Structure elucidation of 9‐S and 9‐R oxirane derivatives of ascomycin, a 23‐membered immunomodulating macrolactam, was performed using NMR spectroscopy. The total 1H and 13C signal assignments required the gradient‐selected versions of COSY (gs‐COSY), heteronuclear multiple quantum‐correlation spectroscopy (gs‐HSQC), heteronuclear multiple‐bond correlation spectroscopy (gs‐HMBC), and nuclear Overhauser methods. The data sets then were used to examine the dependence of ketone–hemiketal and cistrans amide equilibria on the substitution pattern and the absolute configuration of the chiral oxirane. Alopecurin A, an alkaloid with an unprecedented skeleton, was isolated from the seeds of Sophora alopecuroides L. The absolute configuration and structure of this compound was identified as (3S,12R)‐3‐hydroxy‐1,7‐diazatricyclo[10.4.0.13,7]heptadecane‐11,16,17‐trione (=(7S,15aR)‐decahydro‐7‐hydroxy‐6H‐7,11‐methano‐4H‐pyrido[1,2‐a][1,7]diazacyclododecine‐4,15,16(12H)‐trione). The structure and absolute configuration was elucidated by spectroscopic methods, mainly HR‐ESI‐TOF‐MS, IR, 1D‐NMR (1H‐ and 13C‐NMR), 2D‐NMR (COSY, NOESY, HSQC, HMBC), and particularly X‐ray crystal‐diffraction and CD spectral analysis.  相似文献

Structure elucidation of compounds in the benzisoxazole series ( 1 – 6 ) and naphtho[1,2‐d][1,3]‐ ( 7 – 10 ) and phenanthro[9,10‐d][1,3]oxazole ( 11 – 14 ) series was accomplished using extensive 2D NMR spectroscopic studies including 1H–1H COSY, long‐ range 1H–1H COSY, 1H–13C COSY, gHMQC, gHMBC and gHMQC‐TOCSY experiments. The distinction between oxazole and isoxazole rings was made on the basis of the magnitude of heteronuclear one‐bond 1JC2, H2 (or 1JC3, H3) coupling constants. Complete analysis of the 1H NMR spectra of 11 – 14 was achieved by iterative calculations. Gradient selected gHMQC‐TOCSY spectra of phenanthro[9,10‐d][1,3]oxazoles 11 – 14 were obtained at different mixing times (12, 24, 36, 48 and 80 ms) to identify the spin system where the protons of phenanthrene ring at H‐5, H‐6 and at H‐9 and H‐7 and H‐8 were highly overlapping. One new diacylglycoside named lyciumaside ( 1 ) and a new sesquiterpene lactone named lyciumate ( 2 ) were isolated from Lycium shawii Roem . & Schult . The structures of the two new compounds were elucidated based on 1D‐ (1H‐ and 13C‐NMR and NOE) and 2D‐NMR (COSY, HSQC, and HMBC) spectroscopic techniques, and mass spectrometry (ESI‐MS). Preliminary evaluations demonstrated lyciumaside ( 1 ) possesses strong antioxidant activity with an IC50 = 30 μg/ml (80% inhibition) while it was inactive in α‐glucosidase and urease enzymes assays.  相似文献

From the reaction of 2‐acetylcyclopentanone and 2‐acetyl‐2‐methylcyclopentanone with benzohydrazide, the 1‐benzoyl‐6a‐hydroxycyclopentapyrazole derivatives 2a and 2b were obtained as the only reaction products, whereas from the reaction of 2‐acetylcyclohexanone an epimeric cis/trans mixture of the 2‐benzoyl‐3‐hydroxy‐2H‐indazole derivative 3c was formed. The dehydration of the isolated compounds 2a and 3c , as well as the NaBH4 and NaBH3CN reduction products of 2a were studied. The structural assignments of the compounds derived were established by analysis of their NMR spectra (1H, 13C, DEPT, COSY, NOESY, HETCOR C? H, and COLOC C? H). The chemoselectivity of the reactions of 1a and 1c with benzohydrazide was studied by conformational analysis with MM2 and semiempirical (AM1 and PM3) MO calculations.  相似文献

A new alkaloid, 15‐β‐stemmadenine, was isolated from the fruits of Tabernaemontana heyneana Wall. from the ethanolic extract. The complete 1H and 13C NMR assignments of the compound were carried out using COSY, HMQC and HMBC. A new dimeric secoiridoid glycoside, trivially named oleferrugine B ( 1 ), has been isolated from the AcOEt‐soluble part of the MeOH extract of leaves of Olea ferruginea Royle . The structure of the isolated compound was established on the basis of ESI‐MS fragmentation patterns, and 1D‐ and 2D‐NMR techniques, including 1H‐ and 13C‐NMR, HSQC, 1H,1H‐COSY, HMBC, and NOESY experiments, and by comparison with literature data.  相似文献

Cycloaddition of C,N‐diphenylnitrones 1 to N‐aryl maleimides 2 afforded two diastereomeric isoxazolidines with high selectivity. The structure and steric configuration of the adducts have been assigned on the basis of 1H NMR, 1H NMR COSY, 13C NMR and IR spectroscopy. The π–π stacking interactions between maleimide's and nitrone's aromatic rings during the 1,3‐dipolar cycloaddition were assumed to control the exo–endo selectivity of the reaction. Thus, the exo–endo ratio depends upon the position of the substituent present on the C‐phenyl ring of the C,N‐diphenylnitrones.  相似文献

Coagulansins A ( 1 ) and B ( 2 ), two new withanolides, were isolated from the CHCl3‐soluble fraction of Withania coagulans Dunal , along with two known withanolides, coagulin R and withanolide P. This paper describes a method of preparation of new 3,5′‐dioxo‐2′‐phenyl‐1,3‐dihydrospiro[indene‐2,4′‐[1,3]oxazol]‐1‐yl acetate and its 5‐chloro‐ and bromoderivatives as products of interaction of N‐benzoylglycine (hippuric acid) with corresponding ortho‐formylbenzoic acids. The reaction carried out in acetic anhydride media in the presence of piperidine as catalyst. The novel spirocompounds were purified by column chromatography from multicomponent reaction mixtures. The composition of the spiro‐products was confirmed by C, H, N element analysis. The structure was established by IR, MS, 1H‐ and 13C‐NMR analysis including COSY 1H‐13C experiments.  相似文献

An unexpected multicomponent reaction of cyanothioacetamide with aromatic aldehydes and aminopyrazole under MWI was found and reported. It turned out to be an efficient and simple procedure for the preparation of pyrazolo[3,4‐b]pyridine derivatives. The structure of the product was characterized by IR, MS, 1H and 13C NMR, COSY and NOESY spectroscopy, X‐ray crystallographic analysis, and by comparison with the authentic sample prepared with the known procedure. © 2008 Wiley Periodicals, Inc. A facile method for the synthesis of substituted 3‐(2‐furylidene)‐2‐furanones has been developed using cyclofunctionalization reactions of 2,4‐dialkenyl‐1,3‐dicarbonyl compounds and iodine as electrophile in the presence of Na2CO3, in refluxing chloroform. Compounds 4 are obtained in modest to good yields and their structural identification was established by 1H NMR, 1H COSY, 13C NMR and 1H‐13C COSY. A mechanism has been proposed to rationalize the formation of the ylidene furanone.  相似文献

Five new diterpene polyesters, 1 – 5 , with jatrophane skeletons were isolated from the fresh whole plants of Euphorbia serrulata. The structure elucidation was performed by means of UV/VIS spectroscopy, HR‐ESI‐MS, and advanced two‐dimensional NMR methods, including 1H‐NMR, JMOD, 1H,1H‐COSY, NOESY, HMQC, and HMBC experiments. The relative configurations of 1 – 5 and their conformations in solution were analyzed on the basis of NOESY measurements. As a result of detailed NMR studies, complete 1H and 13C chemical‐shift assignments of the compounds were possible. The isolated compounds differ stereochemically and do not comprise a uniform series regarding the configurations at C(2), C(6), and C(13). Compound 5 possesses the new structural feature of a double bond with (Z)‐configuration in the macrocyclic ring of the jatrophane skeleton, while compound 2 has a C?C bond in the five‐membered ring, this being the first observation of this structural feature in the type of macrocyclic Euphorbiaceae diterpenes.  相似文献

The triterpene ester (3β)‐olean‐18‐en‐3‐yl stearate ( 1 ), together with (3β)‐urs‐12‐en‐3‐yl stearate ( 2 ), and (3β)‐lup‐20(29)‐en‐3‐yl stearate ( 3 ) were isolated from leaves of Maytenus salicifolia Reissek (Celastraceae). The structure of 1 , a new compound, including its configuration, was established by 1H, 13C, and DEPT‐135 NMR data, including 2D experiments (HSQC, HMBC, COSY, and NOESY). The molecular mass (692 Da) was confirmed by gas chromatography coupled with mass spectrometry (CG/MS).  相似文献

N‐acetyl‐4‐nitrotryptophan methyl ester (2), N‐acetyl‐5‐nitrotryptophan methyl ester (3), N‐acetyl‐6‐nitrotryptophan methyl ester (4) and N‐acetyl‐7‐nitrotryptophan methyl ester (5) were synthesized through a modified malonic ester reaction of the appropriate nitrogramine analogs followed by methylation with BF3‐methanol. Swainsonine (SW, 1 ), a unique indolizine with poly‐hydroxyl groups, was re‐isolated from the plant endophytic fungus Alternaria oxytropis. The structure (including planar structure and relative configuration) was systematically elucidated by NMR spectra (including 1H, 13C, 1H‐1H COSY, HMQC, HMBC, and NOESY spectra in DMSO‐d6 and in CD3OD); 1H NMR spectra of the modified Mosher's products were first used to determine the absolute configuration of SW. More importantly, the complex coupled features of H‐7α, H‐7β, and H‐6α in the 1H NMR spectrum of ( 1 ) were analyzed in details, which will provide aids for the planar and relative configuration determination of analogs.  相似文献
