowding agent. For short DNAs in a solution of low MW PEGs, however, the change of excluded volume upon melting is very small, leading to no increase in T(m). We demonstrate herein that by attaching 12-mer DNAs to gold nanoparticles, the excluded volume change was significantly increased upon melting, leading to increased T(m) even with PEG 200. Larger AuNPs, higher MW PEGs, and higher PEG concentrations show even larger effects in stabilizing the DNA. This study reveals a unique and fundamental feature at nanoscale due to geometric effects. It also suggests that weak interactions can be stabilized by a combination of polyvalent binding and the enhanced macromolecular crowding effect using nanoparticles.  相似文献
